Wingate test 및 Supramaximal exercise를 이용한 무산소성 운동능력의 측정시 나타난 변인의 관련성을 살펴본 본 연구를 요약하면 다음과 같다. Wingate test시 혈중젖산농도 최고치는 9.04±0.62mM로서 Supramaximal exercise시 보다 유의하게 높았으며, 혈장 LDH 및 CPK활성도는 두가지 운동부하후 모두 유의한 변화는 나타나지 않았다. 근전도상의 적분치, 중간파형빈도 및 평균파형빈도도 유의한 변화는 없었다. 등속성 수축시의 최대토오크는 양쪽 다리 모두에서 초당 60도의 각속도에서가장 높았다. Wingate test시의 최대파워는 등속성 수축시 최대토오크 및 Wingate test시 중간파형빈도의 직접적인 영향을 받는 것으로 나타났으며, 평균파워는 Wingate test시의 적분치 변화, 듣는 쪽 슬관절의 등속성 수축시최대토오크 및 Supramaximal exercise시 혈장 LDH 활성도의 직접적인 영향을 받는 것으로 나타났다. 이상의 결과에서 무산소성 운동능력과 관련지어 그 기전을 살피고자 할 때 체내 화학적 변화, 전기생리적 변화 및 발현능력의 복합성을 고려해야 하며 최대누적 산소결핍량의 활용가능성은 측정강도 결정방법을 중심으로한 계속적인 연구가 요구된다. The present study examined the relationship of variables related to anaerobic capacity with Wingate test and supramaximal exercise. Peak value of blood lactate concentration with Wingate test showeda significantly higher than suprammaximal exercise, and plasma LDH & median power frequency and mean power frequency were not any difference between two tests. Peak torque of isokinetic contractionh on knee joint showed a highest value in 60 degrees of angular velocity among a various angular velocity. Peak power of Wingate test was related to peak torque of isokinetic contraction and median power frequency of EMG with Wingate test, and mean power of Wingate test was related to integrated EMG with Wingate test, peak torque with isokinetic contraction of dominant knee joint and plasma LDH activity with supramaximal exercise. In this result, when it analyzed the mechanism of anaerobic capacity, ti was required the consideration of complexity according to biochemical alteration, electrophysiological alteration and revelation of performance, Appicapability of maximal accumulated O₂deficit was demaned a continuous study with the focus of exercise intensity.

Objectives : This study was carried out to suggest preventive methods for hypertension in cold-exposed workers. Methods : In 11 refs·iteration industries, 68 workers working in refrigerated areas more than one time per day were selected as the exposed group, and 68 workers not exposed to cold were selected as the control group. We interviewed the subjects with a questionnaire covering occupational history, and conducted clinical and laboratory tests including measurements of blood pressure and core temperature. Results : The systolic blood pressure in the exposed group(130.0±13.3 mmHg) was significantly higher than that recorded In the control group(118.3±12.1 mmHg), as was the diastolic blood pressure in the exposed group(82.7±8.5 mmHg) versus the control group(77.4±8.7 mmHg). The core temperature in the exposed group(36.1±0.7℃) was significantly lower than that experienced in the control group (36.4±0.5℃). In logistic regression analysis, age, cold exposure severity and milk intake were significant variables, with odds ratios of 5.204(95 % CI 1.440∼18.812), 2.674(95 % CI 1.080∼6.618), and 0.364(95 % Cl 0.141∼0.942) , respectively. Conclusions : Our study suggests that cold-exposed workers have higher a higher risk of hypertension, and that their core temperature is lower. Risk factors affecting hyper-tension of cold-exposed workers include age, cold exposure severity and milk intake. For the prevention of hypertension, cold-exposed workers should minimize cold-exposure time as much as possible.

Chronic neutrophilic leukemia is a very rare myeloproliferative disorder which is characterized by severe sustained mature neutrophilic leukocytosis in peripheral blood (PB), hepatosplenomegaly, elevated leukocyte alkaline phosphatase, serum uric acid. serun viatmin B 12 and the bleeding tendency despite normal platelet count and coagulation time, absence of philadelphia chromosome, and the absence of fever or underlying infection or disease sufficient to mimic a leukemoid reaction. We have experienced a 34-years-old male patient with atypical chronic neutrophilic leukemia. On admission, the leukocyte count was 56,860/uL. severe neutrophic leukocytosis with 76% neutrophils and rare immature forms on PB smear. Many neutrophils had toxic granules, and vacuoloes. Leukocyte alkaline phosphatase score was decreased. The cytogenetic study showed Philadelphia chromosome negative with normal karyotype.

<P><B>Abstract</B></P> <P>In this study, a system was established to perform an initial environmental risk assessment of hazardous and noxious substances (HNS) spill accidents. Initial environmental risk assessment was performed using exposure and hazard assessments. An integrated hydrodynamic and chemical fate model was used to predict HNS concentrations at harbors, taking into account local environmental conditions. To consider the worst case HNS spill accident, the spill amount of 10,000 tonnages, was used for this study. The results show that highly soluble HNS are fatal to marine organisms during the neap tide. The results were based on a hypothetical worst case HNS spill accident and, not any specific actual HNS spill accident. Nevertheless, the method and system developed in this study, which includes the physical/chemical properties of 158 priority HNS, can be readily used to perform an initial environmental risk assessment for future HNS spill accidents.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A system has been established to perform the initial environmental risk assessment of HNS spill accident. </LI> <LI> The system can predict the HNS concentrations at harbor areas, taking into account local environmental conditions. </LI> <LI> The spilt HNS of high solubility are fatal to marine organism during the neap tide. </LI> </UL> </P>

Retinoids regulate not only various cell functions including proliferation and differentiation but also glucose and lipid metabolism. After we observed a marked up-regulation of cellular retinol-binding protein-I (CRBP-I) in the liver of hepatitis B virus x antigen (HBx)-transgenic (HBx Tg) mice which are prone to hepatocellular carcinoma (HCC) and fatty liver, we aimed to evaluate retinoid pathway, including genes for the retinoid physiology, CRBP-I protein expression, and retinoid levels, in the liver of HBx Tg mice. We also assessed the effect of chronic metformin treatment on HCC development in the mice. Many genes involved in hepatic retinoid physiology, including CRBP-I, were altered and the tissue levels of retinol and all-trans retinoic acid (ATRA) were elevated in the liver of HBx Tg mice compared to those of wild type (WT) control mice. CRBP-I protein expression in liver, but not in white adipose tissue, of HBx Tg mice was significantly elevated compared to WT control mice while CRBP-I protein expressions in the liver and WAT of high-fat fed obese and db/db mice were comparable to WT control mice. Chronic treatment of HBx Tg mice with metformin did not affect the incidence of HCC, but slightly increased hepatic CRBP-I level. In conclusion, hepatic CRBP-I level was markedly up-regulated in HCC-prone HBx Tg mice and neither hepatic CRBP-I nor the development of HCC was suppressed by metformin treatment.

<P><B>Abstract</B></P><P>Phosphatase and tensin homolog deleted on chromosome 10 (PTEN) expressed in <I>Saccharomyces cerevisiae</I> was reversibly oxidized by hydrogen peroxide and reduced by cellular reductants. Reduction of hPTEN was delayed in each of <I>S. cerevisiae</I> gsh1Δ and gsh2Δ mutants. Expression of γ-glutamylcysteine synthetase Gsh1 in the gsh1Δ mutant rescued regeneration rate of hPTEN. Oxidized hPTEN was reduced by glutathione in a concentration- and time-dependent manner. Glutathionylated PTEN was detected. Incubation of 293T cells with BSO and knockdown expression of GCLc in HeLa cells by siRNA resulted in the delay of reduction of oxidized PTEN. Also, in HeLa cells transfected with GCLc siRNA, stimulation with epidermal growth factor resulted in the increase of oxidized PTEN and phosphorylation of Akt. These results suggest that the reduction of oxidized hPTEN is mediated by glutathione.</P>
Genome-Wide Association Study of Lung Cancer in Korean Non-Smoking Women

<P>Lung cancer in never-smokers ranks as the seventh most common cause of cancer death worldwide, and the incidence of lung cancer in non-smoking Korean women appears to be steadily increasing. To identify the effect of genetic polymorphisms on lung cancer risk in non-smoking Korean women, we conducted a genome-wide association study of Korean female non-smokers with lung cancer. We analyzed 440,794 genotype data of 285 cases and 1,455 controls, and nineteen SNPs were associated with lung cancer development (<I>P</I> < 0.001). For external validation, nineteen SNPs were replicated in another sample set composed of 293 cases and 495 controls, and only rs10187911 on 2p16.3 was significantly associated with lung cancer development (dominant model, OR of TG or GG, 1.58, <I>P</I> = 0.025). We confirmed this SNP again in another replication set composed of 546 cases and 744 controls (recessive model, OR of GG, 1.32, <I>P</I> = 0.027). OR and <I>P</I> value in combined set were 1.37 and < 0.001 in additive model, 1.51 and < 0.001 in dominant model, and 1.54 and < 0.001 in recessive model. The effect of this SNP was found to be consistent only in adenocarcinoma patients (1.36 and < 0.001 in additive model, 1.49 and < 0.001 in dominant model, and 1.54 and < 0.001 in recessive model). Furthermore, after imputation with HapMap data, we found regional significance near rs10187911, and five SNPs showed <I>P</I> value less than that of rs10187911 (rs12478012, rs4377361, rs13005521, rs12475464, and rs7564130). Therefore, we concluded that a region on chromosome 2 is significantly associated with lung cancer risk in Korean non-smoking women.</P>
