Anti-inflammatory effects of TP1 in LPS-induced Raw264.7 macrophages

Inflammation is an essential defense mechanism in health; however, excessive inflammation contributes to the patho physiology of several chronic diseases. Although anti‑inflammatory drugs are essential for controlling inflammation, they have several side effects. Recent findings suggest that naturally derived compounds possess physiological activi ties, including anti‑inflammatory, antifungal, antiviral, anticancer, and immunomodulatory activities. Therefore, this study aimed to investigate the anti‑inflammatory effects and molecular mechanisms of 2,5,6‑trimethoxy‑p‑terphenyl (TP1), extracted from the Antarctic lichen Stereocaulon alpinum, using in vitro models. TP1 treatment decreased the production of nitric oxide (NO) and reactive oxygen species (ROS) in LPS‑stimulated Raw264.7 macrophages. Additionally, TP1 treatment significantly decreased the mRNA levels of pro‑inflammatory cytokines (IL‑1β, TNF‑α, IL‑6) and the mRNA and protein levels of the pro‑inflammatory enzymes (inducible nitric oxide synthase and cyclooxyge nase‑2). Moreover, TP1 suppressed lipopolysaccharide‑induced phosphorylation of the NF‑κB and MAPK signaling pathways in Raw264.7 macrophages. Conclusively, these results suggest that TP1 ameliorates inflammation by sup pressing the expression of pro‑inflammatory cytokines, making it a potential anti‑inflammatory drug for the treatment of severe inflammatory diseases.
Afrormosin exerts an anticancer effect via MAPK and AKT signaling pathways in B16F10 cells

Melanoma is a deadly skin cancer with high mortality, and its incidence is increasing every year. Although numerous anticancer drugs have been developed, these treatments have various side effects, such as skin rash, fatigue, diarrhea, cough, and muscle pain. Therefore, there is a need for research on novel anticancer drugs with low cytotoxicity and few side effects. In this study, we investigated whether afrormosin (7-hydroxy-4′,6-dimethoxyisoflavone), a member of the isoflavonoid family, could have the potential as a novel anticancer drug. Afrormosin decreased the viability of B16F10 melanoma cells in a time- and dose-dependent manner. We also found that the afrormosin-induced decrease in cell viability was caused by the reduction of cell proliferation through Go/G1 arrest and the induction of apoptosis in B16F10 melanoma cells. Furthermore, afrormosin decreased the metastatic activity (cell invasion and migration) of B16F10 melanoma cells. At the molecular level, afrormosin reduced the levels of Bcl-2, an anti-apoptotic protein, and augmented the levels of Bax, a pro-apoptotic protein, and p53, a tumor suppressor. Additionally, procaspase-3 levels were reduced by afrormosin treatment. When we examined the signaling pathways affected by afrormosin, we found that the AKT/ERK pathways were inhibited and the p38/JNK pathway was activated by afrormosin. Collectively, these results suggest the potential anticancer effect of afrormosin, making it a prospective candidate for development as an anticancer drug.
The anti-cancer effect by phytochemical in B16F10 cell

Cancer which is defined by abnormal growth of cells, is one of the leading causes of death worldwide. Since the anticancer drugs that are currently available elicit various adverse side effects, a need for discovering new drug candidates has become crucial. Recently, natural compounds such as phytochemicals, are attracting attention as potential anticancer agents owing to their lower adverse effects, compared to synthetic drugs. In our study in order to examine the anti-cancer effect of phytochemical, we have selected the murine melanoma cell line B16F10. Firstly, to verify the cytotoxicity of phytochemical, MTT assay was performed. We found that phytochemical reduced cell viability of B16F10 cells and thus, could have anticancer effect. In addition, we performed wound healing assays to investigate whether phytochemical affected cell migration. We idendified that phytochemical inhibited cell migration in B16F10 cells. Therefore, we suggest that phytochemical has anticancer activity and has the potential to be an anticancer agent.
DB3 from Antarctic lichen inhibits the growth of B16F10 melanoma cells in vitro and in vivo

Malignant melanoma is a fatal disease with an increasing global incidence. Despite numerous studies focused on anti-cancer drugs, a variety of side effects of cancer treatment remain challenging. Thus, there is a pressing need to identify novel anti-cancer agents with minimal cytotoxicity and side effects. DB3 (1,3,7,9-tetrahydroxy-2,8-dimethyl-4,6-di[ethanoyl]dibenzofuran) is a member of the dibenzofuran family and is extracted from Ramalina terebrata (Antarctic lichen). We investigated if DB3 exerted an antitumor effect on B16F10 melanoma cells. The results revealed that DB3 exerted time- and dose-dependent reduction of cell viability by inducing apoptosis and significantly suppressing cell proliferation through cell cycle arrest in the G0/G1 phase in B16F10 melanoma cells. Additionally, DB3 impeded the migration and invasiveness of B16F10 cells. Subsequently, we observed that DB3 decreased the expression levels of Cdk4/Cyclin D1 and the phosphorylation of p38, JNK, ERK, and AKT. Furthermore, DB3 decreased melanoma tumor growth in a mouse tumor syngraft model. Based on these findings, we propose that DB3 possesses potential for use as an anti-cancer agent for melanoma treatment.
ETNA: Stretching Guidance Through Interactive Game Design

The usage rate of laptops is growing considerably over the years, resulting in muscular pains from prolonged laptop usage hours. To address this issue we propose the game ETNA, an interaction-based game with stretching guidance and narrative design as an affordable solution to alleviate such pains and enhance the postural health of laptop users. We conducted user testing to assess if our game design can effectively instruct users and facilitate them to better perform stretching exercises. It was found that the user interaction during the stretching guidance can promote ease of use and perceived usefulness. Also, the narrative element can improve the general interest and satisfaction by enhancing learnability and entertainment that can potentially lead to regular participation of the user. ETNA has the potential as an innovative solution to lower the costs and time boundaries of postural maintenance for laptop users.

This study investigated the microbiological hazards in the manufacturing processes of Naengmyeon. Sanitary indicative bacteria, such as aerobic plate counts and coliforms as well as pathogenic bacteria, were examined from raw materials, manufacturing processes, working area, 17 utensils and equipment. The aerobic plate counts for raw materials and arrowroot starch estimated as 0.77±0.68~5.02±0.28 and 5.02±0.28 log CFU/g, respectively. Coliforms were detected from wheat flour, buckwheat flour, and potato starch. Staphylococcus aureus was detected to be 0.61±1.06 log CFU/g in wheat flour and 0.20±0.35 log CFU/g in buckwheat flour. During the manufacturing process, aerobic plate counts for kneading process were 4.54±0.34 log CFU/g. But after the press out and heat process, contamination of aerobic plate counts and coliforms decreased and remained at a low level until the release process. Aerobic plate counts before washing disinfection of screw were 3.28±0.62 log CFU/100 cm<sup>2</sup>, the level of which was high in utensils and equipment that had contact with employees or water. These results represent not only an important indicator for the hygienic level but also a scientific basis for analyzing biological hazards, which lead to the introduction of HACCP for the production of safe and hygienic cold noodles processed by manufacturers.

Most people can use the existing application, providing walking courses, that focuses on directions. However, people with reduced mobility have difficulties in walking due to various factors. To solve this problem, we designed an application service that helps people with reduced mobility to take a safe walk and promotes their health.
Anti-Cancer Effects of Phytochemical A against B16F10 Melanoma Cells

Melanoma is one of the most common cancer worldwide and is an aggressive form that accounts for most of the deaths of patients with skin cancers. If the invasion of melanoma has occurred, it is hard to be cured and can be life-threatening. Phytochemicals are the metabolic compounds that were found in plants. It has already been known to prevent various chronic diseases including cancer. The chemical “A” is the phytochemical which is one of the isoflavones that are known to inhibit tumor growth and limit the production of cancer-related hormones. We focused on the anti-cancer effects of the phytochemical A that play important roles in B16F10 melanoma cells. The phytochemical A reduced cell viability and cell numbers. We also found that these effects are because of the inhibition of proliferation rather than cytotoxicity. In addition, the phytochemical A induced cell cycle arrest at the G0/G1 phase and inhibited the migration of melanoma cells. Taken together, these results suggest that phytochemical A could exert its effect against metastasis and the cell cycle in melanoma cells. Therefore, it implies that it could work as a chemotherapeutic agent for cancer treatment.
