        Activities of Sulfhydryl-Related and Phenylpropanoid-Synthesizing Enzymes during the Germination of Arabidopsis thaliana Seeds

        Seeds of Arabidopsis thaliana were sowed in a Murashige and Skoog's medium and incubated in a tissue culture chamber at 26.2℃. They appeared to germinate at 60-72 hours after sowing. We measured the tune-course activities of sulfhydryl-related enzymes such as thioredoxin, thiol-transferase (glutaredoxin), thioredoxin reductase, and glutathione reductase. The activities of two enzymes, phenylalanine ammonia-lyase and tyrosine ammonia-lyase, which are involved in phenylpropanoid synthesis, were also measured and compared. Phenylalanine ammonia-lyase activity increased from 48 hours after sowing, whereas tyrosine ammonia-lyase activity increased transiently at the early stage and decreased slightly afterwards. Thioredoxin activity gradually decreased during the germination process. However, thioltransferase activity increased drastically from 60 hours after sowing. Thioredoxin reductase and glutathione reducatase increased rapidly from 36 hours after sowing. Thioredoxin activity was found to be relatively high in the dormant seeds.

      • Structural basis for the dephosphorylating activity of PTPRQ towards phosphatidylinositide substrates.

        <P>Unlike other classical protein tyrosine phosphatases (PTPs), PTPRQ (PTP receptor type Q) has dephosphorylating activity towards phosphatidylinositide (PI) substrates. Here, the structure of the catalytic domain of PTPRQ was solved at 1.56?? resolution. Overall, PTPRQ adopts a tertiary fold typical of other classical PTPs. However, the disordered M6 loop of PTPRQ surrounding the catalytic core and the concomitant absence of interactions of this loop with residues in the PTP loop results in a flat active-site pocket. On the basis of structural and biochemical analyses, it is proposed that this structural feature might facilitate the accommodation of large substrates, making it suitable for the dephosphorylation of PI substrates. Moreover, subsequent kinetic experiments showed that PTPRQ has a strong preferences for PI(3,4,5)P3 over other PI substrates, suggesting that its regulation of cell survival and proliferation reflects downregulation of Akt signalling.</P>

      • Up-regulated transforming growth factor β–inducible gene h3 in rheumatoid arthritis mediates adhesion and migration of synoviocytes through αvβ3 integrin: Regulation by cytokines

        <B>Objective</B><P>To delineate the expression of transforming growth factor β–inducible gene h3 (βIG-H3) in rheumatoid synovitis and to determine the regulatory role of βIG-H3 in the adhesion and migration of fibroblast-like synoviocytes (FLS).</P><B>Methods</B><P>Synovial tissue was obtained from patients with rheumatoid arthritis (RA) during joint replacement surgery, and FLS were isolated using enzymatic treatment. Immunohistochemical staining was performed to show the expression of βIG-H3 within rheumatoid synovium. Synthesis of βIG-H3 from FLS was determined by semiquantitative reverse transcription–polymerase chain reaction, Western blot analysis, and enzyme-linked immunosorbent assay. Cell adhesion and migration assays were performed using the YH18 peptide in the fourth fas-1 domain of βIG-H3 and function-blocking antibodies to integrins.</P><B>Results</B><P>Expression of βIG-H3 was up-regulated in RA synovial tissue compared with synovial tissue from patients with osteoarthritis. FLS isolated from RA synovial tissue constitutively produced βIG-H3, which was up-regulated by transforming growth factor β1, interleukin-1β, and tumor necrosis factor α. Although FLS expressed a variety of integrins, βIG-H3 mediated adhesion and migration of FLS through interaction with αvβ3 integrin. Cytokines failed to affect the βIG-H3–mediated adhesion. However, migration of FLS guided by βIG-H3 was enhanced by interferon-γ and platelet-derived growth factor type BB. The YH18 peptide in the fourth fas-1 domain of βIG-H3 inhibited adhesion and migration in a dose-dependent manner.</P><B>Conclusion</B><P>The results suggest that βIG-H3, which is abundantly expressed in RA synovial tissue, plays a regulatory role in chronic destructive inflammation through the modulation of the adhesion and migration of FLS. This finding indicates the relevance of βIG-H3 and αvβ3 integrin–interacting motifs as potential therapeutic targets in this disease.</P>

        In vivo quantitative measurement of arthritis activity based on hydrophobically modified glycol chitosan in inflammatory arthritis: more active than passive accumulation.

        <P>We demonstrated that arthritis could be visualized noninvasively using hydrophobically modified glycol chitosan nanoparticles labeled with Cy5.5 (HGC-Cy5.5) and an optical imaging system. Activated macrophages expressing Mac-1 molecules effectively phagocytosed HGC-Cy5.5, which formed spherical nanoparticles under physiologic conditions. We estimated the applicability of HGC-Cy5.5 to quantitative analysis of arthritis development and progression. Near-infrared fluorescence images, captured after HGC-Cy5.5 injection in mice with collagen-induced arthritis, showed stronger fluorescence intensity in the active arthritis group than in the nonarthritis group. According to the progression of arthritis in both collagen-induced arthritis and collagen antibody-induced arthritis models, total photon counts (TPCs) increased in parallel with the clinical arthritis index. Quantitative analysis of fluorescence after treatment with methotrexate showed a significant decrease in TPC in a dose-dependent manner. Histologic evaluation confirmed that the mechanism underlying selective accumulation of HGC-Cy5.5 within synovitis tissues included enhanced phagocytosis of the probe by Mac-1-expressing macrophages as well as enhanced permeability through leaky vessels. These results suggest that optical imaging of arthritis using HGC-Cy5.5 can provide an objective measurement of disease activity and, at the same time, therapeutic responses in rheumatoid arthritis.</P>

        太極拳24式이 健康에 미치는 影響에 대한 小考

        Taegeuk is the source and union of the two primary aspects of the cosmos, Eum(陰, Yin) and Yang(陽). The Neo-Confucian philosophers of Chinese Song(宋) dynasty associated Taegeuk with the supreme rational principle of the universe. Taegeuk is a powerful blend of self-discipline, self-defense and healing. It is an art which has grown over the millennia to include hundreds of forms and variations, and can easily take a lifetime to master. Taegeukgwon based on the Taoist philosophies of Eum(陰, Yin) and Yang(陽) is an ancient Eastern form of exercises, characterized by slow reversing movements. There is strong research evidence validating Taegeuk as a safe form of exercise having potential for health promotion and rehabilitation. Today, Taegeukgwon has spread worldwide. The Internal Training of Taegeuk is one of the exercises for regiment. As exercise, Taegeukgwon is designed to provide relaxation in the process of body-conditioning exercise and is drawn from the principles of Taegeuk, notably including the harmonizing of the Eum-yang(陰陽, Yin-yang), respectively the passive and the active principles. It employs flowing, rhythmic, deliberate movements, with carefully individually prescribed stances and positions. In practice, two masters teach the system exactly alike. As a mode of attack and defense, Taegeukgwon resembles Kungfu and is properly considered a martial art. It may be used with or without weapons. Freehand exercise to promote health was practiced in Korea. This training proved to have effect on hypertension, enervation, impotence, premature ejaculation, seminal emission, chronic gastrointestinal diseases, and arteriosclerosis. There have been many schools of Taegeukgwon. Depending on school and master, the number of prescribed exercise forms varies from 24 to 108 or more. The objective of this paper is to reconsider Taegeukgwon and its effect on the body and mind-conditioning in Hwangjenaegyeong(黃帝內經)

        The stay cylinder weld at the steam generator of Korean Standard Nuclear Power Plants is safety class I component and is subjected to be inspected by the volumetric examination such as ultrasonic method. As accessibility of this area is limited due to the narrow space and high radiation, the existing manual inspection method involves various difficulties. Moreover operators may be exposed to internal contamination by contaminated dust during the surface buffing process to improve the inspection reliability of this area. Recently the new automatic inspection system for stay cylinder welds has been developed. The inspection system basically consists of a driving assembly, data acquisition device and signal processing units. The driving assembly is classified by 1) the scanner for inspecting and buffing the weld, 2) pillars for guiding the scanner and 3) the base frame for loading and supporting pillars. The scanner has 4 sensor modules to inspect in 4 refracted angles and 4 incident directions. These components can be inserted into the skirt of the stay cylinder through the manway hole and assembled easily by one-touch in the skirt. Data acquisition device and signal processing units developed in previous works are also newly upgraded for better processing of data analysis and evaluation. The system has been successfully demonstrated not only in the mock-up but also in the field. In this paper, newly developed inspection system for the stay cylinder weld of the steam generator is introduced and their field applications are discussed.

        βig-h3 절편-RGD 재조합단백의 만성염증성 관절염 치료효과

        Objective. βig-h3 is a 68kDa extracellular matrix protein which is overexpressed in synovial tissues of rheumatoid arthritis (RA). Previous results proved that βig-h3 fragments are relevant to adhesion and migration of synovial fibroblast and angiogenesis through interaction with αvβ 3 integrin. We designed a recombinant βig-h3 protein consisting of a fas-1 domain and RGD motif and evaluated the therapeutic efficacy in RA. Methods. Inhibitory effect of adhesion and migration of NIH3T3 cell line was evaluated in 96 well microtiter and transwell plates coated with βig-h3. Clinical arthritis index was evaluated after treating CIA mice with MFK12. Immunohistochemical staining in synovial tissues were performed. Expression of transcripts and proteins of inflammatory mediators were analyzed by semi-quantitative RT-PCR and immunoblotting. Results. Recombinant protein consisted of 4th fas-1 domain truncated for H1 and H2 sequences and RGD peptide (MFK12), had M.W. of 10.4kDa. βig-h3 mediated adhesion and migration of NIH3T3 cell line were significantly inhibited in a dose-dependent manner. Arthritis severity and incidence were efficiently reduced when CIA mice were treated with MFK12 at 30 mg/kg/day compared with the control. Immunohistochemical staining of joint tissues in MFK12 treated mice exhibited reduced angiogenesis. In treated mice, expression of transcripts regarding inflammatory mediators was markedly suppressed and immunoblotting of ICAM-1 and RANKL from whole extract of hind paws also showed a significant reduction. Conclusion. This study shows that MFK12 is effective in treating RA, although further study is warranted to improve the therapeutic efficacy. Key Words.         외래마취시 전처치제로 사용한 Midazolam의 마취회복 및 회복실 퇴실 시간에 미치는 영향

        Background: Midazolam is often used as an anxiolytic premedication before surgery. But preoperatively administered midazolam may contribute to postopertive sedation and delayed recovery from general anesthesia. This study was undertaken to evaluate the effect of midazolam premedication on postoperative recovery and discharge-readiness after brief outpatient surgery. Methods: Sixty healthy ASA physical status I women scheduled for outpatient diagnostic laparoscopic surgery were considered for the study. They were randomly allocated to one of two groups. Group one received normal saline (N/S) 5 ml intravenously (IV), while group two received IV midazolam 0.04 mg/kg. The study drug was prepared in 5 ml of saline and administered 10 minutes before the induction of general anesthesia. General anesthesia was induced with fentanyl, propofol and vecuronium and was maintained with N2O and enflurane. Postanesthetic recovery (PAR) scores were recorded after the arrival of the patients in the postanesthetic recovery room. Sedation was quantified before and after premedication and 60, 120 minutes after arriving in the postanesthetic recovery room, using the symbol-digit-modalities test (SDMT) and trail-making test (TMT). Results: There were no significant differences between the two groups with respect to age, weight and anesthesia time. There were no significant differences in PAR scores or PAR-stay time between two groups. SDMT and TMT scores were significantly different 5 minutes after the study's drug administration, and 60 minutes after arrival in the postanesthetic recovery room between the two groups. The incidence of side effects was similar in both groups. Conclusions: Midazolam premedication proved effective in sedation and anxiolysis without prolonging postanesthetic recovery and discharge times for outpatient general anesthesia.
