The effects of additions/deletions in glycosylated residues of recombinant human EPO (rhEPO) produced in CHO-K1 on their secretion were examined. hEPO cDNA was amplified from human liver mRNA and cloned into the pCR2.1 TOPO. Using overlapping-extension site-directed mutagenesis method, glycosylation sites at 24th, 38th, 83rd, and 126th were respectively or accumulatively removed by substituting its asparagine (or serine) with glutamine. To add novel glycosylation sites, 69 and 105th leucine was mutated to asparagine. Mutant and wild type rhEPO constructs were cloned into the pcDNA3 expression vector with CMV promoter and transfected into CHO cell line, CHO-K1, to produce mutant rhEPO mutant rhEPO proteins. Enzyme-linked immunosorbant assay (ELISA) and Western analysis with monoclonal anti-EPO antibody were performed using supernatants of the cultures showing transient and stable expressions respectively. Addition of novel glycosylation reduced rhEPO secretion dramatically while deletion mutants had little effect except some double deletion mutants (△24/83 and △38/83) and triple mutant (△24/38/83). This fact suggests that not single but combination of changes in glycosyl groups affect secretion of rhEPO in cell culture, possibly via changes in their conformations.
Control of Thermoelectric Properties through the addition of Ag in the Bi0.5 Sb1.5 Te3Alloy

In this study, the thermoelectric properties of the Ag-doped Bi0.5Sb1.5Te3 compounds were investigated in the temperature range from 323 K to 573 K. Ingots were fabricated by a conventional melting process and the powder crushed from ingots was then sintered using a hot-pressing method. The temperature dependence of the Seebeck coefficient and the electrical conductivity of the Ag-doped Bi0.5Sb1.5Te3compound are characteristic of degenerate semiconductors, which is fairly different from the conventional Bi0.5Sb1.5Te3 compound. The power factor (α 2 σ) of the quaternary compound was larger than that of the ternary, which is mainly due to the increase in the electrical conductivity with doping content of Ag. The thermal conductivity was greater than that of the Ag-freeBi0.5Sb1.5Te3compound in the temperature range from 323 K to 523 K. The lattice thermal conductivity showed low values throughout the temperature range. The maximum value of the dimensionless figure of merit (ZT) of the 0.05 wt. % Ag-doped compound and the ternary alloy were 1.2 at 373 K and 0.88 at 323 K, respectively. Each of the maximum peak ZT shifts to a higher temperature region with increases in the doping content of Ag. This is likely due to the control of the lattice thermal conductivity by the twin structure, which had a nano-ordered layer.
스텐실 프린팅에서 스퀴지 속도와 각도가 프린팅 성능에 미치는 영향 분석

본 연구에서는 솔더 페이스트를 이용한 스텐실 프린팅 공정의 해석에 대해 연구를 수행하였다. 스텐실 프린팅 공정의 설계에서 중요한 인자는 프린팅 조건, 스텐실 설계, 그리고 솔더 페이스트의 물성 등이다. 본 연구에서는 이 인자들 중에서, 프린팅 조건인 스퀴지 각도와 스퀴지 속도에 따른 솔더 페이스트 내부에 형성되는 압력 분포에 대해 해석을 수행하여, 프린팅 공정의 성능에 미치는 영향을 파악하였다. 해석 결과, 프린팅 조건은 솔더 페이스트 내부 압력 분포형성에 큰 영향을 줌을 알 수 있었다.
지속형 천식 환자에서 아콜레이트 (R) (Zafirlukast) 투여에 따른 유효성과 안전성 및 내약성 평가를 위한 비대조, 다기관 임상연구

Background and Objective:Accolate, one of the cysteinyl leukotriene receptor antagonists, have anti-inflammatory and bronchodilating effects. The clinical indications for LT antagonist are not yet in consensus. We performed a non-comparative multicenter trial to assess the additional responses of Accolate to the current long-term control regimens in persistent asthma. Materials and Methods: Eighty persistent asthmatics from 10 asthma special care clinics were enrolled. Daily 20 mg of Accolate was added 2 times to the current control regimen for 6 weeks and its effects on symptom scores, morning and evening PEFR, FEV₁ and frequency of the short acting 2 agonist usage for relief of asthma were measured. Results: The morning symptom scores significantly improved by addition of Accolate for 1 week (p$lt;0.01) and its effect persisted to the 6th week (p$lt;0.01) of trial. Nocturnal symptom scores also improved at 2nd week (p$lt;0.05) and the effect continued to the end of this trial. Days with morning asthma symptoms deceased by addition of Accolate at 4th week (p$lt;0.05) and 6th week (p$lt;0.05). The frequency of the short acting β₂- agonist inhalations significantly decreased by addition of Accolate from 1st week to the end of this trial. Morning and evening PEFR significantly improved at 1st week and the effects progressively improved to the end of this trial. The FEV1 not improve by addition of Accolate. However, when the asthmatic with obstructed lung function (FEV₁ $lt;80%) were enrolled only for analysis, the FEV₁ significantly improved at 3rd (p$lt;0.05) and 6th weeks (p$lt;0.05). In 21 asthmatics, oral prednisolone was continuously used at the beginning of this trial. It could be discontinued in 4 patients and the dosage of prednisolone was significantly reduced in another 3 patients by addition of Accolate. Their mean administered dosage of prednisolone decreased from 8.6±0.9 mg/day to 6.8±1.1 mg/day at the end of this trial (p$lt;0.05). Conclusions: With these results, we thought that addition of Accolate as a long term control imen in the symptomatic persistent asthmatics would be an effective strategy. For evaluation of steroid sparing effects, further studies will be needed.

Recently, according to the rapid development of display, many display applications, such as, cellular phone, navigation, monitor and LCD TV have been changed from CRT type to LCD type. BLU(Back Light Unit) is one of main parts in LCD unit and, generally, it consists of a light source, a reflective sheet, a LGP(Light Guide Plate), a diffuser sheet, and two prism sheets. The most important component of BLU is a light guide plate, which diffuses the input light to the TFT-LCD module uniformly. The LGP is usually made by injection molding process, and it has numerous optical micro patterns on the surface. In the present study the micro-patterned stamper which has cylindrical shape was fabricated by using the UV-LiGA process. And the replication characteristics have been compared among three different kinds of injection molding process; general injection molding, injection/compression molding and RHCM(Rapid Heating and Cooling Molding). Average replication ratios of CIM and ICM were 19.1% and 64.6%, respectively. On the other hand, average replication ratio of RHCM process showed the higher value of 98.4% among those. Finally, maintaining the mold surface above Tg could increase the replication ratio of micro patterns substantially.
