AtBAG6, a novel calmodulin-binding protein, induces programmed cell death in yeast and plants

Calmodulin (CaM) influences many cellular processes by interacting with various proteins. Here, we isolated AtBAG6, an Arabidopsis CaM-binding protein that contains a central BCL-2-associated ethnogeny (BAG) domain. In yeast and plants, overexpression of AtBAG6 induced cell death phenotypes consistent with programmed cell death (PCD). Recombinant AtBAG6 had higher affinity for CaM in the absence of free Ca^2+ than in its presence. An IQ motif(IQXXXRGXXXR, where X denotes any amino-acid) was required for Ca^2+ -independent CaM complex formation and single amino-acid changes within this motif abrogated both AtBAG6-activated CaM-binding and cell death in yeast and plants. A 134-amino-acid stretch, encompassing both the IQ motif and BAG domain, was sufficient to induce cell death. Agents generating oxygen radicals, which are known to be involved in plant PCD, specifically induced the AtBAG6 transcript. Collectively, these results suggest that AtBAG6 is a stress-upregulated CaM-binding protein involved in plant PCD.

Primary endometrial squamous cell carcinoma is extremely rare, with only 64 cases repo- rted in the literature. In 1928 Fluhmann proposed three criteria for differentiating primary from secondary endometrial squamous cell carcinoma:(1) no coexisting endometrial adenocarcinoma (2) no connection between the endometrial tumor and the squamous epithelium of the cervix, and (3) no squamous cell carcinoma of the cervix present. Several authors have modified these criteria in their case presentations. Kay accepts the presence of an in situ cervical carcinoma, if there is no connection between cervical carcinoma in situ and the endometrial tumors. We have experienced a case of simultaneous carcinoma in situ of the cervix and endomet- rial squamous cell careinoma. This case was treated with chemotherapy(cisplatin, 5-fluoruracil) and external radiation following extended hysterectomy and bilateral salpingooophorectomy We report this case with a review of the concerned literatures.

Asphyxiating thoracic dysplasia(ATD;Jeunes`s syndrome) is a rare variety of short limb dwarfism. It is characterized by an extremely small thorax when compared to the abdominal circumference, which frequently results in respiratory distress. Other anomalies associated with Jeune`s syndrome are pelvic bone malformations and renal dysplasia. It was first described and namely by Jeune et al. in 1954. Jeune`s syndrome is an autosomal recessive trait and has a 25% recurrence risk. These patients died at early age due to respiratory insufficiency. Death due to uremia has occurred in number of children surviving infancy, following progressive renal failure, hypertension and hepatic failure. About 50 cases have been reported in the world literature. We experienced a case of small thorax with short limb dwarfism on antenatal ultrasound examination and then the baby was delivered by cesarean section. The diagnosis was confirmed to Asphyxiating thoracic dysplasia by clinical features, radiological findings and pathological findings. We reported a case of Asphyxiating thoracic dysplasia with review of literatures.

The cystic teratoma is the most common germ cell tumors in reproductive age women. The malignant change of cystic teratoma is rare, about 1∼2%, and mostly change to squ-amous cell carcinoma. We experienced 4 cases of squamous cell carcinoma arising in cystic teratoma and re- port our cases with a brief review of the literature.

Scar endometriosis is one of the rare conditions among the extrapelvic endometriosis, but it often occurs after pelvic operation and episiotomy. Four cases of scar endometriosis following Cesarean section and episiotomy were exp- erienced and treated with complete surgical excision. Therefore we reported these cases with concerned literatures.
난자 채취 전 시행한 혈청 난포자극호르몬치 및 난포호르몬치와 GnRH 효능제 사용 체외수정시술 예후에 관한 연구

총 172예의 GnRH 효능체와 hMG/FSH 사용 체외 수정 주기에서 생리 제3일의 혈청 FSH, LH, estradiol 농도와 hCG 투여일의 혈청 estradiol 농도를 회수 난자수, 성숙난자수, 수정란의 수, 수정률, 배아 발달 지수, 주기당 임신율과 비교하여 다음의 결과를 얻었다. 1. 생리 제3일의 혈청 FSH 농도는 단일 종목 검사로 난소원을 예측할 수 있는 가장 유용한 검사이다. 2. 생리 제3일의 혈청 LH 및 estradiol 농도는 FSH 농도와 병행하여 사용하는 것이 난소원 예측에 도움을 줄 수 있다. 3. hCG 투여일의 혈청 estradiol 농도 2000 pg/ml 이상인 고응답군은 체외수정으로 좋은 결과를 얻을 가능성이 높다. 4. 수정란의 형태학적 양호도와 세포 분열 정도를 포함하는 배아 발달 지수는 hCG 투여일의 혈청 estradiol 농도 및 단위 난자당 estradiol 농도와 양의 상관관계를 보였다. 이상의 결과로 미루어 생리 제3일의 혈청 FSH 농도는 가용 난소원을 예측할 수 있는 가장 유용한 단일 종목 검사이며 hCG 투여일의 높은 혈청 estradiol 농도는 GnRH 효능제 사용 체외수정 각 단계의 좋은 결과를 예측하게 한다는 결론에 도달하였다. Purpose: To evaluate the prognostic value of serum follicular stimulating hormone (FSH) level and serum estradiol (E2) level before ovum pick up during In Vitro Fertilization-Embryo Transfer cycle. Method: We retrospectively analyzed the 172 cycles underwent oocyte retrieval for IVF after controlled ovarian hyperstimulation with GnRH and hMG/FSH. Serum hormone levels including FSH and LH and E2 on menstrual day 3 (Basal E2), E2 on hCG administraion day (Day 0 E2) were determined using radioimmunoassay. We also analyzed IVF-ET outcome (No. of oocytes, No. of metaphase Ⅱ oocytes, No. of transferable embryos, fertilization rate, index of embryo development, pregnancy rate per cycle) related to the determined serum hormone levels. Results: 1. No. of retrieved oocyte was higher in the low FSH group (<15 mIU/mL) than in the high FSH group (≥15 mIU/mL) (9.7±8.3, 7.0±1.7; P =0.001). 2. Pregnancy rate was significantly different between two groups (13.0 %/cycle in FSH<15 mIU/mL, 0 %/cycle in FSH>15 mIU/mL; P<0.001). 3. Pregnancy rate was higher in the high responder group (E2>2000 pg/ml) than in low responder group (E2<2000 pg/ml) (25.0, 3.7 %/cycle; P<0.001). 4. Pregnancy rate was higher in high E2/Ovum group (>200 pg/ml) than in low E2/Ovum group (<200 pg/ml) (16.0, 5.3 %/cycle; P<0.001). 5. There was a positive correlation between index of embryo development and E2/Ovum(γ=0.265; P<0.05). Conclusion; Day 3 serum FSH is the best single predictor of ovarian reserve and high serum estradiol level of the hCG administration day is an useful predictive parameter for favorable outcome of the IVF cycle treated with GnRH agonist.
