        Molecular and Biochemical Characterization of a Novel Intracellular Low-Temperature-Active Xylanase

        A 990 bp full-length gene (xynAHJ2) encoding a 329- residue polypeptide (XynAHJ2) with a calculated mass of 38.4 kDa was cloned from Bacillus sp. HJ2 harbored in a saline soil. XynAHJ2 showed no signal peptide, distinct amino acid stretches of glycoside hydrolase (GH) family 10 intracellular endoxylanases, and the highest amino acid sequence identity of 65.3% with the identified GH 10 intracellular mesophilic endoxylanase iM-KRICT PX1-Ps from Paenibacillus sp. HPL-001 (ACJ06666). The recombinant enzyme (rXynAHJ2) was expressed in Escherichia coli and displayed the typical characteristics of low-temperatureactive enzyme (exhibiting optimum activity at 35 o C, 62% at 20 o C, and 38% at 10 o C; thermolability at ≥45 o C). Compared with the reported GH 10 low-temperature-active endoxylanases, which are all extracellular, rXynAHJ2 showed low amino acid sequence identities (<45%), low homology (different phylogenetic cluster), and difference of structure (decreased amount of total accessible surface area and exposed nonpolar accessible surface area). Compared with the reported GH 10 intracellular endoxylanases, which are all mesophilic and thermophilic, rXynAHJ2 has decreased numbers of arginine residues and salt bridges, and showed resistance to Ni 2+ , Ca 2+ , or EDTA at 10 mM final concentration. The above mechanism of structural adaptation for low-temperature activity of intracellular endoxylanase rXynAHJ2 is different from that of GH 10 extracellular low-temperature-active endoxylanases. This is the first report of the molecular and biochemical characterizations of a novel intracellular low-temperatureactive xylanase.

        Improved photocatalytic performance of Pd-doped ZnO

        ZnO and Pd-doped ZnO photocatalysts with different molar ratio of Pd/Zn (1/100, 2/100, 3/100 and 4/100) were prepared by a solegel method. The photocatalysts prepared were characterized by BET surface area, X-ray diffraction (XRD), UV/vis diffuse reflectance (DRS) and surface photovoltage spectroscopy (SPS), respectively. The results show that doping Pd into ZnO decreases the BET surface area. The XRD spectra of the Pd-doped ZnO catalysts calcined at 773 K show only the characteristic peaks of wurtzite-type. Doping Pd into ZnO increases the absorbance of ZnO in visible region and enhances the photoinduced charge separation rate. The photocatalytic activity of Pd-doped ZnO photocatalysts for decolorization of methyl orange (MO) solution was evaluated, of all the photocatalysts prepared, the Pddoped ZnO with 3/100 possesses the best photocatalytic activity. The results of further experiments show that increased adsorption ability of light and high separation rate of photoinduced charge carriers all play an important role in promotion of photocatalytic activity of Pd-doped ZnO nanostructure.

        De novo assembly of the transcriptome for Greenbug (Schizaphis graminum Rondani) and analysis on insecticide resistance-related genes

        The greenbug, Schizaphis graminum Rondani is a major pest species of wheat crops. In this study, a transcriptome sequencing, and the expression of the 12 genes related to insecticide resistance were conducted in S. graminum. The sequencing and subsequent bioinformatics analysis outputed 46,593 unigenes, among which 28,289 unigenes were annotated to corresponding functions by blasting with high homologous genes in database, giving annotation rate of 60.72%. To gain insight into the mechanism of insecticide resistance, the expression of the 12 genes related to insecticide resistance for S. graminum was investigated. The expression level of aminopeptidase N (AN), cytochrome P450 (CYP), acetylcholinesterase 1 (AC), catalase (CAT), cytochrome c oxidas (CCC), GABA receptor (GABA), glutathione S-transferase (GST) were highest in the apterous nymphs among different developmental stages; The expression level of AN, CBL, CYP, CA, SD, and GST were relatively more abundant in the abdomen compared to head and throax. The results could give out the key information about the relationship between the expression of these genes in different developmental stages, tissues, treatments and metabolism of insecticides. These genes that were co-up-regulated significantly in third instar nymphs of S. graminum induced by imidacloprid, were consistent with their putative involvement in insecticide resistance. This provides most comprehensive transcriptome data for S. graminum to further study and managenment.

        Plant Virome Analysis by the Deep Sequencing of Small RNAs of Fritillaria thunbergii var. chekiangensis and the Rapid Identification of Viruses

        Thunberg fritillary (Fritillaria thunbergii), a perennial used in traditional Chinese herbal medicine, is a members of the family Liliaceae. The degeneration of germplasm is a severe problem in the production of Fritillaria thunbergii var. chekiangensis. However, no information about viral infections of F. thunbergii var. chekiangensis has been reported. In this study, we sequenced the small RNAs of F. thunbergii var. chekiangensis from leaves and bulbs, and viruses were identified using a phylogenetic analysis and BLAST search for sequence. In addition, multiplex reverse transcriptase-polymerase chain reaction (RT-PCR) was used to rapidly detect viruses in this variety. Our study first reported that five viruses infected F. thunbergii var. chekiangensis. Among them, fritillary virus Y (FVY), lily mottle virus (LMoV), Thunberg fritillary mosaic virus (TFMV), and hop yellow virus (HYV) had been reported in F. thunbergii, while apple stem grooving virus was first reported in the genus Fritillaria. A multiplex RT-PCR method was developed to rapidly test the four viruses FVY, LMoV, TFMV, and HYV in F. thunbergii var. chekiangensis. Our results provide a better understanding of the infection of F. thunbergii var. chekiangensis by viruses and a basic reference for the better design of suitable control measures.

      • Weight Loss Correlates with Macrophage Inhibitory Cytokine-1 Expression and Might Influence Outcome in Patients with Advanced Esophageal Squamous Cell Carcinoma

        Background: Weight loss during chemotherapy has not been exclusively investigated. Macrophage inhibitory cytokine-1 (MIC-1) might play a role in its etiology. Here, we investigated the prognostic value of weight loss before chemotherapy and its relationship with MIC-1 concentration and its occurrence during chemotherapy in patients with advanced esophageal squamous cell carcinoma (ESCC). Materials and Methods: We analyzed 157 inoperable locally advanced or metastatic ESCC patients receiving first-line chemotherapy. Serum MIC-1 concentrations were assessed before chemotherapy. Patients were assigned into two groups according to their weight loss before or during chemotherapy:>5% weight loss group and ${\leq}5%$ weight loss group. Results: Patients with weight loss>5% before chemotherapy had shorter progression-free survival period (5.8 months vs. 8.7 months; p=0.027) and overall survival (10.8 months vs. 20.0 months; p=0.010). Patients with weight loss >5% during chemotherapy tended to have shorter progression-free survival (6.0 months vs. 8.1 months; p=0.062) and overall survival (8.6 months vs. 18.0 months; p=0.022), and if weight loss was reversed during chemotherapy, survival rates improved. Furthermore, serum MIC-1 concentration was closely related to weight loss before chemotherapy (p=0.001) Conclusions: Weight loss both before and during chemotherapy predicted poor outcome in advanced ESCC patients, and MIC-1 might be involved in the development of weight loss in such patients.

        Simulations of the hysteretic behavior of thin-wall cold-formed steel members under cyclic uniaxial loading

        In this paper, the hysteretic behaviors of channel and C-section cold-formed steel members (CFSMs) under cyclic axial loading were simulated with the finite element method. Geometric and material nonlinearities, Bauschinger effect, strain hardening and strength improvement at corner zones were taken into account. Extensive numerical results indicated that, as the width-to-thickness ratio increases, local buckling occurs prematurely. As a result, the hysteretic behavior of the CFSMs degrades and their energy dissipation capability decreases. Due to the presence of lips, the hysteretic behavior of a C-section steel member is superior to that of its corresponding channel section. The intermediate stiffeners in a C-section steel member postpone the occurrence of local buckling and change its shapes, which can greatly improve its hysteretic behavior and energy dissipation capability. Therefore, the CFSMs with a large width-to-thickness ratio can be improved by adding lips and intermediate stiffeners, and can be used more extensively in residential buildings located in seismic areas.

        Nonlinear Optical Properties of Silicon Nanocrystals Grown by Using a SiOx/SiO2 Superlattice Approach

        The nonlinear optical properties have been studied using the Z-scan technique for silicon nanocrystals embedded in a SiO2 matrix formed by high-temperature annealing of SiOx/SiO2 superlattices grown by thermal evaporation. A mode-locked Ti:sapphire laser system producing 140-fs-long pulses at 800 nm was used as the optical source for the Z-scan measurements. The nonlinear refractive index and the nonlinear absorption coefficient of the silicon nanocrystals were found to be 1.2 × 10−13 cm2/W and 1.5 × 10−9 cm/W, respectively, and to be strongly enhanced compared to those of bulk silicon. Such enhancement of the nonlinear optical properties is considered to be due to the quantum confinement effect of silicon nanocrystals.

        Molecular characterization and inhibition analysis of the acetylcholinesterase gene from the silkworm maggot, Exorista sorbillans

        Several organophosphorus (OP) insecticides can selectively kill the silkworm maggot, Exorista sorbillans (Es) (Diptera: Tachinidae), while not obviously affecting the host (Bombyx mori) larvae, but the mechanism is not yet clear. In this study, the cDNA encoding an acetylcholinesterase (AChE) from the field Es was isolated. One point mutation (Gly353Ala) was identified. The Es-353G AChE and Es-353A AChE were expressed in baculovirus- insect cell system, respectively. The inhibition results showed that for eserine and Chlorpyrifos, Es-353A AChE was significantly less sensitive than Es-353G AChE. Meanwhile, comparison of the I(50) values of eserine, dichlorvos, Chlorpyrifos and omethoate of recombinant Es AChEs with its host (Bombyx mori) AChEs indicated that, both Es AChEs are more sensitive than B. mori AChEs. The results give an insight of the mechanism that some OP insecticides can selectively kills Es while without distinct effect on its host, B. mori. [BMB reports 2010; 43(8): 573-578]

        Fermented cottonseed meal improves production performance and reduces fat deposition in broiler chickens

        Objective: This study was conducted to investigate the effect of fermented cottonseed meal (FCSM) on growth performance, carcass traits, and fat deposition in white-feather broiler chickens. Methods: A total of 480 male one-day-old white-feather broiler chickens were selected randomly and divided into four groups with six replicates of 20 chickens in each. The experimental chickens were fed diets including 3%, 6%, or 9% FCSM fermented by Candida tropicalis until 42 days old. In the experiment, the chickens of the control group were fed soybean meal. Results: FCSM supplementation linearly decreased the feed conversion ratio from d 15 to 21 and d 36 to 42, respectively (p<0.05). The percentage of carcass and semi-eviscerate increased in response to dietary FCSM supplementation at d 21 (p<0.05). The percentage of eviscerated and semi-eviscerate of 3FCSM was higher than that in other groups at d 35 (p<0.05). At the age of 42 d, the percentage of carcass increased in a quadratic way among increasing FCSM in diets (p<0.05). The subcutaneous fat thickness linearly decreased with the increasing levels of FCSM at d 21 (p<0.05). Gompertz and Logistic functions provided a better fit on abdominal fat and subcutaneous fat, respectively. The best fitted equation predicted that the maximum growth rate of abdominal fat weight and subcutaneous fat thickness occurred at d 28. FCSM had no significant effects on the shape of growth curve of abdominal fat weight and subcutaneous fat thickness, but reduced the height of the curve. Birds receiving the 6FCSM diet for 21 d had smaller adipocyte surface and lower serum glucose as well as triglyceride concentration. Conclusion: FCSM is beneficial for broiler chickens as it positively affects their growth and carcass in addition to altering their fat deposition.       • Protective Effects of Scutellaria barbata Against Rat Liver Tumorigenesis

        Scutellaria barbata D. Don (S. barbata), a traditional Chinese medicine, is used to treat cancers, inflammation, and urinary diseases. This study aimed to determine any protective effects of S. barbata crude extract (CE-SB) against rat liver tumorigenesis induced by diethylnitrosamine (DENA). Liver malfunction indices in serum were measured by biochemical examination. Hematoxylin and eosin staining was performed to examine liver pathology. Contents of malondialdehyde (MDA) and superoxide dismutase (SOD) were measured in liver homogenates to evaluate oxidative stress. The levels of liver malfunction indices in the CE-SB groups, especially in the CE-SB high dose group, were lower than that of the model group (P<0.05). The results from histological examination indicated that the number of liver nodules in the CE-SB groups decreased compared with the model group (P<0.05). Content of MDA determined in liver was significantly decreased, and level of SOD elevated by CE-SB. CE-SB can inhibit experimental liver tumorigenesis and relieve hepatic injury in rats.
