An protease from Actinidia arguta for improving meat tenderness was purified, characterizedfrom wild A. arguta fruit by ammonium sulfate precipitation, Sephdex G-25 gel filtration chromatography,and DEAE Sepharose Fast Flow ion exchange chromatography, and its activity was investigated. Thepurified protease was subjected to sodium dodecyl sulfate polyacrylamide gel electrophoresis toobtain a single band of protease. The protease was purified successfully, and found to have a molecularweight of 23.8 kDa (mass spectrometry). The specific activity of the purified protease reached 53,428U/mg with a 25.5-fold purification factor and 9% activity recovery. Based on N-terminal sequencingresults, the A. arguta protease was derived from the class of actinidia proteases that have an Nterminalsequence of VLPDY VDWRS AGAVV. The protease was effective for tenderizing beef anddecomposing actomyosin, suggesting the potential application for improving meat tenderness.
Valve core shapes analysis on flux through control valves in nuclear power plants

Control valves are widely used to regulate fluid flux in nuclear power plants, and there are more than1500 control valves in the primary circuit of one nuclear power plant. With their help, the flux can beregulated to a specific level of water or steam to guarantee the energy efficiency and safety of the nuclearpower plant. The flux characteristics of the control valve mainly depend on the valve core shape. In orderto analyze the effects of valve core shapes on flux characteristics of control valves, this paper focuses onthe valve core shapes. To begin with, numerical models of different valve core shapes are established, andresults are compared with the ideal flux characteristics curve for the purpose of validation. Meanwhile,the flow fields corresponding to different valve core shapes are investigated. Moreover, relationshipsbetween the valve core opening and the outlet flux under different valve core shapes are carried out. The purpose of the study is how to educate construction IT convergence for BIM engineers. With the advent of the fourth industry in recent years, BIM has attracted much attention in the construction industry. However, There are not many Construction-IT experts who can link the 4th industry with BIM. There is an increasing demand for construction IT convergence education that connects the 4th industry and BIM. How should we design construction IT convergence training for construction engineers? We surveyed 38 BIM engineers and 88 construction professionals in order to find a direction for construction IT convergence education for BIM engineers.

<P>The thermolysis of a heterobimetallic Sn–Co organometallic precursor resulted in the formation of hollow Sn/CoO<SUB><I>x</I></SUB> nanomaterials through galvanic reaction between metal species. <P>Vps34, the catalytic subunit in the class III phosphatidylinositol 3 kinase complexes, mediates the production of PtdIns3P, a key intracellular lipid involved in regulating autophagy and receptor degradation. Xiao et al. show that DNA damage-activated mitotic arrest and CDK activation lead to the phosphorylation of Vps34. This provides a signal to promote Vps34 ubiquitination and proteasomal degradation mediated by FBXL20, leading to inhibition of autophagy and receptor endocytosis. Importantly, they also find that expression of FBXL20 is regulated by p53-dependent transcription.</P><P>Vacuolar protein-sorting 34 (Vps34), the catalytic subunit in the class III PtdIns3 (phosphatidylinositol 3) kinase complexes, mediates the production of PtdIns3P, a key intracellular lipid involved in regulating autophagy and receptor degradation. However, the signal transduction pathways by which extracellular signals regulate Vps34 complexes and the downstream cellular mechanisms are not well understood. Here we show that DNA damage-activated mitotic arrest and CDK activation lead to the phosphorylation of Vps34, which provides a signal to promote its ubiquitination and proteasomal degradation mediated by FBXL20 (an F-box protein) and the associated Skp1 (S-phase kinase-associated protein-1)–Cullin1 complex, leading to inhibition of autophagy and receptor endocytosis. Furthermore, we show that the expression of FBXL20 is regulated by p53-dependent transcription. Our study provides a molecular pathway by which DNA damage regulates Vps34 complexes and its downstream mechanisms, including autophagy and receptor endocytosis, through SCF (Skp1–Cul1–F-box)-mediated ubiquitination and degradation. Since the expression of FBXL20 is regulated by p53-dependent transcription, the control of Vps34 ubiquitination and proteasomal degradation by FBXL20 and the associated SCF complex expression provides a novel checkpoint for p53 to regulate autophagy and receptor degradation in DNA damage response.</P>
