        牧楡湯이 人體 肺癌細胞의 變化에 미치는 影響에 關한 分子生物學的 硏究

        This experimental study was performed to investigate the effects of Mokyou-Tang on cell cycle and gene expressions of human lung cancer cell. I examined the cell cycle progression and cell cycle-related gene expression in A549 using a flow cytometry and a quantitative PT-PCR analysis. The treatment groups, in accordance with concentration of Mokyou-Tang were classified as 3 group(MYT 100㎕/㎖, MYT 500㎕/㎖, MYT 1,000㎕/㎖), and according to treatment time, were classified as 2 group(24hours, 48hours). The results were summarized as follows; 1. In cell cycle stage, MYT 100㎕/㎖(48hours) and 1000㎕/㎖(48hours) groups were increased as compared with control group, especially 1000㎕/㎖(48hours) group showed eminent increase. 2. In p53 gene, MYT 500㎕/㎖(48hours) and 1000㎕/㎖(48hours) groups showed eminent suppressive effect to cancer cell. 4. CDKNI gene was decreased in all treatment groups, this mean that MYT has suppressive effect to proliferation of cancer cell. 4. In GADD45 gene was decreased in all treatment groups, this mean that MYT has suppressive effect to proliferation of cancer cell. The results suggest that MYT promotes G1 cell cycle arrest and suppresses the proliferation of cancer cell. Thus, it seems likely that MYT has suppressive effect to proliferation of lung cancer cell.

      • 고립성 폐결절의 조직학적 진단에 있어서 경기관지폐생검의 진단적 가치

        Transbronchial lung biopsy(TBLB) is a relatively simple and convenient diagnostic tool in solitary pulmonary nodule(SPN) on chest radiograph, which is cannot be diagnosed through routine tests. To investigate the diagnostic value and diagnostic rate of TBLB according to the variable factor, we analyzed 69 patients with radiologically SPN lesion on chest radiograph which was confirmed histologically. A histologic diagnosis of TBLB was made in 47 of 69 patients(68.1%), in which 26 of 37(70.3%) with malignancy and 21 of 32(65.6%) with benign lesion. There were no significant differences in diagnostic yield according to age, sex and location of the lesion. The diagnostic rate tended to increase with the more than 5 pieces to acquired by TBLB. When the size of SPN was less than 3 ㎝, it cannot be diagnosed. In conclusion, TBLB is a diagnostic procedure with good yield and safety, but percutaneous needle aspiration, transbronchial needle aspiration, and bronchial washing fluid cytology may be a complementary procedure to overcome the limitation of TBLB and get higher diagnostic yield for SPN.

      • 都市街路의 遲滯度推定에 관한 硏究

        This study suggested input variables whose effects have been considered by adjusting various input variables in the TRANSYT-7F and NETSIM, the Simulation programs employed in calculating the delay time at street sections. The results can be summarized as follow. 1. Among the input variables in the TRANSYT-7F needed to calculate the delay time at street sections, the excluded are those which effect the most on time delay, such as bus stops, pedestrian crossings and on/off ramps. Among the input variables the most affecting factors are Saturation flow rate and time delay whose suggested quotients are well summarized in the text of this study. 2. There are variables which affect the change of time delay in the NETSIM, such as bus stop, pedestrian crossing, bus appearance period, and departure time delay. This study did the Simulation program using those variables except pedestrian crossing because, as the result of the correlation analysis suggests, there is a strong correlation between bus stop and pedestrian crossing. In this study, the survey had been done in the morning and afternoon except the Peak time, so heavily congested sections were excluded. And both sides of three street sections were surveyed. Therefore, there should be further studies on heavily congested situations and many other street sections.

      • Enhancement of 2,3-butanediol Production Using Extracted Sugars by Chlorella pyrenoidosa and Biodiesel-derived Crude Glycerol from Klebsiella aerogenes ATCC 29007

        In this study, extracted sugars of microalgae, which cause algal blooms and crude glycerol, which is a biodiesel industry by-product, were used simultaneously to produce 2,3-BDO. The 2,3-BDO production using only extracted algal sugars was about 4.8 g/L at 18 h, and the production of 2,3-BDO using both extracted algal sugar and crude glycerol was about 7 g/L at 18 h. It was confirmed that the main culture with crude glycerol was increased 1.5-fold compared to the case of using only extracted algal sugars. In addition, four components of the main medium (ammonium sulfate, casein hydrolysate, yeast extract and crude glycerol) were statistically optimized and the concentrations of the medium were 12 g/L, 16 g/L, 12 g/L and 13 g/L, respectively. In addition, the final 2,3-BDO production was about 11g/L, which 1.6-fold higher than before optimization process. As a result, it has been confirmed that 2,3-BDO production is possible through the simultaneous use of algal sugars and crude glycerol, which can greatly contribute to the development of zero-waste processes.

        Clinicopathological features of diabetic and nondiabetic renal diseases in type 2 diabetic patients with nephrotic-range proteinuria

        <▼1><P>Supplemental Digital Content is available in the text</P></▼1><▼2><P><B>Abstract</B></P><P>Heavy proteinuria with or without features of nephrotic syndrome is associated with many primary and systemic diseases. For diabetic patients, distinguishing nondiabetic renal disease (NDRD) from diabetic nephropathy (DN) is important in choosing treatment modalities and determining renal prognosis. However, clinical relevance of heavy proteinuria is inconsistent with clinical DN assessments. This study investigated the clinicopathological features and renal outcomes of DN and NDRD in type 2 diabetic patients with nephrotic-range proteinuria.</P><P>We enrolled 220 cases of type 2 diabetic patients who underwent renal biopsy. They were grouped according to the presence of nephritic-range proteinuria and pathological features. Baseline characteristics, laboratory findings, types of pathological diagnosis, and renal outcomes were analyzed in patients with heavy proteinuria.</P><P>Upon kidney biopsy, 129 patients (58.6%) showed nephritic-range proteinuria. Patients with heavy proteinuria (an average urine protein-to-creatinine ratio of 10,008 ± 7307 mg/gCr) showed lower serum albumin levels and higher total cholesterol levels, but did not show any difference in age, duration of diabetes, renal function, or the presence of retinopathy compared with those with mild-to-moderate proteinuria (an average urine protein-to-creatinine ratio of 1581 ± 979 mg/gCr). Renal biopsy revealed that the prevalence of NDRD was 37.2% in patients with heavy proteinuria, which was significantly lower than that in patients with mild-to-moderate proteinuria (63.7%). The most common pathological types of NDRD were membranous nephropathy (41.7%), IgA nephropathy (14.6%), and minimal change disease (10.4%). NDRD patients showed lower prevalence of diabetic retinopathy and better kidney function irrespective of proteinuria. Immunosuppressive treatment was administered more frequently in patients with heavy proteinuria (56.3%) compared with patients with mild-to-moderate proteinuria (20%) because of the pathological differences according to the amount of proteinuria. Renal outcomes were significantly worse in patients with DN than in patients with NDRD.</P><P>DN patients with heavy proteinuria exhibited different prevalence of NDRD and worse prognosis. Renal biopsy in type 2 diabetic patients should be more extensively considered to accurately diagnose NDRD, guide further management, and predict renal outcomes, especially in patients with nephrotic-range proteinuria.</P></▼2>

      • Identification of a serodiagnostic antigen, legumain, by immunoproteomic analysis of excretory-secretory products of Clonorchis sinensis adult worms

        <P>Clonorchis sinensis, the Chinese liver fluke, is the causative agent of clonorchiasis as well as liver and biliary diseases. The excretory-secretory products (ESPs) of the parasites play important roles in host–parasite interactions. In this study, we have investigated the proteome of ESPs obtained from C. sinensis adult worms. Although the full genome database of C. sinensis is not yet available, we have successfully identified 62 protein spots using 2-DE-based mass analysis and EST database of C. sinensis. The proteins identified include detoxification enzymes, such as glutathione S-transferase and thioredoxin peroxidase, myoglobin and a number of cysteine proteases that are expressed abundantly. In order to identify potential targets for the diagnosis and therapy of clonorchiasis, we conducted immunoblot analysis of the ESPs proteome using the sera obtained from clonorchiasis patients and identified legumains and cysteine proteases as antigens present in the ESPs. Although the cysteine proteases were previously reported to elicit antigenicity, the legumains are found herein for the first time as a serological antigen of C. sinensis. To confirm these findings, we expressed recombinant legumain in Escherichia coli and verified that recombinant legumain also functions as a potent antigen against the sera of clonorchiasis patients. Our results illustrate the validity of immuno-proteomic approaches in the identification of serodiagnostic antigens in the parasites.</P>

        Genetic Analyses of the Chimeric <i>CYP11B1</i> / <i>CYP11B2</i> Gene in a Korean Family with Glucocorticoid-Remediable Aldosteronism

        <P>Glucocorticoid-remediable aldosteronism (GRA) is an autosomal-dominant inheritable form of hyperaldosteronism with early onset hypertension. GRA is caused by unequal crossing-over of the steroid 11β-hydroxylase (<I>CYP11B1</I>) and aldosterone synthase (<I>CYP11B2</I>) genes. As a result of chimeric gene duplication, aldosterone is ectopically synthesized in the adrenal zona fasciculata under the control of adrenocorticotropin. Here, we describe three cases of GRA in a Korean family. The proband-a 21-yr-old female-was incidentally found to have high blood pressure (170/108 mmHg). Her 46-yr-old father had been treated twice for cerebral hemorrhage at the ages of 29 and 39 yr. Her 15-yr-old brother had a 2-yr history of hypertension; however, he was never treated. Their laboratory test results showed normokalemia, hyporeninemia, hyperaldosteronism, and a high plasma aldosterone concentration-to-plasma renin activity ratio. Normal saline loading failed to suppress aldosterone secretion. However, dexamethasone administration effectively suppressed their plasma aldosterone concentrations. Following genetic analyses with PCR and direct sequencing to document the chimeric gene and crossover site, respectively, we identified <I>CYP11B1</I>/<I>CYP11B2</I> and determined the breakpoint of unequal crossover to be located between intron 2 of <I>CYP11B1</I> and exon 3 of <I>CYP11B2</I>.</P>
