        The present study was carried out to investigate the viability of frozen-thawed aggregated mouse embryos and to produce the chimeras. Different phenotypic embryos were obtained by mating ICR female mice with ICR or CBA male mice. The aggregated morulae were produced following aggregation of embryos at 4-, 8- and 12- to 16-cell stage. The desirable stage for the aggregation of the mouse embryos was 8- to 16-cell stage. The post-thawed in vitro survival rates of aggregated embryos in glycerol, DMSO and ethylene glycerol were 51.5, 78.6 and 69.4%, respectively. Although the higher survival was obtained in DMSO, there were no significant differences in the survival rates among the three cryoprotectants. A total of 155 frozen-thawed aggregated embryos were transferred to 11 recipient mice, 3 out of 7 offspring were born to overt chimera. These experiments have proven that mouse chimeras can be obtained from forzen-thawed aggregated embryos.

        Objectives : To figure out the factors related to the failure of primary pulmonary tuberculosis(Tb) treatment and to enhance the efficiency of the treatment. Methods and Materials : The subjects were 137 Tb patients composed of the two groups; 44 failure cases and 93 success cases who admitted in National Masan Tuberculosis Hospital during 2001. The failure means no improvement with the standard short term regimen of INH and RFP. Patients' informations were gathered through medical records and face-to-face interview survey, and, the data were analyzed with SPSS(ver 10.0). Results : Mean age of the subjects were 47.6 years. Most cases were diagnosed at clinics or hospitals at first, due to the symptoms. No difference in smoking and alcohol habits between the two groups. The cases who got the informations about the whole process of treatment, and following regular regimens were more in the success group. The proportion of multi-drug resistance to the regular primary Tb regimen was higher in the failure group 8 cases out of 25 detectable failure cases. 0 case among 46 successful cases. And the four regular regimen(INH, RFP, EMB, PZA) was popular type of the resistance. In chest X-ray, multiply involved lesion(≥ two lobes), atelectasis and pleurisy were more frequent in the failure group. Conclusion : The major factors that enhance the efficiency of Tb treatment were the informations and compliance. Enough explanation about the whole process would be essential during Tb treatment, and, in case of MDR-Tb, susceptible regimen and surgical therapy should be considered.

        It has been rapidly given to the importance of the embodiment of the efficient computer systems in university education. Since the recent trend of the adoption of the openess policy in university education systems significantly affects on the realization of the embodiment of the efficient computer systems which is very essential to promote the international competition in many areas of university education. So it is greatly needed that our Soonchunhyang university must have the following principles for the embodiment of the efficient computer systems. (1) It is required that the appropriate computer education and the good quality of research environment must be given. (2) The establishment of the synthetic computer network system for efficient the university administrations is needed. (3) It is needed to establish the computer system for the consideration of the future performance. (4) It is also needed to improve the organization and the administration systems of the university computer center. (5) It is required on the establishment of the local area network(LAN) systems in the Soonchunhyang university, and the LAN must be connected to other outer national and international computer systems. And we propose the following requirement for the establishment of the convienent computer systems in our university and we have been greatly though about the significant change of the environment in university computer system. [1] The subject of the basic computer must become as the required subject for the all students in the curriulum. [2] The new computer systems both in hardware and software must be introduced. [3] It is needed to the change of the present name of the university computer center into the university information and computer center. And its organizations must be furthur developed [4] The training center for the university emploee is also needed. [5] The establishment of the database systems for the automtion of the university administation is needed. [6] It is strongly required on the establishment of the local area network system in the Soonchunhang university to connect the Bitnet and the Internet through the Hankook communication. [7] The computerization for the central library is needed. The indepent local area network system for the library is also needed to establish the own database system. Finally we hope that the LAN will be connected to the national wide library computer system.

        Betulin suppressed interleukin-1β-induced gene expression, secretion and proteolytic activity of matrix metalloproteinase in cultured articular chondrocytes and production of matrix metalloproteinase in the knee joint of rat

        We investigated whether betulin affects the gene expression, secretion and proteolytic activity of matrix metalloproteinase-3 (MMP-3) in primary cultured rabbit articular chondrocytes, as well as <i>in vivo</i> production of MMP-3 in the rat knee joint to evaluate the potential chondroprotective effect of betulin. Rabbit articular chondrocytes were cultured and reverse transcription-polymerase chain reaction (RT-PCR) was used to measure interleukin-1β (IL-1β)-induced gene expression of MMP-3, MMP-1, MMP-13, a disintegrin and metalloproteinase with thrombospondin motifs-4 (ADAMTS-4), ADAMTS-5 and type II collagen. Effect of betulin on IL-1β-induced secretion and proteolytic activity of MMP-3 was investigated using western blot analysis and casein zymography, respectively. Effect of betulin on MMP-3 protein production was also examined<i> in vivo</i>. The results were as follows: (1) betulin inhibited the gene expression of MMP-3, MMP-1, MMP-13, ADAMTS-4, and ADAMTS-5, but increased the gene expression of type II collagen; (2) betulin inhibited the secretion and proteolytic activity of MMP-3; (3) betulin suppressed the production of MMP-3 protein<i> in vivo</i>. These results suggest that betulin can regulate the gene expression, secretion, and proteolytic activity of MMP-3, by directly acting on articular chondrocytes.

      • 전대장 내시경 검사 505예에 대한 임상적 고찰

        대장질환은 지역과 인종에 따라 다소 차이가 있지만 과거에 비해 진단기술의 발달과 더불어 증가하고 있고, 특히 우리나라에서는 대장암의 발생이 현저히 늘고 있다. 이에 필연적인 검사 또한 기기의 발달 및 삽입기술의 향상, 대장암을 조기에 발견할 수 있는 장점으로 인해 많이 이용되고 있다. 저자들은 1990년 3월부터 1996년 2월까지 인제대학교 의과대학 부속 상계백병원에 입원 및 외래를 통하여 시행한 대장내시경 검사 505 예를 후향적 의부기록 분석을 통하여 다음과 같은 결과를 얻었다. 1.30대와 40대가 전체의 38.4%로 가장 많았고 다음으로 60대가 18.2%로 많았다. 남녀비는 1.01:1이었다. 2.환자의 주소는 복통 36.5%, 혈변 18.0%, 설사 17.2%, 복부팽만감 7.5%, 배변습관의 변화 6.7% 순이었다. 3.대장질환은 대장염 26.1%, 대장암 14.8%, 용종 8.3%, 치핵 4.3%. 정상 38.8%였다. 4.용종은 42예로 18예(39.0%)에서 내시경으로 절제하였고 조직검사 결과 관상 선종이 54%로 가장 많았고 2예에서 용종내에 암이 내재되어 있었다. 5.전대장 삽입의 성공률은 대상환자의 95.6%, 실패원인은 불충분한 대장 정결 및 통증이었다. In this report, we analysed the clinical findings of 505 cases that undertook total colonoscopic examination from March, 1990 to March, 1996. The results were followings ; 1) The most common age groups were 30's and 40's which were 38.4% of tonal patients. The sex ratio of male to female was close to one. 2) The patients' chief complaints were abdominal pain(36.5%), bloody stool(18%), diarrhea(17.2%), abdominal bloating(7.5%), bowel habit change(6.7%) in order. 3) Among the 505 cases of total colonoscopic findings, colitis was found in 132 cases(27.1%), colon cancer in 75 cases(14.8%), polyps in 42 cases(8.3%), hemorrhoid in 22 cases(4.3%) and normal in 196 cases(38.8%) . 4) Among the 42 cases of polyps, gastroscopic polypectomy was done in 18 cases(39.0% ). The most common histologic type was tubular adenoma which included 25 cases(54.0%). Two cases out of 25 cases in tubular were colorectal cancer with polyp remnant. 5) The success rate of total colonoscopy was 95.6% with failure in 4.4%. The causes for the failure of total colonoscopic examination turned out to be poor preparation in 12 cases(54.5%) and pain in 9 cases(40.9%).

        Device-to-device-based heterogeneous radio access network architecture for mobile cloud computing

        <P>The emerging heterogeneous mobile network architecture is designed for an increasing amount of traffic, quality requirements, and new mobile cloud computing demands. This article proposes a hierarchical cloud computing architecture to enhance performance by adding a mobile dynamic cloud formed by powerful mobile devices to a traditional general static cloud. A mobile dynamic cloud is based on heterogeneous wireless architecture where device-to-device communication is used for data transmission between user devices. The main advantage of the proposed architecture is an increase in overall capacity of a mobile network through improved channel utilization and traffic offloading from Long Term Evolution-Advanced to device-to-device communication links. Simulations show that the proposed architecture increases the capacity of a mobile network by up to 10 percent depending on the conditions and amount of offloaded data. The offloading probability is also evaluated by taking into consideration the number of devices in the cloudlet and the content matching values. We have gained insight into how content similarity affects offloading probability much more than the number of devices in a cloudlet.</P>

      • Ganglioside GM3 participates in the TGF-β1-induced epithelial-mesenchymal transition of human lens epithelial cells

        TGF-β(transforming growth factor-β)-induced EMT (epithelial-mesenchymal transition) induces the proliferation and migration of the HLE (human lens epithelial) cells. Ganglioside GM3, simple sialic-acid-containing glycosphingolipids on mammalian cell membranes, regulated various pathological phenomena such as insulin resistance and tumour progression. However, the relationship between ganglioside GM3 and TGF-β-induced EMT in the HLE B-3 cells is poorly understood. In the present study we demonstrated that ganglioside GM3 was involved in TGF-β1-induced EMT in HLE B-3 cells. Our results indicated that the expression of ganglioside GM3 and GM3 synthase mRNA were significantly increased in TGF-β1-induced HLE B-3 cells. Reporter gene analysis also demonstrated that transcriptional activation of the GM3 synthase gene was regulated by Sp1 (specificity protein 1) in HLEB-3 cells upon TGF-β1 stimulation. Interestingly, the inhibition of ganglioside GM3 expression by d-PDMP [d-threo-1phenyl-2decanoylamino-3-morpholino-1-propanol] and GM3 synthase shRNA (short hairpin RNA) resulted significantly in the suppression of cell migration and EMT-related signaling in HLEβ-3 cells stimulated by TGF-β. Furthermore, exogenous treatment of ganglioside GM3 rescue the expression of EMT molecules and cell migration suppressed by the depletion of ganglioside GM3 in TGF-β1-induced HLE B-3 cells. We also found that ganglioside GM3 in TGF -β1-induced HLE B-3 cells. We also found that ganglioside GM3 interacted with TGFβRs (TFG-β receptors) in TGF-β1-induced HLE B-3 cells. Taken together, these results suggest that ganglioside GM3 induced by TGF-β1 regulates EMT by potential interaction with TGFβRs.
