Translocation of macromolecules across the nuclear membrane under the normal conditions has been well studied, but the mechanism of nuclear translocation under the heat-shock stress conditions remain elusive. Hikeshi is a nuclear transport receptor required for cell survival after heat-shock stress. It is in charge of heat shock-induced nuclear import of 70-kilodalton heat shock proteins (HSP70s) through interactions with FG repeats in nuclear pore complexes. Hikeshi forms an “asymmetric” homodimer that is a characteristic feature. Asymmetry of Hikeshi arises from the distinct conformation of the C-terminal domain and the flexibility of the linker regions which areas are essential for nuclear import of HSP70. A unique extended-loop (E-loop) in the N-terminal domain controls the interactions of Hikeshi with FG-Nups by opening or closing the F97 binding site. The recently reported X-ray structure of Hikeshi explains how Hikeshi participates in the nuclear import regulation through FG-Nups recognition, and which part of Hikeshi affects its binding to HSP70. In this review, we will discuss the structural insight on the highly unique import receptor Hikeshi in heat-shock stress condition.

<P>Fas-associated factor (FAF)-1 is a multidomain protein that was first identified as a member of the Fas death-inducing signaling complex, but later found to be involved in various biological processes. Although the exact mechanisms are not clear, FAF1 seems to play an important role in cancer, asbestos-induced mesotheliomas, and Parkinson's disease. It interacts with polyubiquitinated proteins, Hsp70, and p97/VCP (valosin-containing protein), in addition to the proteins of the Fas-signaling pathway. We have determined the crystal structure of the ubiquitin-associated domain of human FAF1 (hFAF1-UBA) and examined its interaction with ubiquitin and ubiquitin-like proteins using nuclear magnetic resonance. hFAF1-UBA revealed a canonical three-helical bundle that selectively binds to mono- and di-ubiquitin (Lys48-linked), but not to SUMO-1 (small ubiquitin-related modifier 1) or NEDD8 (neural precursor cell expressed, developmentally down-regulated 8). The interaction between hFAF1-UBA and di-ubiquitin involves hydrophobic interaction accompanied by a transition in the di-ubiquitin conformation. These results provide structural insight into the mechanism of polyubiquitin recognition by hFAF1-UBA.</P>
Low pH-driven folding of WW45-SARAH domain leads to stabilization of the WW45-Mst2 complex

<P>The scaffolding protein Salvador (Sav) plays a key role in the Hippo (Hpo) signalling pathway, which controls tissue growth by inhibiting cell proliferation and promoting apoptosis. Dysregulation of the Hippo pathway contributes to cancer development. Since the identification of the first <I>Sav</I> gene in 2002, very little is known regarding the molecular basis of Sav-SARAH mediating interactions due to its insolubility. In this study, refolding of the first Sav (known as WW45)-SARAH provided insight into the biochemical and biophysical properties, indicating that WW45-SARAH exhibits properties of a disordered protein, when the domain was refolded at a neutral pH. Interestingly, WW45-SARAH shows folded and rigid conformations relative to the decrease in pH. Further, diffracting crystals were obtained from protein refolded under acidic pH, suggesting that the refolded WW45 protein at low pH has a homogeneous and stable conformation. A comparative analysis of molecular properties found that the acidic-stable fold of WW45-SARAH enhances a heterotypic interaction with Mst2-SARAH. In addition, using an Mst2 mutation that disrupts homotypic dimerization, we showed that the monomeric Mst2-SARAH domain could form a stable complex of 1:1 stoichiometric ratio with WW45 refolded under acidic pH.</P>

There are computation time constraints caused by the number and size of particles in the powder packing simulation using DEM. In this paper, newly suggested packing model transforms a general packing sequence –particle generation, stack, and compressioninto particle generation and packing by growing particles. To verify the new packing model, it was compared using three contact models widely used in DEM, in terms of radial distribution function, porosity, and coordination number. As a result, contact between particles showed a similar trend, and the pore distribution was also similar. Using the new packing model can reduce simulation time by 400 % compared to the normal packing model without any other coarse graining methods. This model has only been applied to particle packing simulations in this paper, but it can be expanded to other simulations with complex domain based on DEM.

The West African Power Pool is currently developing the regional electricity market for its member states. As the basic design for the cross-border power trading system for the West African Power Pool, the step-by-step evolution, the Baseline System and the Fullscale System, is estimated. Along with the interconnection project between member countries, the various functions from the data acquisition and processing by the Baseline System to the interchange trading by the Fullscale System will be implemented gradually.

Background : Panax ginseng C.A. Meyer is a representative medicinal plants and it has been used in traditional medicine because the plant have many effective component such as saponins. To obtain a wild-cultivated ginseng (WCG) which is similar to wild ginseng (WG), this method usually performed in a mountain using seeds or seedlings of cultivated ginseng (CG) and WG. WCG is very expensive because it is difficult to cultivate. However, systematic cultivation method have not been developed compared to their high added value. Furthermore, very high price of WCG caused the problem that Panax notoginseng or Panax quinquefolium are sold as WCG in Korean market. This is concerned as a serious problem to consumers. In this study, we analyzed the agronomic and growth characteristics of the WCG cultivated in Korea. Methods and Results : We examined the WCG that was collected and sold by regional groups at the Korean market. The root age, growth conditions, and quality level of the cultivated WCG were confirmed. WCG samples were collected from five areas in Korea (Bucheon, Cheongju, Hoengseong, Judeok and Ulsan). The main root diameter, root shape index, rhizome length, and root weight showed high level of variation and they did not form annual rings. Conclusion : Agronomic and growth characteristics of WCG showed high variations according to cultivating regions.

Background : Wild-cultivated ginseng (WCG) prices are very different according to root ages. Generally, two methods are used to discriminate the root ages of Panax ginseng C.A. Meyer. The first method is the yearly determination method by the ring dyeing method, and the second method is the confirmation the number of stem vestiges in the rhizome. In this study, we analyzed the agronomic and growth characteristics of the WCG cultivated in Korea. In this study, to determine the appropriate root ages discrimination method for the determination of the root ages of WCG, the root ages of WCG and cultivated ginseng was examined. Methods and Results : We examined the cultivated ginseng (CG) and WCG that was collected and sold by regional groups at the Korean market. WCG does not form annual rings, which are clear and regular in wild ginseng. Therefore, it is impossible to identify the age of WCG by using the annual growth rings staining method. However, the age can be estimated by determining the number of stem vestiges in the rhizome. Conclusion : From the results of the Study on identification of root age for quality evealuation in WCG in Korea. Appropriate root ages discrimination method of WCG was confirmation the number of stem vestiges in the rhizome.
