        Inhibition of MicroRNA-92a Improved Erectile Dysfunction in Streptozotocin-Induced Diabetic Rats via Suppressing Oxidative Stress and Endothelial Dysfunction

        Purpose: To determine whether microRNA could be a therapy target of erectile dysfunction (ED) and the underlying mecha-nisms. Materials and Methods: Eight-week-old fasting male SD rats were intraperitoneally injected with streptozotocin to construct diabetic rat models. Diabetic ED rats were treated with miRNA-92a inhibitor. The cavernous nerves were electrically stimu-lated to measure the intracavernous pressure and mean arterial pressure of rats in each group. After the detection, the penile cavernous tissues are properly stored for subsequent experiments. Rat aortic endothelial cells were used in in vitro studies. Results: The expression of miR-92a was significantly increased in the corpus cavernosum of Streptozocin (STZ)-induced di- abetic rats and injection of miR-92a antagomir into the corpus cavernosum of diabetic rats significantly increased eNOS/NO/ cGMP signaling pathway activities, cavernous endothelial cell proliferation, endothelial cell-cell junction protein expression and decreased the levels of oxidative stress. These changes restored erectile function in STZ-induced diabetic rats. Moreover, in vitro study demonstrated that the miR-92a expression increased significantly in endothelial cells treated with high glucose, inhibiting AMPK/eNOS and AMPK/Nrf2/HO-1 signaling pathways in rat aortic endothelial cells via targeting Prkaa2, causing endothelial dysfunction and overactive oxidative stress, miR-92a inhibitor can improve the above parameters. Conclusions: miRNA-92a inhibitor could exert an inhibition role on oxidative stress and endothelial dysfunction to improve diabetic ED effectively.

        Synthesis of (Imido)vanadium(V) Complexes Containing 8-(2,6-Dimethylanilide)-5,6,7-trihydroquinoline Ligands: Highly Active Catalyst Precursors for Ethylene Dimerization

        <P>A series of (imido)vanadium(V) dichloride complexes containing 8-(2,6-dimethylanilide)-5,6,7-trihydroquinoline ligands of the type V(NR)Cl<SUB>2</SUB>[8-(2,6-Me<SUB>2</SUB>C<SUB>6</SUB>H<SUB>3</SUB>)N(C<SUB>9</SUB>H<SUB>10</SUB>N)] (R = Ad (<B>3</B>), 2-MeC<SUB>6</SUB>H<SUB>4</SUB> (<B>4</B>), 2,6-Me<SUB>2</SUB>C<SUB>6</SUB>H<SUB>3</SUB> (Ar, <B>5</B>)) have been prepared and identified, and their structures have been determined by X-ray crystallographic analysis. The ethylene dimerization catalyst generated from complex <B>3</B> upon treatment with an excess amount of MAO exhibited remarkable catalytic activities (e.g. TOF = 9600000 h<SUP>–1</SUP> (2670 s<SUP>–1</SUP>), Al/V = 4000 (molar ratio)), affording 1-butene as the major product (95.0–99.4%). The activities of <B>3</B> and <B>4</B> were higher than those exhibited by the corresponding 2-(anilide)methylpyridine analogues; <B>3</B> showed higher 1-butene selectivity than the others and the activity did not decrease remarkably at 50 °C.         Modelling of effective irradiation swelling for inert matrix fuels

        The results of effective irradiation swelling in a wide range of burnup levels are numerically obtained for an inert matrix fuel, which are verified with DART model. The fission gas swelling of fuel particles is calculated with a mechanistic model, which depends on the external hydrostatic pressure. Additionally, irradiation and thermal creep effects are included in the inert matrix. The effects of matrix creep strains, external hydrostatic pressure and temperature on the effective irradiation swelling are investigated. The research results indicate that (1) the above effects are coupled with each other; (2) the matrix creep effects at high temperatures should be involved; and (3) ranged from 0 to 300 MPa, a remarkable dependence of external hydrostatic pressure can be found. Furthermore, an explicit multi-variable mathematic model is established for the effective irradiation swelling, as a function of particle volume fraction, temperature, external hydrostatic pressure and fuel particle fission density, which can well reproduce the finite element results. The mathematic model for the current volume fraction of fuel particles can help establish other effective performance models.

        Blockage effects on aerodynamics and flutter performance of a streamlined box girder

        Wind tunnel test is one of the most important means to study the flutter performance of bridges, but there are blockage effects in flutter test due to the size limitation of the wind tunnel. On the other hand, the size of computational domain can be defined by users in the numerical simulation. This paper presents a study on blockage effects of a simplified box girder by computation fluid dynamics (CFD) simulation, the blockage effects on the aerodynamic characteristics and flutter performance of a long-span suspension bridge are studied. The results show that the aerodynamic coefficients and the absolute value of mean pressure coefficient increase with the increase of the blockage ratio. And the aerodynamic coefficients can be corrected by the mean wind speed in the plane of leading edge of model. At each angle of attack, the critical flutter wind speed decreases as the blockage ratio increases, but the difference is that bending-torsion coupled flutter and torsional flutter occur at lower and larger angles of attack respectively. Finally, the correction formula of critical wind speed at 0o angle of attack is given, which can provide reference for wind resistance design of streamlined box girders in practical engineering.

        miR-148b-3p, miR-190b, and miR-429 Regulate Cell Progression and Act as Potential Biomarkers for Breast Cancer

        Purpose: Breast cancer is the most frequently diagnosed malignancy in women worldwide. MicroRNAs (miRNAs) are thought to serve as potential biomarkers in various cancers, including breast cancer. Methods: We evaluated the miRNA expression profiles in 1,083 breast cancer samples and 104 normal breast tissues from The Cancer Genome Atlas database. We used the edgeR package of R software to analyze the differentially expressed miRNAs in normal and cancer tissues, and screened survival-related miRNAs by Kaplan-Meier analysis. A receiver operating characteristic curve was generated to evaluate the accuracy of these miRNAs as molecular markers for breast cancer diagnosis. Furthermore, the functional role of these miRNAs was verified using cell experiments. Targets of candidate miRNAs were predicted using 9 online databases, and Gene Ontology (GO) functional annotation and pathway analyses were conducted using Database for Annotation, Visualization and Integrated Discovery online tool. Results: A total of 68 miRNAs showed significantly different expression patterns between the groups (p < 0.001), and 13 of these miRNAs were significantly associated with poor survival (p < 0.05). Three miRNAs with high specificity and sensitivity, namely, miR-148b-3p, miR-190b, and miR-429, were selected. In vitro experiments showed that the overexpression of these 3 miRNAs significantly promoted the proliferation and migration of MDA-MB-468 and T47D cells and reduced the apoptosis of T47D cells. GO and pathway enrichment analyses revealed that the targets of these dysregulated miRNAs were involved in many critical cancer-related biological processes and pathways. Conclusion: The miR-148b-3p, miR-190b, and miR-429 may serve as potential diagnostic and prognostic markers for breast cancer. This study demonstrated the roles of these 3 miRNAs in the initiation and progression of breast cancer.

        The China Spallation Neutron Source (CSNS) complex consists of an H- linear accelerator, a rapid cycling synchrotron accelerating the beam to 1.6 GeV, a solid tungsten target station and instruments for spallation neutron applications. The facility operates at a 25-Hz repetition rate with an initial design beam power of 120 kW and is upgradeable to 500 kW. The primary challenge is to build a robust and reliable user-friendly facility with upgrade potential at a fraction of the \world standard" cost. Success of the project relies on the results of prototyping research & development (R&D) of key technical systems and components. This paper discusses the prototyping experiences of the past two and a half years.
