A new NMR-based metabolomics approach for the diagnosis of biliary tract cancer

<P><B>Background & Aims</B></P><P>Biliary tract cancer is highly lethal at presentation, with increasing mortality worldwide. Current diagnostic measures employing multiple criteria such as imaging, cytology, and serum tumor markers are not satisfactory, and a new diagnostic tool is needed. Because bile is a cognate metabolite-rich bio-fluid in the biliary ductal system, we tested a new metabolomic approach to develop an effective diagnostic tool.</P><P><B>Methods</B></P><P>Biles were collected prospectively from patients with cancer (<I>n</I>=17) or benign biliary tract diseases (<I>n</I>=21) with percutaneous or endoscopic methods. Nuclear magnetic resonance spectra (NMR) of these biles were analyzed using orthogonal partial least square discriminant analysis (OPLS-DA).</P><P><B>Results</B></P><P>The metabolomic 2-D score plot showed good separation between cancer and benign groups. The contributing NMR signals were analyzed using a statistical TOCSY approach. The diagnostic performance assessed by leave-one-out analysis exhibited 88% sensitivity and 81% specificity, better than the conventional markers (CEA, CA19-9, and bile cytology).</P><P><B>Conclusion</B></P><P>The NMR-based metabolomics approach provides good performance in discriminating cancer and benign biliary duct diseases. The excellent predictability of the method suggests that it can, at least, augment the currently available diagnostic approaches.</P>

3-Hydroxy-3-methyl glutaryl coenzyme A reductase (HMGR) is an important rate-limiting enzyme in the mevalonate pathway, which is critical for the synthesis of juvenile hormone and insect development. In this study, a full-length cDNA encoding HMGR (BdHMGR) was cloned from the oriental fruit fly, Bactrocera dorsalis (Hendel). The open reading frame of BdHMGR encoded 928 amino acids and shared a high degree of identity with other known HMGRs. The expression levels of BdHMGR were high during the early and lately pupal stages, and were low at the larval and mid pupal stages. In the adult stages, the highest level of BdHMGR was detected on day-10 of the adult lifecycle, and the expression levels in females were greater than those in males. The highest tissuespecific expression was in the ovary, followed by fat body and other tissues. Injection of double-stranded RNA (dsRNA) of BdHMGR into adult females significantly reduced BdHMGR transcript levels, and inhibited ovarian development and affected the expression of two yolk protein genes. After dissection, we found that one or two of the spheroids of ovaries from the BdHMGR dsRNA-injected flies were amorphous and reduced in size as compared with the controls. The results suggest that BdHMGR is required for ovarian development in B. dorsalis and may be a potential target for pest control.
Performance Evaluation of Ethernet Frame Burst Mode in EPON Downstream Link

We apply IEEE 802.3 frame burst mode (FBM) to the Ethernet passive optical network (EPON) downstream link and compare its performance with non-frame burst mode for various traffic patterns. Although in light traffic loads (ρ<0.5) the efficiency of the FBM mechanism is not significant, it does feature high throughput, small jitter, low queue occupancy, and short queuing delay in optical line terminals under various traffic loads with various numbers of optical network units (ONUs). The FBM performance always approaches that of full-duplex mode, especially under heavy traffic loads (ρ>0.5). Moreover, an increase in number of ONUs will decrease the burst performance. Our work shows that FBM scheme is very useful for EPON transmission and has low design complexity.
Antifungal Activities of Anthocyanins from Purple Sweet Potato in the Presence of Food Preservatives

Effects of anthocyanins from purple sweet potato (APSP) against the fungal growth of Geotrichum candidum and Candida albicans were assessed. Response surface methodology was applied for optimization of proportions of sodium benzoate, potassium sorbate, and APSP. Optimum concentrations against G. candidum were 0.300 mg/mL of sodium benzoate, 0.290mg/mL of potassium sorbate, and 13.9mg/mL of APSP. Optimum concentrations against C. albicans were 0.380 mg/mL of sodium benzoate, 0.240 mg/mL of potassium sorbate, and 3.56 mg/mL of APSP. APSP exhibited enhanced antifungal properties in the presence of food preservatives.

Objective: To investigate whether the expression level of valosin-containing protein (VCP) is correlated with the prognosis of primary orbital mucosa-associated lymphoid tissue (MALT) lymphoma. Methods: VCP expression in 58 samples from primary orbital MALT lymphoma patients was determined by immunohistochemisty using monoclonal antibodies. Correlations between VCP expression level and prognosis were clarified by statistical analysis. Results: It was found that the percentage of VCP positive cells in samples of primary orbital MALT lymphoma ranged from 32% to 95%. The samples were divided into two groups (level 1 and level 2) according to the median value (45%) of the percentage of VCP positive cells. It was found that the expression level of VCP was significantly correlated with recurrence (P=0.003) and tumor size (P=0.008). At the same time, the 5-year disease-free and overall survival rate of patients of level 1 was significantly better than that of level 2 (P=0.001; P=0.032). There was no observed correlation between the expression level of VCP and other clinical features. Conclusion: VCP could be a useful marker for predicting the prognosis of primary orbital MALT lymphoma.

<P>A total of 60 weaned pigs with initial body weight (BW) of 8.51 ± 0.42 kg (28 d of age) were randomly divided into 2 groups of 30 each consisted of 6 pens with 2 gilts and 3 borrows and supplemented (treatment) 5.3×108 cfu/kg Lactobacillus plantarum GB805 in addition to basal diet offered during d 0 to 14 and d 15 to 42 (control). During the total trial period or different phases of trial, pigs fed L. plantarum diet had a higher (P less than 0.05) ADG, ADFI and feed efficiency. At the end of experiment, dietary L. plantarum supplementation improved (P less than 0.05) the apparent total tract digestibility of dry matter, nitrogen and the fecal Lactobacillus populations (P less than 0.05) while the fecal E.coli populations were decreased (P less than 0.05). In conclusion, dietary supplementation of L. plantarum GB805 had positive influence on growth rate, nutrient digestibility and gut microbial balance and its supplementation recommended to weaning pigs.</P>

<P>FeCo alloy nanoparticles were nucleated onto graphitic carbon layers through the pyrolysis of polydopamine (PDA) sub-micrometer spheres to form a highly active electrocatalytic system that exhibits excellent oxygen conversion catalytic activity in both alkaline and acidic media. Owing to the strong metal chelation capability during the chemical modification and high sp<SUP>2</SUP>-dominant carbon yield of PDA, an abundance of non-precious-metal ions were easily trapped and absorbed into the PDA segments at room temperature by catechol and amine functional groups, followed by the <I>in situ</I> nucleation of FeCo alloy nanoparticles on graphitic carbon layers during the pyrolysis. The contents of graphitic nitrogen and pyridinic nitrogen were significantly increased by the presence of the non-precious-metal ions during carbonization as well, which is a result of the chelation effect of non-precious-metal atoms. Meanwhile, the FeCo nanoparticles (diameter < 5 nm) were protected by the multi-layer-graphene-like carbon layer from the harsh acid and uniformly anchored on graphitic carbon sub-microspheres, which can greatly improve the catalytic durability, particularly in acidic media. From the perspective of the whole catalytic system being used as an air electrode in rechargeable Zn-air batteries, the porous nitrogen-doped graphitic carbon framework was functionalised as a continuous conductive framework due to its catalytic activity towards high oxygen conversion and good electrical conductivity.</P>
