        Dimethyl Sulfoxide Suppresses Mouse 4T1 Breast Cancer Growth by Modulating Tumor-Associated Macrophage Differentiation

        Purpose: The universal organic solvent dimethyl sulfoxide (DMSO)can be used as a differentiation inducer of many cancer cells andhas been widely used as a solvent in laboratories. However, itseffects on breast cancer cells are not well understood. The aimof this study is to investigate the effect and associated mechanismsof DMSO on mouse breast cancer. Methods: We appliedDMSO to observe the effect on tumors in a mouse breast cancermodel. Tumor-associated macrophages (TAMs) were tested byflow cytometry. Ex vivo tumor microenvironment was imitated by4T1 cultured cell conditioned medium. Enzyme-linked immunosorbentassays were performed to detect interleukin (IL)-10 andIL-12 expression in medium. To investigate the cytotoxicity ofDMSO on TAMs, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide (MTT) assays were performed. Results: We foundthat DMSO produced tumor retardation when injected into mouseperitoneal cavities in a certain concentration range (0.5–1.0 mg/g). Furthermore, as detected by flow cytometry, TAM subtypeswere found to be transformed. We further imitated a tumor microenvironmentin vitro by using 4T1 cultured cell conditionedmedium. Similarly, by using low concentration DMSO (1.0%–2.0% v/v), TAMs were induced to polarize to the classically activatedmacrophage (M1-type) and inhibited from polarizing intothe alternatively activated macrophage (M2-type) in the conditionedmedium. IL-10 expression in tumors was reduced, whileIL-12 was increased compared with the control. Furthermore, wereported that 2.0% (v/v) DMSO could lead to cytotoxicity in peritonealmacrophages after 48 hours in MTT assays. Conclusion:Our findings suggest that DMSO could exert antitumor effects in4T1 cancer-bearing mice by reversing TAM orientation and polarizationfrom M2- to M1-type TAMs. These data may providenovel insight into studying breast cancer immunotherapy.

        Preparation, Characterization and Properties of High-salt-tolerance Sodium Alginate/Krill Protein Composite Fibers

        Sodium alginate (SA) and krill protein (AKP) were blended to obtain composite solution, and functional SA/AKP composite fibers were prepared via wet spinning. To further improve the salt tolerance, SA/AKP composite fibers were modified with copper sulfate aqueous solution as secondary coagulation bath because of the strong adsorption to copper ions. The CSA/AKP composite fibers with high salt tolerance have been successfully prepared. The intermolecular interaction of SA/AKP composite system and the two-order structure of protein in the composite system were characterized by Fourier transform infrared spectroscopy (FT-IR). Besides, the crystallinity, morphology, mechanical properties, salt tolerance and water resistance and thermal stability of SA/AKP composites were investigated respectively. The results showed that the adsorption rate and the adsorption capacity of the composite solution to copper ion were significantly higher than those to calcium ion. Under the effect of secondary solidification by copper sulfate, the β-sheet chain of the composite fibers increased from 41.48 % to 49.21 %, the intramolecular hydrogen bond increased from 38.18 % to 44.26 %, the intermolecular hydrogen bond decreased from 59.84 % to 54.70 % and free hydroxyl slightly decreased. The water resistance of the modified composite fibers was improved by about 22 %; when the swelling time was 25 min, the salt resistance increased by about 150 %; the number of grooves on the surface of the composite fibers obviously increased, and the grooves on the surface of CSA/AKP composite fibers and the fiber section structure were much denser; Meanwhile, copper sulfate had some influence on the crystallization, thermal stability and mechanical properties of the composite fibers.

        Time-varying H∞ Control for Discrete-time Switched Systems with Admissible Edge-dependent Average Dwell Time

        The problem of H∞ control for discrete-time switched systems is investigated via admissible edgedependent average dwell time (AED-ADT) method in this paper. By virtue of a convex combination of positive definite matrices, a novel multiple piecewise convex Lyapunov function (MPCLF) is designed, which can relax the restricted conditions of Lyapunov functions at switching points and interval interior points. Based on the MPCLF approach, the time-varying H∞ state feedback controllers, guaranteeing that the corresponding closed-loop system is globally uniformly exponentially stable (GUES) with a prescribed H∞ performance, are established for the considered switched system. Finally, three numerical examples are provided to illustrate the effectiveness of the proposed approaches.

      • hARIP2 is a Putative Growth-promoting Factor Involved in Human Colon Tumorigenesis

        Activin is a multifunctional growth and differentiation factor of the growth factor-beta (TGF-${\beta}$) superfamily, which inhibits the proliferation of colon cancer cells. It induces phosphorylation of intracellular signaling molecules (Smads) by interacting with its type I and type II receptors. Previous studies showed that human activin receptor-interacting protein 2 (hARIP2) can reduce activin signaling by interacting with activin type II receptors; however, the activity of hARIP2 in colon cancer has yet to be detailed. In vitro, overexpression of hARIP2 reduced activin-induced transcriptional activity and enhanced cell proliferation and colony formation in human colon cancer HCT8 cells and SW620 cells. Also, hARIP2 promoted colon cancer cell apoptosis, suggesting that a vital role in the initial stage of colon carcinogenesis. In vivo, immunohistochemistry revealed that hARIP2 was expressed more frequently and much more intensely in malignant colon tissues than in controls. These results indicate that hARIP2 is involved in human colon tumorigenesis and could be a predictive maker for colon carcinoma aggressiveness.

      • Ten Year Literature on Psychological and Behavioral Interventions Against Cancer: a Terms Analysis

        We here performed a systematic review of PBIC literature using terms analysis in a hope of both identifying potential trends and patterns and exploring methods leveraging traditional literature reviews in this specific area. Articles meeting inclusion criteria were retrieved from PUBMED and translated into dichotomized article records representing presence or non-presence of MeSH terms and a metric consisting of numbers of times of co-occurrence between all pairs of terms identified using a self-designed program. The occurrence of and relations among the terms were calculated and visualized using Excel2007 and UCINET respectively. A total of 1,742 terms were identified from 997 articles retrieved. Put in a descending order, the lines representing the times of term occurrence formed a typical hyperbolic curve; when plotted along the x-axis of whole MESH terms, the lines clustered within four specific regions. Comparison of term occurrence between 2002 and 2011 revealed priority changes in population and subjects (from general groups to priority groups), intervention approaches (from medicine to exercise and psychotherapy), methodology and techniques (from cohort studies to randomized controlled trials) and outcomes (from health and mental health to quality of life, depression etc.). Networks of the terms featured a number of closely linked groups of topics including method and questionnaires, therapy and outcomes, survival management, psychological assessment and intervention, behavioral intervention (individual and community oriented). Terms analysis revealed interesting trends and patterns about PBIC publications and both the analysis methods and findings have implications for future research and literature reviews.

        LncRNA TMPO-AS1 promotes esophageal squamous cell carcinoma progression by forming biomolecular condensates with FUS and p300 to regulate TMPO transcription

        Esophageal squamous cell carcinoma (ESCC) is one of the most life- and health-threatening malignant diseases worldwide, especially in China. Long noncoding RNAs (lncRNAs) have emerged as important regulators of tumorigenesis and tumor progression. However, the roles and mechanisms of lncRNAs in ESCC require further exploration. Here, in combination with a small interfering RNA (siRNA) library targeting specific lncRNAs, we performed MTS and Transwell assays to screen functional lncRNAs that were overexpressed in ESCC. TMPO-AS1 expression was significantly upregulated in ESCC tumor samples, with higher TMPOAS1 expression positively correlated with shorter overall survival times. In vitro and in vivo functional experiments revealed that TMPO-AS1 promotes the proliferation and metastasis of ESCC cells. Mechanistically, TMPO-AS1 bound to fused in sarcoma (FUS) and recruited p300 to the TMPO promoter, forming biomolecular condensates in situ to activate TMPO transcription in cis by increasing the acetylation of histone H3 lysine 27 (H3K27ac). Targeting TMPO-AS1 led to impaired ESCC tumor growth in a patient-derived xenograft (PDX) model. We found that TMPO-AS1 is required for cell proliferation and metastasis in ESCC by promoting the expression of TMPO, and both TMPO-AS1 and TMPO might be potential biomarkers and therapeutic targets in ESCC.

        Blood-compatible Polyaniline Coated Electrospun Polyurethane Fiber Scaffolds for Enhanced Adhesion and Proliferation of Human Umbilical Vein Endothelial Cells

        The endothelialization and anti-thrombotic abilities of tissue engineered vascular scaffolds are considered to be effective properties for improving the performance small-caliber vascular scaffolds. For this purpose, we designed and developed electrically conductive fibrous scaffolds based on polyaniline coated polyurethane (PANI-PU) electrospun fibersfor vascular tissue engineering applications. The porosity of PANI-PU fibers was 75.27±2.04 %. The obtained PANI-PU fibers were characterized by SEM observations, XPS analysis, water contact angle (WCA) measurement and mechanical property. The PANI functionalization aimed to improve the performance of anticoagulation and endothelialization. The WCA of PAIN-PU decreased to 35 o from 135 o of PU fibers. Blood compatibility and cytocompatibility were compared before and after PANI coating. The adhered platelet cells on PANI-PU was 6.87×105/cm2 and plasma recalcification time was 123 s. Platelet adhesion and plasma recalcification time test showed that the PANI-PU scaffolds had a certain anticoagulant effect. The hemolysis rate of PANI-PU fibers was 0.14 %, which showed that the PANI-PU scaffolds could be used as blood contact materials. The observation of endothelial cell proliferation and morphology in human umbilical vein endothelial cells showed that PANI-PU fibers were more beneficial to cell adhesion, proliferation and extension than that of PU fibers. The results demonstrates the PANI coated electrospun PU fibers have great potential in application as small-diameter vascular grafts and this work shows new insights into conductive scaffolds for vascular tissue engineering.

        Phase separation phenomena and thermodynamics of octylphenol polyoxyethylene ether in micellar solutions: Effect of high concentration of inorganic salts and linear correlation

        The phase separationphenomena or cloud points (CP) behaviorof nonionic octylphenol polyoxyethylene etherwith oxyethylene glycol ether unit of 15 (OP-15) were investigated in aqueous micellar solutions containinghigh concentrations of inorganic salts including NaCl, KCl, MgCl2, CaCl2, BaCl2, AlCl3, FeCl3, Na2SO4and Na2SiO3. The change in CP induced by salts and its mechanism are discussed theoretically. In aqueous solutionscontaining high concentration of salt, for thefirst time the linear relationship between CP and saltconcentrationiswellestablishedtocharacterizethedepressionofCP.Basedonpseudophaseseparationmodel,thermodynamic parameters are estimated and adopted to further describe the phase separation process andthe interactions between OP-15 molecules or the OP-15/water interactions. Thermodynamic parametersincluding standard Gibbs energy change (DG0CP), enthalpy change (DH0CP) and entropy change (DS0CP)indicate that the additions of different salts result in the change in entropy-driven nonspontaneousprocess of phase separation. These data, which are from the compensation temperatures and the changeof molar heat capacity, also prove that different ions induce the change in the interactions betweensurfactant molecules or the surfactant–water interaction and thus affect the depression of CP. Thesefindings help with understanding the effect of inorganic salts, especially the high concentration of salt, onCP of nonionic surfactant and then broadening the application of nonionic surfactant based on CP such asthe extraction of biomaterials in biological samples derived from animals or plants, etc.

        <P><B>Abstract</B></P> <P>Uniform, monodisperse yttrium aluminum garnet (YAG) rhombic dodecahedron micro-crystals were synthesized using coprecipitation and hydrothermal methods. The Na<SUB>2</SUB>SO<SUB>4</SUB> as a surfactant was helpful in improving the dispersity of the crystallites and forming well-faceted, micro-sized dodecahedral YAG crystallites. The effect of the reaction time and the solvent has also been studied. The crystal growth mechanism called dissolution/crystallization was determined by analyzing the experimental and simulated results. And the specific well-developed crystal facets in the {110} family have been demonstrated by the theoretical calculation. A higher integrated emission intensity can be achieved for the YAG:Ce crystallites by improving the phase purity, morphology with better developed dodecahedrons, and dispersion. Furthermore, a rather high luminous efficacy (LE = 104.14 lm/W) for the white LED fabricated using the YAG:Ce<SUP>3+</SUP> phosphor prepared in this work was realized.         Interaction and micellar behavior of ternary mixture of amphoteric amino sulfonate surfactant with traditional anionic and nonionic surfactants: Effect of hydrophilicity

        The micellization and the molecular interaction behaviors for two ternary mixtures constituted by anamphoteric sodium 3-(n-dodecyl ethylenediamino)-2-hydroxypropyl sulfonate (C12AS), an anionicsodium dodecylbenzene sulfonate (SDBS) and a nonionic octylphenol polyoxyethylated ether with thenumber (n) of oxyethylene glycol ethers OP-n (n = 10 or 7) in aqueous solution were investigated usingthe tensiometry and the effect of hydrophilicity on them was also discussed. In the framework of pseudophaseseparation model and based on the regular solution theory, the related micellization parametersincluding the mixed critical micelle concentration (cmc) values in the ideal and real cases, the activitycoefficients and the compositions in mixed micelle, etc. and thermodynamic parameters were estimatedby the Clint’s model and the Rubingh’ model. The mixed cmc value is dependent on the composition inaqueous solution and influenced by the hydrophilicity of nonionic surfactant. With increasing the nonionicin ternary mixture, the mixed cmc value is initially decreased and then slightly rise. An increasingin the hydrophilicity of nonionic will make the minimum value of mixed cmc be increased from1.299 mM to 1.705 mM. The resulting phenomena can be explained reasonably by the electrostatic effect,the steric hindrance, the hydrogen bonding, etc. Thermodynamic data indicate that the contribution ofentropy or enthalpy plays a vital role on the spontaneous process of micellization and the share ofentropy or enthalpy in free energy change is dependent largely on the amount of nonionic surfactantand the hydrophilicity. In ternary mixtures of C12AS/SDBS/OP-10, an increase in an amount of OP-10 willinduce the change from the enthalpy-driven micellization process to the entropically favorable process. Once an abundant amount of OP-7 is added, while, the enthalpy will make a main contribution on themicellization process, which can be described by the drop in the share of entropy at the composition(0.6970/0.0000/0.3030) of ternary mixture from 0.6478 to 0.4901. These findings will help with understandingthe molecular interaction behavior for the ternary surfactant mixture and the effect of the additionof nonionic surfactant and its hydrophilicity.
