        Influence of Intranasal Drugs on Human Nasal Mucociliary Clearance and Ciliary Beat Frequency

        The nasal mucociliary clearance system, which comprises epithelial cilia and mucus from goblet cells, is an important intrinsic defense mechanism of the upper respiratory tract. Intranasal drugs and additives can have a detrimental effect on ciliary activity and mucociliary clearance, and thus impact the integrity of nasal defense mechanisms. This article discusses the current literature on the effects of different classes of intranasal drugs including intranasal corticosteroids, antihistamines, decongestants, antimicrobials and antivirals, as well as various drug excipients and nasal irrigation solutions on human nasal mucociliary clearance and ciliary beat frequency. Available data indicate that some intranasal formulations tend to hamper nasal ciliary function and mucociliary clearance. Therefore, it is of great importance to assess the effects of intranasal drugs and additives on mucociliary function before they are recommended as therapy for different nasal conditions.

        Monoterpene esters and aporphine alkaloids from Illigera aromatica with inhibitory effects against cholinesterase and NO production in LPS-stimulated RAW264.7 macrophages

        Three new monoterpene phenylpropionic acidesters, illigerates A–C (1–3), and one new aporphinealkaloid, illigeranine (4), as well as four known ones,actinodaphnine (5), nordicentrine (6), 8-hydroxy carvacrol(7), and 3-hydroxy-a,4-dimethyl styrene (8), were isolatedfrom the tubers of Illigera aromatica. The structures of 1–4were identified by HRESIMS, 1D and 2D NMR, andelectronic circular dichroism spectra. Compound 1 potentlyinhibited NO production in LPS-stimulated RAW264.7cells with an IC50 value of 18.71 ± 0.85 lM; compound 1,3, and 4 showed moderate butyrylcholinesterase inhibitoryactivities with the IC50 values of 46.86 ± 0.65,53.51 ± 0.71, and 31.62 ± 1.15 lM, respectively. Compound4 showed weak AChE inhibitory activity with anIC50 value of 81.69 ± 2.07 lM, and compounds 5 and 6possessed moderate AChE inhibitory activities with theIC50 values of 47.74 ± 1.66 and 40.28 ± 2.73 lM,respectively. This paper provides a chemical structure andbioactive foundation for using I. aromatica as an herbalmedicine.

        Optimization of Nutrients for Dinactin Production by a Marine Streptomyces sp. from the High Latitude Arctic

        Streptomyces sp. R-527F, which produces the macrotetrolide antibiotic dinactin, was isolated from the sediments of the Arctic Ocean. In this work, optimization of the nutrients required for dinactin production including medium development and precursor stimulation, were investigated. Optimization of the medium and replacement of polar sea water were achieved using a one factor at a time experiment in conjunction with statistical analysis using methods covering Plackett–Burman design, the steepest descent method and central composite design. Dinactin production in the optimized medium was 160.8 mg/L, which was 47 fold higher than the control. Supplementation of the fermentation with exogenous acetate (1.5 mmol/L), succinate (6 mmol/L), malonate (24 mmol/L) and citrate (6 mmol/L) further enhanced dinactin biosynthesis by 42.7, 122.3, 66.7, and 62.1%, respectively. The precursors, in particular succinate, facilitated sugar use and also increased pH levels. Furthermore, a six-pulse feeding of total 6 mmol/L succinate in a 5 L bioreactor fermentation yielded a maximal production of 279.0 mg/L dinactin, 124.1% higher than that without precursor stimulation.         The near-field acoustic levitation for spheres by transducer with concave spherical radiating surface

        To levitate ICF target spheres in the near-field acoustic levitation, a transducer with concave spherical radiating surface and a near-field acoustic levitation system is established. The concave spherical radiating surface of the transducer is designed by the finite element parametric method. Then the levitation height and levitation perturbation of spheres with different mass and diameters in the near-field acoustic levitation system are tested and discussed in the driving voltage at 400V, 500V and 600V, respectively, when the levitation system is under the resonant frequency. Finally, based on the experimental results, the height formula of the near-field acoustic levitation for spheres is deduced by introducing a coupling coefficient.

      • 3D CACT-assisted Radiofrequency Ablation Following Transarterial Chemoembolization for Hepatocellular Carcinoma: Early Experience

        Background: To explored the value of 3D C-arm CT (CACT) guidance system in performing radiofrequency ablation (RFA) following transarterial chemoembolizationon (TACE) for hepatocellular carcinomas. Materials and Methods: RFA of hepatocellular carcinomas (HCC) were performed on 15 patients (21 lesions) with the assistance of CACT guidance system. Technical success, procedure time, complications and patient radiation exposure were investigated. The puncture performance level was evaluated on a five-point scale (5-1: excellent-poor). Complete ablation rate was evaluated after two months follow-up using enhanced CT scans. Results: The technical success rate of RFA procedure under CACT navigation system was 100 %. Mean total procedure time was $24.24{\pm}6.53min$, resulting in a mean effective exposure dose of $15.4{\pm}5.1mSv$. The mean puncture performance level rated for CACT guided RFA procedure was $4.87{\pm}0.35$. Complete ablation (CA) was achieved in 20 (95.2%) of the treated 21 tumors after the first RFA session. None of patients developed intra-procedural complications. Conclusions: 3D CACT guidance system enables reliable and efficient needle positioning by providing real-time intraoperative guidance for performing RFA on HCCs.

        Ontogenetic Expression of Lpin2 and Lpin3 Genes and Their Associations with Traits in Two Breeds of Chinese Fat-tailed Sheep

        Lipins play dual function in lipid metabolism by serving as phosphatidate phosphatase and transcriptional co-regulators of gene expression. Mammalian lipin proteins consist of lipin1, lipin2, and lipin3 and are encoded by their respective genes Lpin1, Lpin2, and Lpin3. To date, most studies are concerned with Lpin1, only a few have addressed Lpin2 and Lpin3. Ontogenetic expression of Lpin2 and Lpin3 and their associations with traits would help to explore their molecular and physiological functions in sheep. In this study, 48 animals with an equal number of males and females each for both breeds of fat-tailed sheep such as Guangling Large Tailed (GLT) and Small Tailed Han (STH) were chosen to evaluate the ontogenetic expression of Lpin2 and Lpin3 from eight different tissues and months of age by quantitative real-time polymerase chain reaction (PCR). Associations between gene expression and slaughter and tail traits were also analyzed. The results showed that Lpin2 mRNA was highly expressed in perirenal and tail fats, and was also substantially expressed in liver, kidney, reproductive organs (testis and ovary), with the lowest levels in small intestine and femoral biceps. Lpin3 mRNA was prominently expressed in liver and small intestine, and was also expressed at high levels in kidney, perirenal and tail fats as well as reproductive organs (testis and ovary), with the lowest level in femoral biceps. Global expression of Lpin2 and Lpin3 in GLT both were significantly higher than those in STH. Spatiotemporal expression showed that the highest levels of Lpin2 expression occurred at 10 months of age in two breeds of sheep, with the lowest expression at 2 months of age in STH and at 8 months of age in GLT. The greatest levels of Lpin3 expression occurred at 4 months of age in STH and at 10 months of age in GLT, with the lowest expression at 12 months of age in STH and at 8 months of age in GLT. Breed and age significantly influenced the tissue expression patterns of Lpin2 and Lpin3, respectively, and sex significantly influenced the spatiotemporal expression patterns of Lpin3. Meanwhile, Lpin2 and Lpin3 mRNA expression both showed significant correlations with slaughter and tail traits, and the associations appear to be related with the ontogenetic expression as well as the potential functions of lipin2 and lipin3 in sheep.

      • Fermi surface reconstruction and multiple quantum phase transitions in the antiferromagnet CeRhIn<sub>5</sub>

        <P><B>Significance</B></P><P>Conventional, thermally driven continuous phase transitions are described by universal critical behavior that is independent of microscopic details of a specific material. An analogous description is lacking for phase transitions that are driven at absolute zero temperature by a nonthermal control parameter. Classification of quantum-driven phase transitions is a fundamental but open problem that arises in diverse contexts and multiple classes of materials. Here we report the first observation, to our knowledge, of a sharp Fermi surface reconstruction while applying a strong magnetic field to suppress an antiferromagnetic transition to zero temperature. These experiments demonstrate that direct measurements of the Fermi surface can distinguish theoretically proposed models of quantum criticality and point to a universal description of quantum phase transitions.</P><P>Conventional, thermally driven continuous phase transitions are described by universal critical behavior that is independent of the specific microscopic details of a material. However, many current studies focus on materials that exhibit quantum-driven continuous phase transitions (quantum critical points, or QCPs) at absolute zero temperature. The classification of such QCPs and the question of whether they show universal behavior remain open issues. Here we report measurements of heat capacity and de Haas–van Alphen (dHvA) oscillations at low temperatures across a field-induced antiferromagnetic QCP (<I>B</I><SUB>c0</SUB> ≈ 50 T) in the heavy-fermion metal CeRhIn<SUB>5</SUB>. A sharp, magnetic-field-induced change in Fermi surface is detected both in the dHvA effect and Hall resistivity at [Formula] ≈ 30 T, well inside the antiferromagnetic phase. Comparisons with band-structure calculations and properties of isostructural CeCoIn<SUB>5</SUB> suggest that the Fermi-surface change at [Formula] is associated with a localized-to-itinerant transition of the Ce-4<I>f</I> electrons in CeRhIn<SUB>5</SUB>. Taken in conjunction with pressure experiments, our results demonstrate that at least two distinct classes of QCP are observable in CeRhIn<SUB>5</SUB>, a significant step toward the derivation of a universal phase diagram for QCPs.</P>

      • Stochastic stability analysis of switched genetic regulatory networks without stable subsystems

        <P><B>Abstract</B></P> <P>This paper is focused on the stability problem of a class of stochastic switched genetic regulatory networks, where the stable property of every subsystem is not imposed. By employing the stabilization effects of switching behaviors and stochastic differential equation theory, a sufficient condition for globally asymptotic stability in mean is derived. Furthermore, inspired by the idea of switching interval segmentation, an easily verifiable criterion is established by means of multiple discretized Lyapunov functions. Then, we extend the attained results to the case with time delays via the multiple discretized Lyapunov-Krasovskii functionals approach. Finally, the results obtained in the paper are illustrated by a numerical example.</P>

      • Microwave Ablation Treatment of Liver Cancer with a 2,450-MHz Cooled-shaft Antenna: Pilot Study on Safety and Efficacy

        To evaluate efficacy of microwave ablation in a primary clinical study, sixty patients (44 men, 16 women; mean age 53 years) with 96, 1-8 cm (mean $3.20{\pm}0.17$ cm) liver cancers were treated with 2,450-MHz internally cooled-shaft antenna. Complete ablation (CA) and local tumor progression (LTP) rates as well as complications were determined. CA rates in small (< 3.0 cm), intermediate (3.1-5.0 cm) and large (5.1-8.0 cm) liver cancers were 96.4% (54/56), 92.3% (24/26) and 78.6% (11/14), respectively. During a mean follow-up period of $17.17{\pm}6.52$ months, LTP occurred in five (5.21%) treated cases. There was no significant difference in the CA and LTP rates between the HCC and liver metastasis patient subgroups (P<0.05). Microwave ablation provides a reliable, efficient, and safe technique to perform hepatic tumor ablation.
