Night shift workers with disordered rhythmic mechanical loading are more prone to intervertebral disc degeneration (IDD). Our results showed that circadian rhythm (CR) was dampened in degenerated and aged NP cells. Long-term environmental CR disruption promoted IDD in rats. Excessive mechanical strain disrupted the CR and inhibited the expression of core clock proteins. The inhibitory effect of mechanical loading on the expression of extracellular matrix genes could be reversed by BMAL1 overexpression in NP cells. The Rho/ROCK pathway was demonstrated to mediate the effect of mechanical stimulation on CR. Prolonged mechanical loading for 12 months affected intrinsic CR genes and induced IDD in a model of upright posture in a normal environment. Unexpectedly, mechanical loading further accelerated the IDD in an Light-Dark (LD) cycle-disrupted environment. These results indicated that intrinsic CR disruption might be a mechanism involved in overloading-induced IDD and a potential drug target for night shift workers.

The line gear is an innovative gear transmission usually manufactured for mini gear boxes in small machines, which is designed according to the meshing theory of threedimensional conjugate contact curves. In a line gear pair, the rotation of driving and driven contact curves will form a couple of virtual revolving bodies, which are defined as pitch patterns. The shapes of pitch patterns indicate the working space of the line gear pair, and different kinds of driving and driven contact curves will commonly lead to diverse pitch patterns. In this paper, the shapes of the pitch patterns are as given constraints to design the line gears applied in confining space. Several types of two-dimensional curves (straight line, sine curve and logarithmic curve) are rolled up to form varying three-dimensional contact curves, which can occupy the same working space accordingly. To further indicate the features of the obtained contact curves, the pressure angles and the strength of line gears are calculated and compared concurrently with an experiment pertaining to their transmission errors. The test results show that the transmission errors of all the line gear pairs are below 3 %, indicating that the design theory proposed in this paper can efficiently deduce contact curves for the line gear in constrained spaces and consequently extend the application of the line gear in industrial use.
Improved design method for line gear pair based on screw theory

The line gear (LG) is a novel gear mechanism based on conjugate space curve meshing theory. It is suitable for micro-mechanical systems due to its small size, light weight, and large transmission ratio. Since its invention, the LG pair has been studied from the perspective of differential geometry. To simplify the design process of the LG pair, we studied the LG pair from the innovative perspective of screw algebra. A unified screw coordinate was created for the LG pair, and velocity screws were established to derive its meshing equations. Moreover, the design equations of the center curves and contact curves were calculated for the driving line teeth and the driven line teeth, respectively. Finally, examples of LG pairs were fabricated for the kinematic performance tests, which indicated that the LG pairs designed according to screw theory were capable of performing a stable transmission.

The clearance of activated hepatic stellate cells (HSCs) by apoptosis is critical for the reversibility of hepatic fibrosis. Mitochondrial homeostasis is regulated by mitophagy, which is an efficient way of clearing injured mitochondria that plays an important role in apoptosis. However, the role of mitophagy in apoptosis in HSCs and hepatic fibrosis is still unclear. Here, we show that mitophagy is enhanced in parallel with increased apoptosis in hepatic stellate cells during the reversal of hepatic fibrosis. The inhibition of mitophagy suppressed apoptosis in HSCs and aggravated hepatic fibrosis in mice. In contrast, the activation of mitophagy induced apoptosis in HSCs. Furthermore, we confirmed that BCL-B, which is a member of the BCL-2 family, is a regulator mediating mitophagy-related apoptosis. The knockdown of BCL-B resulted in increased apoptosis and mitophagy in HSCs, while the overexpression of BCL-B caused the opposite effects. BCL-B inhibited the phosphorylation of Parkin (a key regulator of mitophagy) and directly bound phospho-Parkin. Altogether, enhanced mitophagy promotes apoptosis in HSCs during the reversal of hepatic fibrosis. BCL-B suppresses mitophagy in HSCs by binding and suppressing phospho-Parkin, thereby inhibiting apoptosis. BCLB- dependent mitophagy is a new pathway for the regulation of apoptosis in HSCs during the regression of hepatic fibrosis

To have a better understanding of the torsional mechanism and influencing factors of PC composite box-girder with corrugated steel webs, ultimate torsional strength of four specimens under pure torsion were analyzed with Model Test Method. Monotonic pure torsion acts on specimens by eccentric concentrated loading. The experimental results show that cracks form at an angle of $45^{\circ}$ to the member's longitudinal axis in the top and bottom concrete slabs. Longitudinal reinforcement located in the center of cross section contributes little to torsional capacity of the specimens. Torsional rigidity is proportional to shape parameter ${\eta}$ of corrugation and there is an increase in yielding torque and ultimate torque of specimens as the thickness of corrugated steel webs increases.

Anesthetics are used extensively in surgeries and related procedures to prevent pain. However, there is some concern regarding neuronal degeneration and cognitive deficits arising from regular anesthetic exposure. Recent studies have indicated that brain-derived neurotrophic factor (BDNF) and cyclic AMP response element-binding protein (CREB) are involved in learning and memory processes. Genistein, a plant-derived isoflavone, has been shown to exhibit neuroprotective effects. The present study was performed to examine the protective effect of genistein against isoflurane-induced neurotoxicity in rats. Neonatal rats were exposed to isoflurane (0.75%, 6 hours) on postnatal day 7 (P7). Separate groups of rat pups were orally administered genistein at doses of 20, 40, or 80 mg/kg body weight from P3 to P15 and then exposed to isoflurane anesthesia on P7. Neuronal apoptosis was detected by TUNEL assay and FluoroJade B staining following isoflurane exposure. Genistein significantly reduced apoptosis in the hippocampus, reduced the expression of proapoptotic factors (Bad, Bax, and cleaved caspase-3), and increased the expression of Bcl-2 and Bcl-xL. RT-PCR analysis revealed enhanced BDNF and TrkB mRNA levels. Genistein effectively upregulated cAMP levels and phosphorylation of CREB and TrkB, leading to activation of cAMP/CREB-BDNF-TrkB signaling. PI3K/Akt signaling was also significantly activated. Genistein administration improved general behavior and enhanced learning and memory in the rats. These observations suggest that genistein exerts neuroprotective effects by suppressing isoflurane-induced neuronal apoptosis and by activating cAMP/CREB-BDNF-TrkB-PI3/Akt signaling.
Recent advances of Na3V2(PO4)3 as cathode for rechargeable zinc-based batteries

Rechargeable zinc-based batteries (RZBs) with the advantages of high safety, low cost, abundant resources and environmental friendliness, are considered as advanced secondary battery systems that can be applied to large-scale energy storage. As an important cathode material for RZBs, NASICON-type Na3V2(PO4)3 (NVP) possesses three-dimensional and large-scale ion channels that facilitate the rapid diffusion of Zn2+, and has a higher average operating voltage compared with other vanadium-based compounds, thus exhibiting the possibility of realizing RZBs with high energy density. However, NVP still has some problems, such as poor electronic conductivity and spontaneous dissolution in aqueous solution. The sluggish kinetics of Zn2+ (de)intercalation in NVP and dendritic growth on the Zn anode also contribute to the poor rate performance and short cycle life of the batteries. In this review, optimization strategies for the electrochemical performance of RZBs with NVP as cathode are systematically elaborated, including modification of NVP cathode and optimization of electrolyte. Several mainstream energy storage mechanisms and analysis methods in this battery system are sorted out and summarized. On this basis, the development direction of NVP–RZB system is further prospected.
Variants on ESR1 and their Association with Prostate Cancer Risk: A Meta-analysis

Background: Epidemiological studies evaluating the association of two variants rs9340799 and rs2234693 on estrogen receptor 1 (ESR1) with prostate risk have generated inconsistent results. Methods: A meta-analysis was here conducted to systematically evaluate the relationship of these two variants with prostate cancer susceptibility. Results: For rs9340799, heterozygosity of T/C carriers showed a significant increased prostate cancer risk with a pooled odds ratio (OR) of 1.34 (95% CI = 1.06-1.69) while homozygote C/C carriers showed an increased but not statistically significant association with prostate cancer risk (pooled OR = 1.29, 95% CI = 0.94-1.79). Compared to the homozygous TT carriers, the allele C carriers showed a 31% increased risk for prostate cancer (pooled OR = 1.31, 95% CI = 1.06-1.63). No significant association between the rs2234693 and prostate cancer risk was found with the pooled OR of 1.15 (95% CI = 0.97-1.39, T/C and C/C vs. T/T) under the dominant genetic model. Compared to the homozygote T/T carriers, the heterozygous T/C carriers did not show any significantly different risk of prostate cancer (pooled OR = 1.13, 95% CI = 0.94-1.36) and the homozygous C/C carriers also did not show a significant change for prostate cancer risk compared to the wide-type T/T carriers (pooled OR = 1.26, 95% CI = 0.98-1.62). Conclusion: These data suggested that variant rs9340799, but not rs2234693, on ESR1 confers an elevated risk of prostate cancer.
