        Terrabacter ginsengisoli sp. nov., isolated from ginseng cultivating soil

        A Gram-positive, strictly aerobic, nonmotile, yellowish, coccus-rod-shaped bacterium (designated Gsoil <TEX>$653^T$</TEX>) isolated from ginseng cultivating soil was characterized using a polyphasic approach to clarify its taxonomic position. The strain Gsoil <TEX>$653^T$</TEX> exhibited optimal growth at pH 7.0 on R2A agar medium at <TEX>$30^{\circ}C$</TEX>. Phylogenetic analysis based on 16S rRNA gene sequence similarities, indicated that Gsoil <TEX>$653^T$</TEX> belongs to the genus Terrabacter of the family Humibacillus, and was closely related to Terrabacter tumescens DSM <TEX>$20308^T$</TEX> (98.9%), Terrabacter carboxydivorans <TEX>$PY2^T$</TEX> (98.9%), Terrabacter terrigena <TEX>$ON10^T$</TEX> (98.8%), Terrabacter terrae <TEX>$PPLB^T$</TEX> (98.6%), and Terrabacter lapilli <TEX>$LR-26^T$</TEX> (98.6%). The DNA G + C content was 70.5 mol%. The major quinone was MK-8(<TEX>$H_4$</TEX>). The primary polar lipids were phosphatidylglycerol, diphosphatidylglycerol, phosphatidyl-ethanolamine. The predominant fatty acids were <TEX>$iso-C_{15:0}$</TEX>, <TEX>$iso-C_{16:0}$</TEX>, <TEX>$iso-C_{14:0}$</TEX>, and <TEX>$anteiso-C_{15:0}$</TEX>, as in the case of genus Terrabacter, thereby supporting the categorization of strain Gsoil <TEX>$653^T$</TEX>. However, the DNA-DNA relatedness between Gsoil <TEX>$653^T$</TEX> and closely related strains of Terrabacter species was low at less than 31%. Moreover, strain Gsoil <TEX>$653^T$</TEX> could be both genotypically and phenotypically distinguished from the recognized species of the genus Terrabacter. This isolate, therefore, represents a novel species, for which the name Terrabacter ginsengisoli sp. nov. is proposed with the type strain Gsoil <TEX>$653^T$</TEX> (<TEX>$=KACC\;19444^T=LMG\;30325^T$</TEX>).

        Chemical constituents from the aerial parts of Melandrium firmum

        Two new anthraquinones, melrubiellin C (1) andmelrubiellin D (2), were isolated from the aerial parts of Melandriumfirmum Rohrbach, together with eight known compounds(3–10). The structures of these compounds wereelucidated using 1Dand 2DNMR(COSY,HMQC,HMBCandNOESY) experiments. All isolated compounds were tested fortheir cytotoxicity against NCI-H460, Hep G2, MKN-28 andA-549 cells.Of these 10 compounds,1 and 2 exhibitedmoderatecytotoxicity with IC50 values ranging from 9.54 to 32.41 lM.

      • Stressful life events preceding the onset of depression in Asian patients with major depressive disorder

        <P><B>Background:</B></P><P>Previous studies have identified the significant role of stressful life events in the onset of depressive episodes. However, there is a paucity of cross-national studies on stressful life events that precede depression.</P><P><B>Aims:</B></P><P>We aimed to compare types of stressful life events associated with the onset of depressive episodes in patients with major depressive disorder (MDD) in five Asian countries.</P><P><B>Method:</B></P><P>A total of 507 outpatients with MDD were recruited in China (<I>n</I> = 114), South Korea (<I>n</I> = 101), Malaysia (<I>n</I> = 90), Thailand (<I>n</I> = 103) and Taiwan (<I>n</I> = 99). All patients were assessed with the Mini-International Neuropsychiatric Interview and the List of Threatening Experiences. The prevalence of each type of stressful life events was calculated and compared between each country.</P><P><B>Results:</B></P><P>The type of stressful life event that preceded the onset of a depressive episode differed between patients in China and Taiwan and those in South Korea, Malaysia and Thailand. Patients in China and Taiwan were less likely to report interpersonal relationship problems and occupational/financial problems than patients in South Korea, Malaysia and Thailand.</P><P><B>Conclusion:</B></P><P>Understanding the nature and basis of culturally determined susceptibilities to specific stressful life events is critical for establishing a policy of depression prevention and providing effective counseling services for depressed patients.</P>

        <P>C–H amination of <I>N</I>-aryl benzamides with <I>O</I>-benzoyl hydroxylamines has been achieved with either Pd(II) or Pd(0) catalysts. Furthermore, we demonstrate that secondary amines can be directly used with benzoyl peroxide in a one-pot procedure that proceeds via the in situ generation of the appropriate <I>O</I>-benzoyl hydroxylamines.       • Association of Matrix Metalloproteinase (MMP)-2 and -9 Expression with Extra-gastrointestinal Stromal Tumor Metastasis

        Matrix metalloproteinase (MMP)-2 and MMP-9 are important proteases involved in invasion and metastasis of various tumors. Extra-gastrointestinal stromal tumors (EGISTs) are rare neoplasms. This study was performed to assess MMP-2 and MMP-9 expression in EGIST tissue samples for association with clinicopathological data from the patients. Twenty-one surgical EGIST tissue specimens were collected for analysis of MMP-2 and MMP-9 expression using immunohistochemistry. MMP-2 and MMP-9 proteins were expressed in all of the epithelial cell types of EGISTs, whereas they were only expressed in 75% of the spindle cell type, although there was no statistically significant difference (p>0.05). Expression of MMP-2 and MMP-9 proteins was associated with tumor size, mitotic rate, tumor necrosis, and distant metastasis (p<0.05). MMP-2 expression was linked with MMP-9 levels (p<0.05). However, there was no correlation between MMP-9 expression and age, sex, primary site, or cell morphology in any of these 21 EGIST patients (p>0.05). Moreover, expression of MMP-2 and MMP-9 proteins increased with the degree of EGIST risk. This study provided evidence of an association of MMP-2 and MMP-9 expression with advanced EGIST behavior.

        Preliminary study of genome-wide association identified novel susceptibility genes for thyroid-related hormones in Chinese population

        Background: Thyroid hormones are critical regulators of metabolism, development and growth in mammals. However, the genetic association of thyroid-related hormones in the Chinese Han population is not fully understood. Objective: We aimed to identify the genetic loci associated with circulating thyroid-related hormones concentrations in the healthy Chinese Han population. Methods: Genotyping was performed in 124 individuals using Applied Biosystems™ Axiom™ PMDA, and 796,288 single nucleotide polymorphisms (SNPs) were available for the GWAS analysis. For replication, eleven SNPs were selected as candidate loci for genotyping by Agena MassARRAY platform in additional samples (313 subjects). The values of p < 5 × 10- 6 suggest a suggestively significant genome-wide association with circulating thyroid-related hormones concentrations. Results: We identified that rs11178277 (PTPRB, p = 4.88 × 10- 07) and rs7320337 (LMO7DN-KCTD12, p = 1.22 × 10- 06) were associated with serum FT3 level. Three SNPs (rs4850041 in LOC105373394-LINC01249: p = 3.55 × 10- 06, rs6867291 in LINC02208: p = 2.40 × 10- 06 and rs79508321 in WWOX: p = 3.35 × 10- 06) were related to circulating T3 level. Rs12474167 (LOC105373394-LINC01249, p = 1.65 × 10- 06) and rs1864553 (IWS1, p = 2.00 × 10- 06) were associated with circulating T4 concentration. The association with TGA concentration was for rs17163542 in DISP1 (p = 3.46 × 10- 06) and rs12601151 in NOG-C17orf67 (p = 2.72 × 10- 07). Two genome-level significant SNPs (rs2114707 in LINC01314, p = 1.69 × 10- 06 and rs12601151, p = 1.41 × 10- 07) associated with serum TMA concentration were identified. Moreover, rs6083269 (CST1-CST2, p = 3.36 × 10- 06) was a significant locus for circulating TSH level. In replication, rs12601151 in NOG-C17orf67 was still associated with serum TGA level (p = 0.012). Conclusions: The GWAS reported 11 new suggestively significant loci associated with circulating thyroid-related hormones levels among the Chinese Han population. These findings represented suggestively biological candidates for circulating thyroid-related hormones levels and provided new insights into the mechanisms of regulating serum TGA concentration.

      • Plasma Post-operative miR-21 Expression in the Prognosis of Gastric Cancers

        Tumor-associated microRNAs have been detected in serum or plasma, but whether plasma microRNA-21 (miR-21) could be a potential circulating biomarker for gastric cancer (GC) prognosis in Chinese is still uncertain. Real-time quantitative reverse transcription PCR (qRT-PCR) was employed in this study to compare the relative expression of miR-21 between pre-operative and post-operative paired plasmas from 42 patients with primary GCs. The results showed that the expression levels of miR-21 in the post-operative plasmas were significantly reduced by an average of 18.2 times in all patients when compared to the pre-operative plasmas, and by 22.1 times in the subgroup of patients without family history, while only 1.76 times in the subgroup of patients with a family history. With respect of clinicopathological characteristics, the plasma miR-21 expression was highly associated with differentiation degree and lymph node metastasis rate. The results suggested plasma miR-21 could be a novel potential biomarker for GC prognosis and evaluation of surgery outcomes, especially in patients without a family history.

        The role of the apoptosis-related protein BCL-B in the regulation of mitophagy in hepatic stellate cells during the regression of liver fibrosis

        The clearance of activated hepatic stellate cells (HSCs) by apoptosis is critical for the reversibility of hepatic fibrosis. Mitochondrial homeostasis is regulated by mitophagy, which is an efficient way of clearing injured mitochondria that plays an important role in apoptosis. However, the role of mitophagy in apoptosis in HSCs and hepatic fibrosis is still unclear. Here, we show that mitophagy is enhanced in parallel with increased apoptosis in hepatic stellate cells during the reversal of hepatic fibrosis. The inhibition of mitophagy suppressed apoptosis in HSCs and aggravated hepatic fibrosis in mice. In contrast, the activation of mitophagy induced apoptosis in HSCs. Furthermore, we confirmed that BCL-B, which is a member of the BCL-2 family, is a regulator mediating mitophagy-related apoptosis. The knockdown of BCL-B resulted in increased apoptosis and mitophagy in HSCs, while the overexpression of BCL-B caused the opposite effects. BCL-B inhibited the phosphorylation of Parkin (a key regulator of mitophagy) and directly bound phospho-Parkin. Altogether, enhanced mitophagy promotes apoptosis in HSCs during the reversal of hepatic fibrosis. BCL-B suppresses mitophagy in HSCs by binding and suppressing phospho-Parkin, thereby inhibiting apoptosis. BCLB- dependent mitophagy is a new pathway for the regulation of apoptosis in HSCs during the regression of hepatic fibrosis
