      • WSN Missing Data Imputing Based on Multiple Time Granularity

        Missing data is a common phenomenon in the data collection process of wireless sensor network (WSN), and the missing data imputing is an important issue of WSN stream data mining. Currently WSN missing data imputing method has little considered about the dynamic characteristics of internal data time structure during the data collection process, which makes data imputing difficult to reflect the real monitoring change objectively. In order to analyze the internal structure and dynamics of WSN time sequence data systematically, with the equivalence relation of the monitored object the time domain can be regarded as a series of integral time granule (ie atomic time point set), a wireless sensor network timing information system (WTIS) is established. The system can reason logically at different time granularity, and a multiple optimal time granularity strategy of WTIS based on hierarchical successive approximation approach is proposed. Finally, based on the research, a multiple optimal time granularity WSN missing data clustering imputing algorithm is proposed. Compared with traditional fixed time granularity missing data imputing algorithm, experiments show that the algorithm can lower error rate when imputing WSN missing data.

      • A Novel Dynamic Time Wrapping Similarity Algorithm Optimized by Multi-Granularity

        Dynamic time warping algorithm (DTW) is a method of measuring the similarity of time series. Concerning the problem that DTW cannot keep high classification accuracy when the computation speed improved, a FG-DTW method based on the idea of naive granular computing is proposed. In this method, firstly, better temporal granularity is acquired by calculating temporal variance feature and it is used to replace original time series; Secondly, the elastic size of under comparing time series granularity allow dynamic adjustment through DTW algorithm and optimal time series corresponding granularity is obtained; Finally, DTW distance is calculated by optimal corresponding granularity model. At the same time, the early termination strategy of infimum function is introduced to improve the efficiency of FG-DTW algorithm. Experiments show that the proposed algorithm improves the running rate and accuracy effectively.

        Bombyx mori used as a fast detection model of liver melanization after a clinical drug – Acetaminophen exposure

        Acetaminophen (APAP) is an effective and widely used analgesic. However, APAP overdose is the principal cause of acute liver failure (ALF) in many countries. Here, we report the phenomenon of liver melanization which occurred before APAP-induced ALF in mice. A melanic surface induced by APAP which was time- and dosedependent in the silkworm invertebrate model was observed. In addition, an APAP-induced acute tissue failure model (ATF) was established using a metabolic detoxification tissue fat body which simulated mouse liver. An investigation of the anabolic mechanism of melanin in experimental animals showed that dopaquinone and dopamine which were synthesized from tyrosine via dopa in silkworms were further metabolized to melanin, while in mice, epinephrine was synthesized via the dopamine branch and melanin was only synthesized via the dopaquinone branch. On this basis, it is proposed that melanin-metabolic levels in plasma could be used as an early diagnostic marker of APAP overdose and the black spots on insect epidermis could be used as a fast detection model of toxicity.

        Association of the Porcine Cluster of Differentiation 4 Gene with T Lymphocyte Subpopulations and Its Expression in Immune Tissues

        Cluster of differentiation 4 (CD4) is mainly expressed on $CD4^+$ T cells, which plays an important role in immune response. The aim of this study was to detect the association between polymorphisms of the CD4 gene and T lymphocyte subpopulations in pigs, and to investigate the effects of genetic variation on the CD4 gene expression level in immune tissues. Five missense mutations in the CD4 gene were identified using DNA pooling sequencing assays, and two main haplotypes (CCTCC and AGCTG) in strong linkage disequilibrium (with frequencies of 50.26% and 46.34%, respectively) were detected in the population of Large White pigs. Our results indicated that the five SNPs and the two haplotypes were significantly associated with the proportions of $CD4^-CD8^-$, $CD4^+CD8^+$, $CD4^+CD8^-$, $CD4^+$ and $CD4^+/CD8^+$ in peripheral blood (p<0.05). Gene expression analysis showed the mRNA level of the CD4 gene in thymus was significantly higher than that in lymph node and spleen (p<0.05). However, no significant difference was observed between animals with CCTCC/CCTCC genotype and animals with AGCTG/AGCTG genotype in the three immune tissues (p>0.05). These results indicate that the CD4 gene may influence T lymphocyte subpopulations and can be considered as a candidate gene affecting immunity in pigs.

        CRISPR/Cas9-mediated knockout of the Vanin-1 gene in the Leghorn Male Hepatoma cell line and its effects on lipid metabolism

        Objective: Vanin-1 (VNN1) is a pantetheinase that catalyses the hydrolysis of pantetheine to produce pantothenic acid and cysteamine. Our previous studies have shown that the VNN1 is specifically expressed in chicken liver which negatively regulated by microRNA-122. However, the functions of the VNN1 in lipid metabolism in chicken liver haven’t been elucidated. Methods: First, we detected the VNN1 mRNA expression in 4-week chickens which were fasted 24 hours. Next, knocked out VNN1 via CRISPR/Cas9 system in the chicken Leghorn Male Hepatoma cell line. Detected the lipid deposition via oil red staining and analysis the content of triglycerides (TG), low-density lipoprotein-C (LDL-C), and highdensity lipoprotein-C (HDL-C) after VNN1 knockout in Leghorn Male Hepatoma cell line. Then we captured various differentially expressed genes (DEGs) between VNN1- modified LMH cells and original LMH cells by RNA-seq. Results: Firstly, fasting-induced expression of VNN1. Meanwhile, we successfully used the CRISPR/Cas9 system to achieve targeted mutations of the VNN1 in the chicken LMH cell line. Moreover, the expression level of VNN1 mRNA in LMH-KO-VNN1 cells decreased compared with that in the wild-type LMH cells (p<0.0001). Compared with control, lipid deposition was decreased after knockout VNN1 via oil red staining, meanwhile, the contents of TG and LDL-C were significantly reduced, and the content of HDL-C was increased in LMH-KO-VNN1 cells. Transcriptome sequencing showed that there were 1,335 DEGs between LMH-KO-VNN1 cells and original LMH cells. Of these DEGs, 431 were upregulated, and 904 were downregulated. Gene ontology analyses of all DEGs showed that the lipid metabolism-related pathways, such as fatty acid biosynthesis and long-chain fatty acid biosynthesis, were enriched. KEGG pathway analyses showed that “lipid metabolism pathway”, “energy metabolism”, and “carbohydrate metabolism” were enriched. A total of 76 DEGs were involved in these pathways, of which 29 genes were upregulated (such as cytochrome P450 family 7 subfamily A member 1, ELOVL fatty acid elongase 2, and apolipoprotein A4) and 47 genes were downregulated (such as phosphoenolpyruvate carboxykinase 1) by VNN1 knockout in the LMH cells. Conclusion: These results suggest that VNN1 plays an important role in coordinating lipid metabolism in the chicken liver.

        Driver Preview Model with Dual Far-near Points for Autonomous Vehicles

        This paper proposes a driver preview-based path-following controller to control both the lateral and longitudinal movements of a vehicle. First, a lateral tracking controller with two preview points is established by considering the displacement and heading errors of the two preview points: the far point, determined by the vehicle speed and fixed preview time, and the near point located at the center of the front axle. Depending on different parameters of the road input, the steering wheel angle is calculated, and different weights are assigned to the steering wheel angles corresponding to different road inputs. Next, a longitudinal tracking controller is established, which controls the vehicle velocity based on the road information of the far point. The control objects are the brake and accelerator pedals. Subsequently, the coupling of the lateral and longitudinal motion of the vehicle is analyzed, and an integrated longitudinal and lateral tracking controller is established. To verify the performance of the controller, the controller and vehicle model are established in Simulink and CarSim, respectively, to enable joint simulation. It is observed that the coupling is solved, and the near-point control makes the tracking error converge to zero and enhances the control effect. It demonstrates high adaptability and control accuracy of the proposed controller.

        Tissues Expression, Polymorphisms of IFN Regulatory Factor 6 (IRF6) Gene and Their Associated with Immune Traits in Three Pig Populations

        Interferon regulatory factor 6 (IRF6) gene is a member of the IRF-family, and plays functionally diverse roles in the regulation of the immune system. In this report, the 13,720 bp porcine IRF6 genomic DNA structure was firstly identified with a putative IRF6 protein of 467 amino acids. Alignment and phylogenetic analysis of the porcine IRF6 amino acid sequences with their homologies to other species showed high identity (over 96%). Tissues expression of IRF6 mRNA was observed by RT-PCR, the results revealed IRF6 expressed widely in eight tissues. One SNP (HQ026023:1383 G>C) in exon7 and two SNPs (HQ026023:130 G>A; 232 C>T) in the 5′ promoter region of porcine IRF6 gene were demonstrated by DNA sequencing analysis. A further analysis of SNP genotypes associated with immune traits including IFN-${\gamma}$ and IL10 concentrations in serum was carried out in three pig populations including Large White, Landraces and Songliao Black pig (a Chinese indigenous breed). The results showed that the SNP (HQ026023:1383 G>C) was significantly associated with the level of IFN-${\gamma}$ (d 20) in serum (p = 0.038) and the ratio of IFN-${\gamma}$ to IL10 (d 20) in serum (p = 0.041); The other two SNPs (HQ026023:130 G>A; 232 C>T) were highly significantly associated with IL10 level in serum both at the day 20 (p = 0.005; p = 0.001) and the day 35 (p = 0.004; p = 0.006). Identification of the porcine IRF6 gene will help our further understanding of the molecular basis of the IFN regulation pathway in the porcine immune response. All these results should indicate that the IRF6 gene can be regarded as a molecular marker associated with the IL10 level in serum and used for genetic selection in the pig breeding.

        Experimental Investigation on Permeability Evolution of Limestone Caprock under Coupled THM Processes

        To investigate the gas sealing capacity of limestone as cap rock for gas reservoir at different burial depths, triaxial compression coupled with permeability tests of limestone subjected to different confining pressures and temperatures were carried out using the improved MTS 815 test machine. During the tests, acoustic emission (AE) monitoring system was also employed to estimate the rock damage. Test results demonstrated a degradation of triaxial compression strength with the increase in temperature and decrease of confining pressure. An empirical equation for the peak stress was established considering confining pressure and temperature. Three different permeability evolution patterns of limestone samples can be proposed based on the experimental results under different coupled thermo-hydro-mechanical (THM) conditions. According to the spatial-temporal characteristics of AE signals, permeability evolution was highly related to the fracture growth and connection in limestone. Ultimately, the experimental results revealed that the permeability of limestone gradually decreased with the increasing confining pressure and temperature. In addition, an exponential relationship between initial permeability and confining pressure was established.

        Efficient Expression and Purification of Recombinant Alcohol Oxidase in Pichia pastoris

        In order to improve the production of alcohol oxidase (AOX), a recombinant Pichia pastoris (P. pastoris)system was constructed by transformation of the plasmid pPIC9K-AOX into P. pastoris GS115. The effects of different expression conditions on alcohol oxidase activity in the culture supernatant were investigated in the shake flask scale. The results showed that the highest extracellular activity (562 U/L) of alcohol oxidase was obtained after 56h induction with 4% methanol at OD600 1.0 in the medium containing 50 g/L maltose, which is about 4.2 folds higher than previously reported. High-purity functional recombinant AOX (>90%) was purified from the culture with the Ni-NTA affinity column and Sephadex G-100 chromatographical methods, with a total recovery rate of 68.9%. Further studies showed that the purified rAOX had similar enzymatic characteristics as the native enzyme, except that the thermal stability and resistance to H2O2 inhibition of rAOX were significantly greater compared to the previous report. The purified rAOX was well tolerant to various water-miscible organic solvents. This efficient expression and purification process will be promising for large-scale production of rAOX as an important diagnostic enzyme for alcohol detection in many areas.
