        Tissues Expression, Polymorphisms of IFN Regulatory Factor 6 (IRF6) Gene and Their Associated with Immune Traits in Three Pig Populations

        Interferon regulatory factor 6 (IRF6) gene is a member of the IRF-family, and plays functionally diverse roles in the regulation of the immune system. In this report, the 13,720 bp porcine IRF6 genomic DNA structure was firstly identified with a putative IRF6 protein of 467 amino acids. Alignment and phylogenetic analysis of the porcine IRF6 amino acid sequences with their homologies to other species showed high identity (over 96%). Tissues expression of IRF6 mRNA was observed by RT-PCR, the results revealed IRF6 expressed widely in eight tissues. One SNP (HQ026023:1383 G>C) in exon7 and two SNPs (HQ026023:130 G>A; 232 C>T) in the 5′ promoter region of porcine IRF6 gene were demonstrated by DNA sequencing analysis. A further analysis of SNP genotypes associated with immune traits including IFN-${\gamma}$ and IL10 concentrations in serum was carried out in three pig populations including Large White, Landraces and Songliao Black pig (a Chinese indigenous breed). The results showed that the SNP (HQ026023:1383 G>C) was significantly associated with the level of IFN-${\gamma}$ (d 20) in serum (p = 0.038) and the ratio of IFN-${\gamma}$ to IL10 (d 20) in serum (p = 0.041); The other two SNPs (HQ026023:130 G>A; 232 C>T) were highly significantly associated with IL10 level in serum both at the day 20 (p = 0.005; p = 0.001) and the day 35 (p = 0.004; p = 0.006). Identification of the porcine IRF6 gene will help our further understanding of the molecular basis of the IFN regulation pathway in the porcine immune response. All these results should indicate that the IRF6 gene can be regarded as a molecular marker associated with the IL10 level in serum and used for genetic selection in the pig breeding.

        Tissues Expression, Polymorphisms Identification of FcRn Gene and Its Relationship with Serum Classical Swine Fever Virus Antibody Level in Pigs

        Neonatal Fc receptor (FcRn) gene encodes a receptor that binds the Fc region of monomeric immunoglobulin G (IgG) and is responsible for IgG transport and stabilization. In this report, the 8,900 bp porcine FcRn genomic DNA structure was identified and putative FcRn protein included 356 amino acids. Alignment and phylogenetic analysis of the porcine FcRn amino acid sequences with their homologies of other species showed high identity. Tissues expression of FcRn mRNA was detected by real time quantitative polymerase chain reaction (Q-PCR), the results revealed FcRn expressed widely in ten analyzed tissues. One single nucleotide polymorphism (SNP) (HQ026019:g.8526 C>T) in exon6 region of porcine FcRn gene was demonstrated by DNA sequencing analysis. A further analysis of SNP genotypes associated with serum Classical Swine Fever Virus antibody (anti-CSFV) concentration was performed in three pig populations including Large White, Landrace and Songliao Black pig (a Chinese indigenous breed). Our results of statistical analysis showed that the SNP had a highly significant association with the level of anti-CSFV antibody (At d 20; At d 35) in serum (p = 0.008; p = 0.0001). Investigation of expression and polymorphisms of the porcine FcRn gene will help us in further understanding the molecular basis of the antibody regulation pathway in the porcine immune response. All these results indicate that FcRn gene might be regarded as a molecular marker for genetic selection of anti-CSFV antibody level in pig disease resistance breeding programmes.

        Pd nanoparticles immobilized on TiO2 nanotubes-functionalized ceramic membranes for flow-through catalysis

        A high performance catalytic membrane was fabricated with Pd nanoparticles supported by TiO2 nanotubes, where the TiO2 nanotubes were synthesized on the ceramic membrane via a simple hydrothermal etching. A flow-through catalytic membrane reactor was developed for testing the catalytic properties in the p-nitrophenol reduction. The effect of etching time was investigated in detail and an optimal etching time was determined to be 16 h. The characterization results highlighted that the as-prepared bouquet-like TiO2 nanotubes could significantly improve the loading amount and dispersity of Pd nanoparticles. The fabricated catalytic membrane exhibited considerably improved catalytic activity and stability, with a 100% conversion of p-nitrophenol and no loss in catalytic activity during five reaction cycles. The obtained activation energy was much lower than the values in literatures, implying that the p-nitrophenol reduction could take place more easily on our catalytic membranes compared to other catalysts.

        Enhanced Sliding Mode Control and Online Estimation of Optimal Slip Ratio for Railway Vehicle Braking Systems

        This paper proposes an optimal anti-locking control scheme so as to improve the braking performance of railway vehicles. The controlling effect of sliding mode control is improved, and the optimal slip ratio is achieved by extreme seeking algorithm. Firstly, a substitute function for the conventional sign function is proposed. Secondly, a closed loop observer for braking systems is used to enhance the estimation value of adhesion force, which can also be used for calculating reference speed. Finally Sliding Mode Controlbased controller needs to be entered a reference slip ratio called optimal slip ratio, which is searched by extreme seeking algorithm from the functional relationship between slip ratio and friction coefficient. Thus, the maximum adhesion is achieved despite wheel/ rail surface changes. The simulation result demonstrates the effect of real-time adjustment for braking torque, which guarantees the braking performance.

        Failure Analyses of a Reinforced Embankment by Strength Reduction and Limit Equilibrium Methods Considering Hardening of Soft Clay

        A 7.6 m high reinforced embankment-wall system was constructed in stages on very thick soft clay. The reinforced structure incurred a deep-seated global failure at the end of backfill loading. The Strength Reduction Method (SRM) and the Limit Equilibrium Method (LEM) were employed to analyze the stability of the structure. It was found that the SRM captured the deep-seated global failure of the structure through a coupled mechanical and hydraulic Finite Element (FE) modeling. The stability of the reinforced structure was much underestimated or overestimated by the LEM using unconsolidated-undrained or consolidated-undrained shear strength values. However, the stability of the structure could be still captured by the LEM with the increased shear strength values assigned to the consolidated zone directly beneath the reinforced structure and with the unconsolidated-undrained shear strength values assigned to the unconsolidated zone beyond the structure. In practice, both the SRM and the LEM are recommended for stability analyses of geosynthetic-reinforced structures on soft ground considering hardening of the soil so that the results from the two methods can be cross-examined.

        RC‑qTPWM method with high DC voltage utilization ratio and power balance for three‑phase CHB inverters

        In this paper, a modified method named reconstructed carrier quasi-trapezoidal pulse width modulation (RC-qTPWM) isproposed to improve the DC voltage utilization ratio, decrease the line voltage total harmonic distortion (THD), and solve thepower imbalance among H-bridge units. The modulation method is based on two steps. In the first step, the quasi-trapezoidal wave obtained by an intercepting sine wave is used as the modulation wave to improve the DC voltage utilization ratio. In the second step, the carrier arrangement of the phase disposition is reconstructed to achieve a power balance among the H-bridge units. Carrier reconstruction is achieved by shifting a single triangular carrier in the horizontal and vertical directions. Through simulation and theoretical analysis, the selection method of the optimal sine coefficient δ and the principle of the carrier reconstruction are expounded, which shows that the optimal sine coefficient δ is 0.58 and that the power balance can be achieved within 3Tc. When compared with conventional TPWM methods, this modulation method has a higher DC voltage utilization ratio, a better efficiency, a smaller line voltage, a reduced current THD, and better power balance performance. Experimental results verify the correctness of the simulation and theoretical analysis, and the feasibility of the RC-qTPWM method.

        Scale effect mechanism research of insect-imitating hexapod robot

        The scale parameter is one of the key factors which influence the structure and locomotion characteristics of the legged robot. The scale effect mechanism of animals in nature was revealed, and the influence of scale parameters on animal was analyzed. This paper establishes a dynamic model of insect-imitating hexapod robot. A high-fidelity simulation platform for hexapod robot was established based on Vortex, and the foot-ground interaction mechanics model was established and applied to the developed simulation platform. Based on the existing six-legged robot prototype, the validity of the relevant model and the fidelity of the simulation platform are verified. The influence of a robot’s mass and characteristic size on its feature locomotion was analyzed. The foot force rises with the increase of the whole robot mass, and the foot force of the unit robot mass decreases with the increase of the whole robot mass, eventually tending to a fixed value. The maximum joint torque rises with the increase of the whole robot mass. The system power rises with the increase of the whole robot mass, but the system power of unit robot mass is basically a constant value. The peak system power decreases with the increase of the distance between the front and the rear leg, and the joint torque rises with the increase of the distance between the front and rear leg. The related research results have guiding significance and reference value for the system design of hexapod robots.

        Visible light photocatalytic removal performance and mechanism of diclofenac degradation by Ag3PO4 sub-microcrystals through response surface methodology

        In this study, high visible light response and charge separation efficiency of body-centered cubic Ag3PO4sub-microcrystals were fabricated through ultrasound-precipitation process. Additionally, the parameterswere optimized by photocatalytic (PC) removal degradation of diclofenac based on response surfacemethodology. Results indicated that the PC removal efficiency of diclofenac was significantly affected byAg3PO4 content and pH value. Under the optimized conditions, 99.9% of diclofenac could be degradedwithin 16 min Xenon illumination. Furthermore, three PC removal degradation pathways of diclofenacwere proposed, including OH-adduct to aromatic ring, direct oxidation by photoinduced holes anddecarboxylation of side chain from the parent compound.

        Acoustic emission characterization and cracking characteristics of reinforced concrete pipe

        Acoustic emission (AE) and digital image correlation (DIC) technology were used in this study to monitor the behavior of reinforced concrete pipe (RCP) in the optimized three-edge bearing test (TEBT). At the same time, the AE source location of different pipe sections was simulated. The results show that the whole compressive loading process for RCP could be divided into 3 to 4 stages. Both the AE counts and AE energy could well characterize the cracking of concrete, and AE energy was more sensitive to the breaking of steel bar. The amplitude and duration of AE signal produced by the breaking of steel bar were larger than those produced by the cracking of concrete. The AE frequency for concrete cracking was relatively scattered. The AE frequency for steel bar breaking was small and concentrated, and this frequency distribution showed an exponential decreasing trend. The results of optimized AE source location showed that the generation and development of cracks occur in the initial loading stage. The results of multi-index analysis showed that the cracks in the top (loading position) and the bottom (bearing position) of the RCP show that the cracks extend from the inner wall to the outer wall, while the cracks on the left and right sides perpendicular to the loading direction show that the cracks extend from the outer wall to the inner wall.
