Health-related Quality of Life After Treatment for Malignant Bone Tumors: A Follow-up Study in China

Aim: We conducted the present study to assess health-related quality of life (HRQoL) among bone cancer patients after surgical treatment in one large teaching hospitals in China, and assess the risk factors for improving the physical or mental HRQoL. Methods: 344 eligible adult patients who were admitted to the hospital with malignant bone tumors during the period of Jun. 2008 to Dec. 2011, and a reference group with 361 health cases was recruited in the same hospital during the same period. All 344 patients were followed up for one year. The HRQoL before treatment and after one year was evaluated with the Medical Outcome Short Form 36 (SF-36). Results: All 8 domains of HRQoL had the lowest scores greatly improved over the first year after discharge. However, the patients still had significantly lower scores in every domain than the reference group one year after discharge. Age and type of surgery were associated with HRQoL in the mental domain. Conclusion: The HRQoL of patients with malignant bone tumors greatly improved one year after the treatment. This study also highlighted the utility of HRQoL assessment for prognostic evaluation of patients after surgical treatment for bone cancer.

Background: Sentinel lymph node (SLN) mapping has been recommended as an alternative staging approach to lymphadenectomy for apparent uterine-confined endometrial cancer (EC). However, the prognostic value of SLN mapping alone instead of systematic lymphadenectomy on EC patients remains unclear. Methods: A multi-center, open label, non-inferiority randomized controlled trial has been designed to identify if SLN mapping alone is not inferior to pelvic lymphadenectomy on prognosis of patients with intermediate-high-risk EC clinically confined to uterus. Eligible patients will be 1:1 randomly assigned to accept SLN mapping or pelvic lymphadenectomy. The primary endpoint is the 2-year progression-free survival (PFS). The second points are the 5-year PFS, 5-year overall survival, surgery-related adverse events and life quality. A total of 780 patients will be enrolled from 6 hospitals in China within 3-year period and followed up for 5 years. Trial Registration: ClinicalTrials.gov Identifier: NCT04276532

Objectives: To observed the effects of ginsenoside -Rh2 (GS-Rh2) on proliferation and apoptosis of side population (SP) human gastric cancer SGC-7901 cells. Materials and Methods: SGC-7901 SP and Non-SP cells were sorted by flow cytometry and assessed using the cck-8 method. Expression of apoptosis-related proteins Bax and Bcl-2 of SP before and after the intervention was determined by Western-blotting. Results: It was found that the proliferation of SP was significantly faster than that of NSP (P<0.05). In addition, GS-Rh2 inhibited proliferation of gastric cancer SP cells, induced cell cycle arrest and cell apoptosis, and changed the expression of BAX/Bcl-2 proteins in a time-dependent and concentration-dependent manner (P<0.05). Conclusions: With increase of GS-Rh2 dose, GS-Rh2 gradually inhibit the proliferation of SGC-7901 SP cells, which have high proliferation rate, through G1/G0 phase arrest, followed by apoptosis which involves the up-regulation of Bax and the down-regulation of Bcl-2.

Oxide dispersion-strengthened materials W-1wt%Pr<sub>2</sub>O<sub>3</sub> and W-1wt%La<sub>2</sub>O<sub>3</sub> were synthesized by wet chemical method and spark plasma sintering. The field emission scanning electron microscopy (FE-SEM) analysis, XRD and Vickers microhardness measurements were conducted to characterize the samples. The irradiations were carried out with a 5 keV helium ion beam to fluences up to 5.0 × 10<sup>21</sup> ions/m<sup>2</sup> under 600 ℃ using the low-energy ion irradiation system. Transmission electron microscopy (TEM) study was performed to investigate the microstructural evolution in W-1wt%Pr<sub>2</sub>O<sub>3</sub> and W-1wt%La<sub>2</sub>O<sub>3</sub>. At 1.0 × 10<sup>20</sup> He<sup>+</sup>/m<sup>2</sup>, the average loops size of the W-1wt%Pr<sub>2</sub>O<sub>3</sub> was 4.3 nm, much lower than W-1wt% La<sub>2</sub>O<sub>3</sub> of 8.5 nm. However, helium bubbles were not observed throughout in both doped W materials. The effects of pre-irradiation with 1.0 × 10<sup>21</sup> He<sup>+</sup>/m<sup>2</sup> on trapping of injected deuterium in doped W was studied by thermal desorption spectrometry (TDS) technique using quadrupole mass spectrometer. Compared with the samples without He<sup>+</sup> pre-irradiation, deuterium (D) retention of doped W materials increased after He<sup>+</sup> irradiation, whose retention was unsaturated at the damage level of 1.0 × 10<sup>22</sup>D<sub>2</sub><sup>+</sup>/m<sup>2</sup>. The present results implied that irradiation effect of He<sup>+</sup> ions must be taken into account to evaluate the deuterium retention in fusion material applications.
Metastasis associated genomic aberrations in stage II rectal cancer

Genomic aberrations of rectal carcinoma, especially DNA copy number changes associated with metastasis were largely unclear. We aim to identify the metastasis associated biomarkers in stage II rectal cancer. Formalin-fixed, paraffin-embedded primary tumor tissues of stage II rectal carcinoma were analyzed by array-based comparative genomic hybridization, and genomic aberrations were identified by Genomic Workbench and SAM software. Copy number changes and mRNA expressions were validated by Real-time PCR in an independent rectal cancer samples. The results showed that the most frequent gains in stage II rectal cancer were at 1q21.2-q23.1, 3p21.31, 11q12.2-q23.3, 12q24.11-q24.31, 12q13.11-q14.1 and losses in 18q11.2-q23, 17q21.33-q22, 13q31.1-q31.3, 21q21.1-q21.3, 8p23.3-p23.1 and 4q22.1-q23. Twenty-two amplifications and five homozygous deletions were also identified. We further found that S100A1 (1q21.3-q23.1), MCM7 (7q22.1) and JUND (19p13.11) were amplified and overexpressed in stage II rectal cancer. Interestingly, the genomic aberrations affected 14 signaling pathways including VEGF signaling pathway and fatty acid metabolism. Most importantly, loss of 13q31.1-q34 and gain of 1q44 were associated with distant metastasis. Our results indicated that these metastasis associated genomic changes may be useful to reveal the pathogenesis of rectal cancer metastasis and identify candidate biomarkers.

The displacement modes of retaining walls in engineering are complicated, and the backfill behind retaining walls is often narrow. Failure mechanisms of soils under various displacement modes of retaining walls have not been clarified yet in previous studies. According to results of the finite element limit analysis, multiple sliding surfaces were observed developing reflectively between interfaces on both sides in the active limit state. In rotational displacement modes, some layers of soil near the rotated point maintained in the non-limit state which was not considered in the previous studies. Based on numerical analysis results, failure wedges were divided into differential soil slice elements by an inclined slice method. An analytical model was established to estimate the earth pressure of the narrow backfill under various displacement modes using the limit equilibrium method. The calculated results are consistent with the data gathered from previous tests. Compared with previous studies, the proposed method can reflect failure mechanisms of the narrow backfill and be adapted to various displacement modes of the retaining wall. The accuracy of the calculation results is high in the non-limit state soil layer. In addition, the boundary criterion between the narrow and semi-limit backfills was determined. Based on extensive parametric studies, it can be concluded that increasing the soil-wall interface friction and decreasing the backfill area would be conducive to decreasing the earth pressure and the depth of resultant application point.
