        Ground type-I collagen—a focused study on its fibrillogenesis behavior and bioactivity in vitro

        Take bovine tendon type-I collagen as the raw material, we pretreated it by freezing and then grinding the macromolecule at a low temperature (4–10 °C). Effects of the grinding process on the molecular structure, water solubility, fibrillogenesis behavior in vitro and biological activity of collagen were systematically prospected. Sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS–PAGE) results revealed that the collagen had typical structural characteristics of type-I collagen; moreover, grinding treatment gave rise to partially degraded collagen molecules which took on continuous bands with lower molecular weights different from α1 and α2. More strikingly, the degradation degree positively corresponded to the grinding duration. Circular dichroism (CD) analysis manifested that with extended grinding, intensities of both the positive and negative peaks for collagen samples shrunk. Interestingly, grinding also contributed to improved water solubility of collagen. The water solubility of the sample ground for 20 min (COL20) was 1.91 mg/mL, which is about 6 times higher than that of the natural collagen sample (0.28 mg/mL). Amid the study concerning the fibrillogenesis behavior, turbidity and rheological data collaboratively demonstrated that with prolonged grinding time, the assembly rate and degree of the sample plunged. Assembled gel from the ground collagen with finer fibrils was also observed in scanning electron microscope (SEM), which further embraced the conclusion aforementioned in fibrillogenesis kinetics. Cell experiments corroborated that the grinding process can be harnessed to manipulate the cell proliferation promoting ability of collagen. Simultaneously, further dissection of COL20 with the best biological activity clarified that the water-soluble part and water-insoluble part of COL20 had distinct aptitudes as to promoting cell proliferation as well as inhibiting the growth of liver cancer cells.

        The Belt and Road Initiative(BRI) represents a platform proposed by China to promote global cooperation, enhance economic governance, and facilitate shared development and prosperity worldwide. As of July 2022, over 150 nations and 32 international organizations had signed more than 200 agreements regarding Belt and Road cooperation. Saudi Arabia constitutes a vital nexus for China's Belt and Road development in West Asia. However, Saudi Arabia's domestic security circumstances, encompassing leadership transitions, complex geopolitics, and terrorism, engender risks that could impede Belt and Road advancement in the West Asian region. Mitigating these challenges through proactive diplomacy and security partnerships will be instrumental as China and Saudi Arabia seek to realize their shared vision for greater connectivity and economic integration. To foster BRI advancement in Saudi Arabia, Chan and Saudi Arabia should strengthen political trust, diversifying economic ties, enhancing counterterrorism cooperation, and shaping a favorable public opinion environment through cultural exchanges. In summary, while Saudi Arabia is a vital BRI partner, mitigating risks through proactive diplomacy and security measures is key.

        The Analysis of Beijing Tourism Destination Image Based on the Network Texts

        정보통신기술의 발달로 인해 잠재 관광객은 인터넷으로 여행 정보를 수집한다. 관광 객들은 온라인 플랫폼에서 여행과 관련된 계획, 전략 등을 포스팅하고 여행 경험을 공 유한다. 이러한 온라인 텍스트는 관광객의 경험과 느낌을 주관적으로 설명한다. 본 연 구는 이러한 텍스트를 기반으로 목적지 이미지를 분석하고자 하였다. 본 연구는 Ctrip.com에 게시된 여행 관련 텍스트 중 베이징의 사례를 수집하여 정리하였다. ROST CM6 소프트웨어를 사용하여 빈도가 높은 특징 단어를 추출하고, 네트워크 텍스트 분석 을 사용하여 베이징의 인지적 이미지, 정서적 이미지 및 추천의도를 분석하였다. 연구 결과, 관광객은 목적지의 관광명소에 대한 인지적 이미지가 가장 강하게 나타나 는 것으로 나타났다. 정서적 이미지는 관광객의 긍정적인 감정에 영향을 주지만, 추천 의도를 명확하게 나타내지는 않았다. 분석된 결과를 바탕으로 베이징의 관광지 이미지 제고를 위한 시사점을 제시하였다. With the development of internet, potential tourists began to search travel information of destination through the internet. At the same time, tourists also share their travel experiences by publishing travel notes and tips on online social platforms. These online texts from tourists describe their experiences and feelings of the destination from a subjective perspective, are also providing original data of destination image for researchers. This paper takes Beijing as a case study site, collected travel notes on Ctrip.com, used ROST CM6 to extract high-frequency feature words, and used network text analysis to analyze Beijing’s cognitive image, emotional image, and recommendation intentions. It can be found that the tourists have the strongest perception of the tourist attractions in the cognitive image, and shown a positive emotional image. Otherwise, the recommendation intention is not very clear. According to the results, relevant destination strategies to enhance the image of Beijing’s are proposed.

        Functional significance of cholesterol metabolism in cancer: from threat to treatment

        Cholesterol is an essential structural component of membranes that contributes to membrane integrity and fluidity. Cholesterol homeostasis plays a critical role in the maintenance of cellular activities. Recently, increasing evidence has indicated that cholesterol is a major determinant by modulating cell signaling events governing the hallmarks of cancer. Numerous studies have shown the functional significance of cholesterol metabolism in tumorigenesis, cancer progression and metastasis through its regulatory effects on the immune response, ferroptosis, autophagy, cell stemness, and the DNA damage response. Here, we summarize recent literature describing cholesterol metabolism in cancer cells, including the cholesterol metabolism pathways and the mutual regulatory mechanisms involved in cancer progression and cholesterol metabolism. We also discuss various drugs targeting cholesterol metabolism to suggest new strategies for cancer treatment.

        Optimal sensor placement for cable force monitoring using spatial correlation analysis and bond energy algorithm

        Cable force monitoring is an essential and critical part of the safety evaluation of cable-supported bridges. A reasonable cable force monitoring scheme, particularly, sensor placement related to accurate safety assessment and budget costsaving becomes a major concern of bridge administrative authorities. This paper presents optimal sensor placement for cable force monitoring by selecting representative sensor positions, which consider the spatial correlativeness existing in the cable group. The limited sensors would be utilized for maximizing useful information from the monitored bridges. The maximum information coefficient (MIC), mutual information (MI) based kernel density estimation, as well as Pearson coefficients, were all employed to detect potential spatial correlation in the cable group. Compared with the Pearson coefficient and MIC, the mutual information is more suitable for identifying the association existing in cable group and thus, is selected to describe the spatial relevance in this study. Then, the bond energy algorithm, which collects clusters based on the relationship of surrounding elements, is used for the optimal placement of cable sensors. Several optimal placement strategies are discussed with different correlation thresholds for the cable group of Nanjing No.3 Yangtze River Bridge, verifying the effectiveness of the proposed method.

        A study on the bearing effect of pier column backfilling in the goaf of a thin coal seam

        The pier column backfilling method in the goaf of a thin coal seam introduced in this paper is the latest backfilling technology in China, which has the combined advantages of high efficiency and low cost. In order to fully verify the load bearing effect of this backfilling method, a comprehensive evaluation model which combined the actual measurement, numerical simulation and physical simulation is implemented in this paper. First, the stress bearing formula and backfilling volume rate of a pier column backfilling body is obtained by theory analysis. Then, through field monitoring, it was concluded that the bearing capacity when the backfilling body reached a stable state was similar to the results of theoretical calculation, and no obvious deformation occurred by observation the roadway deformation with the site. Finally, based on the drilling columnar and lithological parameters of the site, corresponding physical and numerical models were established and used to study the bearing effect of this backfilling method. It could be found that the stresses and displacements data that resulted from the simulation relatively confirmed data obtained from the site. After comprehension of above researches, it is concluded that the pier column backfilling method in a goaf has good bearing effect and a good economic benefit for mining thin coal seams.

        Inhibition of mouse SP2/0 myeloma cell growth by the B7-H4 protein vaccine

        B7-H4 is a member of B7 family of co-inhibitory molecules and B7-H4 protein is found to be overexpressed in many human cancers and which is usually associated with poor survival. In this study, we developed a therapeutic vaccine made from a fusion protein composed of a tetanus toxoid (TT) T-helper cell epitope and human B7-H4IgV domain (TT-rhB7-H4IgV). We investigated the anti-tumor effect of the TT-rhB7-H4IgV vaccine in BALB/c mice and SP2/0 myeloma growth was significantly suppressed in mice. The TT-rhB7-H4IgV vaccine induced high-titer specific antibodies in mice. Further, the antibodies induced by TT-rhB7-H4IgV vaccine were capable of depleting SP2/0 cells through complement-dependent cytotoxicity (CDC) in vitro. On the other hand, the poor cellular immune response was irrelevant to the therapeutic efficacy. These results indicate that the recombinant TT-rhB7-H4IgV vaccine might be a useful candidate of immunotherapy for the treatment of some tumors associated with abnormal expression of B7-H4.         Tolerance potential of Chilo suppressalis larvae to fipronil exposure via the modulation of detoxification and GABA responses

        The Asiatic rice borer, Chilo suppressalis Walker, is one of the most destructive rice pests in the world; its physiological adaptability to the sustained application of fipronil was investigated. After topical application, fipronil showed strong lethal effect with LD20 and LD80 values on the fourth instar larvae as 0.00159 and 0.00906 μg per larvae at 48 h- post treatment, and also revealed slight inhibition on the eclosion of pupae and the hatching of eggs. LD20 dosage of fipronil significantly enhanced the activities of carboxylesterase (CarE), glutathione-Stransferase (GST) and cytochrome P450-linked p-nitro O-demethylase (PNOD) in the borers. LD80 dosage of fipronil showed a mild decrease on the activity of CarE, GST and PNOD, and a slight suppression on the activity of glutamate dehydrogenase (GDH), but strongly induced the transform of glutamate to gamma-aminobutyric acid (GABA) and the activity of glutamate decarboxylase (GAD), thereby improved GABA level in the bores. Results suggested that the borer possessed positive tolerance potential to firponil via the modulation of detoxification and GABA responses and provided the basis for further studies on reducing the tolerance of the borer in the control strategies.
