Background

Several studies have suggested potential links of phthalates to obesity in children and adults. Limited evidence, however, has been available for the relations between diethylhexyl phthalate (DEHP) and obesity-related markers or body mass change in early life.

Methods

128 healthy pregnant women were recruited and, after delivery, their newborns’ first urine and umbilical cord blood samples were collected. We measured urinary levels of two DEHP metabolites, mono-(2-ethyl-5-hydroxyhexyl) phthalate (MEHHP) and mono-(2-ethyl-5-oxohexyl) phthalate (MEOHP). We also measured the levels of leptin, total cholesterol and triglyceride (TG) in cord serum, and used them along with weight, length, head circumference and ponderal index (PI, 100 g/cm3) at birth, as obesity-related markers, and estimated the relations between DEHP metabolites and obesity-related markers using generalised linear models.
Significance

An aberrant covalent histone modification which drives dysregulation of transcriptional program is related to human diseases such as cancer. Thus, identifying signaling pathways modulating transcription factors and epigenetic enzymes are coming into view as attractive therapeutic targets. In this study, we provide evidence that KDM3A is tyrosine-phosphorylated by JAK2, and tyrosine-phosphorylated KDM3A acts as a coactivator for STAT3, thereby exerting increased cancer cell growth and motility. We propose that JAK2-dependent tyrosine phosphorylation of KDM3A could be a potential therapeutic target for epigenetic control of oncogenic effect governed by JAK2−STAT3 signaling pathway.

Janus tyrosine kinase 2 (JAK2)−signal transducer and activator of transcription 3 (STAT3) signaling pathway is essential for modulating cellular development, differentiation, and homeostasis. Thus, dysregulation of JAK2−STAT3 signaling pathway is frequently associated with human malignancies. Here, we provide evidence that lysine-specific demethylase 3A (KDM3A) functions as an essential epigenetic enzyme for the activation of JAK2−STAT3 signaling pathway. KDM3A is tyrosine-phosphorylated by JAK2 in the nucleus and functions as a STAT3-dependent transcriptional coactivator. JAK2−KDM3A signaling cascade induced by IL-6 leads to alteration of histone H3K9 methylation as a predominant epigenetic event, thereby providing the functional and mechanistic link between activation of JAK2−STAT3 signaling pathway and its epigenetic control. Together, our findings demonstrate that inhibition of KDM3A phosphorylation could be a potent therapeutic strategy to control oncogenic effect of JAK2−STAT3 signaling pathway.

Objectives To evaluate the feasibility of robot-assisted laparoscopic myomectomy in multiple myomas over 10. Methods A retrospective study was conducted for 662 patients who underwent robot-assisted laparoscopic myomectomy and open myomectomy by a single operator in a tertiary university hospital. Results A total of 30 women underwent removal of 10 or more uterine myomas by robotics and 13 patients were selected for this study. The average number of myomas removed was 13.7 (range 10-20). The maximum diameter of the myomas was 6.8 cm (range 5.0-10.0 cm). The sum of the diameters of each myoma was 34.7 cm (range 20.0-54.5 cm) and the mass of resected myomas for each case was 229.1 g (range 106.8-437.9 g). In no case was the robotic procedure converted into conventional laparoscopy or laparotomy, and all patients recovered without any major complications. In comparison with 13 cases of open myomectomy during the same period, robotic surgery took longer time than open surgery (360.5 vs. 183.8 minutes; P=0.001) but had shorter postoperative hospital days after surgery (mean 2.5 vs. 3.5 days; P=0.003). Conclusion Robot-assisted laparoscopic myomectomy could be an alternative to laparotomic myomectomy for numerous myomas over 10 in number.

B-cell lymphoma 3 (Bc13) is a proto-oncogene upregulated in a wide range of cancers, including breast cancer. Although Bc13 is known to promote cell proliferation and inhibit apoptosis, the molecular mech¬anisms underlying the proto-oncogenic function of Bc13 have not been completely elucidated. To gain insight into the oncogenic role of Bc13, we applied a proteomic approach, which led to the identification of C-terminal binding protein 1 (CtBP1) as a binding partner of Bc13. A PXDLS/R motif embedded in Bc13 was found to mediate the interaction between Bc13 and CtBP1, which caused the stabilization of CtBP1 by blocking proteasome-dependent degradation. Apoptotic stimuli-induced degradation of CtBP1 was sig¬nificantly abolished by the upregulation of Bc13, leading to the sustained repression of pro-apoptotic gene expression and subsequent inhibition of apoptosis. Intriguingly, a strong positive correlation between the protein levels of Bc13 and CtBP1 was detected in breast cancer patient samples. Our study reveals a novel combinatorial role for Bc13 and CtBP1, providing an explanation for the acquisition of resistance to apop¬tosis in cancer cells, which is a major requirement for cancer development.

Kim Sung Beak,Choi Hyunkyung,Kim Chul Sung 한국물리학회 2020 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.77 No.10
Nanoparticles of spinel ferrite (Zn0.05Fe2.95O4) were prepared using the process of high- temperature thermal decomposition (HTTD) and were treated with an Ar plasma for 30 min. The crystal structure was found to be cubic spinel with space group (Fd-3m). The lattice constants of Zn0.05Fe2.95O4 before and after the plasma treatment were found to be 8.3862 and 8.3853 Å, respectively. The saturation magnetization value increased from 69.1 to 73.5 emu/g after the plasma treatment of the Zn0.05Fe2.95O4 nanoparticles. The Mössbauer spectra before and after the plasma treatment of Zn0.05Fe2.95O4 were analyzed at 4.2 K using four sextets. As the temperature was increased, the lines of the Mössbauer spectra broadened due to a fluctuation in the anisotropic hyperfine field. The Mössbauer spectra were found to have the same anisotropic field fluctuation of +H (P+ = 0.5) and -H (P- = 0.5). The relaxation frequency factor, f, was also calculated from the Mössbauer spectra.

We have achieved an efficient solution-phase parallel synthesis of a library of natural piper-amide-like compounds from the bifunctional β-phosphono-N-hydroxy-succinimidyl ester intermediate. The primary important feature in our study is the construction of natural-product-like molecules through the adaptation of sophisticated organic reactions that create water-soluble byproducts for a chromatography-free purification. This simple and efficient method rapidly provides a combinatorial library of high yield and purity. The library was evaluated against GPCR targets to demonstrate its potential use as a tool for drug discovery and in chemical biology.

Graphic Abstract

ACS Electronic Supporting Info

Angioleiomyoma is a rare type of leiomyoma variant and there are a few cases reported to date. Herein, we present a case of angioleiomyoma in a 36-year-old woman with lower abdominal pain, initially diagnosed by degenerated uterine leiomyoma. The transvaginal ultrasonogram showed an ovoid-shaped heterogeneously hyperechoic lesion in left cornual site of uterus and pelvic magnetic resonance image showed an about 5.1 cm sized heterogenous T2 intermediate high mass with poor enhancement. The patient underwent a robot-assisted laparoscopic myomectomy, and final histopathologic diagnosis revealed uterine angioleiomyoma. This case is the first case of angioleiomyoma resected by robotic surgery. The patient is on follow up for over 1 year and shows no evidence of recurrence until now.

Background and Aims: AL amyloidosis can cause various vascular events, which are mainly related to vascular amyloid deposition or thromboembolism. Despite their clinical importance, only few reports have been published on the manifestations of vascular events in these patients. Methods: We retrospective reviewed medical records of patients including various imaging study results such as angiography, Doppler sonography, computed tomography, and magnetic resonance imaging to find out clinically significant vascular stenosis and thromboembolism. Results: Between January 2001 and March 2014, 106 patients (male/female=60/46) with biopsy-proven AL amyloidosis were included from 3 hospitals. The median age was 58 years (range, 23-79 years). During a median follow-up of 12.6 months (range, 0.1-156.8 months), 13 events of thromboembolism developed in 13 patients: 10 patients (9.4%) experienced cerebral infarction, 2 patients (1.9%) pulmonary embolism and 1 patient (0.9%) deep vein thrombosis. Patients with higher free light chain difference suffered significantly more from vascular events than the others in multivariable analysis. (20.2% vs. 8.9%, p=0.022). The median overall survival (OS) from pathologic diagnosis was 20.9 months (95% CI, 5.7-36.2). In multivariable analysis, higher BM plasma cell percentage and higher serum B2 microglobulin levels were poor prognostic factors. (p<0.05) Conclusions: The incidence of vascular events was substantial in patients with AL amyloidosis. Vascular events significantly occurred in patients with higher free light chain difference. Poor prognostic factor for overall survival was higher serum beta-2 microglobulin.
