      • 교육대학생들이 경험한 초등학교의 ‘좋은 수업’

        본 연구는 예비교사이면서 동시에 학습자의 입장에 있는 ‘교육대학생’들을 대상으로, 그들이 초등학교 시절에 경험한 수업을 회고, 반성(reflect)하는 방법을 통하여 ‘좋은 수업’의 특징들을 알아보고자 하였다. 또한 예비교수자라는 대상의 특수성을 살려 비판적 시각을 갖고 수업에서의 개선점을 제안해 보도록 하였다. 좋은 수업의 특징으로는 흥미유발 활동, 학생중심수업, 모둠활동, 직접체험활동, 교사의 유능함과 열성 등의 요소들을 포함하였으며, 과목별로는 사회과와 국어과의 경우 흥미유발 활동, 그리고 과학과의 경우는 모둠별 활동을 가장 큰 특징으로 꼽았다. 개선점으로는 활동중심수업에서의 학습능률저하, 다양한 학습 자료의 부족, 학생중심수업에서의 교사역할축소, 특정학생에 편중된 활동, 활동에 앞선 사전설명의 부재 등을 언급하였다. 과목별로는 사회과와 과학과 모두 활동중심수업에서 나타날 수 있는 학습능률 저하를 가장 큰 개선점으로 지적하였다. 이것은 경험에 근거한 실제적이고 구체적인 것들로서 학교현장에 몸담고 있는 교사들에게 학습자 중심의 수업을 위한 방법적 정보를 제공할 수 있을 것이다. This study is to investigate the features of "good instruction" by the method which reflects studies experienced in elementary school days of teacher's college students being a position to both preliminary teacher and learner. A "request-for-improvements for good instruction" was also investigated with a preliminary teacher's viewpoint in order to improve their instructions. The features for "good instruction" were classified in general features and subject features which sub-classified in social studies, in Korean language studies, and in science studies. The general features were obtained in interesting-evoked activity, instruction that students take the leading role, grouping activity, direct-experiencing activity, and teaching competency and passion. The subject features in social studies and Korean language studies were identified an interesting-evoked activity, and the feature in science studies was showed a grouping activity. The general request-for-improvements for good instruction were identified a drop of learning about the lesson theme, a lack of variety learning materials, a reducing of the teacher's role in instruction which students take the leading role, an activity biased to specific student, and an insufficiency of briefing before activity. And The subject's request-for- improvements for good instruction in society studies and science studies were identified an drop of learning about the lesson theme. These empirical results are expected to propose practical and embodied information for establishing methods for good instruction to a schoolmaster.

      • 소배양법으로 배양한 생쥐췌장 베타세포의 형태학적 검정

        생쥐췌장 베타세포를 보다 순수하게 배양하기 위해 변형 고안한 소배양법을 검정하였다. 이 소배양법에 있어서는 96well plate에 islet cell을 이식하고 decantation한 다음 thimerosal에 노출시켜 많은 viable cell을 얻을 수 있었고, 생후 7일째의 생쥐의 췌장베타세포의 생존율이 가장 높았다. 또 이 소배양법으로 배양한 생쥐췌장 베타세포들은 위상차현미경, FITC-labelled anti-insulin antibody 염색 및 전자현미경으로 검정한 결과 형태학적으로 정상이었다. 기계적 선택방법으로서 생쥐췌장 베타세포의 decantation 최적시간은 세포를 이식한 후 12∼15시간 이었으며, 화학적 선택방법으로 0.6∼0.8㎍/㎖의 thimerosal에 노출시켜 fibroblastoid cell로 부터 생쥐췌장 베타세포의 분리를 증가시킬 수 있었으며, 두가지 방법을 동시에 응용하여 더 좋은 효율을 얻을 수 있었다. 또 이 소배양법에 있어서 cell duplication은 배양후 36시간후에 2배, 72시간후에 3배가 되었으며, 평균 generation시간은 46.8±7.0시간 이었다. 이들 결과로 미루어 보아 이소배양법은 생쥐췌장 베타세포 배양의 유용한 방법으로서, 췌장 베타세포에 연관된 각종 연구에 이용할 수 있을것으로 사료된다.

      • 인터넷 쇼핑몰 이용 소비자의 정보탐색량에 관한 실증연구

        This research provides a comprehensive analysis of consumer information search behavior in e-shopping. It investigates the determinants of amount for consumer information search in e-shopping in order to provide useful information on consumer decision making and consumer related marketing and public polices. Based on the review of literatures, amount of consumer information search is investigated by three different ways: (1) the number of visiting e-shopping, (2) the length of decision making time, and (3) the length of visiting time. And amount of consumer information search is assumed to be explained by several demographic variables(i.e. sex, age, income, education, marital status) and environmental variables(i.e. experience of purchase, perceived risk, types of internet mall, types of product and experience of internet). The empirical result of this study showed that amount of information search in e-shopping is explained by age, marital status, perceived risk, experience of purchase and experience of internet. The empirical results provide useful implications about consumer behavior and consumer related policies. One of the most important of these implications is the importance of providing objective and efficient information system in e-shopping. These results might also be used in teaching and in further research about consumer information behavior in e-shopping, the formulation of marketing strategies and public policies.

      • 관상 동맥 질환에서 아포 E 지단백 유전자 다형성과 혈청 지질치와의 관계

        Background and aims: The Apolipoprotein E is a ligand of both the protein component LDL receptor as well as the apo E LDL-Receptor related protein (LRP). It modulates the receptor binding of lipoproteins, with the apolipoprotein E found on cell surfaces as its component, thus serving an important role in the lipid metabolism by carrying out the intracellular transport of cholesterol in lipoproteins. The gene for apolipoprotein E is the product of three common genotypes as well as many more rare alleles. The common genotypes are ε2, ε3, and ε4, and are expressed in the three phenotype isoforms of E2, E3, and E4. In the event that E4 is the main component, a rise in the cholesterol level, as the result of down-regulation of the LDL receptor, is observed. Therefore, those samples with E4 genotypes are known to be in much higher risk of coronary artery disease than those with ε3/ε3, while those with ε2 are in low risk (with the exception of hypertiglyceremai Ⅲ). The aim of this study is to analyze in patients with ischemic heart disease the role of aplipoprotein E alleles in order to seek its correlation with coronary artery disease, as well as to seek whether the polymorphism of apo E produces any differences in the severity of coronary artery disease according to plasma lipid levels. Methods: The subjects for study were 273 patients admitted to the Internal Cardiology Division of the Soonchunhyang University Hospital form December 1998 to February 1999. The subjects were divided into the two groups of which one was ischemic heart disease (IHD) experiment group totaling 105 (avg.60.1 years of age, male/female ratio = 69/36) and the control group totaling 168 (avg. 59.7 years of age, male/female = 73:95). The coronary angiogram was given to 127 subjects, and of this total, 94 have developed significant stenosis in the coronary artery. The stages of the analyzing of the apo E phenotype was first, the separation of DNA from the blood samples, subjecting it to the PCR from with 228 base pairs of expanded products were obtained. The band was determined by means of the reverse hybridization principle on the nitrocellulose strip. Results: From the 105 patients the distributions of apo E phenotypes were as follows: ε3/2(5.7%), ε4/2(1.9%), ε3/3(70%), ε4/3(20%), ε4/4(1.9%). The relative frequencies of each allele are as follow: ε2 (0.038), ε3 (0.833), ε4 (0.128). The results show as follows: ⅰ) The IHD experiment group to have a higher occurrence of ε4/3 phenotypes as well as ε4 alleles than the control group. ⅱ) Both the control group and IHD group showed the largest distribution of ε3/3 for phenotypes, and ε4 for alleles. ⅲ) The IHD group showed less ε2/3 phenotypes as well as significantly less allele frequency of ε3 in comparison to the control group. ⅳ) the IHD group showed a much lower level of HDL in comparison to the control group, while the LDL was significantly higher; samples including the apo ε2 showed a significantly higher level of HDL than those without. Among the control group, samples including apo ε2 showed a significantly higher level of TG (triglyceride) than samples without. No significant difference was found between the experiment apo ε4 sample and the control plasma lipid sample. ⅴ) No significant correlation was found between an apo E polynorphism and the number of involved arteries of a coronary angiogram. Conclusion: Between the experiment IHD group and control group were found differences in the frequency of alleles. The polymorphism of apo E alleles may contribute as a risk factor to the development of heart disease by involving itself in the metabolism and modulation of plasma lipids.

        The Synergism between Belotecan and Cisplatin in Gastric Cancer

        Purpose: We wanted to demonstrate the anti-cancer effect and interaction between belotecan and cisplatin on gastric cancer cell line and we evaluated the mechanisms of this synergistic effect in vitro. Materials and Methods: The growth inhibitory effect of belotocan and cisplatin against several gastric cancer cell lines (SNU-5, SNU-16 and SNU-601) was estimated by tetrazolium dye assay. The effect of a combination treatment was evaluated by the isobologram method. The biochemical mechanisms for the interaction between the drugs were analyzed by measuring the formation of DNA interstrand cross-links (ICLs) and DNA topo-I activity.Results: Belotecan showed synergism with cisplatin for growth inhibitory effect on the gastric cancer cell lines SNU-5, and SNU-16, but this was subadditive on the SNU- 601 cell line. The formation of DNA ICLs in SNU-16 cells by cisplatin was increased by combination with belotecan, but this was not affected in SNU-601 cells. The topo-I inhibition by belotecan was enhanced at high concentrations of cisplatin in SNU-16, but not in SNU-601 cells.Conclusion: Belotecan and cisplatin show various combination effect against gastric cancer cells. The synergism between cisplatin and belotecan could be the result of one of the following mechanisms: the modulating effect of belotecan on the repair of cisplatin-induced DNA adducts and the enhancing effect of cisplatin on the belotecan-induced topo-I inhibitory effect.         Normal Lymphocyte Subpopulation of the Spleen is Altered after Peripheral Nerve Injury in Mice

        Background: Chronic neuropathic pain is often associated with altered immune function and the modulated immune cell response play a role in neuropathic pain by experimental nerve injury. In order to assess the possible changes in lymphocytes function following peripheral mononeuropathy, this study examined the lymphocyte subpopulation of the spleen using the monoclonal antibodies against the membrane surface markers in neuropathic BALB/c mice by a partial transection of sciatic nerve (PST). Methods: After confirming tactile allodynia by paw withdrawal threshold, the splenic lymphocytes were stained with fluorescein isothiocyanate (FITC)-conjugated anti-mouse CD45R/B220 (B cell) and CD4 (helper/inducer T cell) or with phycoerythrin (PE)-conjugated anti-mouse CD90.2 (total T cell) and CD8 (suppressor/cytotoxic T cell). The proportions of subsets were analyzed using a FACScan laser flow cytometry system on postoperative day 5 and day 18 respectively. Results: PST induced a mechanical allodynia as verified by the von Frey test at both 5 and 8 days postoperatively compared to pre-surgery (P < 0.05). Lymphocyte subpopulation was affected by PST. The proportion of CD4+ subset was significantly larger in the PST group than in the sham operated group on day 5, while the proportion of CD8+ subset was larger on day 18. In the PST group, there were significantchanges in the proportion of CD4+ on day 5 and in the proportion of CD8+ on day 18 (P < 0.05) compared to pre-surgery. There were no significant fluctuations in the proportion of total splenic T cell and B cell subsets of PST group compared to sham operated group. Conclusions: These results suggest that development of mononeuropathy is responsible for the proportional changes in splenic lymphocyte subsets in mice. (Korean J Anesthesiol 2007; 53: S 42∼7)

        Significance of CD133 as a cancer stem cell markers focusing on the tumorigenicity of pancreatic cancer cell lines

        Purpose: The cancer stem cell hypothesis states that the capacity of a cancer to grow and propagate is dependent on a small subset of cells. To determine the significances of the cancer stem cell markers CD133, CD44, and CD24 using a comparative analysis with a focus on tumorigenicity. Methods: Four pancreatic cancer cell lines, Capan-1, Mia-PACA-2, Panc-1, and SNU-410 were analyzed for the expressions of CD133, CD44, and CD24 by flow cytometry. The tumorigenicity was compared using tumor volumes and numbers of tumors formed/numbers of injection in nonobese diabetic severe combined deficiency mice. Fluorescence-activated cell sorting (FACS) analysis was used to confirm that xenograft explants originated from human pancreatic cancer cells. Results: CD133 was positive in only Capan-1, CD44 positive in all, CD24 partially positive in Panc-1. After injecting 2 × 106 cells, all mice administered Capan-1 or Mia-Paca-2 developed tumors, 3 of 5 administered Panc-1 developed tumors, but no mouse administered SNU-410 developed any tumors. The volumes of Capan-1 tumors were seven times larger than those of Mia-Paca-2 tumors. When 2 × 105 or 2 × 104 of Capan-1 or Mia-Paca-2 was injected, tumors developed in all Capan-1 treated mice, but not in Mia-Paca-2 treated mice. Furthermore, xenograft explants of Capan-1 expressed CD133+CD44+ and Capan-1 injected mice developed lung metastasis. FACS analysis showed that xenograft explants originated from human pancreatic cancer cell lines. Conclusion: CD133 positive cells have higher tumorigenic and metastatic potential than CD44 and CD24 positive cells, which suggests that CD133 might be a meaningful cell surface marker of pancreatic cancer stem cells.
