Novel Methylation Biomarker for Non-invasive Diagnostics in Lung Cancer

To identify aberrantly hypermethylated DNA in lung cancer cells we established a genome-wide analysis for hypermethylation sites, namely Methyl DNA Isolation and Amplification (MeDIA) coupled-CpG microarray analysis. In the comprehensive methyaltion profiling analysis between human lung cancer, A549 cells and normal NHBE cells, we observed that several clusters of genes show a significant level of aberrancy in CpG island methylation pattern in cancer cells compared to normal cells. We further identified PCDHGA12 gene as a new marker of non-invasive diagnostics for lung cancer based on followings. 1) Transcription of PCDHGA12 gene is reactivated after treatment of A549 cells with demethylating agent. 2) Bisulfide clonal-sequencing reveals that CpG island of PCDHGA12 shows a distinctive differential methylation between two cell lines. 3) Pyrosequencing-based quantitative methylation assay for such region in tumor and non-tumorous tissues from lung cancer patients shows aberrant hypermethylation in 37 (92%) of the 40 tumor tissues. In clinical validation by pyrosequencingin induced-sputum of lung cancer patients (n=87) and healthy controls (n=51), we observed aberrant hypermethylation incident at significantly elevated level in samples derived from lung cancer patients. According to the optimal threshold calculated by ROC curve analysis, sensitivity and specificity of PCDHGA12 was 86.2% and 82.4%, respectively. PCDHGA12 methylation status could be a potential methylation biomarker alone or combined with others for the screen and the detection of relapse of lung cancer.
한국 재래식 된장과 고추장의 숙성 중 미생물, 효소활성 및 주요 성분의 변화

재래식 된장과 고추장의 가스 발생을 억제하여 저장성을 향상시키기 위한 생물공학적 방법을 개발하고자 먼저 재래식 된장(봄메주 된장과 가을메주 된장)과 고추장(순천식)을 제조하여 4개월간 숙성시키면서 미생물과 효소활성 및 주요 성분의 변화를 조사하였다. 호기성 세균은 된장과 고추장 모두 숙성 60일까지 증가한 후 큰 변화가 없었고 혐기성 세균은 숙성기간 동안 된장에서는 10^7~10^8/g, 고추장에서는 10^5~10^6/g이었으며 효모는 된장에서는 숙성 15일까지 급격히 증가한 후 변화가 없었으나 고추장에서는 숙성 15일에 출현하여 60일까지 10^6/g으로 증가하였다. α-amylase 활성은 가을메주 된장에서, glucoamylase는 고추장에서 숙성기간 전반에 걸쳐 제일 높았고 산성 protease와 중성 protease 활성은 대체로 숙성 15일~30일경에 최대를 보였다. pH는 된장에서는 숙성 60일까지 7.0까지 높아졌지만 고추장은 숙성기간 내내 pH 5.0으로 낮아졌다. 수분은 봄메주 된장과 고추장에서 숙성이 진행됨에 따라 낮아져서 숙성 120일 후 40%이었으며 환원당은 고추장에서 숙성 15일까지 19.9%로 증가한 후 감소하는 경향이었다. Changes in microorganisms, enzyme activities and major components of two types of Doenjang prepared with spring Meju and autumn Meju and Kochujang were investigated during 5 months of fermentation. The viable cell counts of aerobic bacteria in Doenjang and Kochujang were increased up to 60 days of fermentation, but viable cell counts of anaerobic bacteria did not show remarkable changes during fermentation. Viable cell count of yeast showed a rapid increase up to 15 days of fermentation in Doenjang and 60 days in Kochujang. It was found that α-amylase activity of autumn Meju Doenjang and glucoamylase activity of Kochujang were higher than the other. Acidic and neutral protease showed the highest activity during 15~30 days of fermentation. The pH of Doenjang was increased up to pH 7.0 until 60 days of fermentation, but pH of Kochujang gradually decreased during fermentation. Moisture content of spring Meju Doenjang and Kochujang decreased to 40% during fermentation and reducing sugar content of Kochujang increased up to 15 days of fermentation, but decreased after that.
자발성 뇌간 혈종(출혈) 5예 : Especially in Midbrain, in Pons, and in Ponto-mesencephalon

자발성 뇌간 혈종은 흔하지 않으며 특히 중뇌에 생긴 자발성 혈종은 대단히 드물다. 뇌간에 생긴 혈종은 생긴 장소와 크기에 따라서 다양한 신경학적 증상과 vital sign의 변화를 보인다. ocular sign 및 vital sign의 변화와 의식상태 등에 의해서 뇌간 혈종의 위치를 식별할 수도 있다. 뇌간 혈종이 있는 환자를 치료하는 중에 환자의 의식상태는 혈압과 밀접한 관계를 보이며 수축기 혈압이 140mmHg이거나 이보다 약간 높은 상태에서 비교적 안정될 때는 환자의 의식이 호전되었고 강하시에는 의식의 악화를 보였으며 또한 혈압에 따라 이외의 신경학적 증상의 변화를 보였다. 본 교실에서는 Brain CT에 의해서 자발성 뇌간 혈종 5예, 즉 중뇌 혈종 1예, 뇌교 혈종 2예 및 뇌교-중뇌 혈종 2예를 경험하였다. Spontaneous brain stem hematoma is not common, especially spontaneous midbrain hematoma is extremely rare. Brain stem hematoma leads to variable neurological deficits and wide variations of vital signs correspending to its location and volume. It is possible to localize the hematoma in the brain stem by means of changes of ocular signs and vital signs, and the level of consciousness. During treatment of the patients with brain stem hematomas we found that the level of patient's consciousness was closely related to the level of blood pressure. At or slightly above 140mmHg of systolic pressure the patient's consciousness was markedly improved, but the decreased systolic pressure caused its deterioration. Also according to level of blood pressure other neurological findings were slightly variable. We hove experienced five cases of spontaneous brain stem hematoma which were diagnosed with brain CT. They were a case of midbrain hematoma, two cases of pontine hematoma and two cases of pontomesencephalic hematoma.

Reduction and sulfidation reactivities of mixed metal oxides were investigated by using a TGA. Mixed metal oxides consisted of CuO as the main active material, SiO₂ as the support material and both MoO₃ and Fe₂O₃ as the additives. Each material was chosen based on the literature survey. CS (CuO/SiO₂) sorbents were prepared to investigate the effect of support material, and consisted of various composition of CuO and SiO₂. CMS (CuO-MoO₃/SiO₂) and CFS (CuO-Fe₂O₃/SiO₂) sorbents were prepared to investigate the effect of additives and consisted of various composition of CuO and additive. In CMS and CFS sorbents, a support material was 25wt% SiO₂ because the previous study showed that 25wt% SiO₂ was needed to maintain the activity in multicycle experiment. Simple physical mixing method was chosen as the sorbent preparation method. Chatterjee-Conrad method was used to evaluate activation energies of sorbents from TGA data. The activation energy of CS sorbents decreased in reduction reaction and increased in sulfidation reaction as SiO₂ content increased. Fe₂O₃ was more favorable than MoO₃ as an additive material in terms of reduction reaction rate.
