Insertion of reporter constructs into the mammalian genome leads to variable gene expression due to position effects at the site of integration. This random integration has limited the gene therapy of human genetic disorders by its undesirable effects. We report here the newly constructed plasmid vector(pIRES-neo-YJ) based on the concepts of homologous recombination and position-independent promoter enhancing of beta-globin matrix attachment region(Glb-MAR). chromosome 7 centromere-specific alpha satellite(alphoid) DNA sequence was cloned into pIRES-neo-YJ for homologous recombination of the cloned gene with the centromeric region of chromosome 7, which is genetically silent. Beta Glb-MAR sequence that allows high levels of transcription independent of the chromosomal site of integration was also inserted into pIRES-neo-YJ to ensure the stable and higher expression of the cloned genes. We expect that pIRES-neo-YJ would provide a valuable tool to eliminate random integration of cloned genes into the undesirable chromosomal region and their short-lived expression which often encounters during the construction of transgenic animals and human gene therapy.

In order to develop the functional muffin prepared with different levels of sprouted brown rice flour (SBRF), the mixing conditions of SBRF, butter and water were optimized by response surface methodology(RSM), using central composite design with 3 variables and 5 levels. The result of three-dimensional plot was showed that decrease of SBRF content increased the volume, maximum length, crumb grain, moistness and overall quality of muffin. The overall quality of the functional muffin was maximized under the preparing condition of 31.49 g SBRF(15% of flour used), 85.36 g butter, and 123.86 g water. Moreover, the optimum conditions for preparing muffin were as follow : 29.95 g, 81,24 g, 120.45 g for volume ; 23.67 g, 88.18 g, 118.24 g for maximum length ; 38.44 g, 63.38 g, 117.26 g for flavor ; 29.86 g, 93.04 g, 126.42 g for taste ; and 55.43 g, 94.40 g, 133.61 g for moistness. The range of optimum preparing conditions for the functional muffin in consideration of the physical and sensory characteristics were shown to the 30~40 g as SBRF, 80~90 g as butter, and 120~130 g as water.
NIHSS 활용을 통한 뇌졸중 환자 간호의 질향상 전략

문제: 뇌졸중 환자를 돌보는 간호현장에서는 급성기 뇌졸중 환자의 증상악화에 대한 조기사정과 적절한 대처가 매우 중요한 문제이다. 그러나 임상현장에서는 이를 객관화하고 수치화하여 모든 의료진이 의사소통할 수 있는 유용한 도구를 사용하고 있지는 못하는 실정이다. 이러한 문제를 해결하기위해 NIHSS(National Institute of Health Stroke Scale)를 신경과 진료의사를 중심으로활용하고 있지만 아직 간호현장에서는보편적으로사용하지 않고있다. 이에 뇌졸중환자간호에 NIHSS를 활용하여 뇌졸중 증상악화의 조기사정과 빠른 대처로 궁극적으로는 뇌졸중환자 간호의 질을보다 향상시키기 위하여 본활동이 시도되었다. 목적: 첫째, 간호사의 NIHSS 측정 신뢰도의 현수준을 점검하고 이를향상시키기 위한 프로그램을 개발·적용한다. 둘째, 간호사가급성기 뇌졸중환자에게 NIHSS 측정을 통하여 뇌졸중악화를 얼마나 예측할 수 있는지 분석하고 이를 향상시킬수있는 방법을 모색한다. 셋째, 궁극적으로 급성기 뇌졸중 환자의 증상악화의 조기 발견과치료를위한의료진의 올바른 대처지침을 마련하여 이를 적용한다. 의료기관: 경기도에 소재한 대학병원의 뇌졸중 집중치료실 질 향상 활동: 첫째, 간호사의 NIHSS 측정 신뢰도 향상을 위해 「NIHSS 간호사 교육 프로그램」을 마련하였다.
사육수의 pH변화가 숭어(Mugil cephalus)에 미치는 생리적 영향

We examined changes in the physiological responses of gray mullet Mugil cephalus exposed to acidic seawater (pH 6.0, 6.5, 7.0) and normal seawater (pH 8.0, control) for 15 days. As pH decreased, survival rate and body weight also decreased. Levels of aminotransferase, total protein and triglycerides also differed significantly with changes in pH, presumably due to stress caused by exposure to acidic water. The level of osmotic pressure was significantly higher in the pH 6.0 group than in other groups. Superoxide dismutase was significantly higher in the pH 6.5 and 7.0 groups than in the pH 8.0 group, and glutathione level was lowest in the pH 6.0 group. We conclude that decreasing the pH level of seawater induces a stress response in fish, damaging their ability to control their hematological and osmotic pressure. Antioxidant enzymes are generally sensitive to osmotic stress; in this study, antioxidant activity significantly changed with pH level. These results indicate that physiological stress induced by exposure to acidification reduces survival rates and inhibits growth in M. cephalus.
CaO (oxide calcium)가 첨가된 사료 공급이 넙치 (Paralichthys olivaceus)에 미치는 생리적 변화에 대한 연구

This study investigated the physiological effects of a diet containing calcium oxide (CaO) on the olive flounder, Paralichthys olivaceus. The results indicate that the amount of calcium in the blood was higher in the calcium group compared with the controls. The aspartate amino transferase (AST) and alanine amino transferase (ALT) levels were lower in the calcium group, while lysozyme activity was higher in the calcium group. Histologically, fish in the cal-cium group had more hepatocytes in the liver and more intact scales. In addition, an attack experiment using Vibrio anguillarum (KCTC-2711) and Edwardsiella tarda (KCTC-3657) showed that the survival was higher in the calcium group compared with the controls. These results suggest that feeding fish CaO when they require it, such as during periods of gonadal maturation and rapid growth, will increase their immunity and resistance to pathogens.

A method for the determination of anionic drug, mefenamate with ion-selective electrode using Fe(II)-1,10-phenanthroline chelate as a counter ion was developed. Benzyl nitrophenyl ether (BNPE) plasticized membrane was more selective and sensitive than the other tested membranes. This membrane electrode exhibits a linear response for 10-2 M~5 ×10-5 M of mefenamic acid with a slope of - 61.4mV/dec. in borate buffer sloutions (Ph 9.0). Potentiometric selectivity measurements revealed negligible interferences from various organic and ionorganic anions. Direct potentiometry and potentiometric titration method of mefenamic acid in capsule preparations are presented and compared.
