Background: Genetic alterations have been identified in melanomas according to different levels of sun exposure. Whereas the conventional morphology-based classification provides a clue for tumor growth and prognosis, the new classification by genetic alterations offers a basis for targeted therapy. Objective: The purpose of this study is to demonstrate the biological behavior of melanoma subtypes and compare the two classifications in the Korean population. Methods: A retrospective chart review was performed on patients found to have malignant melanoma in Severance Hospital from 2005 to 2012. Age, sex, location of the tumor, histologic subtype, tumor depth, ulceration, lymph node invasion, visceral organ metastasis, and overall survival were evaluated. Results: Of the 206 cases, the most common type was acral melanoma (n=94, 45.6%), followed by nonchronic sun damage-induced melanoma (n=43, 20.9%), and mucosal melanoma (n=40, 19.4%). Twenty-one patients (10.2%) had the chronic sun-damaged type, whereas eight patients (3.9%) had tumors of unknown primary origin. Lentigo maligna melanoma was newly classified as the chronic sun-damaged type, and acral lentiginous melanoma as the acral type. More than half of the superficial spreading melanomas were newly grouped as nonchronic sun-damaged melanomas, whereas nodular melanoma was rather evenly distributed. Conclusion: The distribution of melanomas was largely similar in both the morphology-based and sun exposure-based classifications, and in both classifications, mucosal melanoma had the worst 5-year survival owing to its tumor thickness and advanced stage at the time of diagnosis. Web-Shear Capacity of Thick Precast Prestressed Hollow-Core Slab Units Produced by Extrusion Method

The prestressed hollow-core slab (PHCS) is a precast concrete member that can maximize productivity and structural performance efficiency of concrete cross-section. For the PHCS members produced by extrusion method, however, it is difficult to provide the shear reinforcement due to its unique production method to form the hollow-cores in concrete section. The recently revised ACI318 Building Code Requirements stipulate that web-shear capacity of thick hollow-core member over 315 mm depth without the minimum shear reinforcement should be reduced in half, which may result in an excessively conservative shear design for the PHCS member compared to conventional prestressed concrete members with thin webs. In this study, shear tests on PHCS members, which have typical thicknesses widely used in precast construction industry, were conducted, and a large number of shear test data were additionally collected from previous studies to evaluate the current shear design criteria for PHCS in detail.
Two Lignans from the Stem Bark of Styrax japonica

Two lignans, 1R,2R,5S,6R-2-(4'-hydroxy-3'-methoxyphenyl)-6-(3'',4''-dimethoxyphenyl)-3,7-dioxabicyclo[3.3.0]octane 4'-O-${\beta}$-D-glucopyranoside (1) and styraxlignolides F (2), were isolated from the EtOAc-soluble fraction of the stem bark of Styrax japonica (Styracaceae). The structures of lignans (1-2) were determined by spectroscopic means. The absolute configuration for styraxlignolides F (2), (2S,3S)-2-(3'',4''-dimethoxybenzyl)-3-(4'-hydroxy-3'-methoxybenzyl)-${\gamma}$-butyrolactone 4'-O-(${\beta}$-D-glucopyranoside), was unambiguously confirmed by X-ray diffraction analysis.

The Gold-spotted pond frog, Rana plancyi chosenica, designated as a vulnerable species by IUCN Red list. This species is a typical example facing local population threats and extinction due to human activities in South Korea. A strategic conservation plan for this endangered species is urgently needed. In order to provide information for future conservation planning, accurate information on the genetic diversity and taxonomic status is needed for the establishment of conservation units for this species. In this study, we used a molecular genetic approach using the mitochondrial cytochrome b gene and control region sequences to find the genetic diversity of gold-spotted pond frogs within South Korea. We sequenced the mitochondrial DNA cytochrome b gene and control region of 77 individuals from 11 populations in South Korea, and one from Chongqing, China. A total of 15 cytochrome b gene haplotypes and 34 control region haplotypes were identified from Korean gold-spotted pond frogs. Mean sequence diversity among Korean gold-spotted pond frogs was 0.31% (0.0-0.8%) and 0.51% (0.0-1.0%), respectively. Most Korean populations had at least one unique haplotype for each locus. The Taean, Ansan and Cheongwon populations had no haplotypes shared with other populations. There was a sequence divergence between Korean and Chinese gold-spotted pond frogs (1.3% for cyt b; 2.9% for control region). Analysis of genetic distances and phylogenetic trees based on both cytochrome b and control region sequences indicate that the Korean gold-spotted pond frog are genetically differentiated from those in China.
Acetylcholinesterase Inhibitory Effect of Lignans Isolated from Schizandra chinensis

The hexane extract of the fruit of Schizandra chinensis (Schisandraceae) was found to show significant inhibition of the activity of acetylcholinesterase enzyme (AChE). In further studies, fourteen lignans were isolated, and evaluated for their inhibitory effect on AChE. The compounds having both aromatic methylenedioxy and hydroxyl groups on their cyclooctadiene ring, such as gomisin C (6), gomisin G (7), gomisin D (8), schisandrol B (11) and gomisin A(13), entirely inhibited AChE in dose dependent manners, with IC$_{50}$ values of 6.71 ${\pm}$ 0.53, 6.55 ${\pm}$ 0.31, 7.84 ${\pm}$ 0.62, 12.57 ${\pm}$ 1.07 and 13.28 ${\pm}$ 1.68 ${\mu}$M, respectively. These results indicate that the lignans could potentially be a potent class of AChE inhibitors.

<P><B>Abstract</B></P> <P>Seed aging is one of the major events, affecting the overall quality of agricultural seeds. To analyze the effect of seed aging, soybean seeds were exposed to controlled deterioration treatment (CDT) for 3 and 7days, followed by their physiological, biochemical, and proteomic analyses. Seed proteins were subjected to protamine sulfate precipitation for the enrichment of low-abundance proteins and utilized for proteome analysis. A total of 14 differential proteins were identified on 2-DE, whereas label-free quantification resulted in the identification of 1626 non-redundant proteins. Of these identified proteins, 146 showed significant changes in protein abundance, where 5 and 141 had increased and decreased abundances, respectively while 352 proteins were completely degraded during CDT. Gene ontology and KEGG analyses suggested the association of differential proteins with primary metabolism, ROS detoxification, translation elongation and initiation, protein folding, and proteolysis, where most, if not all, had decreased abundance during CDT. Western blotting confirmed reduced level of antioxidant enzymes (DHAR, APx1, MDAR, and SOD) upon CDT. This in-depth integrated study reveals a major downshift in seed metabolism upon CDT. Reported data here serve as a resource for its exploitation to metabolic engineering of seeds for multiple purposes, including increased seed viability, vigor, and quality.</P> <P><B>Biological significance</B></P> <P>Controlled deterioration treatment (CDT) is one of the major events that negatively affects the quality and nutrient composition of agricultural seeds.
