Antibacterial and Antibiofilm Activities of Weissella Cibaria CHK903 Against Proteus mirabilis

This study aimed to identify and characterize lactic acid bacteria strains with antimicrobial activity against Proteus mirabilis, a gram-negative bacterium associated with Parkinson’s disease, Crohn’s disease, and nosocomial infections. Among the 987 lactic acid bacteria strains isolated from various sources, strain no. CHK903 showed high antimicrobial activity against P. mirabilis. Phylogenetic tree analysis based on the 16S rRNA gene and scanning electron microscope analysis identified the selected strain as the rod-shaped Weissella cibaria. The culture supernatants of W. cibaria CHK903 showed antimicrobial activity against some pathogens. Two antimicrobial compounds with molecular weights 189 and 365 Da were partially purified from the culture supernatants of W. cibaria CHK903 using Biogel P2 gel permeation column chromatography. The culture supernatants of W. cibaria CHK903 also showed significant antibiofilm properties, inhibiting biofilm formation by 90% and removing pre-formed biofilms by 60%. These findings suggest the potential therapeutic use of W. cibaria CHK903 as a natural antimicrobial against P. mirabilis-related infections.
알코올로 유도된 간손상에 대한 감꼭지 메탄올 추출물의 보호효과

본 연구에서는 감꼭지 추출물이 알코올 자극을 받은 HepG2 세포의 보호효과와 ROS 생성량, MDA 및 GSH 함량에 미치는 영향을 살펴보았고, 감꼭지 추출물의 페놀산 및 플라보노이드 함량을 LC-MS/MS로 정량 분석하였다. 감꼭지 추출물은 HepG2 세포에서 10 μg/mL 농도까지 우려할만한 세포독성을 나타내지 않았고, 알코올 처리군에 비해 농도 비례하여 세포생존율을 증가시켰다. 감꼭지 추출물은 1, 5, 10 μg/mL 모든 농도에서 ROS의 생성을 억제하였다. 또한, 감꼭지 추출물은 모든 농도에서 세포의 MDA 생성을 억제하였는데 양성 대조군인 100 μM 농도의 esculetin보다 효과적으로 MDA 생성을 억제하였다. GSH 함량 분석 결과 감꼭지 추출물은 GSH를 증가시켰다. 감꼭지 추출물의 정량분석 결과에서 gallic acid 629.66±32.41 μg/g, catechin 1,894.89±105.10 μg/g, kaempferol 371.88±30.61 μg/g, quercetin 230.03±19.21 μg/g, rutin 167.98±42.15 μg/g 등 다양한 페놀산 및 플라보노이드가 검출되었으며, 이들이 감꼭지 추출물의 주요 간세포 보호 활성에 기여하는 성분일 가능성이 있다. 이러한 결과들은 감꼭지 추출물이 알코올로 유도된 산화적 스트레스부터 간세포를 보호할 수 있는 기능성 소재로 활용될 수 있음을 시사한다. In this study, the calyx of Diospyros kaki was evaluated for antioxidative activity using HepG2 cells treated with alcohol-induced oxidative stress and quantitatively analyzed for phenolic acids and flavonoid concentration by LC-MS/MS. The extract of the calyx of Diospyros kaki showed no significant toxicity to HepG2 cells till a concentration of 10 μg/mL was reached and displayed increased cell viability compared to the 3% ethanol-treated group. The extract inhibited the production of reactive oxygen species at all concentrations. Besides, the extract inhibited malondialdehyde production at all concentrations and the inhibitory effect was higher than the positive control (esculetin 100 μM). Also, the extract increased the concentration of glutathione. The major phenolic acids and flavonoid in the extract were vanillic acid (729.01±42.48 μg/g) and catechin (1,894.89±105.10 μg/g), respectively. This study shows that the calyx of Diospyros kaki can be used as a functional material that exhibits antioxidative and hepatoprotective effects.
Comparison of conventional lateral cephalograms with corresponding CBCT radiographs

Purpose: This study was performed to assess the compatibility of cone beam computed tomography (CBCT) synthesized cephalograms with conventional cephalograms, and to find a method for obtaining normative values for threedimensional (3D) assessments. Materials and Methods: The sample group consisted of 10 adults with normal occlusion and well-balanced faces. They were imaged using conventional and CBCT cephalograms. The CBCT cephalograms were synthesized from the CBCT data using OnDemand 3D software. Twenty-one angular and 12 linear measurements from each imaging modality were compared and analyzed using paired-t test. Results: The linear measurements between the two imaging modalities were not statistically different (p¤0.05) except for the U1 to facial plane distance. The angular measurements between the two imaging modalities were not statistically different (p¤0.05) with the exception of the gonial angle, ANB difference, and facial convexity. Conclusion: Two-dimensional cephalometric norms could be readily used for 3D quantitative assessment, if corrected for lateral cephalogram distortion.
