      • Thiocholine ester 기질을 이용한 Acetylcholinesterase 활성부위의 구조특성

        The inhibition pattern of three inhibitors(tacrine, decamethonium and propidium) on the hydrolysis of various thiocholine ester substrates by eel acetylcholinesterase was comparatively examined. When the substrate was acetylthiocholine, it showed a similar competitive inhibition by tacrine inhibitor, and a mixed type inhibition by decamethonium and propidium inhibitors. When the substrate was pentanoylthiocholine, it showed an uncompetitive inhibition by tacrine, and a noncompetitive inhibition by decamethonium. When the substrate was laurylthiocholine, it showed mixed type and uncompetitive inhibition by tacrine, and a competitive inhibition by decamethonium and propidium. Those results suggest that the active of acetylcholinesterase has the existence of hydorphobic site besides the anionic and esteratic site.

        A new ferulic acid ester, 1'-methyl-2'-hydroxyethyl ferulate (1), together with methylcaffeate (2), 4-hydroxy cinnamic acid (3), ferulic acid (4), caffeic acid (5), diosmetin (6), luteolin (7), 5,3',4'-trihydroxy-3,7-dimethoxyflavone (8), eriodictyol (9), kaempferol (10), quercetin (11), acacetin-7-O-glcopyranoside (12), 4-($\beta$-glucopyranosyloxy) benzoic acid (13), luteolin-7-O-(6"-feruloyl) glucopyranoside (14), luteolin-7-O-glucopyranoside (15), kaempferide-3-O-rhamnopyranoside (16), quercitrin (17), kaempferol-3-O-glucopyranoside (18), prunasin (19), quercetin-7-O-glucopyranoside (20), quercetin-3-O-glucopyranoside (21), plantaginin (22), linarin (23), luteolin-7-O-rutinoside (24), and chlorogenic acid (25) were isolated from the aerial parts of Dacocephalum peregrinum. The structure of 1 was elucidated on the basis of spectroscopic and HR-ESI-MS analyses. In addition, compound 1 exhibited mild inhibitory effect on NO production in LPS-stimulated RAW264.7 cells.

        Activated carbon supported VN, Mo2N, and W2N as catalysts for acetylene hydrochlorination

        The development of green catalysts for the calcium carbide acetylene method synthesis of vinyl chloridemonomer (VCM), is essential material to industrial polyvinyl chloride production. The original catalystHgCl2 easy to volatile during the reaction lead to environmental pollution. Here we present threedifferent transition metal nitride load into activated carbon (AC): VN/AC, Mo2N/AC, and W2N/AC as novelcatalysts. W2N/AC and Mo2N/AC was a relatively stable catalyst to compared with VN/AC. All of themVCM selectivity can maintain 98%. More HCl ratio for Mo2N/AC lead its acetylene conversion reach 80%. The former two catalysts are promising candidates for acetylene hydrochlorination.

        Construction of Novel Bifunctional Chimeric Proteins Possessing Antitumor and Thrombolytic Activities

        Based on their respective antitumor and thrombolytic activities, the superantigen staphylococcal enterotoxin C2 (SEC2) and staphylokinase (Sak) were chosen for the construction of the novel chimeric proteins Sak-linker- SEC2 and SEC2-linker-Sak using a linker composed of nine Ala residues. Both chimeric proteins possessed nearly the same PBMC proliferation stimulating activity and antitumor activity as SEC2 and thrombolytic activity as Sak. Neither the SEC2 or Sak component of each chimeric protein affected the activity of the other component. The results presented in this study provide a possible strategy to prevent and cure tumor thrombus.

        중국 농촌 체험관광 요인이 농,특산물 구매 및 재방문의도에 미치는 영향

        최근 들어 많은 중국 관광객들이 여행목적지로 농촌을 선택하고 다양한 농촌관광활동이 일어나고 있다. 농어촌 마을의 전통문화·경관·어메니티 등을 관광객에게 휴식과 레크리에이션·체험·교육을 제공함과 동시에 농산물, 먹을거리 판매 및 숙박 제공 등을 통하여 농촌경제의 활성화에 기여한다. 따라서 농촌 특산물의 구매에 어떠한 체험요인들이 영향을 미치는 연구하는 것은 관광객이 다시 찾는 농촌, 지속적인 거래를 희망하는 농산물을 어떻게 구입하는지 그 행위에 관한 분석이 필요하다. 본 연구에서는 농촌체험관광을 통해 관광객들은 농촌의 자원요소와 농촌의 생활, 체험 등에 얼마나 만족하고, 체험과 농촌 특산물 안전성의 신뢰를 바탕으로 안심하고 먹을 수 있는 농촌 특산물 구매와 재방문 의도에 어떠한 영향을 미치는지를 실증적으로 분석하고자 한다. 실증 분석을 통해 본 연구에서 제시한 연구문제를 검증한 결과는 다음과 같다. 농촌체험관광은 ‘농·특산물의 특성``, ‘교육성과 프로그램’, ‘휴양·일탈성’, ‘시설편리성’, ‘문화·자연환경’ 등의 다섯 요인으로 도출되었다. 농촌 체험관광의 “농·특산물의 특성”과 “교육성과 프로그램”요인들이 농·특산물구매에 유의한 정(+)의 영향을 미치는 것으로 나타났다. 따라서 농산물의 안전성에 대한 소비자의 우려와 관심이 높아지고 있는 시점에서 농산물생산과정의 신뢰성, 우수한 맛 등에 대한 정보를 소비자에게 효과적으로 전달하는 것이 필요하다. 농촌관광객이 지역의 농·특산물을 경험할 수 있는 다양한 프로그램을 개발하고 홍보하여야 하며, 생산자가 소비자와의 관계를 구축할 수 있도록 하는 정책적 지원이 필요하다. 농촌 체험관광의 “농·특산물의 특성”, “교육성과 프로그램”과 “휴양·일탈성”요인들이 재방문의도에 유의한 정(+)의 영향을 미치는 것으로 나타났다. 이 결과를 바탕으로 볼 때 본 연구는 농촌체험 관광활동을 활성화시키기 위해 농촌의 생태자원, 생활 및 생업 자원을 활용한 차별화되고 고급화된 다양한 체험프로그램을 개발하고 방문객들의 특성을 고려한 마케팅 전략을 수행하여 안정적인 수익창출을 위한 기반을 조성하여야 할 것이다. Recently, rural tourism is booming in China since Chinese tourists have increasingly visited rural areas for vacation. Rural tourism provides various opportunities for urban residents to experience traditional culture, landscape, and environment of rural villages and to relax and learn. At the same time, rural tourism contributes to local economy by selling their special agricultural products and food and providing accomodations to tourists. Hence, it is important to identify the major factors of tourist experience that influence the purchase of special agricultural products of the village because it resulted in sustainable development of rural tourism. Therefore, this study aims to examine rural tourist experiences and how these experiences influence their intentions to purchase special agricultural products of the village and to revisit there. The results show that the rural tourist experience consists of the five main factors: "characteristics of special agricultural products", "education & program", "recreation & excape", "conveniences", and "cultural and natural environments". Among these factors, two factors, namely "characteristics of special agricultural products" and "education & program" positively influence responedents`` intention to purchase special agricultural products. This implies that it is necessary to inform consumers about the trustworthiness of the products as consumers have increasingly paid more attention to the safety of special agricultural products. Moreover, it is also needed to develop and promote various programs through which consumers can experience the special agricultural products and to support the relationship between manufactures and customers. Also, respondents`` intention to revisit the rurla village is positively influenced by three factors, namely "characteristics of special agricultural products", "education & program", and "recreation & excape". This indicates that rural villages should develop distinguished and advanced experiencce programs and perform effective marketing strategies considering characteristics of visitors in order to make stable profits.

        Causes and classification of male infertility in Korea

        Objective: The aim of this study is to investigate the various causes of male infertility using multiple approaches. Methods: Nine-hundred-twenty infertile male patients were analyzed at their first visit with one physician between January 1 and December 31, 2009. All patients were subjected to physical examination and semen analysis and azoospermic patients underwent hormonal testing, chromosomal tests, and testicular biopsy. Semen analysis was based on the definition of the World Health Organization. Results: Among the 920 patients, 555 patients (60.3%) had semen results within the normal range, 269 patients (29.2%) within the abnormal range, and 96 (10.5%) were diagnosed with azoospermia. Varicoceles were diagnosed in 84 of the 555 normal-range patients (15.1%) and in 113 of the 269 abnormal-range patients (42.0%). Of the 96 patients with azoospermia, 24 patients (25%) were diagnosed with obstructive azoospermia, 68 patients (71%) with non-obstructive azoospermia, and 4 patients (4%) with retrograde ejaculation. Conclusion: Various causes of male infertility have been reported and diverse treatment methods can be adopted for each cause. In this regard, research must be conducted on a larger number of patients to accurately assess the various causes of infertility in Korean patients and to investigate various infertility treatment methods.

        Antioxidant Properties of Proanthocyanidins Attenuate Carbon Tetrachloride (CCl4)–Induced Steatosis and Liver Injury in Rats via CYP2E1 Regulation

        Liver steatosis is characterized by lipid dysregulation and fat accumulation in the liver and can lead tooxidative stress in liver. Since proanthocyanidins are present in plant-based foods and have powerful antioxidant properties,we investigated whether proanthocyanidins can prevent oxidative stress and subsequent liver injury. Carbon tetrachloride(CCl4) treatment can cause steatosis in rats that models both alcoholic and non-alcoholic fatty liver disease in humans. Wepre-treated rats by oral administration of proanthocyanidins extracted from grape seeds 7 days prior to intragastricallyadministering CCl4. Proanthocyanidin treatment continued for an additional 2 weeks, after which time liver and serum were harvested, and mediators of liver injury, oxidative stress, and histological features were evaluated. CCl4-treated rats exhibited significant increases in the following parameters as compared to non-treated rats: fat droplets in the liver, liver injury (ALT, AST), and DNA damage (8-OHdG). Additionally, CCl4 treatment decreased antioxidant enzymes SOD, GSH, GPX, and CAT in the liver due to their rapid depletion after battling against oxidative stress. Compared to CCl4-treated rats, treatment with proanthocyanidins effectively suppressed lipid accumulation, liver injury, DNA damage, as well as restored antioxidant enzyme levels. Further investigation revealed that proanthocyanidins treatment also inhibited expression of CYP2E1 in liver, which prevented the initial step of generating free radicals from CCl4. The data presented here show that treatment with orally administered proanthocyanidins prevented liver injury in the CCl4-induced steatosis model, likely through exerting antioxidant actions to suppress oxidative stress and inhibiting the free radical–generating CYP2E1 enzyme.
