      • 白花蛇舌草 메탄올 抽出物의 抗腫瘍 效果 및 抗癌 棋戰에 關한 硏究

        Objectives: This experimental study was carried out to evaluate the effects of aqueous and methanol extracts of Hedyotis diffusa which has long been used for cancer treatment in oriental medicines on the induction of apoptotic cell death in human lymphoid leukemia cell line, HL-60. Methods: Cells were treated with various concentrations (200 to 0.4㎍) and periods (6 to 30 hr) of H_2O and methanol extracts of Hedyotis diffusa. Then, cells were tested for viability by MTT assay. Cells wrere treated with 200㎍/ml of methanol extract fork various periods. Genomic DNA was isolated, separated, on 1.5% agarose gels, stained with ethidium bromide and visualized under UV light. Cells were treated with 200㎍/ml of each extract for 16hr.Then, cells were treated with various doses of each extract for 12 hr and 100㎍/ml of methanol extract for various periods. Lysate from the cells used to measure the activity of caspase-1 and-3 proteases by using fluorogenic peptide substrates including acetyl-YVAD-AMC and acetyl-DEVD-AMC, respectively, Cells were treated with 200㎍/ml of each extract for various periods. Cell lysates were immunoprecipated with anti-JNKl antibodies. The immune complex was reacted with 32^p-ATP and c-Jun as a substrate. The phosphotransferase activity of JNKI was measured by suing Phosphoimage analyzer (Fuji Co., Japan). Nuclear extracts were isolated and incubated with oligonucleotide probe of NF-kB was measured by using EMSA and visualized by PhosphoImage analyzer (Fuji Co, Japan). Cell lysates were prepared and analyzed by Western blotting with anti-Bc12 antibodies and anti-Bax antibodies. Cells were pretreated with various doses of methanol extract for 2 hr. Then, the extract was removed by centrifugation. Cells were resuspended with RPMI-1640 media containing 0.3% agarose, 10% FBS, overlayred onto bottom layer agarose and incubated at CO_2 incubator for 6 days. The number of colony was cunted under light microscopy (×100). Results: The death of HL_60 cells was markedly induced by the addition of methanol extract of Hedyotis diffusa in a dose and time-dependent manners. The apoptotic characteristic ladder pattern of DNA strand break was observed in death of HL_60 cells. In addition, it was shown nucleus chromatin condensation under Hoechst staining. Therefore, Hedyotis diffusa extract-induced death of HL-60 cells is mediated by apoptotic signaling processes. The activity of Caspaxe 3-like proteases remained in a basal level in HL-60 cells treated with aqueous extract of Hedyotis diffusa. However, it was markedly increased in HL-60 cells treated with methanol extract of Hedyotis diffusa. In addition, the phosphotransferase activity of JNKl was increased in HL-60 cells treated with methanol extract of Hedyotis diffusa. Furthermore, the activation of transcriptional activator, NF-kB was markedly induced by methanol extract of Hedyotis diffusa. Anti-apoptotic Bc12 was cleaved into 23Kda fragment by treatment of methanol extract of Hedyotis diffusa. However, expression of proapoptotic Bax protein was increased by treatment of methanol extract of Hedyotis diffusa in a time-dependent manner. Furthermore, methanol extract markedly inhibited the colony forming efficiency of HL-60 cells in semisolid agar culture. Conclusions: Above results suggest that methanol extract of Hedyotis diffusa induces the apoptotic death of human leukemic HL-60 cells via activations of Caspase-3 proteases, JNKI, transcriptional activator NF-κB, In addition, our results also suggest that methanol exthanol extract of Hedyotis diffusa reduces the malignant potential of HL-60 cells via down regulation of colony forming effciency through cleavage of Bc12 as well as induction of Bax.

        괴산군 지역 농업 생태계의 곤충 다양성 비교 분석

        Agricultural ecosystem biodiversity monitoring and community variation analysis of insects were conducted from 2016 to 2018 in selected conventional and organic farming fields in Goesan district, Chungcheongbuk-do, South Korea. The total number of 1,125 species in 16 orders and 207 families were identified. The numbers of species collected in the locations practicing organic farming were greater than the conventional farming both in the paddy fields (564 vs. 383 species) and the upland fields (471 vs. 365 species). Among them, Hemiptera had the most abundant of species, followed by Diptera, Hymenoptera, Coleoptera and Araneae. We calculated various index values of biodiversity (diversity index H’, richness index R, evenness index J’, dominance index D, and similarity index QS) based on quantitative measurements of species and individuals collected over three years of field monitoring. Variations in biodiversity index values in different agricultural systems show that the positive effect of organic farming is to produce more biodiversity than conventional farming systems. When compared to other index results reported in Korea, Japan and China, the richness index was higher and other index values were at similar levels.

        The roots of Rumex crispus was extracted with 70% MeOH, and the MeOH extract was fractionated with dichloromethane, ethylacetate and n-butanol successively. The relatively active dichloromethane fraction was divided into several fractions with silicagel column chromatography using by MAO inhibition assay. A non-glycosidic anthraquinone, emodin was isolated from the active fraction. The structure of emodin (1,6,8-trihydroxy-3-methyl anthraquinone) was determined based on NMR and MS spectroscopic data and other chemical evidence.

        두 장 소매의 패턴메이킹에 관한 비교 연구

        The purpose of this study was to Investigate the differences in the method of patternmaking, difficulty in draft of patternmaking, fit in sleeve, aesthetic view, fashionableness in the five types of two-piece sleeve under the controled situation. Five types of two piece sleeve that were originated from five countries including domestic companies which called A type, B type, C type, D type, and E type were used. In order to conduct the experiment for the study, 10 persons who were well organized about the patternmaking drafted 5 types of sleeve and were tested for the differences in the method of patternmaking, difficulty in draft of patternmaking. Another panel were tested for fit in sleeve, aesthetic view, fashionableness in the five types of two-piece sleeve in jacket under the controled situation. The results of this study were as followed : 1.C type was needed height of armhole in bodice and D type was needed height of armhole and width of armhole in bodice as well as all parts of sleeve measurements to draft pattemmaking. 2.There were significant differences of the items such as cap height, ease amount around sleeve cap, width of sleeve, forwardness of sleeve and the length of elbow line of the five types patternmaking drafted. 3.There were significant differences of difficulty in draft of the five types of patternmaking. C type and E type were easier to draft. 4.There were significant differences in fitness of the sleeves in jacket. C type and E type were more fit. 5.There were significant differences in aesthetic view. A type, B type and E type were more aesthetic under the situation. 6.There were significant differences in fashionableness in jackets. A type and E type were more fashionable under the situation.       • 측백나무 추출물의 Monoamine Oxidase 저해활성

        Monoamine oxidase (MAO) exists in the outer membrane of mitochondria and contained a flavin adenine dinucleotide (FAD). MAO is classified into two forms, A-type (MAO-A) and B-type (MAO-B) on the basis of their different specifications toward substrates and inhibitors. This study was carried out to identify inhibitory components on MAO from Thuja orientalis seed extract by bioassay- guided isolation. The seeds of Thuja orientalis was extracted with 80% MeOH, and the MeOH extract was fractionated with n-hexane, chloroform, ethylacetate and n-butanol successively. The n-hexane and chloroform fraction showed relatively high inhibitory activity on MAO. Among the fractions from n-hexane layer through silicagel column chromatography, the fractions 2, 3 and 4 showed strong inhibitory activity on MAO

      • 점성·비점성 유동장에서의 Solution-Adaptive Grid 기법연구

        유동장 해석에 있어 격자가 얼마나 잘 구성되어 있는가는 해의 정확도와 수렴성에 큰 영향을 미친다. 가장 이상적인 격자는 모든 격자점에서 오차가 같은 값으로 분배되어 있어야한다. 그러기 위해서는 오차가 크게 생길만한 충격파나 경계층 등에서는 그만큼 많은 수의 격자가 필요하게 된다. 그러나 이를 미리 예측하고 격자를 만들기가 어렵기 때문에 Solution-adaptive grid 생성방법이 사용된다. 이것은 해의 변화가 심하게 나타나 오차가 큰 곳에 격자를 모아주고 상대적으로 오차가 작은 곳은 격자를 펴줌으로써 같은 수의 격자로 효율적인 격자를 생성하는 방법이다. 본 논문은 Elliptic Type의 격자생성법을 이용한Solution-adaptive Grid 방법과 Parameter 영역을 이용한 Solution-adaptive grid 방법을 이용하여 유동장을 계산하였다. One of the most important things for improving solution accuracy and convergency is grid refinements. Ideal grids make the same error distributions in all grid points. For this reason, grid rearrangement is sometimes necessary in some cases. If the flow fields have shock waves or boundary layers, there may be a lot of errors in those regions. Therefore, we should have more grid points in those regions to overcome this defect. Unfortunately, it is very difficult to estimate the regions which needs more grid points unless we have a final solution. So solution-adaptive grid methods are introduced. It is one of the grid refinement methods for making an effective grid in the flow fields. It makes grids be denser in the larger flow gradient regions and be coarser in smaller regions. Generally, two solution-adaptive grid methods are very popular. One of these is a solution adaptive grid generation method using elliptic type partial differential equation and the other is a solution adaptive grid generation methods using bilinear interpolation between physical domain and parametric domain.

        학습구조차트를 이용한 학습 활동이 수학 학습에 미치는 효과

        Teachers tend to believe that their students may not only fully understand what they have been taught in the classroom by self-studying and solving that problems but also perceive the structural relations in all the contents of learning. What they are studying at school, however, is totally new and strange to them. The relations, in the learning elements and contents, are difficult to understood. The purpose of this study is to show the effects of Professor Saito Noboroo's Climbing Learning Method using Learning Element Structure Chart. This method, in ither words, is the way of making learners form basic concepts and learn the relations of concept with handing out the concrete charts of the conceptional structures which are in teacher's mind. We have ben trying to find out the gradual effects to the Mathematics study after this method was applied to the two modeling classes of second year girls' middle school for six months or so.

      • 강상판 접합부의 피로균열 진전 시 STOP HOLE의 효과에 관한 연구

        Orthotropic steel decks are used in long span and moving bridges because of their high strength to weight ratio. However the decks are subjectde to high dynamic stresses due to wheel loading by heavy goods vehicles so that they have to be assessed for fatigue cracking. The fatigue assessment of orthotropic decks is excluded from most bridge design codes because of the complexity of the stress distribution and the fact that bending stress may predominate whereas weld classification tables are based on axially. Orthotropic steel decks are very sensitive to fatigue damage because they are directly subjected to the actions of wheel loads which give rise to stress ranges which are high compared to the dead load stresses especially if the surfacing is thin. The cracking that devel9oped was caused by large imperfections that were fabricated in short transverse groove connections. To stop the crack development, Stop hole should be excavated. The purpose of this paper, serviceability was approved by comparing surveyed data and stress distribution by structural analysis around stop hole and confirmed by minimized stress due to the size of stop hole. Also, stress distribution around scallop was computed by structural analysis modelled as actual steel bridge, Gul-Hyun Grand Bridge.
