        Correlation between DNA methylation and Thymic Stromal Lymphopoietin expression in asthmatic airway epithelial cells

        Background: The overexpression of TSLP and DNA methylation in asthma were both risk factors the relationship was not clear. Objective: This study aimed to investigate the relationship between methylation status of TSLP promoter and mRNA/protein expression in asthmatic airway epithelial cells. Methods: Human bronchial epithelial cells were cultured in vitro and divided into: Control group, treated with PBS, model group, sensitized with LPS (10 μg/mL) for 12 h (37 °C, 5% CO2). Other groups were cultured with the pCMV3 plasmid (M + NC/pCMV), pGPH1 plasmid (M + NC/pGPH), DNMT1/pCMV3 plasmid (M + DNMT1/pCMV), and DNMT1/pGPH1 plasmid (M + DNMT1/pGPH) for 48 h. The expression of DNA methyltransferase 1 and TSLP were measured using real-time PCR and western blotting. Results: Compared with the control group, TSLP mRNA (1.00 ± 0.00 vs. 2.82 ± 0.81 vs. 1, P < 0.001) and protein (1.07 ± 0.04 vs. 1.46 ± 0.11, P < 0.01) were significantly greater, and the methylation of promoter was lower (92.75 ± 1.26 vs. 58.57 ± 3.34, P < 0.05) in the model group. Compared with the model group, TSLP mRNA (2.82 ± 0.81 vs. 1.17 ± 0.10, P < 0.001) decreased, but TSLP promoter methylation increased (58.57 ± 3.34 vs. 92.58 ± 7.30, P < 0.05) in M + DNMT1/pCMV. TSLP mRNA and protein were higher (2.82 ± 0.81 vs. 5.32 ± 0.21, P < 0.001; 1.46 ± 0.11 vs. 1.94 ± 0.11, respectively, P < 0.01), TSLP promoter methylation was lower (58.57 ± 3.34 vs. 33.57 ± 4.29, P < 0.05) in M + DNMT1/pGPH. Conclusions: Overexpression of TSLP in asthmatic airway epithelial cells may be regulated by DNA demethylation.

        Identification of the Genes Involved in the Fruiting Body Production and Cordycepin Formation of Cordyceps militaris Fungus

        A mutant library of Cordyceps militaris was constructed by improved Agrobacterium tumefaciens-mediated transformation and screened for degradation features. Six mutants with altered characters in in vitro and in vivo fruiting body production, and cordycepin formation were found to contain a single copy T-DNA. T-DNA flanking sequences of these mutants were identified by thermal asymmetric interlaced-PCR approach. ATP-dependent helicase, cytochrome oxidase subunit I and ubiquitin-like activating enzyme were involved in in vitro fruiting body production, serine/threonine phosphatase involved in in vivo fruiting body production, while glucose-methanol-choline oxidoreductase and telomerase reverse transcriptase involved in cordycepin formation. These genes were analyzed by bioinformatics methods, and their molecular function and biology process were speculated by Gene Ontology (GO) analysis. The results provided useful information for the control of culture degeneration in commercial production of C. militaris.

      • No association Between Calcium Channel Blockers and Survival in Patients with Cancer: A systematic Review and Meta-analysis

        Background: The association between calcium channel blockers (CCBs) and survival in cancer patients remains unclear and the results of related studies are conflicting. The objective of the study was to investigate the association between calcium channel blockers (CCBs) use and survival in cancer patients. Materials and Methods: We searched PubMed, EMBASE, Web of Science and Cochrane Library for studies published before January 2016 with the terms related to CCBs and survival in cancer patients. The information was reviewed and extracted by two evaluators independently. Data of publications was extracted and calculated into hazard ratios (HRs) for overall survival (OS). Statistical analysis was performed by using Review Manager 5.3. Results: There were 11 studies included in our meta-analysis. Analysis of all studies showed that CCBs use was not associated with survival in cancer patients (HR=1.07; 95% CI: 0.91-1.25; P=0.42). No association between CCBs use and overall survival in cancer patients was existed whether in Asian (HR=1.18, 95% CI: 0.72-1.93; P=0.52) or Caucasian population (HR=1.03, 95% CI: 0.89-1.20; P=0.66). Conclusions: There is no evidence that CCBs use is associated with a better or worse outcome of survival in cancer patients.

        Expression, Purification, and Biological Characterization of The Amino-Terminal Fragment of Urokinase in Pichia pastoris

        Urokinase (uPA) and its receptor (uPAR) play an important role in tumor growth and metastasis. Targeting the excessive activation of this system as well as the proliferation of the tumor vascular endothelial cell would be expected to prevent tumor neovasculature and halt the tumor development. In this regard, the amino-terminal fragment (ATF) of urokinase has been confirmed as effective to inhibit the proliferation, migration, and invasiveness of cancer cells via interrupting the interaction of uPA and uPAR. Previous studies indicated that ATF expressed in Escherichia coli was mainly contained in inclusion bodies and also lacked posttranslational modifications. In this study, the biologically active and soluble ATF was cloned and expressed in Pichia pastoris. The recombinant protein was purified to be homogenous and confirmed to be biologically active. The yield of the active ATF was about 30 mg/l of the P. pastoris culture medium. The recombinant ATF (rATF) could efficiently inhibit angiogenesis, endothelial cell migration, and tumor cell invasion in vitro. Furthermore, it could inhibit in vivo xenograft tumor growth and prolong the survival of tumor-bearing mice significantly by competing with uPA for binding to cell surfaces. Therefore, P. pastoris is a highly efficient and cost-effective expression system for large-scale production of biologically active rATFs for potential therapeutic application.

        Outcomes of Microendoscopic Discectomy and Percutaneous Transforaminal Endoscopic Discectomy for the Treatment of Lumbar Disc Herniation: A Comparative Retrospective Study

        Study Design: Retrospective, case control evaluation of 86 patients who underwent microendoscopic discectomy (MED) and percutaneous transforaminal endoscopic discectomy (PTED) for the treatment of lumbar disc herniation (LDH). Purpose: To evaluate the safety and the outcomes of MED and PTED for the treatment of LDH. Overview of Literature: MED and PTED are minimally invasive surgical techniques for lower back pain. Studies to date have shown that MED and PTED are safe and effective treatment modalities for LDH. Methods: A retrospective study was performed in patients with LDH treated with MED (n=50) and transforaminal endoscopic discectomy (PTED; n=36) in our hospital. All patients were followed-up with self-evaluation questionnaires, Oswestry disability index (ODI), medical outcomes study 36-item short form health survey and MacNab criteria. All the patients in both groups were followed up to 12 months after the operation. Results: ODI questionnaire responses were not statistically different between the MED and PTED groups (53.00 vs. 48.72) before treatment. Average scores and minimal disability after 5 days to 12 months of follow-up were 4.96 in the MED group and 3.61 in the PTED group. According to MacNab criteria, 92.0% of the MED group and 94.4% of the PTED group had excellent or good results with no significant difference. Conclusions: There was no significant difference between MED and PTED outcomes. Further large-scale, randomized studies with long-term follow-up are needed.

      • A SANDWICH-INJECTION METHOD FOR MICROCHIP ELECTROPHORESIS

        In microchip electrophoresis (μ-CE), sample injection is generally achieved through cross, double-T, or T-form injector structures. In these reported approaches, the separation efficiency and detection sensitivity of μ-CE is significantly influenced by the shape and size of the sample plug introduced into the separation channel or sample leakage in separation phase. Here, we present a sandwich-injection method for controlling discrete sample injection in μ-CE. This method involves four accessory arm channels in which symmetrical potentials are loaded to form a unique parallel electric field distribution at the intersection of sample and separation channels. The parallel electric field effectuate a virtual wall to confine the shape of a sample plug and depress the spreading of the sample plug at the junction of sample and separation channels, and also prevent sample leakage during separation step. The key features of this method are the ability to inject well-defined sample plugs at the original sample concentration and the ability to control the sample plug size. The virtues of the novel injection technique were demonstrated with numerical models and validated with fluorescence visualizations of electrophoretic experiments.

        Novel high‑power triple line‑voltage cascaded unity power factor rectifier

        A novel high-power triple line-voltage cascaded three-phase unity power factor rectifier is proposed to address the complex topology and control of the three-phase rectifier stage in traditional high-power charging power supply modules. The topology is composed of three line-voltage cascaded traditional three-phase single-switch boost-type voltage rectifier modules. With the triple line-voltage cascaded structure, in the proposed topology, the number of the fully controlled power switches and the voltage stresses of each power switch are both effectively reduced. In addition, the proposed rectifier topology can operate at a unity power factor with a sinusoidal input current. In this paper, the circuit structure and the operation principle including the DC-link voltages output characteristics of the proposed topology are described, and a corresponding system control strategy based on one cycle control (OCC) is presented. Simulation and experimental results are given which verified the feasibility of the proposed topology and control strategy.

        Metal organic frameworks template-directed fabrication of rod-like hollow BiOClxBr1x with adjustable band gap for excellent photocatalytic activity under visible light

        Developing an efficient, environmentally friendly, and pollution-free catalyst with excellent visible light catalyticactivity is a promising strategy for dye wastewater treatment. Herein, the rod-like hollow BiOClxBr1x (x=1, 0.75, 0.5,0.25, 0), with an adjustable band gap, was successfully prepared using Bi-based metal-organic framework as template. The corresponding hollow assembly and introduction of Br imparted valuable structural advantages and intrinsic characteristicsfor improved photocatalytic activity. Significantly, the degradation efficiency of BiOCl0.5Br0.5 for the RhodamineB (RhB) solution reached 92% under visible light illumination for 90 min, which is considerably higher than that ofCAU-17-derived Bi2O3 and BiOCl. Overall, these findings shed fundamental insight on constructing novel photocatalystswith excellent visible light driven photocatalytic activity and offered a new method for treating dye wastewater.
