        This paper deals with stress analysis and deflections for probe stage systems connected to semi-conductor apparatus. Most of the real probe stage systems are composed of three mechanical parts such as slider, X-base and Y-base. In order to calculate the maximum stress and deflection for probe stage systems, three-dimensional finite element model is applied. Numerical results are obtained by changing various design parameters related to the above three parts. Through the numerical simulation, optimal design shape satisfying von Mises yield criterion is presented. The analysis was performed by using commercial COSMOS/M FEA code.

      • Characterization of agronomic traits of insect resistant GM Kimch cabbage

        A Transgenic Kimch cabbage has been developed harboring T-DNAs expressing delta-endotoxin insecticidal protein, herbicide (basta) resistant protein, and antisense transcript of AsMADS2 gene. Three transgenic lines, #24, #45, and #51, originated from the same T0 plant were analyzed in terms of molecular characterization, phenotype, and agronomic traits. Flanking sequence analysis confirmed that T-DNA, with 7132 bp intact structure, was inserted onto the pseudochromosome A10 of B. rapa and all the genes in T-DNA were functionally active. Three of GM cabbage showed 69.2~75.3% of plant height and 81.8~89.7% of diameter to those of the isogenic variety ‘Nowon’, respectively. Curving upward leaf lamina attitude was observed on GM cabbage, while straight or slight concave on non-GM cabbage. In addition, an average range of 86~91.5% of head height and 87.4~94.8% of head diameter were observed on GM cabbage to those of the isogenic variety ‘Nowon’, respectively Moreover, curled inwards or slight overlap of head-forming leaf overlap at terminal region was observed on GM cabbage, but curled outwards or erect on non-GM cabbage. AsMADS2, a transcription factor reported to be involved in early flowering, was stably expressed to RNA in the GM cabbage, but it was not shown the significant influences to flowering time.

        Identification of SNPs and microRNAs in the massively wholegenome sequencing data of Korean native cattle

        This study aims to enhance the accuracy of genome selection for economic traits in Korean native cattle through the identification of single nucleotide polymorphisms (SNPs) located in microRNA sequences associated with lipid metabolism. We assembled and mapped the Hanwoo genome using whole-genome sequencing data from the NCBI's Sequence Read Archive and the National Institute of Animal Science. This allowed us to discover a total of 25,982,458 variants (21,795,577 SNPs and 4,186,881 Indels). Annotation revealed that a significant number of these variants were located in intergenic and intronic regions, while others were found to alter the DNA, creating amino acid substitutions, premature stop codons, or silent mutations. A high proportion of variants were observed in the protein coding regions and miRNA, with 24 and 74,205 variants identified in Impact_LOW and Impact_MODIFIER, respectively. In addition, we identified 18 putative miRNAs that were differentially expressed in subcutaneous and intramuscular fat tissues. Especially, bta-mir-143 and bta-mir-145 were highly expressed in intramuscular fat tissue. These results could support the impact on the potential regulatory roles of miRNAs in bovine adipogenesis, and could prove beneficial for improving the accuracy of Hanwoo breeding programs.

      • RTK-GPS를 이용한 시공중 발생하는 구조물변위의 웨이브릿, 칼만필터 해석

        본 연구에서는 조석간만의 차로 설치중인 교각의 기초부분에서 발생하는 구조물의 변위를 RTK-GPS를 사용하여 구조물 거동의 건전도 평가에 사용 하였다. 측정 시에 사용한 필터링방법은 웨이브릿 대역필터와 칼만필터를 사용하였다. RTK-GPS 관측 에는 24시간 관측과 8시간 관측 2가지를 나누어 관측하였다. 획득한 RTK-GPS 관측 값을 칼만 필터와 웨이브릿 대역필터를 이용하였으며 그 밖의 과대오차를 소거하기위하여 3σ공식을 사용하여 필터링 하였다. 필터링을 통하여 시간대별 구조물의 거동을 추정하는 정확도가 향상되었음을 확인할 수 있다. In this study, the tidal bridge is being installed ebb and high tide that is raised at the base of the displacement of the structure using RTK-GPS Integrity Assessment of the behavior of structures were used. When measured using the wavelet band filtering method was used to filter and Kalman filter. RTK-GPS observations and a 24-hour observation for eight hours into two kinds of observations were observed. RTK-GPS observation values obtained with the Kalman filter was used for wavelet bandpass filters and other blunder to clear the filter using the official was 3σ. Hourly through filtering to estimate the behavior of the structure can be found that improves the accuracy.

        제조물책임에 있어서 쇠고기 이력제의 역할과 법개정의 필요성

        만약 쇠고기 등의 1차 농축산물로 인하여 소비자에게 피해가 발생하였다고 한다면, 과실책임을 원칙으로 하고 있는 일반불법행위책임법하에서는 그 피해에 대한 효율적 구제를 기대하기 어려울 것이다. 그 이유는 안전하지 않은 소나 쇠고기로 인하여 손해를 입은 소비자로서는 그것을 유통시킨 축산농가나 공급업자(또는 수입업자)를 상대로 하여 그 손해발생에 대한 과실책임을 추궁하기가 쉽지 않기 때문이다. 그러나 그 상황은 무과실책임을 원칙으로 하고 있는 제조물책임법의 적용이 가능하게 된다면 충분히 달라질 수 있다. 하지만 현행 제조물책임법은 미가공의 1차 농축산물을 본법의 적용대상인 제조물의 범위에서 제외시키고 있기 때문에 그 적용이 거의 불가능한 상황이다(제2조 1호 참조). 따라서 그 문제의 해결을 위해서는 미가공 1차 농축산물에 대해서도 무과실책임이 적용될 수 있도록 본법이 정하고 있는 제조물의 개념을 개정할 필요가 있다. 그러나 이와 같은 법개정은 생산자표시의 불명확성과 유통과정의 복잡성 때문에 그 책임소재가 불명확하다는 1차 농축산물에서의 근본적인 문제가 해결되지 않는 이상 큰 실효를 거두기가 어려울 것이다. 이러한 관점에서 볼 때, 최근 도입된 쇠고기 이력제는 그와 같은 문제를 해결하는 데 있어 중요한 역할을 할 수 있을 것이라 기대되며, 나아가 제조물책임법상 제조물개념의 규정을 개정하려는 시도에도 긍정적인 영향을 미칠 수 있을 것이라 생각된다. Product liability is the area of law in which manufacturers, distributors, suppliers, retailers, and others who make products available to the public are held responsible for the injuries those products cause. Article 2(1) of product liability provides that products mean manufactured or processed movables including other movables or a part of immovable properties. According to this rule, unprocessed primary agricultural products is not included. However, aspects of the liability without fault relating to consumer protection and the introduction of new traceability system call for the reform of Article 2(1). The beef traceability system makes consumers feel safe for the occurrence of any problems by taking an immediate action along with informations on the birth, slaughter, processing and sales. Give an identification number to every cattle with an ear tag to keep track of an entire distribution process from its birth to consumer's table at home.

        과거 탈공업화 정책으로 인해 경제 주도권을 서비스업에 내주었던 제조업이 21세기 경제 성장과 고용창출의 중심으로 재조명되고 있다. 과거의 제조업이 기계화, 자동화, 집중화로 경제 성장과 고용창출에 기여를 했다면, 현재의 제조업은 생산 설비와 ICT 인프라가 융합된 서비스화, 디지털화라는 새로운 가능성을 제시하면서 선진국을 중심으로 제조업 부활의 가능성을 예고하고 있다. 특히 ICT 기반의 화학 산업은 국내 경제침체를 극복하는 중요한 성장 동력으로 산업으로, 최근 기술동향과 ICT 융합 시장의 변화에 대한 정부정책 및 민간투자에 대한 기초자료를 제공할 필요가 있으므로, ICT 기반 화학산업의 경제 및 산업 환경에 따른 최근 ICT-화학 융합사례를 분석하고 향후 ICT 기반 화학소재산업이 나아가야 할 방향을 제시하고자 한다.

        Event-specific Qualitative and Quantitative Polymerase Chain Reaction Methods for Detection of Insect-resistant Genetically Modified Chinese Cabbage Based on the 3'-junction of the Insertion Site

        Transgenic Chinese cabbage 416-3 was developed in Korea by a transformation event involving modified insectresistant gene (cry1Ac1). To monitor unintended release of genetically modified (GM) Chinese cabbage in the future, as well as to meet GM-labeling requirements, the development of a reliable method for detection of GM cabbage is requisite. To develop qualitative and quantitative polymerase chain reaction methods for the insect-resistant GM Chinese cabbage, a cytosolic glutathione reductase (BcgGR1) gene was used as the endogenous reference gene. Primer pairs CGR-1/-2, amplifying the Chinese cabbage endogenous gene, yielded an expected amplicon of 121bp, whereas no amplified product was observed when DNA samples from seven non-cabbage plants were used as templates. The event-specific primer pairs amplifying the junction site between the endogenous genome sequence and the transferred DNA of GM event 416-3, produced amplicons of desired size by qualitative polymerase chain reaction (PCR) assay. An eventspecific quantitative PCR detection method was established using a TaqMan probe and a standard plasmid as a reference molecule,which contained both endogenous and event-specific sequences. For the validation of this method, three different compositions of w/w mixed samples (containing transgenic DNA at 5, 1, and 0.5%of total DNA in the control samples) were quantified. The precision, expressed as standard deviation (SD) and relative standard deviations (RSD), deviated by 0.03–0.26% and 4.75–8.06%, respectively. These results clearly demonstrate that the PCR methods developed herein can be used for event-specific qualitative and quantitative testings of insect-resistant GM Chinese cabbage.

      • Genome-wide identification and characterization of novel laccase genes of Flammulina velutipes

        The aim of this study was to identify and characterize new Flammulina velutipes laccases from its whole-genome sequence. Of the 15 putative laccase genes detected in the F. velutipes genome, four new laccase genes (fvLac-1, fvLac-2, fvLac3, and fvLac-4) were found to contain four complete copper-binding regions (ten histidine residues and one cysteine residue) and four cysteine residues involved in forming disulfide bridges. fvLac-1, fvLac-2, fvLac3, and fvLac-4, encoding proteins consisting of 516, 518, 515, and 533 amino acid residues, respectively. Potential N-glycosylationsites(Asn-Xaa-Ser/Thr) were identified in the cDNA sequence of fvLac-1 (Asn-454), fvLac-2(Asn- 437andAsn-455), fvLac-3(Asn-111andAsn-237), and fvLac4 (Asn-402andAsn-457). In addition, the first 19–20 amino acid residues of these proteins were predicted to comprise signal peptides. Laccase activity assays and reverse transcription polymerase chain reaction(RT-PCR) analyses clearly reveal that CuSO4 affects the induction and the transcription level of these laccase genes.
