        검정소 검정돈의 품종 및 환경요인의 효과 추정

        This study was carried out to estimate the effects of breed and environment such as sex, test station, test year, test season, parity, initial and final weight on average daily gain, age at 90㎏, backfat thickness, feed efficiency, lean percent and selection index on the basis of the performance data collected from 25,790 pigs of Duroc, Yoskshire and Landrace breeds which were performance-tested at the Korea Swine Testing Station from 1991 to 2002. The results obtained in the study are summarized as follows; 1. The means of the major economic traits were estimated as 959.95 ± 0.699g for average daily gain, 138.36 ± 0.072days for age at 90㎏, 1.41 ± 0.001㎝ for backfat thickness, 2.33 ± 0.001 for feed efficiency, 56.71 ± 0.018% for lean percent and 221.65 ± 0.113 for selection index. 2. The effect of breed was statistically significant for all studied traits. Briefly, Duroc showed the best per-formance for the average daily gain and age at 90㎏. Landrace had the best performances for the backfat thickness and lean meat percent. In feed efficiency and selection index, Yorkshire had a better score than other breeds. 3. The least-squares means of female and male for the traits studied were 923.05 ± 1.289g and 974.53 ± 0.856g for average daily gain, 139.74 ± 0.145days and 137.21 ± 0.097days for age at 90㎏, 1.49 ± 0.002㎝ and 1.39 ± 0.002㎝ for backfat thickness, 2.43 ± 0.002 and 2.28 ± 0.002 for feed efficiency, 56.43 ± 0.034% and 56.81 ± 0.023% for lean percent and 211.37 ± 0.194 and 224.61 ± 0.129 for selection index. Therefore, males were superior to females for all traits examined. 4. The effect of test station was statistically significant for all traits except for selection index. Performances for age at 90㎏, backfat thickness, feed efficiency and lean meat percent collected from Test station 2 were higher than those from Test station 1. However. Test station 1 showed better average daily gain. 5. The initial weight and final weight included as a covariate in this study had a significant influence on average daily gain, age at 90㎏, backfat thickness, feed efficiency and selection index. From the absolute values of the estimated regression coefficients, it was inferred that the final weight had greater effect for the investigated traits than the initial weight.

      • Ethanol extract of Kalopanax septemlobus leaf inhibits HepG2 human hepatocellular carcinoma cell proliferation via inducing cell cycle arrest at G<sub>1</sub> phase

        <P>Objective: To investigate the effects of an ethanol extract of Kalopanax septemlobus (Thunb.) Koidz. leaf (EEKS) on cell proliferation in human hepatocellular carcinoma cells and its mechanisms of action. Methods: Cells were treated with EEKS and subsequently analyzed for cell proliferation and flow cytometry analysis. Expressions of cell cycle regulators were determined by reverse transcriptase polymerase chain reaction analysis and Western blotting, and activation of cyclin-associated kinases studied using kinase assays. Results: The EEKS suppressed cell proliferation in both HepG2 and Hep3B cells, but showed a more sensitive anti -proliferative activity in HepG2 cells. Flow cytometry analysis revealed an association between the growth inhibitory effect of EEKS and with G, phase cell cycle arrest in HepG2 cells, along with the dephosphorylation of retinoblastoma protein (pRB) and enhanced binding of pRB with the E2F transcription factor family proteins. Treatment with EEKS also increased the expression of cyclin-dependent kinase (CDK) inhibitors, such as p21WAF 1/CIP 1 and p27KIPl, without any noticeable changes in G(1) cyclins and CDKs (except for a slight decrease in CDK4). Treatment of HepG2 cells with EEKS also increased the binding of p21 and p27 with CDK4 and CDK6, which was paralleled by a marked decrease in the cyclin D- and cyclin E -associated kinase activities. Conclusions: Overall, our findings suggest that EEKS may be an effective treatment for liver cancer through suppression of cancer cell proliferation via. G, cell cycle arrest. Further studies arc required to identify the active compounds in EEKS.</P>

        Flavonoids Identified from Korean Scutellaria baicalensis Induce Apoptosis by ROS Generation and Caspase Activation on Human Fibrosarcoma Cells

        The effects of flavonoids from Korean Scutellaria baicalensis on fibrosarcoma HT1080 cells and their underlying molecular mechanism were investigated in this study. Flavonoids affected HT1080 cell proliferation by interrupting cell cycle progress, obviously augmenting the proportion of sub-G1 and diminishing that of G1 phase, and undergoing apoptosis at the tested dosage (100-400 mu g/mL). In addition, the mediated apoptosis was mainly caused by total reactive oxygen species (ROS) generation and by up-regulating the ratio of Bax/Bcl-xL, triggering caspase cascades (caspase-3, -9 and -8), and inactivating PARP, dose-dependently. The proteomics results showed that AP-4, ARID 5B, HNRNP K, PLOG, Prdx6, and myosin-1, associated with cell growth, differentiation and development, and overexpressed in gastric cancer, colorectal cancer, pancreatic cancer, etc., were statistically down-regulated after the flavonoids treatment. Taken together, our data demonstrated that flavonoids from Korean S. baicalensis induced apoptosis in HT1080 cells, which involved a hierarchy of cellular pathways and multiple signal proteins, and might be a potential anticancer therapeutic agent.

        20대 여성의 신발종류에 따른 족저압 영역별 비교 연구

The purpose of this study was to investigate the test-retest of plantar pressures using the F-Scan system over speeds and plantar regions. 6 healthy female subjects in 20's were recruited for the study. Plantar pressure measurements during locomotor activities can provide information concerning foot function, particularly if the timing and magnitude of the loading profile can be related to the location of specific foot structures such as the metatarsal heads. The Tekscan F-Scan system consists of a flexible, 0.18mm thick sole-shape having 1260 pressure sensors, the sensor insole was trimmed to fit the subjects' right. left shoes - sneakers shoes & dress shoes. It was calibrated by the known weight of the test subject standing on one foot. The Tekscan measurements show the insole pressure distribution as a function of the time. This finding has important implications for the development of plantar pressure test protocols where the function of the forefoot is important. According to the result of analysis it is as follows : 1) Center of force trajectory in women's dress shoes display direct movement, compare with center of force trajectory in Sneaker shoes displays a little bit curved slow pronation movement. Sneaker shoes in forefoot part display very quick supination movement, therefore, this shoes effects negative effectiveness for ankle's stability. Considering center of force trajectory analyzing, the more center of force close straight line, the more movement can be quick movement for locomotion. For foot pressure distribution, center of force trajectory in locomotion is better to curved trajectory with pronation movement. So sneaker shoes style is good shoes considering center of pressure distribution trajectory compare with women's dress shoes. 2) Women's dress shoes increased peak pressure in medial, this is effected by high hill's height. The more increased women's dress shoes's height, the more women's peak pressure will increase, pronation can increase compare with before. Supination movement increase, this focused pressure in lateral, also, supination increased more. If the supination movement increased, foot pressure focused in lateral, therefore, it is appeared force distribution in gait direction. This is bad movement in foot's stability. 3) Women's dress shoes in landing phase displayed a long time, this is when women's dress shoes wear, gait movement is unbalance, so, landing phase displayed a long time. For compensation in gait, swing phase quick movement. 4) Women's dress shoes displayed peak pressure distribution in lateral of rearfoot part, Sneakers shoes displayed peak pressure distribution in medial of forefoot part. Its results has good impact absorption compare with women's dress shoes. In forefoot part, sneakers shoes has good propulsive force compare with women's dress shoes.

      • Different culture conditions used for arresting the G0/G1 phase of the cell cycle in goldfish (Carassius auratus) caudal fin-derived fibroblasts

        One of the most important factors determining the success of the development of cloned embryos is the cell cycle stage of the donor cells. We investigated the effects of serum starvation, culturing to confluence and roscovitine treatment on the cell cycle synchronization of goldfish caudal fin-derived fibroblasts by flow cytometric analysis. The results show that culturing the cells to confluence (85.5%) and roscovitine treatment (82.71%) yield a significantly higher percentage of cells arrested in the G0/G1 (P<0.05) phase than serum starvation (62.85%). Different concentrations of roscovitine (5, 10, or 15μM) induce cell cycle arrest at the G0/G1 phase.

        Purpose: The purpose of this study was to figure out general characteristics, health perception, self-efficacy, and family support influencing on health promotion behavior of middle-aged male. Method: In this descriptive correlation design study, 320 middle-aged male were recruited through convenient sampling between October and December, 2015. Questionnaires used for this study were HPLP(Health Promotion Lifestyle Profile), HPS(Health perception Scale), Self-Efficacy Scale developed by Hong, and Family Support Scale developed by Park. Data were statistically analyzed using SPSS 22.0 program with descriptive statistics, independent t-test, one way ANOVA with Scheffe's test, and multiple linear regression. Result: It had significant influence to correlation among health promotion behavior, health perception, self-efficacy, and family support of middle-aged male. Also, the most influenced factor to health promotion behavior showed health perception. Conclusion: The result of this study suggested that health perception significantly influenced to health promotion behavior. Therefore, health perception educating program for health promotion behavior of middle-aged male. And further studies are required with more sample of various group and systematic data collecting methods to confirm these results.

        A genetic variation in microRNA target site of <i>KRT81</i> gene is associated with survival in early-stage non-small-cell lung cancer

        <P>In this study, <I>KRT81</I> rs3660G>C was associated with survival of patients with NSCLC after surgical resection. Mechanistic study suggested that the G-to-C change caused reduced binding efficiency of miRNA, leading to decreased translational repression, thereby increased <I>KRT81</I> expression. The <I>KRT81</I> rs3660G>C may be a useful prognostic biomarker in early-stage NSCLC patients.</P><P><B>Background</B></P><P>MicroRNAs (miRNAs) have a key role in carcinogenesis through negative regulation of their target genes. Therefore, genetic variations in miRNAs or their target sites may affect miRNA–mRNA interactions, thereby result in altered expression of target genes. This study was conducted to investigate the associations between single-nucleotide polymorphisms (SNP) located in the miRNA target sites (poly-miRTSs) and survival of patients with early-stage non-small-cell lung cancer (NSCLC).</P><P><B>Methods</B></P><P>Using public SNP database and miRNA target sites prediction program, 354 poly-miRTSs were selected for genotyping. Among these, 154 SNPs applicable to Sequenom's MassARRAY platform were investigated in 357 patients. A replication study was carried out on an independent patient population (<I>n</I> = 479). <I>Renilla</I> luciferase assay and reverse transcription-polymerase chain reaction were conducted to examine functional relevance of potentially functional poly-miRTSs.</P><P><B>Results</B></P><P>Of the 154 SNPs analyzed in a discovery set, 14 SNPs were significantly associated with survival outcomes. Among these, <I>KRT81</I> rs3660G>C was found to be associated with survival outcomes in the validation cohort. In the combined analysis, patients with the rs3660 GC + CC genotype had a significantly better overall survival compared with those with GG genotype [adjusted hazard ratio (aHR) for OS, 0.65; 95% confidence interval (CI) 0.50–0.85; <I>P</I> = 0.001]. An increased expression of the reporter gene for the C allele of rs3660 compared with the G allele was observed by luciferase assay. Consistently, the C allele was associated with higher relative expression level of <I>KRT81</I> in tumor tissues.</P><P><B>Conclusion</B></P><P>The rs3660G>C affects KRT81 expression and thus influences survival in early-stage NSCLC. The analysis of the rs3660G>C polymorphism may be useful to identify patients at high risk of a poor disease outcome.</P>
