        A Modified Extraction Method of Circulating Free DNA for Epidermal Growth Factor Receptor Mutation Analysis

        Purpose: Circulating free DNA (cfDNA) in plasma is promising to be a surrogate for tumor tissue DNA. However, not all epidermal growth factor receptor (EGFR) mutations in tumor tissue DNA has been detected in matched cfDNA, at least partly due to inefficient cfDNA extraction method. The purpose of this study was to establish an efficient plasma cfDNA extraction protocol. Materials and Methods: The yield of plasma cfDNA extracted by our modified phenol-chloroform (MPC) method from non-small-cell lung cancer (NSCLC) patients was compared with that by QIAamp MinElute Virus Spin kit (Qiagen kit) as control, using the Wilcoxon rank-sum test. TaqMan quantitative polymerase chain reaction (qPCR) assays were used to quantify the plasma cfDNA extracted. Both Mutant-enriched PCR (ME-PCR) coupled sequencing and DxS EGFR mutation test kit were used to evaluate the impact of extraction method on EGFR mutation analysis. Results: MPC method extracted more plasma cfDNA than Qiagen kit method (p=0.011). The proportion of longer fragment (≥202 bp) in cfDNA extracted by MPC method was significantly higher than by Qiagen kit method (p=0.002). In the sequencing maps of ME-PCR products, a higher mutant peak was observed on plasma cfDNA extracted by MPC method than by Qiagen kit method. In DxS EGFR mutation test kit results, plasma cfDNA extracted by MPC method contained more tumor-origin DNA than by Qiagen kit method. Conclusion: An improved plasma cfDNA extraction method of MPC is provided, which will be beneficial for EGFR mutation analysis for patients with NSCLC.

        In this paper, we establish some sufficient conditions for the existence of positive solutions for a class of multi-point boundary value problem for fractional functional differential equations involving the Caputo fractional derivative. Our results are based on two fixed point theorems. Two examples are also provided to illustrate our main results.

        The mechanical design and fabrication of 162.5 MHz buncher for China accelerator driven sub-critical system injector II

        A buncher is one of the main pieces of equipment in the medium energy beam transport line (MEBT) for China accelerator driven sub-critical system (C-ADS) Injector II. To focus the beam longitudinally and match the beam for the acceptance of the superconducting linac section, two room temperature quarter wave resonator (QWR) bunchers with frequency of 162.5 MHz have been designed as parts of the MEBT. According to the beam transmission matching of the MEBT and the geometric parameters requirements of bunchers, the unique mechanical structure and the main processing technology of buncher cavities and their couplers and tuners are described in this paper. The fabrication of bunchers and their parts have been completed and tested at high power, the test results agree well with the design requirements. These bunchers work well for about two years in Institute of Modern Physics, Chinese Academy of Sciences.

      • Representing Topic-Comment Structures in Chinese

        Shi (2000) claims that topics must be related to a syntactic position in the comment, thus denying the existence of dangling topics in Chinese. Under Shi``s analysis, the dangling topic sentences in Chinese are not topic-comment but subject-predicate sentences. However, Shi``s arguments are not without problems. In this paper we argue that topics in Chinese can be licensed not only by a syntactic gap but also by a semantic gap/variable without syntactic realization. Under our analysis, all the dangling topics discussed in Shi (2000) are, in fact, not subjects but topics licensed by a semantic gap/variable that can turn the relevant comment into an open predicate, thus licensing dangling topics and deriving well-formed topic-comment constructions. Our analysis fares better than Shi``s in not only unifying the licensing mechanism of a topic to an open predicate without considering how the open predicate is derived, but also unifying the treatment of normal and dangling topics in Chinese.

        Effect of Tea Polysaccharides on Thermal and Pasting Properties of Wheat Starch

        The thermal and pasting properties of wheat starch (WS)/tea polysaccharides (TPS) composite system were investigated. Significant increases of transition temperatures (To, Tp, Tc) and the enthalpy of gelatinization (ΔH1) of WS/TPS (2 wet% and 4 wet%) were found as TPS content increased. The lower ΔH2 (endothermic enthalpy under reheating) and R2 (degree of retrogradation of WS) were also detected in WS/TPS in storage of 3,7,14 days when compared with those of WS. The agreements were shown by RVA with the lower total setback and by Texture Analyzer with lower gel firmness. The results of TPS affecting the thermal and pasting properties could be generally explained with Avrami equation of WS/TPS. The Avrami equation exposed that TPS affect the primary nucleation in early stage and the decreased nucleation rate announced by the constant k. The overall results demonstrated that the TPS has an inhibitory effect on the recrystallization of pasting WS in storage.

        ONE-DIMENSIONAL ANALYSIS OF THERMAL STRATIFICATION IN THE AHTR COOLANT POOL

        It is important to accurately predict the temperature and density distributions in large stratified enclosures both for design optimization and accident analysis. Current reactor system analysis codes only provide lumped-volume based models that can give very approximate results. Previous scaling analysis has shown that stratified mixing processes in large stably stratified enclosures can be described using one-dimensional differential equations, with the vertical transport by jets modeled using integral techniques. This allows very large reductions in computational effort compared to three-dimensional CFD simulation. The BMIX++ (Berkeley mechanistic MIXing code in C++) code was developed to implement such ideas. This paper summarizes major models for the BMIX++ code, presents the two-plume mixing experiment simulation as one validation example, and describes the codes’ application to the liquid salt buffer pool system in the AHTR (Advanced High Temperature Reactor) design. Three design options have been simulated and they exhibit significantly different stratification patterns. One of design options shows the mildest thermal stratification and is identified as the best design option. This application shows that the BMIX++ code has capability to provide the reactor designers with insights to understand complex mixing behavior with mechanistic methods. Similar analysis is possible for liquid-metal cooled reactors.

        中國經常帳戶調整是危機沖擊還是結構性轉型驅動?

        The reversal of the growing trend of China`s current account surplus has drawn the attention from various fields since US sub-prime lending crisis. By adopting the industry-level trade data, the paper reveals the industrial structural feature of China`s current account surplus. Further, the paper figures out firms` leading forces in each industry by using the firm-level trade data. The results show: (1) Labor-intensive industries are the main sources of China`s trade surplus while the foreign exchange-earning ability of technology-intensive industries is still relatively weak. (2) In most industries, trade surplus is mainly attributed to the quantity-expansion effect of existing products, and the effect of industrial upgrading out of high-tech enterprises and new products are negligible. (3) The quantity-expansion effect drives the rapid recovery growth of China`s trade surplus after the 2008 global financial crisis, and it can be properly inferred that the adjustment of China`s current account structure should be attributed to Chinese economy structural transformation other than crisis shock.

        4D Imaging and Diffraction Dynamics of Single-Particle Phase Transition in Heterogeneous Ensembles

        <P>In this Letter, we introduce conical-scanning dark-field imaging in four-dimensional (4D) ultrafast electron microscopy to visualize single-particle dynamics of a polycrystalline ensemble undergoing phase transitions. Specifically, the ultrafast metal–insulator phase transition of vanadium dioxide is induced using laser excitation and followed by taking electron-pulsed, time-resolved images and diffraction patterns. The single-particle selectivity is achieved by identifying the origin of all constituent Bragg spots on Debye–Scherrer rings from the ensemble. Orientation mapping and dynamic scattering simulation of the electron diffraction patterns in the monoclinic and tetragonal phase during the transition confirm the observed behavior of Bragg spots change with time. We found that the threshold temperature for phase recovery increases with increasing particle sizes and we quantified the observation through a theoretical model developed for single-particle phase transitions. The reported methodology of conical scanning, orientation mapping in 4D imaging promises to be powerful for heterogeneous ensemble, as it enables imaging and diffraction at a given time with a full archive of structural information for each particle, for example, size, morphology, and orientation while minimizing radiation damage to the specimen.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/nalefd/2014/nalefd.2014.14.issue-2/nl404354g/production/images/medium/nl-2013-04354g_0008.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/nl404354g'>ACS Electronic Supporting Info</A></P>
