        20(S)-ginsenoside Rg3 exerts anti-fi brotic effect after myocardial infarction by alleviation of fi broblasts proliferation and collagen deposition through TGFBR1 signaling pathways

        Background: Myocardial fibrosis post-myocardial infarction (MI) can induce maladaptive cardiacremodeling as well as heart failure. Although 20(S)-ginsenoside Rg3 (Rg3) has been applied to cardiovasculardiseases, its efficacy and specific molecular mechanism in myocardial fibrosis are largely unknown. Herein, we aimed to explore whether TGFBR1 signaling was involved in Rg3's anti-fibrotic effectpost-MI. Methods: Left anterior descending (LAD) coronary artery ligation-induced MI mice and TGF-b1-stimulated primary cardiac fibroblasts (CFs) were adopted. Echocardiography, hematoxlin-eosin andMasson staining, Western-blot and immunohistochemistry, CCK8 and Edu were used to study the effectsof Rg3 on myocardial fibrosis and TGFBR1 signaling. The combination mechanism of Rg3 and TGFBR1 wasexplored by surface plasmon resonance imaging (SPRi). Moreover, myocardial Tgfbr1-deficient mice andTGFBR1 adenovirus were adopted to confirm the pharmacological mechanism of Rg3. Results: In vivo experiments, Rg3 ameliorated myocardial fibrosis and hypertrophy and enhanced cardiacfunction. Rg3-TGFBR1 had the 1.78 10 7 M equilibrium dissociation constant based on SPRi analysis,and Rg3 inhibited the activation of TGFBR1/Smads signaling dose-dependently. Cardiac-specific Tgfbr1knockdown abolished Rg3's protection against myocardial fibrosis post-MI. In addition, Rg3 downregulatedthe TGF-b1-mediated CFs growth together with collagen production in vitro through TGFBR1signaling. Moreover, TGFBR1 adenovirus partially blocked the inhibitory effect of Rg3. Conclusion: Rg3 improves myocardial fibrosis and cardiac function through suppressing CFs proliferationalong with collagen deposition by inactivation of TGFBR1 pathway.

        In the big data era, with the parallel evolution of computer architecture, computing changes and modifications of industrial application mode resource expansion capability, we need to explore a new parallel computing model, to reflect the properties and large data applications form the current parallel machines, and a variety of mainstream big data processing system for unified theoretical analysis to guide large data applications tuning. Currently, despite the large data programming model study made many achievements, and is widely used in the TB level or even PB-class data processing and analysis, but the corresponding computational model study has just begun. From traditional parallel computing model, research big data programming model and large data computation model, summed up the three basic problems of large data model, in theory, need to be addressed: the three elements of the problem model, scalability and fault tolerance issues and performance optimization. Around these three questions, on the one hand and performance optimization model to calculate the theoretical study of data from a large, on the other hand these performance optimization methods in case of an actual big data.

      • Computational thermal stability and critical temperature buckling of nanosystem

        Many of small-scale devices should be designed to tolerate high temperature changes. In the present study, the states of buckling and stability of nano-scale cylindrical shell structure integrated with piezoelectric layer under various thermal and electrical external loadings are scrutinized. In this regard, a multi-layer composite shell reinforced with graphene nano-platelets (GNP) having different patterns of layer configurations is modeled. An outer layer of piezoelectric material receiving external voltage is also attached to the cylindrical shell for the aim of observing the effects of voltage on the thermal buckling condition. The cylindrical shell is mathematically modeled with first-order shear deformation theory (FSDT). Linear elasticity relationship with constant thermal expansion coefficient is used to extract the relationship between stress and strain components. Moreover, minimum virtual work, including the work of the piezoelectric layer, is engaged to derive equations of motion. The derived equations are solved using numerical method to find out the effects of temperature and external voltage on the buckling stability of the shell structure. It is revealed that the boundary condition, external voltage and geometrical parameter of the shell structure have notable effects on the temperature rise required for initiating instability in the cylindrical shell structure.

        Point In Triangle Testing Based Trilateration Localization Algorithm In Wireless Sensor Networks

        Localization of sensor nodes is a key technology in Wireless Sensor Networks(WSNs). Trilateration is an important position determination strategy. To further improve the localization accuracy, a novel Trilateration based on Point In Triangle testing Localization (TPITL)algorithm is proposed in the paper. Unlike the traditional trilateration localization algorithm which randomly selects three neighbor anchors, the proposed TPITL algorithm selects three special neighbor anchors of the unknown node for trilateration. The three anchors construct the smallest anchor triangle which encloses the unknown node. To choose the optimized anchors, we propose Point In Triangle testing based on Distance(PITD) method, which applies the estimated distances for trilateration to reduce the PIT testing errors. Simulation results show that the PIT testing errors of PITD are much lower than Approximation PIT(APIT) method and the proposed TPITL algorithm significantly improves the localization accuracy.

        A Receiver-Aided Seamless And Smooth Inter-RAT Handover At Layer-2

        The future mobile networks consist of hyper-dense heterogeneous and small cell networks of same or different radio access technologies (RAT). Integrating mobile networks of different RATs to provide seamless and smooth mobility service will be the target of future mobile converged network. Generally, handover from high-speed networks to low-speed networks faces many challenges from application perspective, such as abrupt bandwidth variation, packet loss, round trip time variation, connection disruption, and transmission blackout. Existing inter-RAT handover solutions cannot solve all the problems at the same time. Based on the high-layer convergence sublayer design, a new receiver-aided soft inter-RAT handover is proposed. This soft handover scheme takes advantage of multihoming ability of multi-mode mobile station (MS) to smooth handover procedure. In addition, handover procedure is seamless and applicable to frequent handover scenarios. The simulation results conducted in UMTS-WiMAX converged network scenario show that: in case of TCP traffics for handover from WiMAX to UMTS, not only handover latency and packet loss are eliminated completely, but also abrupt bandwidth/wireless RTT variation is smoothed. These delightful features make this soft handover scheme be a reasonable candidate of mobility management for future mobile converged networks.

        High Systemic Inflammation Response Index (SIRI) Indicates Poor Outcome in Gallbladder Cancer Patients with Surgical Resection: A Single Institution Experience in China

        Purpose The systemic inflammation response index (SIRI) has been reported to have prognostic ability in various solid tumors but has not been studied in gallbladder cancer (GBC). We aimed to determine its prognostic value in GBC. Materials and Methods From 2003 to 2017, patients with confirmed GBC were recruited. To determine the SIRI’s optimal cutoff value, a time-dependent receiver operating characteristic curve was applied. Univariate and multivariate Cox analyses were performed for the recognition of significant factors. Then the cohort was randomly divided into the training and the validation set. A nomogram was constructed using the SIRI and other selected indicators in the training set, and compared with the TNM staging system. C-index, calibration plots, and decision curve analysis were performed to assess the nomogram’s clinical utility. Results One hundred twenty-four patients were included. The SIRI’s optimal cutoff value divided patients into high (≥ 0.89) and low SIRI (< 0.89) groups. Kaplan-Meier curves according to SIRI levels were significantly different (p < 0.001). The high SIRI group tended to stay longer in hospital and lost more blood during surgery. SIRI, body mass index, weight loss, carbohydrate antigen 19-9, radical surgery, and TNM stage were combined to generate a nomogram (C-index, 0.821 in the training cohort, 0.828 in the validation cohort) that was significantly superior to the TNM staging system both in the training (C-index, 0.655) and validation cohort (C-index, 0.649). Conclusion The SIRI is an independent predictor of prognosis in GBC. A nomogram based on the SIRI may help physicians to precisely stratify patients and implement individualized treatment.

        Hepatoprotective effect of phenylethanoid glycosides from Incarvillea compacta against CCl<sub>4</sub>-induced cytotoxicity in HepG2 cells

        The extraction and solvent partition of roots of Incarvillea compacta, a traditional Tibetan folk medicine, and repeated column chromatography and preparative high-performance liquid chromatography for n-butanol fraction yielded four phenylethanoid glycosides, crenatoside (1), 3'''-O-methylcrenatoside (2), leucoseceptoside A (3), and martynoside (4). The chemical structures were identified on the basis of spectroscopic data analyses including NMR and MS. All compounds were isolated for the first time from the plant. Compound 1 exerted better 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity. In addition, compounds 1-4 were evaluated for their hepatoprotective activity on carbon tetrachloride ($CCl_4$)-induced liver injury in HepG2 cells. Pretreatment of HepG2 cells with compound 1-4 significantly increased the viability on $CCl_4$-induced cell death. Furthermore, compounds 1-4 also alleviated $CCl_4$-induced hepatotoxicity by enhancement of the antioxidant enzyme activities of superoxide dismutase and reduction of the malondialdehyde content, intracellular ROS as well as NF-${\kappa}B$ transactivation. Our results suggest that phenylethanoid glycosides ameliorate $CCl_4$-induced cell injury, and this protection was likely due to antioxidative activity and down-regulation of NF-${\kappa}B$.

        Comparison of the Prognostic Value of Platelet-Related Indices in Biliary Tract Cancer Undergoing Surgical Resection

        Purpose Platelet-related indices, including mean platelet volume (MPV) and plateletocrit (PCT), have been reported as new prognostic factors of overall survival (OS) in many cancers, but not yet in biliary tract cancer (BTC). We intended to assess these indices in predicting OS in BTC patients with the aim to build a new prognostic model for patients with BTC after surgical resection. Materials and Methods Survival analysis and time receiver operating characteristic analysis were applied to screen the platelet indices. Univariate and multivariate Cox analyses were used to identify independent prognostic factors and develop a new prognostic model. Harrell’s C-statistics, calibration curves, and decisive curve analysis were used to assess the model. Results MPV and platelet distribution width (PDW)/PCT showed the best prognostic accuracy among the platelet indices. In multivariable analysis, factors predictive of poor OS were presence of nodal involvement, Non-radical surgery, poor tumor differentiation, carbohydrate antigen 19-9 > 100 U/mL, MPV > 8.1 fl, and PDW/PCT > 190. The new model was found to be superior to the TNM staging system and our new staging system showed higher discriminative power.Conclusion MPV and PDW/PCT have high prognostic value in BTC patients, and the novel staging system based on these two indices showed good discrimination and accuracy compared with the American Joint Committee on Cancer 7th TNM staging system.

        The Effect of H2O2/Fe2+ Catalytic Oxidation System on the Morphology, Structure and Properties of Flake-Like Poly(2,3-dimethylaniline)

        In this work, flake-like poly(2,3-dimethylaniline) (P(2,3-DMA)) with enhanced thermal stability andanticorrosive ability was synthesized by in situ polymerization using H2O2/Fe2+ catalytic oxidation system, comparingwith traditional oxidant ammonium persulfate (APS) synthetic method. The structure and morphology of thesamples were characterized Fourier transform infrared (FTIR) spectra, X-ray diffraction (XRD) and field-emissionscanning electron microscope (FESEM). The experimental results demonstrated that using H2O2/Fe2+ catalytic oxidationsystem was more inclined to form the two-dimensional P(2,3-DMA) flakes. The enhancement in thermostabilityand corrosion resistance was attributed to the formation of phenazine-like structures in the polymer chains,which could serve as templates to form the flake-like morphology. In addition, using H2O2/Fe2+ catalytic oxidationsystem is more environmental friendly than the APS method that can avoid ammonium pollution on aquatic life aswell as waters.
