중합효소연쇄반응을 이용한 도말양성 폐결핵 환자에서의 결핵균 DNA 검출방법에 관한 연구

Polymerase chain reaction (PCR) is the most sensitive and rapid method for the diagnosis of M.tuberculosis infection. Many reports have shown different methods for mycobacterial DNA extraction, but revealed many differences in simplicity and time-consumption. We studied which method was the best among 7 different ones for DNA extraction from 32 smear-positive sputa, using PCR of targeting 123bp DNA fragment of IS6110. The methods were the following : SDS-Microwave oven method, NaOH Lysis method, Triton X-100, Proteinase K method, Lysis buffer method. SDS-Proteinase K method, Bead Beater method, and Bead Beater/CTAB method. The amplification products were identified by electrophoresis on ethidium bromide-stained agarose gels, and confirmed by restriction enzyme, Southern blot hybridization, and DNA sequencing. All 32 cases were positive (100%) by the 7 different methods. It took 1 hour or more to detect DNA in SDS-Microwave oven method, NaOH Lysis method, and Bead beater/CTAB method. It took 2 hours or less in the others. SDS-Proteinase K method and Bead Beater/CTAB method were simpler and more convinient than the others. These results suggest that SDS-Proteinase K method and Bead Beater/CTAB method might be better than the others for the extraction of mycobacterial DNA in terms of simplicity and time-consumption, although all the methods were sensitive.
Cu/GDC Cermet Anodes by Sintering via Surface Modification with MgO Sol

<P>CuO/GDC composite powder with 50 wt% of CuO was prepared by surface modification of ~60 nm GDC powder with Cu precursors. Since copper oxide melts at lower temperature than GDC sintering temperature, fabrication procedure was modified by inducing infiltration of molten copper oxide via capillary force and then followed by heat treatment at ~1000. Surface modification was carried out with a MgO sol to suppress agglomeration of GDC. Such prepared Cu/GDC cermets showed uniform microstructure and excellent electronic conductivity of ~8500 S/cm for the Cu/GDC cermet and ~10200 S/cm for the modified one at 800.</P>
Understanding the Teacher`s Self-efficacy and Middle School Classroom Social Dynamics

The purpose of this study was to examine the relationship between the teacher’s self-efficacy and classroom social dynamics in a middle-school setting. Seven middle-school classes were extracted from a larger dataset collected in 2017 as part of the ClassNet program. A total of seven homeroom teachers and 157 students in their classrooms were analyzed for this study. Peer nominations were used to measure classroom social dynamics, while a self-report measure was used to assess teacher self-efficacy both at the beginning and at the end of the first semester. This study found that students who were perceived as receiving the teacher’s positive support were more likely to be liked, perceived as popular, pro-social, defenders of bullied students, and leaders by their classmates. For classrooms where the teacher’s self-efficacy increased, popular students were likely to be considered as relationally aggressive at the beginning of the semester but by the end of the semester, this correlation was no longer significant. For classrooms where the teacher’s self-efficacy decreased over the semester, by the end of the semester, on average more students exhibited aggressive behavior and were victims of aggression. In contrast, for classrooms where the teacher’s self-efficacy increased over the semester, by the end of the semester, on average fewer students were considered to exhibit aggressive behaviors and were perceived as victims of aggression.
