        Comparative Transcriptome Analysis of Rhizoctonia solani-resistant and -Susceptible Rice Cultivars Reveals the Importance of Pathogen Recognition and Active Immune Responses in Host Resistance

        Rice sheath blight (SB), caused by Rhizoctoniasolani (R. solani), is a major threat to rice productionworldwide. The molecular mechanisms of the SB resistancein rice are poorly understood. The transcriptomes of the SBresistantrice cultivar YSBR1 and the susceptible cultivarLemont were analyzed after R. solani infection. A total of7624 differentially expressed genes (DEGs) were identifiedat one or more timepoints in a cultivar. 5526 and 5618 DEGswere differentially expressed in Lemont and YSBR1,respectively. YSBR1 exhibited stronger and earlier transcriptionalresponse to R. solani than Lemont. Gene ontology enrichmentanalysis revealed that genes that encode cell wall-modifyingand glycosyl-degrading enzymes or anti-microbial proteinswere specifically induced in YSBR1 at 6 hpi. MapMananalysis revealed that more DEGs related with cell wall, β-glucanses, respiratory burst, phenylpropanoids and ligninwere highly induced by R. solani in YSBR1 than in Lemont. The results also showed that receptor-like kinases and jasmonicacid signaling may play important roles in host resistance toR. solani. This study highlights potential candidate genes andsignaling pathways involved in rice sheath resistance and canhelp to further clarify the mechanistic events underlyingresistance and susceptibility to R. solani.

        Robust Stabilization of Singular Markovian Jump Systems with Uncertain Switching

        This paper focuses on the robust stabilization problem for a class of singular Markovian jump systems with uncertain switching probabilities. Based on a slack matrix method on transition probabilities, a new criterion for quadratically stochastic admissibility of such an uncertain system is established. Then, two new sufficient conditions for the existence of mode-dependent controller are given as linear matrix inequalities. Especially, a more practical controller named as mode-independent controller is derived by a mode-dependent Lyapunov function. Finally, a numerical example is used to demonstrate the effectiveness of the proposed methods.

        A new model and testing verification for evaluating the carbon efficiency of server

        To cope with the risks of climate change and promote the realization of carbon peaking and carbon neutrality, this paper first comprehensively considers the policy background, technical trends and carbon reduction paths of energy conservation and emission reduction in data center server industry. Second, we propose a computing power carbon efficiency of data center server, and constructs the carbon emission per performance of server (CEPS) model. According to the model, this paper selects the mainstream data center servers for testing. The result shows that with the improvement of server performance, the total carbon emissions are rising. However, the speed of performance improvement is faster than that of carbon emission, hence the relative carbon emission per unit computing power shows a continuous decreasing trend. Moreover, there are some differences between different products, and it is calculated that the carbon emission per unit performance is 20-60KG when the service life of the server is five years.

        Molecular Characterization and Expression Pattern of Gene IGFBP-5 in the Cashmere Goat (Capra hircus)

        Insulin-like growth factor-binding protein-5 (IGFBP-5) is one of the six members of IGFBP family, important for cell growth, apoptosis and other IGF-stimulated signaling pathways. In order to explore the significance of IGFBP-5 in cells of the Inner Mongolian Cashmere goat (Capra hircus), IGFBP-5 gene complementary DNA (cDNA) was amplified by reverse transcription polymerase chain reaction (RT-PCR) from the animal's fetal fibroblasts and tissue-specific expression analysis was performed by semi-quantitative RT-PCR. The gene is 816 base pairs (bp) in length and includes the complete open reading frame, encoding 271 amino acids (GenBank accession number JF720883). The full cDNA nucleotide sequence has a 99% identity with sheep, 98% with cattle and 95% with human. The amino acids sequence shares identity with 99%, 99% and 99%, respectively. The bioinformatics analysis showed that IGFBP-5 has an insulin growth factor-binding protein homologues (IB) domain and a thyroglobulin type-1 (TY) domain, four protein kinase C phosphorylation sites, five casein kinase II phosphorylation sites, three prenyl group binding sites (CaaX box). The IGFBP-5 gene was expressed in all the tested tissues including testis, brain, liver, lung, mammary gland, spleen, and kidney, suggesting that IGFBP-5 plays an important role in goat cells.

        Cardiac-targeted delivery of nuclear receptor RORα via ultrasound targeted microbubble destruction optimizes the benefits of regular dose of melatonin on sepsis-induced cardiomyopathy

        Background Large-dose melatonin treatment in animal experiments was hardly translated into humans, which may explain the dilemma that the protective effects against myocardial injury in animal have been challenged by clinical trials. Ultrasound-targeted microbubble destruction (UTMD) has been considered a promising drug and gene delivery system to the target tissue. We aim to investigate whether cardiac gene delivery of melatonin receptor mediated by UTMD technology optimizes the efficacy of clinically equivalent dose of melatonin in sepsis-induced cardiomyopathy. Methods Melatonin and cardiac melatonin receptors in patients and rat models with lipopolysaccharide (LPS)- or cecal ligation and puncture (CLP)-induced sepsis were assessed. Rats received UTMD-mediated cardiac delivery of RORα/cationic microbubbles (CMBs) at 1, 3 and 5 days before CLP surgery. Echocardiography, histopathology and oxylipin metabolomics were assessed at 16–20 h after inducing fatal sepsis. Results We observed that patients with sepsis have lower serum melatonin than healthy controls, which was observed in the blood and hearts of Sprague–Dawley rat models with LPS- or CLP-induced sepsis. Notably, a mild dose (2.5 mg/kg) of intravenous melatonin did not substantially improve septic cardiomyopathy. We found decreased nuclear receptors RORα, not melatonin receptors MT1/2, under lethal sepsis that may weaken the potential benefits of a mild dose of melatonin treatment. In vivo, repeated UTMD-mediated cardiac delivery of RORα/CMBs exhibited favorable biosafety, efficiency and specificity, significantly strengthening the effects of a safe dose of melatonin on heart dysfunction and myocardial injury in septic rats. The cardiac delivery of RORα by UTMD technology and melatonin treatment improved mitochondrial dysfunction and oxylipin profiles, although there was no significant influence on systemic inflammation. Conclusions These findings provide new insights to explain the suboptimal effect of melatonin use in clinic and potential solutions to overcome the challenges. UTMD technology may be a promisingly interdisciplinary pattern against sepsis-induced cardiomyopathy.

        Application of Intraoperative Ultrasonography for Guiding Microneurosurgical Resection of Small Subcortical Lesions

        Objective: We wanted to evaluate the clinical value of intraoperative ultrasonography for real-time guidance when performing microneurosurgical resection of small subcortical lesions. Materials and Methods: Fifty-two patients with small subcortical lesions were involved in this study. The pathological diagnoses were cavernous hemangioma in 25 cases, cerebral glioma in eight cases, abscess in eight cases, small inflammatory lesion in five cases, brain parasite infection in four cases and the presence of an intracranial foreign body in two cases. An ultrasonic probe was sterilized and lightly placed on the surface of the brain during the operation. The location, extent, characteristics and adjacent tissue of the lesion were observed by high frequency ultrasonography during the operation. Results: All the lesions were located in the cortex and their mean size was 1.3 ± 0.2 cm. Intraoperative ultrasonography accurately located all the small subcortical lesions, and so the neurosurgeon could provide appropriate treatment. Different lesion pathologies presented with different ultrasonic appearances. Cavernous hemangioma exhibited irregular shapes with distinct margins and it was mildly hyperechoic or hyperechoic. The majority of the cerebral gliomas displayed irregular shapes with indistinct margins, and they often showed cystic and solid mixed echoes. Postoperative imaging identified that the lesions had completely disappeared, and the original symptoms of all the patients were significantly alleviated. Conclusion: Intraoperative ultrasonography can help accurately locate small subcortical lesions and it is helpful for selecting the proper approach and guiding thorough resection of these lesions.         Experimental Study of Cooling Speed for Ultra-Thick Steel Plate during the Jet Impinging and Quenching Process

        The quenching temperature drop curve for Q345B steel plate with 84 mm and 170 mm thickness was tested to analyze the distributing regularities and influencing factors of cooling speed for ultra-thick steel plate during the jet impinging and quenching process. The influences for temperature drop, temperature gradient and cooling speed were analyzed under the conditions of 60~100 m3/h water amount, 0.4~1.0 MPa water pressure, transient switching of quenching mode and the distribution of heat exchanger. Threedimensional heat anti transfer model, surface heat transfer coefficient model and thermal physical parameter model were built up by finite element and optimization. The results showed that the deviation of calculated and measured values was less than 4% for temperature drop curve model. The cooling speed of vertical section for 84 mm-thick steel plate was approximately proportional to surface heat transfer coefficient. The influence of surface heat transfer to cooling speed became weak when the thickness was increased. The influences of temperature effect when switching different quenching modes and temperature gradient of vertical section to cooling speed were stronger. The minimum value of cooling speed was about 1.0~1.8oC/s, between H/6 and H/3 region. These data provide the key information for increasing the cooling speed and uniformity.

        Proteomic Profiles of Mouse Neuro N2a Cells Infected with Variant Virulence of Rabies Viruses

        We characterized the proteomes of murine N2a cells following infection with three rabies virus (RV) strains, characterized by distinct virulence phenotypes (i.e., virulent BD06, fixed CVS-11, and attenuated SRV9 strains), and identified 35 changes to protein expression using twodimensional gel electrophoresis in whole-cell lysates. The annotated functions of these proteins are involved in various cytoskeletal, signal transduction, stress response, and metabolic processes. Specifically, a-enolase, prx-4, vimentin, cytokine-induced apoptosis inhibitor 1 (CIAPIN1) and prx-6 were significantly up-regulated, whereas Trx like-1 and galectin-1 were down-regulated following infection of N2a cells with all three rabies virus strains. However, comparing expressions of all 35 proteins affected between BD06-, CVS-11-, and SRV9-infected cells, specific changes in expression were also observed. The up-regulation of vimentin, CIAPIN1, prx-4, and 14-3-3 θ/δ, and downregulation of NDPK-B and HSP-1 with CVS and SRV9 infection were ≥2 times greater than with BD06. Meanwhile, Zfp12 protein, splicing factor, and arginine/serine-rich 1 were unaltered in the cells infected with BD06 and CVS- 11, but were up-regulated in the group infected with SRV9. The proteomic alterations described here may suggest that these changes to protein expression correlate with the rabies virus`` adaptability and virulence in N2a cells, and hence provides new clues as to the response of N2a host cells to rabies virus infections, and may also aid in uncovering new pathways in these cells that are involved in rabies infections. Further characterization of the functions of the affected proteins may contribute to our understanding of the mechanisms of RV infection and pathogenesis.
