        Cloning and Expression of amyE Gene from Bacillus subtilis in Zymomonas mobilis and Direct Production of Ethanol from Soluble Starch

        Two proven secretion signal zmo130 and zmo331 native to Zymomonas mobilis were fused to the N terminal of α-amylase from Bacillus subtilis and transformed into 5 different strains of Z. mobilis separately. It was found that the signal zmo130 could direct the extracellular secretion of the expressed α-amylase with high activity, but zmo331 could not. Fermentation experiments demonstrated that the recombinant Z. mobilis CICC 10225(p130A) exhibited the highest level of ethanol production, which is nearly 50% of the theoretical yield of ethanol from soluble starch,but another recombinant Z. mobilis ATCC 31821(p130A)took the shortest fermentation time of approximately 3days, with the second high level of ethanol yield. The recombined strains in our study could be an important target for the following genetic engineering of next amylase in order to hydrolyze starch completely.

        Meridian Studies in China: A Systematic Review

        Meridian theory is a major part of Chinese medicine and has guided acupuncture and clinical practice for thousands of years. Meridian theory describes many important concepts about the rules of human body function and regulation, but has comparatively huge differences with the basic concepts of modern medicine. These differences have caused deep concern and attracted attention from scholars, both inside and outside of China. The interest in meridian theory lies in determining the structural nature of meridians. Not only is this information still unclear, it is very difficult to achieve clear results in a short period of time. Despite this, the phenomena of meridians can be used as the entry point for meridian studies. After many years of effort, although the physical structure of meridians has not been found, the existence of the meridian phenomena has been fully confirmed. Although there is a lack of morphological evidence for the existence of the meridian,concluding non-existence may be incorrect as morphology techniques develop and structures previously not determined are being found. Since the phenomenon of meridians exists, some biological basis behind its occurrence must be present. This implies that research on meridians needs to continue as research techniques advance and may eventually reveal the biological basis of the meridian phenomenon. In the present review, we analyze the history of meridian studies in China.

      • Macrophage-secreted Exosomes Delivering miRNA-21 Inhibitor can Regulate BGC-823 Cell Proliferation

        Exosomes, membranous nanovesicles, naturally carry bio-macromolecules or miRNA and play impoetant roles in tumor pathogenesis. Here, we showed that macrophages cell-derived exosomes can function as vehicles to deliver exogenous miR-21 inhibitor into BGC-823 gastric cancer cells. Exosomes loaded with miR-21inhibitor significantly increased miR-21 levels in BGC-823, but miR-21inhibitor loaded in exosomes exerted an opposite effect. miRNA transfected with exosomes had less cellular toxicity to host cells compared to conventional transfection methods. The miR-21inhibitor loaded exosomes promoted the migration ability and reduced apoptosis of BGC-823 gastric cancer cells. These observations indicate that miR-21 acts as a tumor promoter by targeting the PDCD4 gene and preventing apoptosis of gastric cancer cells through inhibition of PDCD4 expression. Furthermore, exosome -mediated miR-21 inhibitor delivery resulted in functionally more efficient inhibition and less cellular toxicity compared to conventional transfection methods. Similar approaches could be useful in modification of target biomolecules in vitro and in vivo. These findings contribute to our understanding of the functions of miR-21 and exosomes as a carrier for therapy of gastric cancer.

        Partial oxidation reforming of biomass fuel gas over nickel-based monolithic catalyst with naphthalene as model compound

        With naphthalene as biomass tar model compound, partial oxidation reforming (with addition of O2) and dry reforming of biomass fuel gas were investigated over nickel-based monoliths at the same conditions. The results showed that both processes had excellent performance in upgrading biomass raw fuel gas. Above 99% of naphthalene was converted into synthesis gases (H2+CO). About 2.8 wt% of coke deposition was detected on the catalyst surface for dry reforming process at 750 oC during 108 h lifetime test. However, no coke deposition was detected for partial oxidation reforming process, which indicated that addition of O2 can effectively prohibit the coke formation. O2 can also increase the CH4 conversion and H2/CO ratio of the producer gas. The average conversion of CH4 in dry and partial oxidation reforming process was 92% and 95%, respectively. The average H2/CO ratio increased from 0.95 to 1.1 with the addition of O2, which was suitable to be used as synthesis gas for dimethyl ether (DME) synthesis.

        Preliminary Application of High-Definition CT Gemstone Spectral Imaging in Hand and Foot Tendons

        Objective: To assess the feasibility of visualizing hand and foot tendon anatomy and disorders by Gemstone Spectral Imaging (GSI) high-definition CT (HDCT). Materials and Methods: Thirty-five patients who suffered from hand or foot pain were scanned with GSI mode HDCT and MRI. Spectrum analysis was used to select the monochromatic images that provide the optimal contrast-to-noise ratio (CNR) for tendons. The image quality at the best selected monochromatic level and the conventional polychromatic images were compared. Tendon anatomy and disease were also analyzed at GSI and MRI. Results: The monochromatic images at about 65 keV (mean 65.09 ± 2.98) provided the optimal CNR for hand and foot tendons. The image quality at the optimal selected monochromatic level was superior to conventional polychromatic images (p = 0.005, p < 0.05). GSI was useful in visualizing hand and foot tendon anatomy and disorders. There were no statistical differences between GSI and MRI with regard to tendon thickening (χ2 = 0, p > 0.05), compression (χ2 = 0.5, p > 0.05), absence (χ2 = 0, p > 0.05) and rupture (χ2 = 0, p > 0.05). GSI was significantly less sensitive than MRI in displaying tendon adhesion (χ2 = 4.17, p < 0.05), degeneration (χ2 = 4.17, p < 0.05), and tendinous sheath disease (χ2 = 10.08, p < 0.05). Conclusion: GSI with monochromatic images at 65 keV displays clearly the most hand and foot tendon anatomy and disorders with image quality improved, as compared with conventional polychromatic images. It may be used solely or combined with MRI in clinical work, depending on individual patient disease condition.       • Recent Progress in Methods of Generating Water Mist for Fire Suppression

        To prevent the ozonosphere from being destroyed by Halon, it is an urgent task to find out Halon replacement. As one of the replacements water mist have showed broad applications by its advantages: little pollution to environment (not destroying the ozone layer or bring green house effect), extinguishing fire quickly, consuming a small quantity of water and having little damage to the protected objects. The methods of generating water mist strongly influence fire suppression effectiveness, which determine the cone angle, drop size distribution, flux uniformity, and momentum of the generating spray. The traditional water mist nozzle included pressure jet nozzles, impingement nozzles and twin-fluid nozzles. All of them have more or less disadvantages for fire suppression. Therefore, many research institutes and corporations are taking up with innovations in mist generation. This article provided some recent studies in State Key Laboratory of Fire Science (SKLFS) of University of Science and Technology of China. SKLFS have investigated new methods of generating water mist (i.e. effervescent atomization and ultrasonic atomization), and self developed a series of nozzles and developed advanced DPIVS (Digital Particle Image Velocimetry and Sizing) technique. Characteristics of water mist (the distribution of droplet sizes, flux density, spray dynamics and cone angle) produced by these nozzles were measured under different conditions (work pressure, nozzle geometry, etc.) using LDV/APV and DPIVS systems. A series of experiments were performed to study the fire suppression effectiveness in different fire scenario (different kinds of the fuel, fire size and ventilation conditions). The fire extinguishing mechanisms of water mist was also discussed.

        CONTROL STRATEGY FOR SHIFT SCHEDULE CORRECTION BASED ON DRIVING HABITS FOR VEHICLES WITH AUTOMATIC TRANSMISSION

        A shift schedule modification program is an intelligent system for automatic transmission. This program can adjust shift points to cater to drivers with different driving habits. An important prerequisite in designing a personalized shift schedule is identifying the driving habits of drivers. In this study, we developed an identification algorithm based on wavelet neural network and Bayesian fusion decision-making. First, a system for identifying driving styles was established based on the wavelet neural network. Second, the results were integrated by Bayesian fusion decision-making to obtain the driving habits. Finally, different correction coefficients were selected based on driving habits to satisfy the requirements of drivers. Experimental results show that the driving habits can be accurately identified based on wavelet neural network and Bayesian fusion decision-making, and the correction control strategy can rectify the shift schedule effectively. The correction control strategy satisfies the requirements of different drivers for vehicle performance and enhances the intelligence of automatic transmission.

      • The Modification of Exocyclic Ketone on Methyl(Pyro) pheophorbide-a and Influence with Visible Spectra

        The methyl pheophorbide-a (MP-a) and methyl pyropheophorbide-a (MPP-a) were modified by reaction of exocyclic ketone in E-ring with nucleophilic reagent and several chlorin derivatives were synthesized. The change of the structure in E-ring served an expanding conjugation region and introduction of electron-withdrawing group, which strongly influenced the visible spectra. The Qy bands of synthesized compounds were affected by the substituents on the Qy axis(N$\sub$21/-N$\sub$23/).

        Synthesis, Structure and Biological Properties of a Novel Copper (II) Supramolecular Compound Based on 1,2,4-Triazoles Derivatives

        A novel mononuclear supramolecule of copper(II) has been synthesized with Ippyt ligand (Ippyt=3-(4'- imidazole phenyl)-5-(pyrid-2''-yl)-1,2,4-triazole) (1). Compound 1, namely [Cu(Ippyt)2(H2O)2], has been characterized by single-crystal X-ray diffraction, IR spectrum, elemental analysis and thermogravimetric analysis. Structure determination reveals that the elongated-octahedral geometry is formed in the vicinity of the copper (II) atom being coordinated by four nitrogen atoms from two Ippyt ligands occupying the equatorial position and two oxygen atoms from two coordinated water molecules in the axial position, which together form the N4O2 donor set. Hydrogen bonding interactions between nitrogen and oxygen atoms result in the set up of a supramolecular network architecture. Biological properties including antibacterial activity and superoxide dismutase (SOD) mimetic activity of compound 1 have been investigated by agar diffusion method and the modified Marklund method, respectively. The results indicate that compound 1 exhibits a stronger antibacterial efficiency than the parent ligand and it also has a certain radical-scavenging activity.
