Water plays a crucial role in the synthesis mechanism of metal organic framework-5 (MOF-5). Synthesized MOF-5 with good phase structure and large specific surface area is largely determined by an important synthesis factor: the total water concentration of the initial synthesis solution (Ctw). An understanding of the effects of different and high Ctw on the synthesis of MOF-5 and the investigation of the maximum Ctw suitable for the synthesis of MOF-5 are important to guide the synthesis of MOF-5. Through the research of the maximum Ctw, a favorable synthetic approach was established which could realize the synthesis of MOF-5 with fine performance on CO2 adsorption and separation. The research results show that the maximum Ctw could be as high as 1,440mmol/L, and synthesized MOF- 5 still has a good phase structure and a large specific surface area of 2,136m2/g (BET). Synthesized MOF-5 by the maximum Ctw exhibits a high CO2 adsorption capacity of 2.5mmol/g and a low N2 adsorption capacity of 0.2mmol/g at 298 K and 100 kPa. More importantly, synthesized MOF-5 by the maximum Ctw exhibits a high selectivity for CO2/N2 of 18-22 at 298 K and 20-130 kPa in simulated flue gas.

Backgrounds: Polymorphisms of OPRM1 A118G and ABCB1 C3435T have been suggested to contribute to inter-individual variability regarding pain sensitivity, opioid usage, tolerance and dependence and incidence of adverse effects in patients with chronic pain. This study aimed to investigate the association of both two polymorphisms with opioid requirements in Chinese patients with cancer pain. Methods: The genotypes of rs1799971 (OPRM1) and rs1045642 (ABCB1) were determined by PCR-RFLP and direct sequencing methods respectively in 112 patients with cancer-related pain. Comparisons between the different genotype or allele groups were performed with t-tests or one-way ANOVA tests, as appropriate. The potential relationship of allele number with opioid response was performed with a trend Jonckheere-Terpstra test. Results: In the 112 subjects, the frequencies of variant 118 G and 3435T allele were 38.4% and 37.9%, respectively. Significant higher 24h-opioid doses were observed in patients with GG (P=0.0004) and AG + GG (P=0.005) genotypes than the AA carriers. The dominant mutant 118G allele tended to be associated with progressively increasing 24h-opioiddoses (P=0.001). Compared with CC/CT, patients with ABCB1 TT genotype received higher 24h- and weight-surface area-adjusted-24h- opioids doses (P=0.057 and 0.028, respectively). Conclusions: The OPRM1 A118G single nucleotide polymorphism (SNP) is a key contributor for the inter-individual variability in opioidrequirements in Chinese cancer pain patients. This may possibly extend to the ABCB1 C3435T SNP.

Purpose: The main treatment options of neurogenic bladder remains catheterization and long-term oral medications. Metabolic interventions have shown good therapeutic results in many diseases. To date, no studies have characterized the metabolites of the detrusor muscle during neurogenic bladder. Using metabolomics, new muscle metabolomic signatures were identified to reveal the temporal metabolic profile of muscle during disease progression. Methods: We used 42 Sprague-Dawley rats (200±20 g, males) for T10 segmental spinal cord injury modeling and collected detrusor tissue and performed nontargeted metabolomics after sham surgery, 30-minute, 6-hour, 12-hour, 24-hour, 5-day, and 2-week postmodelling, to identify the dysregulated metabolic pathways and key metabolites. Results: By comparing mzCloud, mzVault, MassList, we identified a total of 1,271 metabolites and enriched a total of 12 metabolism-related pathways with significant differences (P<0.05) based on Kyoto Encyclopedia of Genes and Genomes analysis. Metabolites in several differential metabolic pathways such as ascorbate and aldarate metabolism, Steroid hormone biosynthesis, and carbon metabolism are altered in a regular manner before and after ridge shock. Conclusions: Our study is the first time-based metabolomic study of rat forced urinary muscle after traumatic spinal cord injury, and we identified multiple differential metabolic pathways during injury that may improve long-term management strategies for neurogenic bladder and reduce costs in long-term treatment.

Cadmium (Cd) stress can cause system disorders in plants, affect the absorption of some nutrients. The roles and their relationship of salicylic acid (SA) and hydrogen peroxide (H2O2) in adventitious root formation (ARF) of cucumber (Cucumis sativus L.) under Cd stress were studied. The results showed that the exogenous SA and H2O2 in enhancing ARF in cucumber explants under Cd stress was concentration-dependent, with an optimal biological effect at 0.1 µM SA or 50 µM H2O2. Under Cd stress, SA-induced ARF was significantly reversed by H2O2 scavenger and inhibitor, indicating that endogenous H2O2 may be involved in SA-induced ARF under Cd stress. In addition, endogenous H2O2 levels were also promoted by SA in cucumber explants. The application of SA promoted pectin content and decreased pectin methylesterification degree under Cd stress. Moreover, we founded that SA and SA + H2O2 up-regulated the expression level of CsQUA and CsPME under Cd stress, respectively. However, the application of H2O2 scavenger and inhibitor could significantly reverse the effects of SA on the expression of the above genes. These results suggested that SA might play an important role in ARF in cucumber under Cd stress via enhancing endogenous H2O2 levels.

Berberine is one of active alkaloids from Rhizomacoptidis in traditional Chinese medicine. The pharmacokineticsof berberine in rat plasma were compared betweennormal and chronic visceral hypersensitivity irritable bowelsyndrome rats (CVH-IBS) established by mechanical colonirritation using angioplasty balloons for 2 weeks after oraladministration of berberine hydrochloride (25 mg/kg) withthe equivalent dose of 22 mg/kg for berberine according tobody weight. Immunohistochemical analysis of c-fos andmyosin light chain kinase (MLCK) and immunofluorescenceanalysis of MLCK in rat colon were conducted. Quantificationof berberine in rat plasma was achieved by using a sensitiveand rapidUPLC-MS/MSmethod. Plasma samples werecollected at 15 different points in time and the pharmacokineticparameters were analyzed by WinNonlin software. Thegreat different pharmacokinetic behavior of berberine wasobserved between normal and CVH-IBS model rats. Compared with normal group, T1/2 and AUC(0–t) of berberinein the model group were significantly increased, respectively(573.21 ± 127.53 vs 948.22 ± 388.57 min; 8,657.19 ±1,562.54 vs 11,415.12 ± 1,670.72 min.ng/ml). Cl/F of berberinein the model group significantly decreased, respectively(13.89 ± 1.69 vs 9.19 ± 2.91 L/h/kg). Additionally,the expressions of c-fos and MLCK in model group werehigher than those in normal group. The pharmacokinetic behaviorof berberine was significantly altered in CVH-IBSpathological conditions, which indicated the dosage modificationof berberine hydrochloride in CVH-IBS were necessary. Especially, improved exposure to berberine in ratplasma inCVH-IBSmodel rats was attributed to increased theexpression of MLCK.

Glutathione peroxidase (GPX) is one of the important members of the antioxidant enzyme family. It can catalyze the reduction of hydroperoxides with glutathione to protect cells against oxidative damage. Single-chain variable fragment (scFv) can be converted into seleniumcontaining single-chain variable fragment (Se-scFv) by chemical modification of the hydroxyl groups in scFv, thus Se-scFv possesses GPX activity and becomes a prodrug. To improve the expression of scFv and simplify its purification steps, Single-protein production (SPP) system was used to express scFv and chemical modification was used to synthesize Se-scFv. Therefore, we must construct a new scFv-WCD1-lessACA gene, which can express its mRNA not containing any ACA sequences and express its amino acid sequence of target protein (scFv) being same to scFv-WCD1. In this way, the scFv-WCD1-lessACA can be only expressed in SPP system and no other background proteins in the cells could be expressed. The expression results showed that high level of scFv-WCD1-lessACA synthesis was at least sustained for 96 h in the virtual absence of background protein synthesis. Then, selenocysteine (Sec) was incorporated into the scFv-WCD1-lessACA by chemical modification and resulted in Se-scFv-WCD1-lessACA. The enzymatic characteristics of Se-scFv-WCD1-lessACA were determined. GPX activity was 2,563 U/μmol,its binding constant for GSH was 0.687 ×105/mol. Moreover,Se-scFv-WCD1-lessACA was confirmed to have a strong antioxidant ability to protect mitochondria against oxidative damage induced by Vc/Fe2+ (mitochondrial damage model),suggesting that Se-scFv-WCD1-lessACA has potential application for protection of mitochondrial damage induced by reactive oxygen species (ROS).
A Target-Oriented Path Generation Method based on Concolic Testing

The space explosion caused by loop structure in path exploration makes it difficult to find a target-oriented path during code coverage testing. In this paper, a method named target-oriented concolic testing(TOCT) is introduced to tackle the problem of generating a path to cover a target in a loop structure. First, the static analysis method static prefix path search (SPSS) is used to generate a prefix path. Solving the constraints on the prefix path obtains a test case, with which concrete execution is carried out as an input. Meanwhile the execution trace is captured. The trace is used to unfold the loop into a feasible path. Finally SPPS is adopted again for the next prefix-path until a feasible path containing the target is achieved. Experimental results show that the proposed strategy reached a higher coverage when the targets are in loop structure and demonstrate that it is applicable in engineering.

Background/Aims A large number of studies have shown that function constipation (FC) has an extremely high incidence of mental and psychological disorders. Cranial electrotherapy stimulation (CES) was applied to the treatment of psychological disorders such as anxiety and depression. We explored the effects of CES combined with biofeedback therapy (BFT) on the psychological state, clinical symptoms, and anorectal function in patients with FC. Methods A total of 74 patients with FC were randomly divided into 2 groups. The control group received BFT. CES combined with BFT was carried out in the experiment group. All patients were assessed using the self-rating anxiety scale (SAS), self-rating depression scale (SDS), and Wexner constipation score at baseline and the end of each course. Anorectal manometry and balloon expulsion tests were performed before and after treatment. Results After treatment, the participants in the experiment group had significantly lower score SAS, SDS, and Wexner constipation scores than the control group (all P < 0.05). The number of successful expulsion in the experiment group was larger than the control group (P = 0.016). Conclusions CES combined with BFT was effective in improving the psychological status of anxiety, depression, and bowel symptoms in patients with FC. (J Neurogastroenterol Motil 2016;22:497-508)
