To evaluated the effect of recombinant adenovirus Ad-ODC-AdoMetDCas which can simultaneously express both antisense ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (AdoMetDC) on cell cycle distribution in colorectal cancer cell and investigated underlying regulatory responses, human colorectal cancer cells HT-29 were cultured in RPMI 1640 medium and infected with Ad-ODC-AdoMetDCas. Cell cycle progression was detected by flow cytometry analysis. The expression levels of cell cycle regulated proteins were measured by Western blot analysis. The mRNA level of cyclin D1 was measured by RT-PCR. And a luciferase reporter plasmid of cyclin D1 promoter was constructed to observe the effect of Ad-ODC-AdoMetDCas on cyclin D1 promoter activity. The results showed that recombinant adenovirus Ad-ODC-AdoMetDCas significantly induced $G_1$ arrest, decreased levels of cyclin D1 protein and mRNA and suppressed the promoter activity. Ad-ODC-AdoMetDCas also inhibited nuclear translocation of $\beta$-catenin. In conclusion, downregulation of ODC and AdoMetDC mediated by Ad-ODC-AdoMetDCas transfection induces $G_1$ arrest in HT-29 cells and the arrest was associated with suppression of cyclin D1 expression and inhibition of $\beta$-catenin nuclear translocation. As a new anticancer reagent, the recombinant adenovirus Ad-ODC-AdoMetDCas holds promising hope for the therapy of colorectal cancers.

Background/Aims There are increasing evidences for gastrointestinal motility disorder (GIMD) and gastric stress ulcer induced by noise stress. The present study was to investigate the reversed effect of melatonin on GIMD and gastric stress ulcer induced by noise stress and potential mechanism. Methods Noise stress was induced on rats, and melatonin (15 mg/kg) was administered to rats by intraperitoneal injection. Differences were assessed in gastric residual rate (GRR), small intestine propulsion rate (SPR), Guth injury score, cortisol, gastrointestinal hormones (calcitonin-gene-related peptide and motilin) and oxidative stress markers (superoxide dismutase and malondialde hyde) in blood plasma as well as gastric mucosa homogenate with or without melatonin. The pathological examination of gastric mucosa was also performed. Results The GRR and SPR were improved by noise stress compared with control (P < 0.05). The pathological examination and Guth injury score revealed gastric stress ulcer. Moreover, the levels of cortisol, motilin and malondialdehyde in blood plasma and malondialdehyde in gastric mucosa homogenate were increased by noise stress (P < 0.05). CGRP and superoxide dismutase activity in both of blood plasma and gastric mucosa homogenate were significantly decreased (P< 0.05). Furthermore, melatonin reversed changes in GRR, SPR, pathological examination, Guth injury score, cortisol, motilin, CGRP, superoxide dismutase activity and malondialdehyde (P < 0.05). Conclusions Melatonin is effective in reversing the GIMD and gastric stress ulcer induced by noise stress. The underlying mechanism may be involved in oxidative stress and gastrointestinal hormones. Identification a novel de novo RUNX2 frameshift mutation associated with cleidocranial dysplasia

Background: Cleidocranial dysplasia (CCD) is a rare genetic disorder affecting bone and cartilage development. Clinical features of CCD comprise short stature, delayed ossification of craniofacial structures with numerous Wormian bones, underdeveloped or aplastic clavicles and multiple dental anomalies. Several studies have revealed that CCD development is strongly linked with different mutations in runt-related transcription factor 2 (RUNX2) gene. Objective: Identification and functional characterization of RUNX2 mutation associated with CCD. Methods: We performed genetic testing of a patient with CCD using whole exome sequencing and found a novel RUNX2 frameshift mutation: c.1550delT in a sporadic case. We also compared the functional activity of the mutant and wild-type RUNX2 through immunofluorescence microscopy and osteocalcin promoter luciferase assay. Results: We found a novel RUNX2 frameshift mutation, c.1550delT (p.Trp518Glyfs*60). Both mutant RUNX2 and wild-type RUNX2 protein were similarly confined in the nuclei. The novel mutation caused abrogative transactivation activity of RUNX2 on osteocalcin promoter. Conclusions: We explored a novel RUNX2 deletion/frameshift mutation in a sporadic CCD patient. This finding suggests that the VWRPY domain may play a key role in RUNX2 transactivation ability.

Purpose: This study investigated the role of natriuretic peptide receptor 2 (NPR2) on cell proliferation and testosterone secre-tion in mouse Leydig cells.Materials and Methods: Mouse testis of different postnatal stages was isolated to detect the expression C-type natriuretic peptide (CNP) and its receptor NPR2 by quantitative reverse transcription polymerase chain reaction (RT-qPCR). Leydig cells isolated from mouse testis were cultured and treated with shNPR2 lentiviruses or CNP. And then the cyclic guanosine mono-phosphate production, testosterone secretion, cell proliferation, cell cycle and cell apoptosis in mouse Leydig cells were ana-lyzed by ELISA, RT-qPCR, Cell Counting Kit-8, and flow cytometry. Moreover, the expression of NPR2, cell cycle, apoptosis proliferation and cell cycle related gene were detected by RT-qPCR and Western blot. Results: Knockdown of NPR2 by RNAi resulted in S phase cell cycle arrest, cell apoptosis, and decreased testosterone secre-tion in mouse Leydig cells.Conclusions: Our study provides more evidences to better understand the function of CNP/NPR2 pathway in male reproduc-tion, which may help us to treat male infertility.

To achieve the clean and efficient utilization of low-rank coal, the combustion and pollutant emission characteristics of chars from low-temperature and fast pyrolysis in a horizontal tube furnace were investigated in a TG-MS analyzer. According to the results, the combustion characteristic of chars was poorer than its parent coals. The temperature range of gaseous product release had a good agreement with that of TGA weight loss. Gaseous products of samples with high content of volatile were released earlier. The NO and NO₂ emissions of chars were lower than their parent coals. Coals of high rank (anthracite and sub-bituminous) released more NO and NO₂ than low rank coals of lignite, so were chars from coals of different ranks. SO₂ emissions of char samples were lower than parent coals and did not show obvious relationship with coal ranks.
Ringing Frequency Extraction Method Based on EMD and FFT for Health Monitoring of Power Transistors

Condition monitoring has been recognized as an effective and low-cost method to enhance the reliability and improve themaintainability of power electronic converters. In power electronic converters, high-frequency oscillation occurs during theswitching transients of power transistors, which is known as ringing. The ringing frequency mainly depends on the values of theparasitic capacitance and stray inductance in the oscillation loop. Although circuit stray inductance is an important factor thatleads to the ringing, it does not change with transistor aging. A shift in either the inside inductance or junction capacitance is animportant failure precursor for power transistors. Therefore, ringing frequency can be used to monitor the health of powertransistors. However, the switching actions of power transistors usually result in a dynamic behavior that can generate oscillationsignals mixed with background noise, which makes it hard to directly extract the ringing frequency. A frequency extractionmethod based on empirical mode decomposition (EMD) and Fast Fourier transformation (FFT) is proposed in this paper. Theproposed method is simple and has a high precision. Simulation results are given to verify the ringing analysis and experimentalresults are given to verify the effectiveness of the proposed method.
Selective and Fast-Response Fluorescent Probes for Hypochlorite and their Application

It is extremely important to develop selective and sensitive fluorescent probes for the detection of hypochlorite (HClO/ClO?). Herein, two ESIPT-based fluorescent probes (BS1 and BS2) were designed and prepared for the fast determination of HClO. The probes exhibited excellent specificity towards HClO and showed the rapid and huge fluorescent responses. Moreover, the probes BS1 and BS2 could sensitively detect HClO in the range of 0?100??M with the low detection limits of 87 and 106?nM, especially. Besides, the success of practical application in water samples and test strips suggested that the synthetic probes may be effective tools for the detection of hypochlorite in environmental samples.
Novel Buck Mode Three-Level Direct AC Converter with a High Frequency Link

A novel family of Buck mode three-level direct ac converters with a high frequency link is proposed. These converters can transfer an unsteady high ac voltage with distortion into a regulated sinusoidal voltage with a low THD at the same frequency. The circuit configuration is constituted of a three-level converter, high frequency transformer, cycloconverter, as well as input and output filters. The topological family includes forward, push-pull, half-bridge, and full-bridge modes. In order to achieve a reliable three-level ac-ac conversion, and to overcome the surge voltage and surge current of the cycloconverter, a phase-shifted control strategy is introduced in this paper. A prototype is presented with experimental results to demonstrate that the proposed converters have five advantages including high frequency electrical isolation, lower voltage stress of the power switches, bi-directional power flow, low THD of the output voltage, and a higher input power factor.
