      • Endoplasmic Reticulum Stress-Mediated Apoptosis of EBV-Transformed B Cells by Cross-Linking of CD70 Is Dependent upon Generation of Reactive Oxygen Species and Activation of p38 MAPK and JNK Pathway

        CD70 is expressed in normal activated immune cells as well as in several types of tumors. It has been established that anti-CD70 mAb induces complement-dependent death of CD70^+ tumor cells, but how anti-CD70 mAb affects the intrinsic signaling is poorly defined. In this report, we show that ligation of CD70 expressed on EBV-transformed B cells using anti-CD70 mAb induced production of reactive oxygen species (ROS) and subsequent apoptosis. We observed an early expression of endoplasmic reticulum (ER) stress response genes that preceded the release of apoptotic molecules from the mitochondria and the cleavage of caspases. CD70-induced apoptosis was inhibited by pretreatment with the ER stress inhibitor salubrinal, ROS quencher N-acetylcysteine, and Ca^(2+) chelator BAPTA. We supposed that ROS generation might be the first event of CD70-induced apoptosis because N- acetylcysteine blocked increases of ROS and Ca^(2+), but BAPTA did not block ROS generation. We also round that CD70 stimu¬lation activated JNK and p38 MAPK. JNK inhibitor SP600125 and p38 inhibitor SB203580 effectively blocked upregulation of ER stress-related genes and cleavage of caspases. Inhibition of ROS generation completely blocked phosphorylation of JNK and p38 MAPK and induction of ER stress-related genes. Taken together, we concluded that cross-linking of CD70 on EBV-transformed B cells triggered ER stress-mediated apoptosis via ROS generation and JNK and p38 MAPK pathway activation. Our report reveals alternate mechanisms of direct apoptosis through CD70 signaling and provides data supporting CD70 as a viable target for an Ab-based therapy against EBV-related tumors.

        Cross-linking of CD80 and CD86 Diminishes Expression of CD54 on EBV-transformed B Cells through Inactivation of RhoA and Ras

        Background: Epstein Barr virus (EBV) infected B cells are transformed into lymphoblastoid cell lines. Some researchers suggested some a few similarities between this process and carcinogenesis. We observed the expression of CD80 and CD86, co-stimulatory molecules on EBV-transformed B cells and changes of CD54 expression after stimulation of CD80 and CD86. Methods: CD80 and CD86 were stimulated using anti-CD80 and anti-CD86 monoclonal antibodies. To assess apoptosis and surface protein expression, flow cytometric analysis was performed. Intracellular signal molecules were evaluated by RT-PCR and immunoblot. Morphology and localization of proteins were examined using inverted or confocal microscope. Results: Cross-linking of CD80 and CD86 induced apoptosis and interfered with proliferation of EBV-transformed B cells, and dispersion of clumped cells. We also examined that their stimulation induced ROS accumulation and reduced CD54 expression. Interestingly, we observed that CD80 and CD86 diminished the expression of CD54 in different methods. Both CD80 and CD86 downregulated activation of focal adhesion kinase. CD80 stimulus inhibited CD54 expression through mainly RhoA inactivation, while CD86 down-regulated Ras and JNK phosphorylation. Conclusion: These results suggest that co-stimulatory CD80 and CD86 molecules, expressed EBV-transformed B cells, may play a role in apoptosis and cell adhesion.

      • Microwave Processing for Recovery of C-14 From Spent Ion Exchange Resin

        Ion exchange resins are commonly employed in the treatment of liquid radioactive waste generated in nuclear power plants (NPP). The ion exchange resin used in NPP is a mixed-bed ion exchange resin known as IRN-150, which is of nuclear grade. This resin is a mixture of cation exchange resin and anion exchange resin. The cation exchange resin removes cationic radionuclides such as Cs and Co, while anion exchange resin handles anions (e.g., H14CO3 -), effectively purifying the liquid waste. Spent ion exchange resins (spent resin) containing C-14 are classified as low and intermediate level radioactive waste, and their radioactivity needs to be reduced as it exceeds the disposal limit regulated by law. Therefore, the microwave technology for the removal of C-14 from spent resin has been investigated. Previous studies have successfully developed a method for the effective removal of C-14 during the resin treatment process. However, it was observed that, in this process, functional groups in the resin were also removed, resulting in the generation of off-gases containing trimethylamine. These off-gases can dissolve in water from process, increasing its pH, which can subsequently hinder the recovery of C-14. In this study, we investigated the high-purity recovery of C-14 by adjusting the moisture content within the reactor following microwave treatment. Mock spent resins, consisting of 100 g of resin with HCO3 - ion-exchanged and 0, 25, or 50 g of deionized water, were subjected to microwave treatment for 40 or 60 minutes. Subsequently, the C-14 desorption efficiency of the mock spent resins was evaluated using an acid stripping process with H3PO4 solution. The functional group status of the mock spent resins was analyzed using 15N NMR spectroscopy. The results showed that the mock spent resins exhibited efficient C-14 recovery without significant functional group degradation. The highest C-14 desorption efficiency was achieved when 25 g of deionized water was used during microwave treatment.

        Accuracy of administrative claim data for gastric adenoma after endoscopic resection

        Background/Aims: Administrative databases provide valuable information for large-cohort studies. This study aimed to evaluate the diagnostic accuracy of an administrative database for resected gastric adenomas. Methods: Data of patients who underwent endoscopic resection for benign gastric lesions were collected from three hospitals. Gastric adenoma cases were identified in the hospital database using International Classification of Diseases (ICD) 10-codes. The non-adenoma group included patients without gastric adenoma codes. The diagnostic accuracy for gastric adenoma was analyzed based on the pathological reports of the resected specimen. Results: Among 5,095 endoscopic resections with codes for benign gastric lesions, 3,909 patients were included in the analysis. Among them, 2,831 and 1,078 patients were allocated to the adenoma and non-adenoma groups, respectively. Regarding the overall diagnosis of gastric adenoma with ICD-10 codes, the sensitivity, specificity, positive predictive value, and negative predictive value were 98.7%, 88.5%, 95.2%, and 96.8%, respectively. There were no significant differences in these parameters between the tertiary and secondary centers. Conclusions: Administrative codes of gastric adenoma, according to ICD-10 codes, showed good accuracy and can serve as a useful tool to study prognosis of these patients in real-world data studies in the future.

      • Selective Treatment of C-14 in Spent Ion Exchange Resin From HWR Using the Microwave

        Mixed-bed ion exchange resin consist of anion exchange resin and cation exchange resin is used to treat liquid radioactive waste in nuclear power plants. C-14 from heavy water reactors (HWR) is adsorbed on the anion exchange resin and is considered intermediate-level radioactive waste. The total amount of radioactivity of C-14 in spent ion exchange resin exceeds the activity limits for the disposal facility. Therefore, it is necessary to reduce the radioactivity through pre-treatment. There are thermal and non-thermal methods for the treatment of spent ion exchange resin. However, destructive methods have the problem of emitting off-gas containing radionuclides. To solve this challenge, various methods have been developed such as acid stripping, PLO process, activity stripping, thermal treatment and others. In this study, spent ion exchange resin (spent resin) was treated using microwave. The reaction characteristics of the resin to microwave were used to selectively remove the C-14 on the functional groups. Simulated spent anion exchange resin and spent resin from Wolseong NPP were treated with the microwave method, and the desorption rate was over 95%. An integrated process system of 1 kg/batch was built to produce operating data. After the operation of the process, characterization and evaluation of post-treatment for condensate water and adsorbent used in the process were performed. When the process system was applied to treat simulated spent resin and real spent resin, both showed a desorption rated of more than 97%. It means that the C-14 was successfully removed from the radioactive spent resin.

      • Optimization of passion fruit filling formulation using passion fruit, sugar, and pectin by mixture design

        The objective of this study was the optimization of passion fruit (Passiflora edulis) filling formulation using mixture design. The experiment was designed based on the mixture design which includes 9 experimental points with 3 independent variables such as passion fruit, sugar, and pectin. The dependent variables (Brix°, water activity, viscosity, L, a, and b) were measured and used to optimize the formulation. The experimental data of dependent variables were explained with the mixture regression model (R square= 0.88, 0.83, 0.72, 0.98, 0.97, and 0.99, respectively). The optimization results were found to contain passion fruit 56.33%, sugar 35.67%, and pectin 1.00% with desirability equals to 0.8308. These results demonstrate the potentials of applying passion fruit in filling for baking, and present the primary data for application.

        Pre-stimulation of CD81 expression by resting B cells increases proliferation following EBV infection, but the overexpression of CD81 induces the apoptosis of EBV-transformed B cells

        <P>Hepatitis C virus (HCV) E2 protein binds to CD81, which is a component of the B cell co-stimulatory complex. The E2-CD81 interaction leads to B cell proliferation, protein tyrosine phosphorylation and to the hypermutation of immunoglobulin genes. Epidemiological studies have reported a high prevalence of B cell non-Hodgkin lymphoma (NHL) in HCV-positive patients, suggesting a potential association between HCV and Epstein-Barr virus (EBV) in the genesis of B lymphocyte proliferative disorders. In the present study, in order to investigate the association between EBV and HCV in B cells, we created an <I>in vitro</I> EBV-induced B cell transformation model. CD81 was gradually overexpressed during transformation by EBV. B cells isolated from HCV-positive patients grew more rapidly and clumped together earlier than B cells isolated from healthy donors following EBV infection. Pre-stimulation of CD81 expressed by resting B cells with anti-CD81 monoclonal antibody (mAb) or HCV E2 accelerated the generation of lymphoblastoid cell lines (LCLs) by EBV infection. These cells proliferated prominently through the early expression of interleukin-10 and intracellular latent membrane protein (LMP)-l. By contrast, the overexpression of CD81 on EBV-transformed B cells by anti-CD81 mAb or HCV E2 protein induced apoptosis through reactive oxygen species (ROS)-mediated mitochondrial dysfunction. These results suggest that the engagement of CD81 expressed by B cells has differential effects on B cell fate (proliferation or apoptosis) according to EBV infection and the expression level of CD81.</P>

        부산지역 중ㆍ고등학생의 김치 섭취 실태 및 기능성 김치 개발에 관한 태도 조사

        본 연구는 부산지역 중ㆍ고등학교 남녀 학생 833명을 대상으로 김치 섭취 실태 및 기능성 김치 개발에 대한 태도를 설문조사 하였다. 조사대상자는 중학생이 65.2%, 고등학생이 34.8%, 남학생이 51.3%, 여학생이 48.7%였다. 김치의 가장 선호하는 맛으로는 ‘매운맛’, 가장 선호하지 않는 맛으로는 ‘신맛’이라고 하였다. 주로 김치를 섭취하는 장소로는 집이 64.3%, 학교가 33.7%였다. 배추김치의 숙성 정도로는 ‘잘 익은 김치’, 김치 섭취량은 ‘하루 두 번 섭취’, 한 끼에 섭취하는 김치의 양은 ‘3~4조각’이 가장 높게 나타났다. 김치를 섭취하는 이유는 ‘맛이 있어서’가 가장 높았고, ‘습관적으로’, ‘건강에 좋아서’ 순으로 높게 나타났으며, 섭취하지 않는 이유로 ‘입맛에 맞지 않아서’라고 하였다. 학교급식용 김치에 대해 전체 응답자 중 65.1%가 ‘학생들의 입맛을 고려한 김치 개발’을 요구하였고, 향후 개발 김치에 대한 섭취의향으로 맛이 개선된 김치를 제공하면 김치 섭취량이 ‘증가할 것이다’라는 응답이 80.6%, 기능성이 개선된 김치를 제공하면 김치 섭취량이 ‘증가할 것이다’라는 응답이 65.2%로 나타났다. 또한 김치에 첨가하고 싶은 과일 부재료로 배(41.3%), 사과(24.6%), 파인애플(10.3%)로 나타났고, 학생들이 원하는 기능성 증진의 종류로는 ‘성장 발달’이 59.5%, ‘체중 감량’이 40.4%, ‘학습능력 개선’이 32.8%, ‘질병 예방’이 15.5%였다. 이상에서 청소년의 기호와 요구에 맞는 김치개발을 통해 청소년의 김치 섭취와 기능성 김치에 대한 관심이 점차적으로 증가될 것으로 기대된다. The intake patterns and attitudes towards development of functional kimchi among middle and high school students in Busan area were studied. Nine hundred respondents, consisting of middle and high school students, were surveyed. A total of 833 completed copies of the questionnaire were used in this research. Respondents preferred kimchi due to its ‘hot taste’ (49.2%) and ‘salty taste’ (17.9%), but they did not like kimchi due to its ‘sour taste’ (33.8%) and ‘jeotgal taste’ (22.9%). Most students (64.3%) consumed kimchi at home, 68.1% of students ate kimchi more than twice a day, and 27.4% of students consumed 3∼4 pieces (size of a piece: 3×4 cm) of kimchi per meal. Students showed a strong desire to develop kimchi with a good taste at school food services (65.1%). They also showed intentions to increase intake of kimchi if ‘taste of kimchi’ (80.6%), and ‘health functionality of kimchi’ (65.2%) are strengthened. Students wanted kimchi to contain the following fruits: pears (41.3%), apples (24.6%) and pineapples (10.3%). Finally, students showed a desire to develop kimchi that helps growth and development (59.5%), weight loss (40.4%), and learning ability improvement (32.8%). This study concludes that it is necessary to increase kimchi intake in school foodservices by developing new kimchi recipes that meet students’ desires and by educating students about the health benefits of kimchi.
