<P>Glioma cells with stem cell properties, termed glioma stem-like cells (GSCs), have been linked to tumor formation, maintenance, and progression and are responsible for the failure of chemotherapy and radiotherapy. Because conventional glioma treatments often fail to eliminate GSCs completely, residual surviving GSCs are able to repopulate the tumor. Compounds that target GSCs might be helpful in overcoming resistance to anticancer treatments in human brain tumors. In this study, we showed that 5-bromo-3-(3-hydroxyprop-1-ynyl)-2H-pyran-2-one (BHP), a new 2-pyrone derivative, suppressed the maintenance of the GSC population in both a glioma cell line and patient-derived glioma cells. Treatment of GSCs with BHP effectively inhibited sphere formation and suppressed the CD133(+) cell population. Treatment with BHP also suppressed expression of the stemness-regulating transcription factors Sox2, Notch2, and β-catenin in sphere-cultured glioma cells. Treatment of GSCs with BHP significantly suppressed two fundamental characteristics of cancer stem cells: self-renewal and tumorigenicity. BHP treatment dramatically inhibited clone-forming ability at the single-cell level and suppressed in vivo tumor formation. BHP markedly inhibited both phosphoinositide 3-kinase/Akt and Ras/Raf-1/extracellular signal-regulated kinase signaling, which suggests that one or both of these pathways are involved in BHP-induced suppression of GSCs. In addition, treatment with BHP effectively sensitized GSCs to chemotherapy and radiotherapy. Taken together, these results indicate that BHP targets GSCs and enhances their sensitivity to anticancer treatments and suggest that BHP treatment may be useful for treating brain tumors by eliminating GSCs.</P>

Arachnoid cysts are usually asymptomatic and discovered incidentally. However, cysts may occasionally rupture because of minor head trauma. We describe the radiologic follow-up of 5 patients with ruptured arachnoid cysts featuring spontaneous resolution, subdural hygroma formation, and cystic and subdural hemorrhage. From January 2004 through July 2020, 5 patients (1.3%) with ruptured arachnoid cysts were evaluated out of 388 patients with arachnoid cysts encountered at our institution at that time. The 5 patients were all male, and they ranged in age from 6-17 years (median, 12 years). The median duration of radiologic follow-up was 3.5 years (range, 2.3-10.1 years). All of the ruptured arachnoid cysts were overlying the temporal lobe with Galassi type II. The median cyst diameter was 4.9 cm (range, 4.4-8.9 cm). Four patients had a history of recent minor head trauma. There were no particular neurologic symptoms in their past medical history in all patients. In the follow-up, two patients’ cysts resolved spontaneously without hemorrhage. One patient’s cyst resolved post-burr-hole drainage for chronic subdural hemorrhage. Another patient, whose cyst led to a hemorrhage and chronic subdural hemorrhage, recovered following a craniotomy, hematoma removal, and cyst fenestration. Another patient, presenting with hygroma, cystic hemorrhage, and chronic subdural hemorrhage, was treated with burr-hole drainage. Three patients recovered postoperatively. Arachnoid cysts rarely rupture, and surgical intervention is required for some cases associated with hemorrhage. Postoperatively, all patients had good outcomes without complications in this series.

<P><B>Abstract</B></P> <P>In this study, bicarbonate was proposed as an alternative carbon source to overcome exceedingly low CO<SUB>2</SUB> fixation efficiency of conventional microalgae cultivation system. 5gL<SUP>−1</SUP> of sodium bicarbonate was found to well support the growth of <I>Dunaliella salina</I>, showing 2.84-fold higher specific growth rate than a bicarbonate-free control. This bicarbonate-fed cultivation also could yield biomass productivity similar to that of CO<SUB>2</SUB>-based system as long as pH was controlled. While the supplied CO<SUB>2</SUB>, because of its being a gas, was mostly lost and only 3.59% of it was used for biomass synthesis, bicarbonate was effectively incorporated into the biomass with 91.40% of carbon utilization efficiency. This study showed that the bicarbonate-based microalgae cultivation is indeed possible, and can even become a truly environment-friendly and workable approach, provided that a CO<SUB>2</SUB> mineralization technology is concomitantly established.</P> <P><B>Highlights</B></P> <P> <UL> <LI> To utilize bicarbonate as carbon source for microalgae cultivation was investigated. </LI> <LI> The specific growth rate of <I>Dunaliella</I> sp. was significantly increased with NaHCO<SUB>3</SUB>. </LI> <LI> pH control was important to obtain high biomass productivity with bicarbonate. </LI> <LI> NaHCO<SUB>3</SUB> positively affected not only for growth but also for carotenoid production. </LI> <LI> With bicarbonate, carbon utilization efficiency was apparently higher than with CO<SUB>2</SUB>. </LI> </UL> </P>

<P><B>Abstract</B></P> <P>This study investigated if an effluent from anaerobic digestion (AD) system can be used as a nutrients source for the microalgae cultivation, and in so doing, if the effluent can be properly treated. Nitrogen and phosphorus in the AD effluent well supported microalgal growth, and their removal efficiency reached >97.9% and 99.2%, respectively. Among four different algal species tested, <I>Micractinium inermum</I> particularly stood out, showing the highest biomass and FAME productivity: 0.16gL<SUP>−1</SUP> d<SUP>−1</SUP> with 3.23gL<SUP>−1</SUP> of dry cell weight, and 0.04gL<SUP>−1</SUP> d<SUP>−1</SUP> with 27.54% (w/w) of FAME contents, respectively. As the concentrations of the nutrients decreased over time, the FAME contents were increased and its quality as well, satisfying several biodiesel quality standards. This study supports that the AD effluent can indeed serve as a cheap and nutrient-rich medium for microalgae cultivation, and equally importantly, microalgae can be a workable treatment option for it.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Wastewater treatment was synergistically combined with biodiesel production. </LI> <LI> Anaerobic digester effluent was used as a nutrients source for microalgae. </LI> <LI> Nitrogen and phosphorus were successfully removed by microalgae. </LI> <LI> Among four algal species tested, <I>Micractinium inermum</I> showed the best efficiency. </LI> <LI> Both quantity and quality of microalgal lipid were improved with the passage of time. </LI> </UL> </P>

Erwinia amylovora is a causal agent of fire blight which is the most devastating disease of apple, pear and other rosaceous plants. In the course of developing for new antibiotics to control E. amylovora, we discovered that the culture filtrate of soil bacterium KRICT-B035 exhibits a promising antibacterial activity. The 16S rDNA based phylogenetic analysis revealed that this strain was most closely related to Paenibacillus elgii. Through a series of chromatographic procedures, active compounds were isolated. The ESI-MS and NMR analyses were performed on the purified compounds. In combination with literature data, the chemical structures of active compound were identified pelgipeptins. Pelgipeptins showed the potent antibacterial activity against E. amylovora with minimum inhibitory concentration values ranged from 12.5 to 25μg/ml. These results indicated that pelgipeptins could be used as agricultural antibiotics for fire blight disease control.
Existence of glioma stroma mesenchymal stemlike cells in Korean glioma specimens.

<P>It was presented that mesenchymal stem cells (MSCs) can be isolated from western glioma specimens. However, whether MSCs exist in glioma specimens of different ethnicities is unknown. To verify the existence of MSCs in an independent cohort, we undertook studies to isolate MSCs from a group of Korean patients. We hypothesized that cells resembling MSCs that were deemed mesenchymal stemlike cells (MSLCs) exist in an independent cohort of Korean gliomas. We cultured fresh glioma specimens using the protocols used for culturing MSCs. The cultured cells were analyzed with fluorescence-activated cell sorting (FACS) for surface markers associated with MSCs. Cultured cells were exposed to mesenchymal differentiation conditions. To presume possible locations of MSLCs in the glioma, sections of glioma were analyzed by immunofluorescent labeling for CD105, CD31, and NG2. From nine of 31 glioma specimens, we isolated cells resembling MSCs, which were deemed Korean glioma stroma MSLCs (KGS-MSLCs). KGS-MSLCs were spindle shaped and adherent to plastic. KGS-MSLCs had similar surface markers to MSCs (CD105(+), CD90(+), CD73(+), and CD45(-)). KGS-MSLCs were capable of mesenchymal differentiation and might be located around endothelial cells, pericytes, and in a disorganized perivascular area inside glioma stroma. We found that cells resembling MSCs indeed exist in an independent cohort of glioma patients, as presented in western populations. We could presume that the possible location of KGS-MSLCs was in perivascular area or in glioma stroma that was a disorganized vascular niche. It might be possible that KGS-MSLCs could be one of constituent of stroma of glioma microenvironment.</P>
