      • Activation of Ets‐2 by oxidative stress induces Bcl‐xL expression and accounts for glial survival in amyotrophic lateral sclerosis

        <P>Amyotrophic lateral sclerosis (ALS) is an adult-onset neurodegenerative disease characterized by selective degeneration of motor neurons and glial activation. Cell-specific transcriptional regulation induced by oxidative stress may contribute to the survival and activation of astrocytes in the face of motor neuron death. In the present study, we demonstrate an age-dependent increase in Bcl-xL and Ets-2 immunoreactivity that correlates with an increase of glial fibrillary acidic protein (GFAP)-positive cells in the ventral horn of the spinal cord in both ALS transgenic mice [mutant SOD1 (G93A)] and affected humans. Chromatin immunoprecipitation (ChIP) analysis verified that Ets-2 preferentially occupies the Ets-2 binding element in the promoter of Bcl-xL in primary astrocytes under oxidative stress conditions as well as in G93A spinal cords. Ets-2 small-interfering RNA down-regulated the transcriptional activity of Bcl-xL. In primary glial cultures, Bcl-xL overexpression and mutant SOD1 (G93A) both conferred resistance to oxidative stress-induced cell death. Our findings suggest that Ets-2 transcription factor activation of Bcl-xL gene may protect glia from constitutive oxidative stress that is thought to be a key mechanism contributing to the pathogenesis of ALS. This survival pathway may contribute to the glial survival and activation seen in the spinal cord of ALS patients.</P>

        The Burden of Rhinitis and Rhinoconjunctivitis in Adolescents

        Purpose: Rhinitis and conjunctivitis are common diseases worldwide that are frequently associated. Nevertheless, the risk factors for rhinoconjunctivitis are not well-described and the impact of conjunctivitis on rhinitis and asthma in children remains unknown. This study explored the different risk factors and evaluated the burden of rhinoconjunctivitis among adolescents. Methods: This was a cross-sectional study conducted on a random sample of schoolchildren, aged 10-17 years, using skin prick tests and a self-administered questionnaire on respiratory health investigating the impact of rhinitis and rhinoconjunctivitis on daily activities. Results: A complete evaluation was obtained for 2,150 children. The prevalence of rhinitis alone was 18.2% and rhinitis associated with conjunctivitis was 20.5%. Rhinoconjunctivitis was more frequently associated with females, a parental history of atopy, domestic exposure to mold/dampness, passive smoke exposure, and reported truck traffic in residential streets. Moreover, rhinoconjunctivitis was associated with a higher level of allergic sensitization. The prevalence of current asthma was 1.7% in subjects without rhinitis or rhinoconjunctivitis, 5.1% in rhinitis and 10.7% in rhinoconjunctivitis. In a logistic model, rhinoconjunctivitis yielded a 2-fold risk for current asthma with respect to rhinitis. Subjects with rhinoconjunctivitis had poorer quality of life (QoL); there was an impact on daily activities in 4.6% of rhinitis and 10.7% of rhinoconjunctivitis. Conclusions: Ocular symptoms increase the role of rhinitis as a risk factor for asthma and its impact on daily activities in children.

        Increasing the functional quality of Crocus sativus L. by-product (tepals) by controlling spectral composition

        Crocus sativus L. is a crop grown for spice production, and large amounts of residues from the flowers are produced during the process. The underutilized by-product from saffron spice production, the C. sativus tepals, was investigated as a promising raw material of natural bioactive compounds using light spectrum manipulation in controlled environments. The plants were grown under either light-emitting diodes (LEDs) or natural light (NL, greenhouse). LED experiments were performed in controlled-environment chambers (120 µmol m–2 s–1of photosynthetically active radiation, 18 °C, 16-h photoperiod). The LED treatments used were as follows: (i) red ʎ = 660 nm (62%) and blue ʎ = 450 nm (38%) (RB); and (ii) red ʎ = 660 nm (50%), green ʎ = 500–600 nm (12%), and blue ʎ = 4 50 nm (38%) (RGB). Flower growth parameters, total phenols, total flavonoids, flavonols, flavonol glycosides, and antioxidant properties were measured in harvested tepals. Floral by-products from plants grown under the two LED treatments accumulated higher amounts of antioxidant compounds compared to those of plants grown under NL. The total flavonoids content was significantly enhanced in the RGB LED treatment, while the corolla fresh weight significantly declined in the same treatments. The higher content of bioactive secondary metabolites in plants grown under both RB and RGB light environments resulted in increased antioxidant capacity measured by DPPH free-radical scavenging capacity and the ferric reducing antioxidant power method. These results indicate that manipulation of LED spectra could boost secondary metabolites and antioxidant capacity to obtain phytochemically enriched floral by-products with superior functional quality.

        An efficient methodology for the simulation of nonlinear irregular waves in computational fluid dynamics solvers based on the high order spectral method with an application with OpenFOAM

        An efficient methodology for simulating nonlinear irregular waves in a Computational Fluid Dynamics (CFD) solver is proposed. The High Order Spectral (HOS) method is used to generate nonlinear irregular waves in an open ocean and a numerical wave tank. The inverse Fast Fourier Transforms (FFTs) and multi-dimensional interpolation from the HOS simulation results are used for the efficient reconstruction of nonlinear waves in the CFD solver. The proposed procedure is published as an open-source project called Grid2Grid, which is developed to interface with a generic CFD solver. It provides the function Application Programming Interface (API), which can communicate with different programming languages. Extreme wave events were used to validate the proposed procedure. The predicted wave breaking events are reported both in OpenFOAM CFD and HOS simulations, and the wave elevations of CFD during simulations show good agreement with experiments and with HOS simulation.

        Smartphone App in Stroke Management: A Narrative Updated Review

        The spread of smartphones and mobile-Health (m-health) has progressively changed clinical practice, implementing access to medical knowledge and communication between doctors and patients. Dedicated software called Applications (or Apps), assists the practitioners in the various phases of clinical practice, from diagnosis to follow-up and therapy management. The impact of this technology is even more important in diseases such as stroke, which are characterized by a complex management that includes several moments: primary prevention, acute phase management, rehabilitation, and secondary prevention. This review aims to evaluate and summarize the available literature on Apps for the clinical management of stroke. We described their potential and weaknesses, discussing potential room for improvement. Medline databases were interrogated for studies concerning guideline-based decision support Apps for stroke management and other medical scenarios from 2007 (introduction of the first iPhone) until January 2022. We found 551 studies. Forty-three papers were included because they fitted the scope of the review. Based on their purpose, Apps were classified into three groups: primary prevention Apps, acute stroke management Apps, and post-acute stroke Apps. We described the aim of each App and, when available, the results of clinical studies. For acute stroke, several Apps have been designed with the primary purpose of helping communication and sharing of patients’ clinical data among healthcare providers. However, interactive systems Apps aiming to assist clinicians are still lacking, and this field should be developed because it may improve stroke patients’ management.

        ^(237)Np(n,f) Cross Section: New Data and Present Status

        In this document, we present the final result obtained at the n_TOF experiment for the neutron-induced fission cross section of the ^(237)Np, from the fission threshold up to 1 GeV. The method applied to get this result is brie y discussed. n_TOF data are compared to the last experimental measurements using other TOF facilities or the surrogate method, reported experiments performed with monoenergetic sources and the FISCAL systematic, including a discussion about the existing discrepancies.

        High-energy Neutron-induced Fission Cross Sections of Natural Lead and Bismuth-209

        The CERN Neutron Time-Of-Flight (n_TOF) facility is well suited to measure small neutron-induced fission cross sections, as those of subactinides. The cross section ratios of ^(nat)Pb and ^(209)Bi relative to ^(235)U and ^(238)U were measured using PPAC detectors. The fragment coincidence method allows to unambiguously identify the fission events. The present experiment provides the first results for neutron-induced fission up to 1 GeV for ^(nat)Pb and ^(209)Bi. A good agreement with previous experimental data below 200 MeV is shown. The comparison with proton-induced fission indicates that the limiting regime where neutron-induced and proton-induced fission reach equal cross section is close to 1 GeV.

      • Does Healthy Food Access Matter in a French Urban Setting?

        <P>Limited access to healthy food is commonly regarded as a contributing factor to poor dietary choices. The objective of this article is to test this hypothesis in a French context given France's increasing obesity rates and incidence of poor dietary habits. We use data on fruit and vegetable consumption frequency and different food retail availability measures, for example the number of food stores, food surface area, and a dispersion measure based on store numbers, store types, and food area surface, from several data sources in France. We also employ different types of geographic units when measuring the food retail environment and instrumental variable model specifications to test the robustness of our results, which indicate that fewer but larger retail outlets increase the odds of consuming the recommended level of fruit and vegetables. We also find that an increase in food supply dispersion will improve fruit and vegetable consumption in Paris, but not in its suburbs.</P>

        Antioxidant Effects of Garlic in Young and Aged Rat Brain In Vitro

        Oxidative stress plays a key role in the pathogenesis of age-related neurodegeneration, and the nonenzymatic production of 8-iso-prostaglandin F2α (8-iso-PGF2α) may represent a reliable index of cellular oxidative damage. Garlic (Allium sativum) has been associated with peripheral antioxidant activities and therefore might prevent or reverse 8-iso-PGF2α production, but scant data are available on its possible neuroprotective effects. Therefore, we have studied the possible antioxidant effects of a garlic extract in rat brain synaptosomes obtained from young (3-month-old) and aged (14-month-old) male Wistar rats that were perfused, in vitro, with graded concentrations of a garlic extract (10–500μg/mL). Release in the effluent was evaluated, both in the basal state and after hydrogen peroxide-induced oxidative stress. In young rats, we observed a concentration-dependent inhibitory effect of the garlic extract on brain 8-iso-PGF2α production, both basally and after hydrogen peroxide-induced oxidative stimulus. In aged rats, 8-iso-PGF2α production was not affected by the garlic extract in the basal state, whereas, after hydrogen peroxide-induced oxidative stimulus, an antioxidant effect of the garlic extract appeared only at the higher concentration tested. In conclusion, garlic supplementation could be effective in preventing brain oxidative damage in young animals, whereas the aging brain seems to be resistant to the antioxidant effects of garlic, in vitro.
