        Artificial diet development for mass rearing and its effect on the reproduction of yellow peach moth, Conogethes punctiferalis (Guenée)

        Yellow peach moth, Conogethes punctiferalis(Guenée) (Lepidoptera: Crambidae), is a very destructive fruit borer in Eastern Asia and Australia. A successful artificial diet and rearing protocol are urgently needed for its biological control and research. In this study, we developed and assessed four artificial diets: AD-1, AD-2, AD-3 and AD-4, for rearing yellow peach moth larvae. AD-1 is effective supplements, leading to the larval survival rate of 82.05%, percentage of pupation 79.48%, mean pupa weight of 76.53 mg, emergence rate of 95.25%, and an adult fecundity rate of 121 eggs/female. Using it to feed can shorten the whole generation developmental period to 33 days. Based on this diet AD-1, we also develop a suitable mass rearing method. During the larval stages providing three- to four-time fresh diet can keep a healthy larval colony.

        Research on Fixture Hole Drilling Quality of Printed Circuit Board

        Printed circuit board (PCB) fixture hole is used for installing PCB board or fixing PCB board during drilling process. The drilling quality parameters involve burr size, hole wall roughness et al. In order to improve drilling quality, proper drilling parameters and tool geometries are important especially the tool geometries. In this study, the drilling machinability of PCB fixture hole is studied,effects of drilling parameters on thrust force, drilling temperature and quality are discussed. The experimental results show that resin will be melted during the drilling process. The increase in spindle speed will lead to drilling temperature rise. The fuzzing and spalling of glass fibers result in the generation of hole wall roughness. The exit burrs and enter burrs are generated from drilling copper foils which located in the upper and lower surfaces. The heights of exit burrs are increased with the elevated feed. Drilling experiments are carried out with 9 drill bits which have different point angles, helix angles and length of chisel edges. Analysis of variance (ANOVA) is carried out for hole quality parameters. Desirability function method is used in multiple response optimization to obtain the optimal tool geometry parameters.

      • Mining and Verification of Cold Stress-responsive Genes Correlated with the Cryogenic Autolysis of Volvariella volvacea

        In Volvariella volvacea, a species of edible mushroom, cryogenic autolysis is a typical part of abnormal metabolism. Previous functional annotation cluster analyses of cold-induced gene expression profiles have shown that the ubiquitin-conjugating enzyme E2 (UBE2) and the cyclin-like F-box domain (FBDC) form the functional clusters. Among them, a specific FBDC gene has been found to be in response to cold stress and correlated with the cryogenic autolysis of V. volvacea. In this study, analysis of gene expression profiling showed that only one type of UBE2 in V. volvacea (UBEV2) was significantly up-regulated. Further quantitative real-time PCR analysis confirmed that the expression of UBEV2 was significantly up-regulated (P < 0.05) after cold-treatment lasting 4, 6, and 8 h. Western-bolt analyses confirmed that the protein expression of UBEV2 was in response to cold stress. This provided evidence that UBEV2 was a cold stress-responsive gene and closely correlated with cryogenic autolysis. The specific distribution of UBEV2 in recently diverged herb decay fungi indicated that UBEV2 was not evolutionarily correlated with early diverging fungi. Phylogenetic analysis indicated that UBEV2 was generated by horizontal gene transfer (HGT) from the ancestry of Selaginella moellendorffii UBE2. Further relative time estimation and detection of natural selection showed that there has been recent positive selection after HGT in UBEV2. Molecular modeling and logo analysis showed that the cysteine-cysteine motif is the characteristic of the UBEV2 family. These observations indicate that UBEV2 and FBDV1 are the newly discovered cold stress-responsive genes correlated with the cryogenic autolysis of V. volvacea.

      • Preparation and Characterization of Paclitaxel-loaded PLGA Nanoparticles Coated with Cationic SM5-1 Single-chain Antibody

        The purpose of this study was to develop paclitaxel-loaded poly(lactide-co-glycolide) (PLGA) nanoparticles coated with cationic SM5-1 single-chain antibody (scFv) containing a polylysine (SMFv-polylys). SM5-1 scFv (SMFv) is derived from SM5-1 monoclonal antibody, which binds to a 230 kDa membrane protein specifically expressed on melanoma, hepatocellular carcinoma and breast cancer cells. SMFv-polylys was expressed in Escherichia coli and purified by cation-exchange chromatography. Purified SMFv-polylys was fixed to paclitaxel-loaded PLGA nanoparticles to form paclitaxel-loaded PLGA nanoparticles coated with SMFv-polylys (Ptx-NP-S). Ptx-NP-S was shown to retain the specific antigen-binding affinity of SMFv-polylys to SM5-1 binding protein-positive Ch-hep-3 cells. Finally, the cytotoxicity of Ptx-NP-S was evaluated by a non-radioactive cell proliferation assay. It was demonstrated that Ptx-NP-S had significantly enhanced in vitro cytotoxicity against Ch-hep-3 cells as compared with non-targeted paclitaxel-loaded PLGA nanoparticles. In conclusion, our results suggest that cationic SMFv-polylys has been successfully generated and may be used as targeted ligand for preparing cancer-targeted nanoparticles.

        Molecular identification and functional analysis of Niemann-Pick type C2 protein in Macrocentrus cingulum Brischke (Hymenoptera: Braconidae)

        Niemann-Pick type C2 (NPC2) proteins in arthropods have been extensively differentiated and possibly dupli cated according to environmental conditions and are probable to have different functions. The participation of NPC2 proteins in chemical communication in arthropods brings new objectives in environmental-friendly stra tegies for pest population control. In this study, NPC2 gene in Macrocentrus cingulum (McinNPC2) was newly identified by rapid amplification cDNA ends (RACE) technology. McinNPC2 amino acid sequence alignment with other representative NPC2 annotates to evaluate the highly conserved consensus amino acids, but with odorant binding proteins in M. cingulum show that only one consensus amino acid. Primary six-cysteine structures that are same to odorant binding proteins in M. cingulum were observed in McinNPC2. Phylogenetic analysis of McinNPC2 indicated that the nearest monophyletic group forming one clade with high posterior probability values clusters as Cyphomyrmex costatus (CcosNPC2) whereas the nearest evolutionary relation group as some odorant binding proteins. Moreover, quantitative real-time PCR (qPCR) measurements show that the McinNPC2 gene expression level in various tissues of the female is significantly and ubiquitously higher than in male, whereas the highest expression level in female antennae. We further explore the binding characterization of recombinant McinNPC2 to candidate odor molecules and did the modeling and docking simulations. The results showed ligands binding specificity and docking tests results indicate that β-ionone, an aroma compound commonly found in essential oils, can strongly bind with McinNPC2. In conclusion, we proposed that McinNPC2 may be involved in chemical communication and play roles in perception of plant volatiles.

        High-Precision Tracking Control of a Piezoelectric Micro-nano Platform Using Sliding Mode Control with the Fractional-Order Operator

        In this paper, a sliding mode control using a fractional-order operator (SMCFO) method is presented for the hysteresis of a piezoelectric (PZT) micro-nano platform. At first, the hysteresis of the PZT micro-nano platform is described using the Duhem model. With respect to the sliding mode controller design, we introduce a fractional-order operator into the design of the sliding mode surface and enhance the performance of the proposed controller. The nonlinear switching action of the controller is eliminated through a smooth control action by applying an uncertainty and disturbance estimation technique. The stability of the proposed controller is analyzed using the Lyapunov theorem. Simulations and experimental results demonstrate that the SMCFO method achieves a no-load high precision tracking control of the PZT micro-nano platform and is also robust against load perturbations of the platform

        IN SITU FORMATION OF COBALT NANOCLUSTERS IN SOL–GEL SILICA FILMS FOR SINGLE-WALLED CARBON NANOTUBE GROWTH

        A facile method was developed for in situ formation of Co nanoclusters in sol–gel silica thin films spin-coated on Si wafers. The size and density of Co nanoclusters can be controlled by spin-coating speeds, annealing methods, reduction temperatures under H2, and metal precursor concentrations in tetraethylorthosilicate solutions. The optimized preparation condition, spin-coating speed of 9000 rpm, annealing at 500°C in air followed by reduction at 800°C in H2, resulted in silica films as thin as 60 nm and Co nanoclusters with a mean diameter of 1.5 nm. Morphological and chemical characteristics of thin films and nanoclusters were studied by atomic force microscopy and X-ray photoelectron spectroscopy, respectively. Subsequently, these Co nanoclusters were successfully used to grow SWCNTs by CO decomposition. Film containing Co monometallic clusters produced SWCNTs of 1.3 nm in diameter, whereas film having Co/Mo bimetallic clusters produced SWCNTs of 0.9 nm. This sol–gel approach allowed not only easy catalyst patterning on a thin film but also a fine-tuning of SWCNT properties, e.g., diameter.

        Ameliorative effects of atractylodin on intestinal inflammation and co-occurring dysmotility in both constipation and diarrhea prominent rats

        Intestinal disorders often co-occur with inflammation and dysmotility. However, drugs which simultaneously improve intestinal inflammation and co-occurring dysmotility are rarely reported. Atractylodin, a widely used herbal medicine, is used to treat digestive disorders. The present study was designed to characterize the effects of atractylodin on amelioration of both jejunal inflammation and the co-occurring dysmotility in both constipation-prominent (CP) and diarrhea-prominent (DP) rats. The results indicated that atractylodin reduced proinflammatory cytokines TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 in the plasma and inhibited the expression of inflammatory mediators iNOS and NF-kappa B in jejunal segments in both CP and DP rats. The results indicated that atractylodin exerted stimulatory effects and inhibitory effects on the contractility of jejunal segments isolated from CP and DP rats respectively, showing a contractile-state-dependent regulation. Atractylodin-induced contractile-state-dependent regulation was also observed by using rat jejunal segments in low and high contractile states respectively (5 pairs of low/high contractile states). Atractylodin up-regulated the decreased phosphorylation of 20 kDa myosin light chain, protein contents of myosin light chain kinase (MLCK), and MLCK mRNA expression in jejunal segments of CP rats and down-regulated those increased parameters in DP rats. Taken together, atractylodin alleviated rat jejunal inflammation and exerted contractile-state-dependent regulation on the contractility of jejunal segments isolated from CP and DP rats respectively, suggesting the potential clinical implication for ameliorating intestinal inflammation and co-occurring dysmotility.

        Theoretical Prediction Model for Surface Settlement Caused by the Excavation of New Tunnels Undercrossing Existing Tunnels Based on Modified Stochastic Medium Theory

        The deformation pattern of the stratum caused by constructing a new metro tunnel crossing an existing tunnel is different from the deformation pattern caused by general tunnel construction. However, the prediction results by the ordinary surface settlement prediction model often bring significant errors because the complex influence of existing tunnels on the surface settlement caused by the excavation of new tunnels is always neglected. Based on the equivalent layered method and stochastic medium theory, a prediction model for the surface settlement due to excavating a new tunnel under an existing tunnel in the heterogeneous stratum was established. By equating the bending stiffness of an existing tunnel before and after applying the equivalent layered method, the layer index was determined. The critical parameters of the stochastic medium theory were derived based on the relationship between the critical parameters of both the Peck empirical formula and the stochastic medium theory. The surface settlement of some typical projects was predicted and compared by the prediction model in this paper and the ordinary prediction model. The comparison shows that the proposed prediction model and parameter determination method in this paper had high accuracy and applicability. The results of the prediction model in this paper fit the results of numerical calculation. The research of this paper can provide a new method for the theoretical prediction of surface settlement caused by the excavation of a new tunnel under an existing tunnel in the heterogeneous stratum and the determination of critical parameters of the stochastic medium theory.problem in the construction industry, and helps reducing the material waste and budget cost.

      • Modeling and Evaluation of Inertia Emulation Control for DFIG Wind Turbine

        With the increasing penetration of renewable energy generators in power grid, many inertia emulation control strategies have been raised to solve the problem of reduced power system inertia. A co-simulation research platform is built based on RTDS and GH Bladed, detailed comparisons are made in terms of inertia response performance, weak grid operation stability, secondary frequency drop and wind tower load analysis. Research results point out that different control schemes are similar in inertia response while virtual synchronous generator control (VSG) and inertial synchronization control (ISynC) can improve weak grid operation stability, VSG strategy is also effective in reducing frequency secondary drop and wind tower fatigue load, the combination of VSG and ISynC can get the best control results.
