        신수혈의 침자극과 황기약침이 실험용 생쥐의 면역활성물질인 IL-1 α의 遺傳子發現에 미치는 影響

        체내의 환경을 항상적으로 유지하는 생체방어체계에는 면역활성물질이 있으며, 이러한 면역활성물질의 적절한 발현이 생체의 항상성을 유지하는데 작용하게 된다. 이에 한의학에서는 생체의 항상성 유지와 체내의 면역력 증가를 위해 많은 요법들이 시행되어 왔는데, 그 중 자침요법과 약침요법이 많이 응용되고 있다. 그러나 침자극과 약침의 효과에 대한 기전은 기대하는 만큼 제시되지 못하고 있다. 이에 본 논문은 침자극과 약침의 효과에 대한 기전을 밝히는 방법의 일환으로서 면역활성물질인 IL-1α가 補腎臟, 振氣化, 祛水濕, 强腰脊, 益水壯火 등의 穴性이 있는 신수혈(BL23)의 침자극과 황기약침처치를 통해 어떻게 발현하는가를 관찰하였다. 그 결과, 면역 증강 작용을 나타내는 cytokine IL-1α 에 대한 침자극의 반응은 관찰되었고, 황기약침에 의한 IL-1α 의 발현은 명확하지 않았다. 그러므로 신수혈을 이용한 cytokine IL-1α의 추적은 황기약침의 효과보다 침자극의 효과에 대한 근거를 추측할 수 있을 것이며, 뿐만 아니라 침자극에 의한 면역반응정도를 통해 질병의 진행상태를 나타내는 진단지표로서 활용가능성이 있을 것이다. Acupuncture and Radix Astragali aqua-acupuncture stimuli have long been used to cure human diseases. However, it still remains to be unknown on its action mechanism, physiological and biochemical aspects. Thus, many attempts were made to show the scientific background covering the above mentioned mechanisms. Most recent studies show that these tests improve blood circulatory system and increase leucocyte counts. In this study, we have applied the acupuncture stimuli to mouse Sinsu(BL23), which is a stimulative point of oriental medicine, to see if cytokine such as IL-1α can be detected. Mice were treated with lipopolysaccharide(LPS) for inflammation induction, and then reverse transcriptase-polymerase chain reaction (RT-PCR) using each primer set was performed to trace the amounts of mRNA. The results are as follows ; 1. The RT-PCR product using IL-1α primers was detected as 450 bp on agarose gels and the IL-1α gene was not expressed in normal mice at 15 min after the removal of acupuncture. However, it was gradually increased indicating a faint expression after 30 min. 2. IL-1α was expressed in LPS-treated mice 30 min after the acupuncture was pulled out, and expressed most highly at 3 h after the removal of acupuncture. The gene expression of LPS-treated mice were more effective than those of normal mice. 3. IL-1α was not expressed in normal mice and LPS-treated mice from 30 min to 3hs. after Radix Astragali aqua-acupuncture stimulation. Therefore, a tracing the gene expression of cytokine IL-1α can be used more not only for a basis of the effect of acupuncture stimulation but a diagnosis guide through the immunological action than the effect of Radix Astragali aqua-acupuncture. Moreover, we need to further study the cytokine's expression in Radix Astragali aqua-acupuncture stimulation.

        Effect of Iron-nanoparticles and Iron-microparticles on Erythropoiesis and Iron-storage in Iron-deficiency Anemic Mice

        Iron is an essential trace element for normal body’s functions. The restriction of iron availability directly limits erythropoiesis. The objective of this experiment was to compare the bioavailability of iron nanoparticles (Fe-NPs) with iron-microparticles (Fe-MPs) in anemic mice. There were four experimental groups including normal control group, iron-deficiency anemia (IDA) group, Fe-NPs group, and Fe-MPs group. Animals in normal group fed on an adequate iron-containing diet (45 ppm Fe). Meanwhile, the animals in the other three groups fed on a low Fe diet (4.5 ppm Fe) for 7 weeks. Double deionized water was supplied as drinking water ad libitum. After feeding for 3 weeks with the low Fe diet, the animals in Fe-NPs and Fe-MPs groups were orally treated with Fe-NPs or Fe-MPs at the daily dose of 40 mg/kg for 4 weeks. IDA group showed a markedly decreased red blood cell (RBC) count, hematocrit (Hct), and hemoglobin (Hb) values compared with normal group throughout experimental periods. Treatments of Fe-NPs or Fe-MPs for 4 weeks restored the decreased RBC count and hematological values similar to normal values. Fe-NPs group showed the faster restoration in the values than Fe-MPs as time passes. The iron contents of the upper small intestine in Fe-NPs and Fe-MPs groups were higher than normal group at weeks 2 and 4. Hepatic iron contents and lipid peroxidation were significantly increased by Fe-NPs and Fe-MPs treatments compared with IDA group as time increase. The iron contents in liver and ferritin deposits in spleen were identified in Fe-NPs and Fe-MPs groups similarly to normal group. These results indicate that oral administration of both Fe-NPs and Fe-MPs can recover anemia and Fe-NPs is more absorbable and available in the body than Fe-MPs.

        Detection of Mitochondrial ATP-Sensitive Potassium Channels in Rat Cardiomyocytes

        Mitochondrial ATP-sensitive potassium (mitoK<SUB>ATP</SUB>) channels play a role in early and late ischemic preconditioning. Nevertheless, the subunit composition of mitoK<SUB>ATP</SUB> channels remains unclear. In this study, we investigated the subunit composition of mitoK<SUB>ATP</SUB> channels in mitochondria isolated from rat cardiac<FONT COLOR=RED><SPAN STYLE= font-size:11pt; > </SPAN>myocytes. Mitochondria were visualized using the red fluorescence probe, Mitrotracker Red, while mitoK<SUB>ATP</SUB> channels were visualized using the green fluorescence probe, glibenclamide-BODIPY. The immunofluorescence confocal microscopy revealed the presence of Kir6.1, Kir6.2 and SUR2 present in the cardiac mitochondria. Western blot analysis was carried to further investigate the nature of mitoK<SUB>ATP</SUB> channels. For SUR proteins, a 140-kDa immunoreactive band that corresponded to SUR2, but no SUR1 was detected. For Kir6.2, three bands (∼44, ∼46, and ∼30 kDa) were detected, and a specific ∼46-kDa immunoreactive band corresponding to Kir6.1 was also observed. These observations suggest that the subunits of mitoK<SUB>ATP</SUB> channels in rat myocytes include Kir6.1, Kir6.2, and a<FONT COLOR=RED><SPAN STYLE= font-size:11pt; > </SPAN>SUR2-related sulfonylurea-binding protein.

        Bioavailability of Iron-nanoparticles with Ascorbic Acid in Anemic Mice

        Recently, iron nanoparticles (Fe-NPs) have been used for cancer diagnosis and therapy for imaging contrast and drug delivery. However, there was no report on nutritional bioavailability of the Fe-NPs in the body. Ascorbic acid (AA) is known to help the absorption of iron in stomach by reducing Fe (III) to Fe (II). In this study, we investigated the bioavailability of Fe-NPs with AA in iron-deficiency-anemic mice in comparison with non-nano iron particles. Iron-deficient anemia was induced by feeding iron-deficient diet (4.5 mg Fe/kg) and ocular bleeding from retro-orbital venous plexus for 4 weeks. Normal control mice were given normal diet (45 mg Fe/kg). After induction of anemia in mice, Fe (40 mg/kg B.W.) + AA (5 g/kg B.W) and Fe-NPs (40 mg/kg B.W) + AA (5 g/kg B.W) were orally administered daily to the anemic mice. After sacrifice, liver and spleen tissues were observed by haematoxylin & eosin stain. Amount of trace iron in liver and upper small intestine was investigated using an inductively coupled plasma-atomic emission spectrometer. Red blood cell (RBC), hematocrit (Hct), hemoglobin (Hb), and total iron binding capacity were also measured. The concentrations of iron in Fe-NPs + AA group were significantly higher in liver and in upper small intestine than that in Fe + AA group. The values of RBC, Hct, and Hb in Fe-NPs + AA group were more rapidly increased to normal values compared with Fe + AA group with increasing time. These results suggest that Fe-NPs in the presence of AA may be more bioavailable than non-nano Fe in Fe-deficient anemic mice.

        Identification of a Marker Protein for Cardiac Ischemia and Reperfusion Injury by Two-Dimensional Gel Electrophoresis and Matrix-Assisted Laser Desorption Ionization Mass Spectrometry

        The purpose of the present study was to evaluate the expression of cardiac marker protein in rabbit cardiac tissue that was exposed to ischemic preconditioning (IPC), or ischemiareperfusion injury (IR) using two-dimensional gel electrophoresis (2DE) and matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS). We compared 2DE gels of control (uninjured) cardiac tissue with those of IPC and IR cardiac tissue. Expression of one protein was detected in IR heart tissue, however the protein was not detected in the samples of control and IPC tissue. To further characterize the detected protein molecule, the protein in the 2D gel was isolated and subjected to trypsin digestion, followed by MALDI-MS. The protein was identified as myoglobin, which was confirmed also by Western blot analysis. These results are consistent with previous studies of cardiac markers in ischemic hearts, indicating myoglobin as a suitable marker of myocardial injury. In addition, the present use of multiple techniques indicates that proteomic analysis is an appropriate means to identify cardiac markers in studies of IPC and IR.

        A disposable light exposure detector kit has been developed by UV curing of a hydrogel material. The devised light exposure detector kit consisted of light sensitive structures, bottom plate, character sheet and sticky back plate. A light exposure detector kit has a serial light sensitive structures that contain various light sensitive dyes such as rhodamine and fluorescein. The light sensitive structure composed of UV curable hydrogel polymer material as a supporing material and photosensitive dye in a certain concentration. The fabrication procedure of the ligh exposure detector kit is very simple and fast due to UV curing procedure of a photopolymerizable hydrogel material such as poly(ethylene glycol) methyl ether acrylate (PEGMEA) and poly(ethylene glycol) diacrylate (PEGDA) with a photosensitive dye. By the proposed fabrication method, various size and shape of a light exposure detector kit could be fabricated using a flexible elastomer mold. Due to a fast and inexpensive fabrication method, the light exposure detector kit could be use a single use for various industrial applications. According to light irradation, the light sensitive structure on a light exposure detector kit could be lose its color by decomposition of a photosensitive dye chemical in the structure. Thus the amount of the exposed light on a substrate could easily be recognised by changing color or transparency of the structure.

      • 통합교육 기관의 긍정적 행동중재지원(PBIS) 사례

        이 연구는 통합교육을 실시하는 초등학교에서 긍적적 행동중재지원(PBIS)이 특수교육대상학생의 문제행동에 미치는 영향을 검증하고자 하였다. 연구 참여 학생은 초등학교의 특수교육대상학생 6명이고, 1차 예방, 2차 예방, 3차 예방을 위한 연속적인 행동지원 통해 공격행동과 파괴행동을 보이는 지적장애학생 1명을 대상으로 준실험적 설계인 AB 설계를 사용하여 행동중재를 실시하였다. 연구 참여 학생의 공격행동과 파괴행동이 자주 발생하는 일반학급과 특수학급에서 기능적 행동평가 및 예방 전략, 대체 행동 교수, 후속 결과 중재 전략을 포함하는 다요소 행동지원 중재를 실행하였다. 연구 결과 협력적 행동중재지원은 연구 참여 학생의 수업 참여행동, 규칙 지키기 행동, 공격행동과 파괴행동 감소에 효과적이었다. 본 연구는 통합교육 기관에서 초등학생의 문제행동의 기능을 파악하여 협력적 행동중재지원을 적용함으로써 다요소 행동 중재 적용이 효과적인 중재 방법이 될 수 있음을 논의하였다. 또한 통합교육기관의 효과적인 행동중재지원 사례를 통해 통합교육 기관에서 특수교사의 주도하에 실천할 수 있는 현실적인 긍정적 행동중재지원(PBIS)의 적용 방안을 제안하였다. The purpose of this study was to examine the effects of positive behavior intervention support (PBIS) on the problem behaviors of students with special education needs in elementary schools implementing inclusive education. Using an AB design, continuous behavioral interventions were implemented for a student with intellectual disabilities who exhibited aggressive and destructive behaviors during primary, secondary, and tertiary prevention phases. The Multi-component behavioral intervention support including functional behavioral assessment and prevention strategies, alternative behavior instruction strategies, consequence interventions in both general and special education classrooms where the student’s aggressive and destructive behaviors frequently occurred. The results indicated that collaborative behavior intervention support was effective in improving student engagement, rule-following behavior, and reducing aggressive and destructive behaviors. This study discusses the application of multi-component behavioral intervention support through collaborative behavior intervention support, emphasizing the role of special education teachers in implementing practical and positive PBIS approaches within inclusive educational settings.

        The boundary evolution based Level Set Method has been widely used in medical image segmentation. In this paper, we have developed a new algorithm that extends the Fast Level Set Method. The main contributions of this work are to formulate a new level set function, define relationships between layers and use an alternative elimination criterion in interface evolution. The experimental results validate the effectiveness and efficiency of our approach, compared to the classical Level Set Method and the original Fast Level Set Method.
