        MiRNA320a Inhibitor-Loaded PLGA-PLL-PEG Nanoparticles Contribute to Bone Regeneration in Trauma-Induced Osteonecrosis Model of the Femoral Head

        BACKGROUND: This study aimed to explore the effect of a nanomaterial-based miR-320a inhibitor sustained release system in trauma-induced osteonecrosis of the femoral head (TIONFH). METHODS: The miR-320a inhibitor-loaded polyethylene glycol (PEG)- Poly(lactic-co-glycolic acid) (PLGA)- Poly-L-lysine (PLL) nanoparticles were constructed using the double emulsion method. The TIONFH rabbit model was established to observe the effects of miR-320a inhibitor nanoparticles in vivo. Hematoxylin–eosin staining and microcomputed tomography scanning were used for bone morphology analysis. Bone marrow mesenchymal stem cells (BMSCs), derived from TIONFH rabbits, were used for in vitro experiments. Cell viability was determined using the MTT assay. RESULTS: High expression of miR-320a inhibited the osteogenic differentiation capacity of BMSCs in vitro by inhibiting the expression of the osteoblastic differentiation markers ALP and RUNX2. MiR-320a inhibitor-loaded PEG-PLGA-PLL nanoparticles were constructed with a mean loading efficiency of 1.414 ± 0.160%, and a mean encapsulation efficiency of 93.45 ± 1.24%, which released 50% of the loaded miR-320a inhibitor at day 12 and 80% on day 18. Then, inhibitor release entered the plateau. After treatment with the miR-320a inhibitor nanoparticle, the empty lacunae were decreased in the femoral head tissue of TIONFH rabbits, and the osteoblast surface/bone surface (Ob.S/BS), osteoblast number/bone perimeter (Ob.N/B.Pm), bone volume fraction, and bone mineral density increased. Additionally, the expression of osteogenic markers RUNX2 and ALP was significantly elevated in the TIONFH rabbit model. CONCLUSION: The miR-320a inhibitor-loaded PEG-PLGA-PLL nanoparticle sustained drug release system significantly contributed to bone regeneration in the TIONFH rabbit model, which might be a promising strategy for the treatment of TIONFH.         Effect of Crystal Structure and Hydrogen Bond of Thermoplastic Oxidized Starch on Manufacturing of Starch-Based Biomass Composite

        Starch-based biomass composites were prepared with starch and plant fiber. In order to improve the properties of the compos ites, oxidized starches (OS) were prepared using hydroxyl peroxide as oxidizer, then glycerol was added into OS as plasticizer to produce thermoplastic oxidized starches (TPOS). Crystal structure, hydrogen bonds of TPOS were researched to gain comprehensive views on the mechanism of the property differences between the starch-based composites with native starch, OS and TPOS. The changes of crystal structure were analyzed by X-ray diffraction analysis. Starch intermolecular hydroxyl changes were investigated using the method of infrared spectrum analysis. It was found that the new hydrogen bonds were formed between the plasticizer, oxidizer and starch, and the starch intermolecular bonding force was weakened. The structure of starch crystallization was destructed in TPOS. Differential scanning calorimetry showed that cross reaction with oxidization and plasticization changed the crystal structure, glass transition process was not observed in TPOS. The tensile and compressive strengths of composite based on TPOS were improved greatly and mechanical properties were not weakened obviously with water content increased. All the results indicated that TPOS could improve the properties of starch-based composites.

      • Interpreting and Analyzing the Character 得 in Mencius : Insights from Four Commentaries

        According to traditional philological materials such as Shuowen Jiezi and oracle bone inscriptions, the original meaning of the character 得 is “get”. This article takes the Book of Mencius as closed materials, exhaustively categorizing all entries of 得 in Mencius, totaling 114 examples. Then we analyze semantic content and grammatical features references to the four commentaries from Zhao Qi , Sun Shi, Zhu Xi, Jiao Xun. We find that all examples of 得 are used as verbs, but there are 12 examples where it is used as a delexical verb, exemplifies the gradual trend of semantic attenuation of 得. After conducting linguistic analysis of the character 得, we explore its manifestation of the philosophical ideas in Mencius, including aspects of personal cultivation, interpersonal relationships, and governance wisdom. Through the interaction of language and cultural analysis, we further deepen our study of the language theory and ancient classics culture.

        Cloning and Expression of the Duck Leptin Gene and the Effect of Leptin on Food Intake and Fatty Deposition in Mice

        Leptin is the adipocyte-specific product of the obese gene and plays a major role in food intake and energy metabolism. Leptin research was mainly focused on mammalian species, but understanding of leptin and its function in poultry is very poor. In this study, the duck leptin gene was amplified using the reverse transcription-polymerase chain reaction (RT-PCR) from duck liver RNA. The cDNA fragment was inserted into the pET-28a expression vector, and the resulting plasmid was expressed in Escherichia coli BL21 (DE3). Experimental mice were given an intraperitoneal injection of 10 mg/kg leptin dissolved in phosphate buffered saline (PBS), while the control mice were injected with PBS. The effect of leptin on food intake, body weight and fatty deposition in mice was detected. Sequence analysis revealed that duck leptin had a length of 438 nucleotides which encoded a peptide with 146 amino acid residues. The sequence shares highly homology to other animals. The coding sequence of duck leptin was 84 and 86% identical to human and pig leptin nucleotides sequence. Highest identity was with the rat coding sequence (95%). The identity of the amino acid sequence was 84, 82 and 96% respectively compared to that of the human, pig and rat. Results of SDS-PAGE analysis indicated that a fusion protein was specifically expressed in E. coli BL21 (DE3). The purified product was found to be biologically active during tests. Continuous administration of recombinant duck leptin inhibited food intake. Despite the decrease of food intake, leptin significantly induced body weight and fatty deposition. These changes were accompanied by a significant down-secretion of plasma glucose, cholesterol, triglyceride and insulin levels in mice. The observations provide evidence for an inhibitory effect of leptin in the regulation of food intake and for a potential role of duck leptin in the regulation of lipogenesis.

        Optical Characters Measurement and Simulation of 2D Cross Grating Microstructures of Butterfly Wings

        In this paper, the reflectance of the butterfly wings are measured by using the microspectrophotometer as the optical character with the reflectance on several scales on the wings. Reflectance spectra from different wing scale zones with the same color are dramatically different. And it suggests that the tiny structural difference has the great influence on the reflectance property. In addition, the microstructures of the butterfly wings can be simplified to a 2D crossing grating. By simulating a two-dimensional model using rigorous coupled-wave analysis technique, the optical properties of the butterfly wings were investigated. The simulation results depend strongly on the structural parameters and refractive index. The research will reveal the mechanisms of the structural color of 2D crossing grating butterfly wings.

        Research on a Multi-level Space Vector Modulation Strategy in Non-orthogonal Three-dimensional Coordinate Systems

        A novel space vector modulation strategy in the non-orthogonal three-dimensional coordinate system for multi-level three-phase four-wire inverters is proposed in this paper. This new non-orthogonal three-dimensional space vector modulation converts original trigonometric functions in the orthogonal three-dimensional space coordinate into simple algebraic operations, which greatly reduces the algorithm complexity of three-dimensional space vector modulation and preserves the independent control of the zero-sequence component. Experimental results have verified the correctness and effectiveness of the proposed three-dimensional space vector modulation in the new non-orthogonal three-dimensional coordinate system.

        Modelling of effective irradiation swelling for inert matrix fuels

        The results of effective irradiation swelling in a wide range of burnup levels are numerically obtained for an inert matrix fuel, which are verified with DART model. The fission gas swelling of fuel particles is calculated with a mechanistic model, which depends on the external hydrostatic pressure. Additionally, irradiation and thermal creep effects are included in the inert matrix. The effects of matrix creep strains, external hydrostatic pressure and temperature on the effective irradiation swelling are investigated. The research results indicate that (1) the above effects are coupled with each other; (2) the matrix creep effects at high temperatures should be involved; and (3) ranged from 0 to 300 MPa, a remarkable dependence of external hydrostatic pressure can be found. Furthermore, an explicit multi-variable mathematic model is established for the effective irradiation swelling, as a function of particle volume fraction, temperature, external hydrostatic pressure and fuel particle fission density, which can well reproduce the finite element results. The mathematic model for the current volume fraction of fuel particles can help establish other effective performance models.

        Tumor Promoting Function of DUSP10 in Non-Small Cell Lung Cancer Is Associated With Tumor-Promoting Cytokines

        Lung cancer, particularly non-small cell lung cancer (NSCLC) which contributes more than 80% to totally lung cancer cases, remains the leading cause of cancer death and the 5-year survival is less than 20%. Continuous understanding on the mechanisms underlying the pathogenesis of this disease and identification of biomarkers for therapeutic application and response to treatment will help to improve patient survival. Here we found that a molecule known as DUSP10 (also known as MAPK phosphatase 5) is oncogenic in NSCLC. Overexpression of DUSP10 in NSCLC cells resulted in reduced activation of ERK and JNK, but increased activation of p38, which was associated with increased cellular growth and migration. When inoculated in immunodeficient mice, the DUSP10-overexpression NSCLC cells formed larger tumors compared to control cells. The increased growth of DUSP10- overexpression NSCLC cells was associated with increased expression of tumor-promoting cytokines including IL-6 and TGFβ. Importantly, higher DUSP10 expression was associated with poorer prognosis of NSCLC patients. Therefore, DUSP10 could severe as a biomarker for NSCLC prognosis and could be a target for development of therapeutic method for lung cancer treatment.

        An integrated blade optimization approach based on parallel ANN and GA with hierarchical fair competition dynamic-niche

        This work presented an available multi-point blade optimization procedure for better aerodynamic performances. Based on the proposed Parallel ANN and GA with hierarchical fair competition dynamic-niche (GA-HFCDN), an integrated approach for the blade optimization design was put forward by combining Bezier parameterization with FINE/TURBO solver. In the optimization design, parallel ANN was employed to build a more proper approximate model. GA-HFCDN was proposed to obtain the global optimization solution more efficiently for the blade design. Two research cases, including plane cascades blade optimization and the optimization and experimental study of a low specific speed centrifugal blower blade, were performed by using the above approach. The conclusions showed the rationality and validity of the optimization approach.
