        Study on the local damage of SFRC with different fraction under contact blast loading

        The steel fiber reinforced concrete (SFRC) shows better performance under dynamic loading than conventional concrete in virtue of its good ductility. In this paper, a series of quasi-static experiments were carried out on the SFRC with volume fractions from 0 to 6%. The compressive strength increases by 38% while the tension strength increases by 106% when the fraction is 6.0%. The contact explosion tests were also performed on the Ф40×6 cm circular SFRC slabs of different volume fractions with 20 g RDX charges placed on their surfaces. The volume of spalling pit decreases rapidly with the increase of steel fiber fraction with a decline of 80% when the fraction is 6%, which is same as the crack density. Based on the experimental results, the fitting formulae are given, which can be used to predict individually the change tendencies of the blast crater volume, the spalling pit volume and the crack density in slabs with the increase of the steel fiber fraction. The new formulae of the thickness of damage region are established, whose predictions agree well with our test results and others. This is of great practical significance for experimental investigations and engineering applications.

        Three-Dimensional Analysis of Chloroplast Structures Associated with Virus Infection

        <P>Three-dimensional visualization identifies structural remodeling in chloroplasts during barley stripe mosaic virus infection.</P><P>Chloroplasts are multifunctional organelles whose morphology is affected by environmental stresses. Although the three-dimensional (3D) architecture of thylakoid membranes has been reported previously, a 3D visualization of chloroplast under stress has not been explored. In this work, we used a positive-strand RNA ((+)RNA) virus, barley stripe mosaic virus (BSMV) to observe chloroplast structural changes during infection by electron tomography. The analyses revealed remodeling of the chloroplast membranes, characterized by the clustering of outer membrane-invaginated spherules in inner membrane-derived packets. Diverse morphologies of cytoplasmic invaginations (CIs) were evident with spherules at the periphery and different sized openings connecting the CIs to the cytoplasm. Immunoelectron microscopy of these viral components verified that the aberrant membrane structures were sites for BSMV replication. The BSMV αa replication protein localized at the surface of the chloroplasts and played a prominent role in eliciting chloroplast membrane rearrangements. In sum, our results have revealed the 3D structure of the chloroplasts induced by BSMV infection. These findings contribute to our understanding of chloroplast morphological changes under stress conditions and during assembly of plant (+)RNA virus replication complexes.</P>

        Effects of carboxylesterase gene silence on wheat aphid Sitobion avenae (Fabricius)

        Multifunctional carboxylesterase (CarE) has been found in all animals, plants and microbes, and belongs to a superfamily enzyme of serine hydrolase involved in detoxification, allelochemical tolerance and some specific hormone or pheromone metabolism. Insects usually utilize carboxylesterases to detoxify xenobiotics, and positively correlated with insect resistance to some insecticides. Despite the importance of CarEs in insects, carboxylesterases and their functions in wheat aphid Sitobion avenae (Fabricius) have not been clear. In this study, a sequence that encodes a carboxylesterase protein from S. avenae (SaCarE) was sequenced and cloned. After aligning the encoded amino acid sequence of the SaCarE gene with other known CarEs of insects, we found that the CarE gene was highly conserved in insects. The SaCarE mRNA levels at different developmental stages of S. avenae were gradually increased from the first instar of nymphs to adult stage. RNAi was employed to further explore its functions, in which oral ingestion of SaCarE double-stranded RNA from the third instar nymph significantly knocked down SaCarE expression, and significantly decreased ecdysis index in S. avenae. These results indicate that SaCarE is functional in S. avenae and could serve one of the potential target genes for management of S. avenae.

        Coordinated Hospital-Home Fecal Microbiota Transplantation via Percutaneous Endoscopic Cecostomy for Recurrent Steroid-Dependent Ulcerative Colitis

        Since its introduction as an alternative intestinal microbiota alteration approach, fecal microbiota transplantation (FMT) has been increasingly used as a treatment of choice for patients with ulcerative colitis (UC), but no reports exist regarding FMT via percutaneous endoscopic cecostomy (PEC). This report describes the case of a 24-year-old man with a 7-year history of recurrent, steroid-dependent UC. He received FMT via PEC once per day for 1 month in the hospital. After the remission of gastrointestinal symptoms, he was discharged from the hospital and continued FMT via PEC twice per week for 3 months at home. The frequency of stools decreased, and the characteristics of stools improved soon thereafter. Enteral nutrition was regained after 1 week, and an oral diet was begun 1 month later. Two months after the FMT end point, the patient resumed a normal diet, with formed soft stools once per day. The follow-up colonoscopy showed normal mucus membranes; then, the PEC set was removed. On the subsequent 12 months follow-up, the patient resumed orthobiosis without any gastrointestinal discomfort and returned to work. This case emphasizes that FMT via PEC can not only induce remission but also shorten the duration of hospitalization and reduce the medical costs; therefore, this approach should be considered an alternative option for patients with UC.         Research on Action Strategies and Simulations of DRL and MCTS-based Intelligent Round Game

        The reinforcement learning problem of complex action control in multiplayer online battlefield games has brought considerable interest in the deep learning field. This problem involves more complex states and action spaces than traditional confrontation games, making it difficult to search for any strategy with human-level performance. This paper presents a deep reinforcement learning model to solve this problem from the perspective of game simulations and algorithm implementation. A reverse reinforcement-learning model based on high-level player training data is established to support downstream algorithms. With less training data, the proposed model is converged quicker, and more consistent with the action strategies of high-level players’ decision-making. Then an intelligent deduction algorithm based on DDQN is developed to achieve a better generalization ability under the guidance of a given reward function. At the game simulation level, this paper constructs Monte Carlo Tree SearchIntelligent Decision Model for turn-based antagonistic deduction games to generate next-step actions. Furthermore, a prototype game simulator that combines offline with online functions is implemented to verify the performance of proposed model and algorithm. The experiments show that our proposed approach not only has a better reference value to the antagonistic environment using incomplete information, but also accurate and effective in predicting the return value. Moreover, our work provides a theoretical validation platform and testbed for related research on game AI for deductive games.

        Regulatory Roles of MAPK Phosphatases in Cancer

        The mitogen-activated protein kinases (MAPKs) are key regulators of cell growth and survival in physiological and pathological processes. Aberrant MAPK signaling plays a critical role in the development and progression of human cancer, as well as in determining responses to cancer treatment. The MAPK phosphatases (MKPs), also known as dual-specificity phosphatases (DUSPs), are a family of proteins that function as major negative regulators of MAPK activities in mammalian cells. Studies using mice deficient in specific MKPs including MKP1/DUSP1, PAC-1/DUSP2, MKP2/DUSP4, MKP5/DUSP10 and MKP7/DUSP16 demonstrated that these molecules are important not only for both innate and adaptive immune responses, but also for metabolic homeostasis. In addition, the consequences of the gain or loss of function of the MKPs in normal and malignant tissues have highlighted the importance of these phosphatases in the pathogenesis of cancers. The involvement of the MKPs in resistance to cancer therapy has also gained prominence, making the MKPs a potential target for anti-cancer therapy. This review will summarize the current knowledge of the MKPs in cancer development, progression and treatment outcomes.

        Moderating effect of regulatory focus on public acceptance of nuclear energy

        Public acceptance has become the most critical question for sustainable development of nuclear energy in recent decades. Many researches concentrated on risk and benefit perception, which were deemed as the most influential factors of Public Acceptance of Nuclear Energy (PANE). But few researches focused on psychological factors including regulatory focus. Therefore, this paper aimed to explore the moderating effect of regulatory focus on PANE based on Regulatory Focus Theory in order to find ways to increase/decrease PANE. An Internet-based survey had been carried out in China nationwide. The results indicated that trust in government was positively related to PANE and this relationship was mediated by risk and benefit perception. In addition, the strength of the associations between risk and benefit perception and PANE were moderated by regulatory focus, consisting of prevention focus and promotion focus. Pre-vention focus strengthened the negative relationship between risk perception and PANE, while pro-motion focus weakened. Moreover, promotion focus weakened the positive relationship between benefit perception and PANE, but no significant moderating effect of prevention focus was founded on the relationship between benefit perception and PANE. Some policy implications were also proposed on the basis of above-mentioned findings.
