        Megaloblastic anemias are disorders caused by impaired DNA synthesis and chara-cterized by the presence of megaloblastic cells. Clinical data on these disorders in Korea are limited. The aim of this study was to analyze clinical characteristics of megaloblastic anemias caused by vitamin B12 deficiency. We identified 40 cases who were diagnosed as megaloblastic anemia from January 1993 to December 2002 in a retrospective study on the basis of chart review. We investigated the annual distribution of diagnosed patients, clinical manifestations and response to treatment. 19 cases were complicated with a previous total gastrectomy(gastrectomy group) and 21 cases had no history of surgical operation(non-gastrectomy group). The most common presenting complaint was generalized weakness. The median age at diagnosis was 7(range: 38-79) years in non-gastrectomy group. The mean pos-toperative duration was 7(range: 4-25)years in group of total gastrectomy and mean level of serum vitamin B12 was lower than 100 pg/mL in both group. Peripheral blood revealed thrombocy-topenia, macrocytic anemia and hypersegmented neutrophil in all cases. After treatment of pareliteral vitamin B12, Their clinical symptoms and hematologic findings improved except two patients who had bone marrow with lower cellularity. Cobalamin replacement after total gastrectomy is still missing. Diagnoses are made too late in a population of patients, resulting in irreversible neurological sequale, and we suggest that Pernicious anemia seems not to be very rare in Korea.

        Effect of microgrooves and fibronectin conjugation on the osteoblast marker gene expression and differentiation

        PURPOSE. To determine the effect of fibronectin (FN)-conjugated, microgrooved titanium (Ti) on osteoblast differentiation and gene expression in human bone marrow−derived mesenchymal stem cells (MSCs). MATERIALS AND METHODS. Photolithography was used to fabricate the microgrooved Ti, and amine functionalization (silanization) was used to immobilize fibronectin on the titanium surfaces. Osteoblast differentiation and osteoblast marker gene expression were analyzed by means of alkaline phosphatase activity assay, extracellular calcium deposition assay, and quantitative real-time PCR. RESULTS. The conjugation of fibronectin on Ti significantly increased osteoblast differentiation in MSCs compared with non-conjugated Ti substrates. On the extracellular calcium deposition assays of MSCs at 21 days, an approximately two-fold increase in calcium concentration was observed on the etched 60-μm-wide/10-μm-deep microgrooved surface with fibronectin (E60/10FN) compared with the same surface without fibronectin (E60/10), and a more than fourfoldincrease in calcium concentration was observed on E60/10FN compared with the non-etched control (NE0) and etched control (E0) surfaces. Through a series of analyses to determine the expression of osteoblast marker genes, a significant increase in all the marker genes except type I collagen α1 mRNA was seen with E60/10FN more than with any of the other groups, as compared with NE0. CONCLUSION. The FN-conjugated, microgrooved Ti substrate can provide an effective surface to promote osteoblast differentiation and osteoblast marker gene expression in MSCs.

        Development and Evaluation of Fixation Equipment for Transporting Unit Modules

        The unit modular construction system is a building technique in which unit modules are manufactured at a factory and then assembled at the construction site. It has many benefits, including reduction of the construction period and improvement in quality. For this reason, it is utilized for various purposes in Japan and England. While it has been introduced in Korea, there have been few Korean studies conducted on the unit modular system. In particular, little research has been done on the method of safely fixating the unit modules to a truck. Therefore, this study reviewed the fixation methods of unit modules for transport, analyzed the problems, and designed a fixation device for unit modules. In addition, a device for the fixation of unit modules to a truck was developed, and a structural simulation was implemented for a safety test by considering the maximum stress generated during the transport of the unit modules fixed on a truck. When the device for the fixation of unit modules is manufactured based on the results of the structural simulation done in this study, it is expected to aid the development of a more practical fixation device for unit modules.

        Active Mechanical Vibration Control of Rotary Compressors for Air-conditioning Systems

        Recent power electronics and variable-frequency motor drive technologies have been applied to air conditioners to improve efficiency and power density. However, the mechanical vibrations and acoustic noise resulting from the compressor still remain as a serious problem. This paper presents the development and implementation of an online disturbance state-filter for the suppression of multiple unknown and time-varying vibrations of air conditioning systems. The proposed design has a form of the state-filter based on a Luenburger-style closed-loop speed observer. An active vibration decoupling strategy with an estimated disturbance is provided, which manipulates a motor torque command. Since the proposed estimation does not require any additional transducers or hardware for obtaining real-time information upon disturbances, it is suitable for retrofitting industrial air conditioners.

        Transcriptome analysis of astringent ‘Cheongdo-Bansi’ persimmon fruit treated with ethylene for removal of astringency

        <P><B>Abstract</B></P> <P>This research was conducted to study the gene expression related to the removal of astringency and ripening in astringent ‘Cheongdo-Bansi’ persimmon. Fruit treated with ethylene were used to compare the differentially expressed genes against fruit ripened naturally without ethylene treatment (control). We sequenced total mRNAs using Illumina high-throughput sequencing platform and constructed the transcriptome gene set by de novo assembly. We identified 93,601 unigenes with an average length of 643.2 bp in transcriptome contigs. Differential gene expression analysis was performed and a total of 12,374 unigenes were differentially expressed in the ethylene vs. control. Of these 12,374 unigenes, 6072 were up-regulated and 6302 were down-regulated in the treated fruit. Compared with the control, the number of genes that induced more than 2 fold expression were 2647 and the number of genes whose expression was inhibited more than 2 fold were 2804. We also identified 38 genes showing significantly different expression, 26 of which were up-regulated and the rest 12 genes were down-regulated. The identified genes were categorized as genes related to astringency removal, softening and other ripening-related changes. The present study will add the information on the effect of ethylene treatment for astringency removal, softening and other ripening-related changes of persimmon fruit at genomic level. This study will also contribute important resources for further study of the genes related to astringent substance for persimmon breeding and improvement.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Astringent type persimmon fruit was treated with ethylene for astringency removal. </LI> <LI> Transcriptome sequencing analysis was done and compared with the control. </LI> <LI> Genes showing significantly different expression were identified. </LI> <LI> Information was provided on astringency removal and ripening related changes. </LI> </UL> </P>

        Bioconversion of Piceid to Piceid Glucoside Using Amylosucrase from Alteromonas macleodii Deep Ecotype

        Resveratrol, or its glycoside form piceid, is a dietary antioxidant polyphenolic compound, found in grapes and red wine that has been shown to have protective effects against cardiovascular disease. However, very low water solubility of the compound may limit its application in the food and pharmaceutical industries. The amylosucrase (AMAS) of Alteromonas macleodii Deep ecotype was expressed in Escherichia coli and showed high glycosyltransferase activity to produce the glucosyl piceid when piceid was used as an acceptor. The conversion yield of piceid glucoside was 35.2%. Biotransformation using culture of the E. coli harboring the amas gene increased the yield up to 70.8%. The transfer product was purified by reverse phase chromatography and recycling preparative HPLC, and the molecular structure of the piceid glucoside was determined using NMR spectroscopy. The piceid glucoside was identified as glucosyl-α-(1→4)-piceid. The solubility of glucosyl piceid was 5.26 and 1.14 times higher than those of resveratrol and piceid, respectively. It is anticipated that dietary intake of this compound is more effective by enhancing the bioavailability of resveratrol in the human body because of its hydrophilic properties in the intestinal fluid.
