        Endophytic bacterial and fungal microbiota in different cultivars of cassava (Manihot esculenta Crantz)

        Endophytes colonize tissues of healthy host plants and play a crucial role in plant growth and development. However, little attention has been paid to the endophytes of tuber crops such as cassava, which is used as a staple food by approximately 800 million people worldwide. This study aimed to elucidate the diversity and composition of endophytic bacterial and fungal communities in different cassava cultivars using high-throughput sequencing. Although no significant differences in richness or diversity were observed among the different cassava cultivars, the community compositions were diverse. Two cultivars (SC124 and SC205) tolerant to root rot exhibited similar community compositions, while two other cultivars (SC10 and SC5), which are moderately and highly susceptible to root rot, respectively, harboured similar community compositions. Proteobacteria, Firmicutes, and Ascomycota dominated the endophyte assemblages, with Weissella, Serratia, Lasiodiplodia, Fusarium, and Diaporthe being the predominant genera. The differentially abundant taxonomic clades between the tolerant and susceptible cultivars were mainly rare taxa, such as Lachnoclostridium_5, Rhizobium, Lampropedia, and Stenotrophomonas. These seemed to be key genera that affected the susceptibility of cassava to root rot. Moreover, the comparison of KEGG functional profiles revealed that ‘Environmental adaptation’ category was significantly enriched in the tolerant cultivars, while ‘Infectious diseases: Parasitic’ category was significantly enriched in the susceptible cultivars. The present findings open opportunities for further studies on the roles of endophytes in the susceptibility of plants to diseases.

        Cell proliferation can be modulated by receptor tyrosine kinase-like orphan receptor 2 in the silkworm, Bombyx mori

        The receptor tyrosine kinase-like orphan receptor 2 (Ror2) is involved in the Wnt/β-catenin signaling pathway which regulates cell proliferation, polarity, differentiation, migration, metabolism and survival. However, the function of Ror2 in the silkworm Bombyx mori is still undisclosed. In the present investigation, we have made an effort to clone the silkworm Ror2 gene (BmRor2). The sequence analysis showed that the open reading frame (ORF) was 1914 bp in size and encoded a protein with the conserved domains of Ror2 protein. The qRT-PCR results indicated that the BmRor2 gene expression level was the highest in the head among all identified tis sues on 3rd day of the fifth instar larvae. In the gonads of the different development stages, the BmRor2 gene expression level was highest on the 4th day of the fourth instar larvae. The immunofluorescence assay indicated that the BmRor2 protein was located at the cytomembrane. The effects of BmRor2 protein on the expression levels of genes related to TGF-β, Hippo, JAK-STAT and Notch signaling pathways were investigated by qRT-PCR. The expression levels of crumbs (crb), warts (wts), α-catenin (cat), four-jointed (fj), decapentaplegic (dpp), kibra ortholog (kibra), serrate (serr) and c-myc (myc) genes were down-regulated, whereas, suppressor of cytokine signaling 2 (socs 2) gene expression was up-regulated in the cultured BmN cells after the BmRor2 expression level was up-regulated. Further, cell proliferation was demoted and the size of cells was decreased when BmRor2 expression level was elevated. Our current finding recommended that BmRo2 can regulate TGF- β, Hippo, JAKSTAT, and Notch signaling pathways, and affect cell proliferation and size.

        Association of Apolipoprotein E Polymorphisms with White Matter Lesions and Brain Atrophy

        Objective Apolipoprotein E (ApoE) is mainly synthesized in the liver. So far, it is unknown the relationship among APOE gene polymorphisms and WML, brain atrophy. Therefore, the aim of the study was to assess the associations of APOE gene polymorphisms in patients with WML and brain atrophy. Methods A total of 58 patients with WML, 128 patients with brain atrophy, 112 patients with co-occurrence of WML and brain atrophy and 95 healthy elderly volunteers were recruited from Renmin Hospital of WuHan University. Results Allele E3 was the most common allele. The alleles E2 had significantly higher levels of ApoB and lower age in WML group. The alleles E2 was associated with the lower level of ApoB, LDL-Ch, TCh, and sdLDL in co-occurrence group. The E3/E3 genotype has higher level of sdLDL, but lower age and female frequency in WML. The E3/E4 genotype had higher level of TG, but lower age in WML. Gender, Age, E2, Hyperhomocysteinemia and UA were also significantly associated with disease progression. Conclusion This study found that clinical data, lipids and metabolic complications were closely related to ApoE genotypes and alleles, and also disease progression and type.

        RCLSTMNet: A Residual-convolutional-LSTM Neural Network for Forecasting Cutterhead Torque in Shield Machine

        During tunneling process, it is of critical importance to dynamically adjust operation parameters of shield machine due to changes of geological conditions. Cutterhead torque is one of the key load parameters, and its accurate prediction could adjust operational parameters including cutterhead rotational speed and tunneling speed in advance and avoid potential cutterhead jamming. Based on operation and state data collected by the monitoring system, we propose a residual-convolutional-LSTM neural network (RCLSTMNet) for forecasting cutter head torque in shield machine. On the basis of correlation analysis, parameters closely related to cutter head torque are selected as inputs by employing cosine similarity, which significantly reduces input dimension. Convolutional-LSTM neural network is fused and constructed for extracting deep useful features, while residual network module is utilized to avoid gradient disappearing and improve regression performance. Comparisons with recent data-driven cutterhead torque prediction methods are made on the actual engineering datasets, which demonstrate the presented RCLSTMNet outperforms the other data driven models in most cases. Moreover, the predicted curves of cutterhead torque using the proposed RCLSTMNet coincide with the actual curves much better than predicted curves using the other models. Meanwhile, the highest and average accuracy of RCLSTMNnet reach 98.1% and 95.6%, respectively.

        Transient propagation of BmLV and dysregulation of gene expression in nontarget cells following BmLV infection

        Bombyx mori latent virus (BmLV), a novel positive-strand RNA virus was first identified in the B. mori cultured BmN cell line. Whether the infectivity of BmLV to silkworm larvae and non-silkworm cells is connected with dysregulation of gene expression are not well understood. A complete sequence of BmLV genomic RNA was identified and revealed that a fragment with 495 nt in length was deleted from the RNA-dependent RNA poly merase (RdRp) gene in some BmLV genomic RNAs. Studies on the infectivity of BmLV to nontarget cells showed that BmLV can infect silkworm larvae, Spodoptera frugiperda Sf-9 and H1299 lung cancer cells with transient propagation. The dysregulation of gene expression of Sf-9 cells followed by BmLV infection was analyzed. Out of 743 differentially expressed genes (DEGs), 300 were upregulated and 443 were downregulated. Gene Ontology (GO) analysis indicated the DEGs were enriched into oxidoreductase activity for CH-NH 2 group donors, gluta mate biosynthetic process, response to stress and proteasome core complex. KEGG enrichment analysis showed that DEGs were mainly enriched into sulfur metabolism, RNA degradation, proteasome, pentose and glucuronate interconversions. Undesirable nutrients and temperature factors contributed to the propagation of BmLV in Sf-9 cells. Additionally, the Imd and RNAi pathways were activated by BmLV infection without stimulating Toll and JAK-STAT pathways. Therefore, it is suggested that BmLV is originated from plants, which can enter nontarget cells with transient propagation. The transient infection of BmLV may not only be regulated by Imd and RNAi immune pathways but also mediated by dysregulation of gene expression.

        Parametric Analysis on Elastic Buckling Performance of Low Yield Point Steel Plate Shear Wall with Two-Side Connections

        The steel plate shear wall (SPSW) with two-side connections can be arranged fl exibly in the structure, and has no additional infl uence on the column, but its buckling capacity is diffi cult to be deduced directly by thin plate theory. The low yield point SPSW (LYP-SPSW) with two-side connections is proposed in this paper, including unstiff ened LYP-SPSW, cross-stiff ened LYP-SPSW and diagonally-stiff ened LYP-SPSW. The elastic buckling performance and buckling capacity of these three types of LYP-SPSWs are studied by numerical simulation. On the basis of the accurate model, a series of parametric analysis on the key factors were conducted, including the height-to-thickness ratio, the stiff ness ratio of the stiff ener, and the stiff ness ratio of the edge member, which have eff ects on the elastic buckling stress of the LYP-SPSW with two-side connections. The elastic buckling stress of the SPSW can be eff ectively increased by adding stiff eners and edge members, but it is unnecessary to use oversized stiff eners and edge members. Based on a large amount of parametric analysis, simplifi ed formulas for calculating the elastic buckling stresses of the three types of LYP-SPSWs mentioned above are proposed in this paper.

        Parametric Analysis on Hysteresis Performance and Restoring Force Model of LYP Steel Plate Shear Wall with Two-Side Connections

        Parametric analysis on the hysteresis performance of the low yield point steel plate sheer wall (LYP SPSW) with three different stiff ening confi gurations, including unstiff ened, cross stiff ened and diagonal stiff ened, was conducted using the fi nite element models which have been verifi ed by the test results. The parameters which have eff ects on the hysteretic behavior of LYP SPSW include ratio of height-to-thickness, stiff ness ratio of stiff ener, and the stiff ness ratio of the edge member. The hysteretic curves, skeleton curves and viscous damping coeffi cient-displacement curves of SPSW with diff erent stiff ening confi gurations were obtained. According to the numerical results, it can be seen that the shear resistance and hysteretic performance of LYP SPSW decrease with the increase of the ratio of height-to-thickness. The energy dissipation capacity and shear capacity of LYP SPSW can be improved eff ectively by setting cross and diagonal stiff eners, but when the stiff - ness ratio of stiff ener is greater than 10, the increase is no longer obvious. Furthermore, for steel plate shear walls without edge members, the diagonal cross-stiff ening steel shear wall can achieve better energy consumption. For steel plate shear walls with edge members, when the ratio of height-to-thickness is less than 100, a better energy dissipation can be achieved by using cross stiff ened steel plates; when the ratio of height-to-thickness is greater than or equal to 200, a better energy dissipation can be refl ected by adopting cross stiff ened steel plates. Finally, a restoring force model of the LYP SPSW with two-side connections is proposed.

        Optimization and Control of Cable Tensions for Hyper-redundant Snake-arm Robots

        Based on the feedback linearization of joints motion and the tension optimization of cables, a hyperredundant snake-arm robot control strategy is presented to solve the problems caused by the joint motion coupling and the cable drive redundancy. First, a hierarchical control system architecture of snake-arm robot is developed. Subsequently, the computed torque control method is utilized to decouple the motion in the joint space, and the tension distribution satisfying the constraint condition is obtained in the cable space by quadratic programming. Since it is hard and also expensive to feedback joint motion and cable tension by sensors, the cable tensions are obtained by the system dynamics equation and the position and speed of joints motion is calculated approximately from the driving electric motors’ position. Finally, the control performance under three typical conditions is studied by numerical simulation. The results demonstrate that the presented method can effectively limit the cable tensions within the range of the minimum preload tension and the maximum allowable tension.

        Precise Cutterhead Clogging Detection for Shield Tunneling Machine Based on Deep Residual Networks

        During the construction process of tunnels, the cutterhead of shield tunneling machines may get cloggeddue to clay adhesion, which may seriously affect the efficiency of the project. Therefore, finding an intelligentdiagnosis method to detect the clogging status is of great importance. In this study, a deep residual network-basedmethod for diagnosing cutterhead clogging on shield tunneling machines is proposed. First, working state data ofthe shield tunneling machine is screened out, and parameters reflecting the clogging state are selected for furtheranalysis. After eliminating extreme outliers, an empirical formula is proposed to label the data. At the same time,several time-domain features of the selected excavation parameters within every five minutes are extracted. Thesefeatures are then fed into the proposed model as the input data to realize clogging detection. Because the originaldataset is imbalanced, the combination of f1-score and accuracy is used to evaluate the performance of the proposedmodel. The results show that the accuracy of the proposed algorithm reaches 95.71%, which is 1.21%, 2.84%,9.84%, 6.04%, and 0.86% higher than the support vector machine-based, random forest-based, AdaBoost-based,extreme gradient boosting-based and deep neural network-based methods. The f1 score of the proposed modelis 0.923, which is also 0.038, 0.042, 0.269, 0.169 and 0.02 higher than those compared methods. Therefore, theproposed deep residual network-based method can accurately detect cutterhead clogging conditions.
