      • Vitexicarpin Induces Apoptosis in Human Prostate Carcinoma PC-3 Cells through G2/M Phase Arrest

        Vitexicarpin (3', 5-dihydroxy-3, 4', 6, 7-tetramethoxyflavone), a polymethoxyflavone isolated from Viticis Fructus (Vitex rotundifolia Linne fil.), has long been used as an anti-inflammatory herb in traditional Chinese medicine. It has also been reported that vitexicarpin can inhibit the growth of various cancer cells. However, there is no report elucidating its effect on human prostate carcinoma cells. The aim of the present study was to examine the apoptotic induction activity of vitexicarpin on PC-3 cells and molecular mechanisms involved. MTT studies showed that vitexicarpin dose-dependently inhibited growth of PC-3 cells with an $IC_{50}{\sim}28.8{\mu}M$. Hoechst 33258 staining further revealed that vitexicarpin induced apoptotic cell death. The effect of vitexicarpin on PC-3 cells apoptosis was tested using prodium iodide (PI)/Annexin V-FITC double staining and flow cytometry. The results indicated that vitexicarpin induction of apoptotic cell death in PC-3 cells was accompanied by cell cycle arrest in the G2/M phase. Furthermore, our study demonstrated that vitexicarpin induction of PC-3 cell apoptosis was associated with upregulation of the proapoptotic protein Bax, and downregulation of antiapoptotic protein Bcl-2, release of Cytochrome c from mitochondria and decrease in mitochondrial membrane potential. Our findings suggested that vitexicarpin may become a potential leading drug in the therapy of prostate carcinoma.

        Experimental Study on Shear Behavior of Bolted Cement Mortar Blocks under Constant Normal Stiffness

        In this paper, the influence of bolting on shear mechanical behavior of cement mortar specimens were studied. First, a shear test apparatus which could provide CNS (constant normal stiffness) condition was designed. Then, more than 140 bolted and non-bolted cement mortar blocks (100 × 100 × 100 mm) were prepared. The strengthening effect of bolts with diameters of 0 − 10 mm on cement mortar specimens was studied under different initial normal loads ranging from 20 kN to 60 kN and constant normal stiffness of 420 kN/mm. Two simplified models of shear behavior of reinforced cement mortar blocks were proposed. The most important influence of bolt on the cement mortar blocks was to strengthen the post-peak shear strength rather than the peak shear strength, and the strengthening effect was reduced under high initial normal force. The cohesion of cement mortar blocks was reduced while internal friction angle was increased in residual sliding stage by bolts. The dilation of reinforced cement mortar specimens in postpeak shear stage was significantly reduced due to bolting, and the larger the bolt diameter, the greater the reduction in the block dilation.

      • PV2JAVA: Automatic Generator of Security Protocol Implementations Written in Java Language from the Applied PI Calculus Proved in the Symbolic Model

        In order to get the security protocol implementations written in programming language from formal languages in secure way, firstly, the model of implementation generation from security protocol implementations written in formal language is presented; Apart from that, an automatic generator PV2JAVA is developed, which can transform security protocol implementations written in the Applied PI calculus proved in the symbolic model into security protocol implementations written in Java language ; Finally, the method of software testing is used to provide a strong confidence in the correctness of the automatic generator PV2JAVA through five typical security protocols.

        관광관련학과 대학생의 성취동기가 자기효능감 및 인턴십 만족도에 미치는 영향

        This study examined the relations among achievement motivation, self-efficacy and satisfaction. Total of 320 questionnaires were distributed and 282 collected, analysed by factor analysis, reliability analysis, and regression analysis. The regression equations generally supported the hypothesized relationships among study constructs. The results showed that sub-factors of achievement motivation have generally significant influences on sub-factors of self-efficacy. Especially, in the achievement motivation factors, task-preferred and future interest were found to be relatively strong sub-factors effecting on the self-efficacy. In the relationship between achievement motivation/self-efficacy and satisfaction, all the factors were found to be strong factors to predict internship`s satisfaction. In addition, following Baron and Kenny`s guidelines, the current study also tested a mediating effect of relationship achievement motivation on satisfaction related to tourism internship. Self-efficacy was found to be a partial mediator in the relationship between achievement motivation and satisfaction. Implications and suggestions for future research are discussed.

        Sulfonation of a hypercrosslinked polymer adsorbent for microwave-assisted desorption of adsorbed benzene

        Optipore V503, a commercial hypercrosslinked hydrophobic polymer adsorbent, was sulfonated to become hydrophilic to allow more efficient microwave-assisted desorption of adsorbed nonpolar volatile organic compounds (VOCs) such as benzene under humidified conditions. Compared to Optipore V503, the hydrophilic adsorbents showed reduced adsorption capacities towards benzene, but significantly improved microwave-assisted desorption efficiencies under a semi-continuous 600 W microwave irradiation process. The increased desorption efficiency was mainly attributed to adsorbed water, which indirectly allowed heating of the polymer adsorbents to higher temperatures upon irradiation by microwaves.

      • Effects of Allogeneic Blood Transfusion in Patients with Stage II Colon Cancer

        The aim of the present study was to determine whether allogeneic red blood cell transfusions showed a deleterious effect and what might be preoperative risk factors for blood transfusion in patients with TNM stage II colon cancer. Total 470 patients who fulfilled inclusion criteria were selected for a further 10-year follow-up study. We found that there were statistical significance between non-transfused and transfused group in mortality (P=0.018), local recurrence (P=0.000) and distant metastasis (P=0.040). Local recurrence and distant metastasis between 1 to 3 units and more than 3 units group did not show any significant differences. There was no difference in survival rate between non-transfused and 1 to 3 units group (log rank=0.031, P=0.860). The difference between different blood transfusion volume in transfused patients was found (78.77% vs 63.83%, P=0.006). Meanwhile, the significant difference of survival rate was existed between non-transfused group and more than 3 units group (84.83% vs 63.83%, P=0.002 ). Univariate analysis showed the following 3 variables to be associated with an increased risk of allogeneic blood transfusions: preoperative CEA level (P<0.05), location of tumor (P<0.01) and diameter of tumor (P<0.01). Multivariate analysis revealed that location of tumor and diameter of tumor are two independent factors for requirement of perioperative transfusions. Therefore, allogeneic transfusion increase the postoperative tumor mortality, local recurrence and distant metastasis in patients with stage II colon cancer. The postoperative tumor mortality, local recurrence and distant metastasis were not associated with the blood transfusion volume. The blood transfusion volume was associated with the survival rate. Location of tumor and diameter of tumor were the independent preoperative risk factors for blood transfusion.

        Column-loss response of RC beam-column sub-assemblages with different bar-cutoff patterns

        Static loading tests were carried out in this study to investigate the effect of bar cutoff on the resistance of RC beam-column sub-assemblages under column loss. Two specimens were designed withcontinuous main reinforcement. Four others were designed with different types of bar cutoff in the mid-span and/or the beam-end regions. Compressive arch and tensile catenary responses of the specimens under gravitational loading were compared. Test results indicated that those specimens with approximately equal moment strength at the beam ends had similar peak loading resistance in the compressive arch phase but varied resistance degradation in the transition phase because of bar cutoff. The compressive bars terminated at one-third span could help to mitigate the degradation although they had minor contribution to the catenary action. Among those cutoff patterns, the K-type cutoff presented the best strength enhancement. It revealedthat it is better to extend the steel bars beyond the mid-span before cutoff for the two-span beams bridgingover a column vulnerable to sudden failure. For general cutoff patterns dominated by gravitational and seismic designs, they may be appropriately modified to minimize the influence of bar cutoff on the progressive collapse resistance.

        As the economy in China enters the new normal, Public-Private Partnerships have been an important measure for alleviating financial tensions and improving the administrative efficiency of the government. Based on the mode of open tendering for the selection of social capital partners for the new profit project, the open tendering procedures were analyzed. In addition, based on an improved TOPSIS model, the evaluation index system for the selection conducts a comprehensive evaluation of six aspects: reputation and performance, water pricing factor, financing program, construction program, operation and maintenance program, and transfer program.

        MicroRNA-708-3p as a potential therapeutic target via the ADAM17-GATA/STAT3 axis in idiopathic pulmonary fibrosis

        MicroRNAs (miRNAs) are important diagnostic markers and therapeutic targets for many diseases. However, the miRNAs that control the pathogenesis of idiopathic pulmonary fibrosis (IPF) and act as potential therapeutic targets for the disease are rarely studied. In the present study, we analyzed the function and regulatory mechanism of microRNA-708-3p (miR-708-3p) and evaluated this marker’s potential as a therapeutic target in IPF. The clinical and biological relevance of fibrogenesis for miR-708-3p was assessed in vivo and in vitro, specifically in matching plasma and tissue samples from 78 patients with IPF. The data showed that the miR-708-3p levels decreased during fibrosis and inversely correlated with IPF. The experiments showed that the decreased miR-708 promoter activity and primer-miR-708(pri-miR-708) expression were the potential causes. By computational analysis, a dual luciferase reporter system, rescue experiments and a Cignal Finder 45-Pathway system with siADAM17 and a miR-708-3p mimic, we identified that miR-708-3p directly regulates its target gene, a disintegrin and metalloproteinase 17 (ADAM17), through a binding site in the 3′ untranslated region, which depends on the GATA/STAT3 signaling pathway. Finally, an miR-708-3p agomir was designed and used to test the therapeutic effects of the miR-708-3p in an animal model. Small-animal imaging technology and other experiments showed that the dynamic image distribution of the miR-708-3p agomir was mainly concentrated in the lungs and could block fibrogenesis. In conclusion, the miR-708-3p– ADAM17 axis aggravates IPF, and miR-708-3p can serve as a potential therapeutic target for IPF.
