        Proangiogenic Hypoxia-Mimicking Agents Attenuate Osteogenic Potential of Adipose Stem/Stromal Cells

        BACKGROUND: Insufficient vascularization hampers bone tissue engineering strategies for reconstructing large bone defects. Delivery of prolyl-hydroxylase inhibitors (PHIs) is an interesting approach to upregulate vascular endothelial growth factor (VEGF) by mimicking hypoxic stabilization of hypoxia-inducible factor-1alpha (HIF-1α). This study assessed two PHIs: dimethyloxalylglycine (DMOG) and baicalein for their effects on human adipose tissue-derived mesenchymal stem/stromal cells (AT-MSCs). METHODS: Isolated AT-MSCs were characterized and treated with PHIs to assess the cellular proliferation response. Immunostaining and western-blots served to verify the HIF-1α stabilization response. The optimized concentrations for long-term treatment were tested for their effects on the cell cycle, apoptosis, cytokine secretion, and osteogenic differentiation of AT-MSCs. Gene expression levels were evaluated for alkaline phosphatase (ALPL), bone morphogenetic protein 2 (BMP2), runt-related transcription factor 2 (RUNX2), vascular endothelial growth factor A (VEGFA), secreted phosphoprotein 1 (SPP1), and collagen type I alpha 1 (COL1A1). In addition, stemness-related genes Kruppel-like factor 4 (KLF4), Nanog homeobox (NANOG), and octamer-binding transcription factor 4 (OCT4) were assessed. RESULTS: PHIs stabilized HIF-1α in a dose-dependent manner and showed evident dose- and time dependent antiproliferative effects. With doses maintaining proliferation, DMOG and baicalein diminished the effect of osteogenic induction on the expression of RUNX2, ALPL, and COL1A1, and suppressed the formation of mineralized matrix. Suppressed osteogenic response of AT-MSCs was accompanied by an upregulation of stemness-related genes. CONCLUSION: PHIs significantly reduced the osteogenic differentiation of AT-MSCs and rather upregulated stemness-related genes. PHIs proangiogenic potential should be weighed against their longterm direct inhibitory effects on the osteogenic differentiation of AT-MSCs.

        The effect of alpha-lipoic acid supplementation on anthropometric, glycemic, lipid, oxidative stress, and hormonal parameters in individuals with polycystic ovary syndrome: a systematic review and meta-analysis of randomized clinical trials

        This systematic review and meta-analysis aimed to examine the effect of the antioxidant alpha-lipoic acid (ALA) on various cardiometabolic risk factors and hormonal parameters in patients with polycystic ovary syndrome (PCOS). We searched PubMed, EMBASE, SCOPUS, Cochrane Library, and Web of Science databases without language restrictions until May 2023 to find randomized controlled trials (RCTs) that assessed the impact of ALA supplementation on anthropometric, glycemic, lipid, oxidative stress, and hormonal parameters in women with PCOS. Outcomes were summarized using the standardized mean difference (SMD) and 95% confidence interval (CI) in a random-effects model. An I2 statistic of >60% established significant between-study heterogeneity. The overall certainty of the evidence for each outcome was determined using the grading of recommendations, assessment, development, and evaluations system. Seven RCTs met the inclusion criteria. The ALA group had significant reductions in fasting blood sugar (fasting blood sugar (FBS), n=7 RCTs, SMD, -0.60; 95% CI, -1.10 to -0.10; I2=63.54%, moderate certainty of evidence) and homeostatic model assessment for insulin resistance (homeostatic model assessment of insulin resistance (HOMA-IR), n=4 RCTs, SMD, -2.03; 95% CI, -3.85 to -0.20; I2=96.32%, low certainty of evidence) compared with the control group. However, significant differences were observed between the groups in body mass index, insulin, estrogen, follicle-stimulating hormone, luteinizing hormone, testosterone, low-density lipoprotein, highdensity lipoprotein, triglyceride, total cholesterol, malondialdehyde, or total antioxidant capacity profiles. ALA supplementation improves FBS and HOMA-IR levels in women with PCOS. ALA consumption is an effective complementary therapy for the management of women with PCOS.

        Water hyacinth (Eichhornia crassipes fractions potentially normalize the lead (Pb) poisoning and enhance in vitro thrombolysis

        Water hyacinth (Eichhornia crassipes) has been used in phytoextraction to uptake heavy metals and trace elements in various experiments. This research investigated the protective effects of Water hyacinth extracts on Lead (Pb)-induced toxicity in the albino rat. Forty-eight six-weeks-old Wistar albino male rats (average weight, 180 ± 6.45 g) were divided into six groups: normal control (NC), Pb control (PbC), Chloroform extract (ChEx), Ethyl acetate extract (EAEx), Methanol extract (MeEx) and Ascorbic acid treated groups (AA, positive control). All animals except NC group have been administered with Lead acetate (Pb(CH3COOH)2) before the therapeutic dose. Thrombolytic and cytotoxic effects were evaluated by modified clot lysis and Brine shrimp lethality tests respectively. Biochemical analyses reports revealed that ChEx significantly (P ≤ 0.05) reduced the elevated alkaline phosphatase (ALP) and aspartate amino transferase (AST) whereas MeEx did the alanine amino transferase (ALT) in comparison to positive control. ChEx significantly inhibited the Pb deposition in kidney and liver than two other extracts. ChEx significantly increased the liver protein compared to PbC group. In the thrombolytic assay, EAEx showed the pronounced clot lysis (49.24 %) while ChEx and MeEx showed 45.18 % and 29.13 % of clot lysis respectively. In Brine shrimp lethality bioassay, the LC50 value of the ChEx, EAEx and MeEx were 4.16, 4.47 and 9.27 μg/mg respectively and values were statistically significant (p < 0.05) compared to that of reference cytotoxic agent, Vincristine sulfate (LC50’ 0.55). Histopathological screening of kidney, liver and spleen showed that ethyl acetate extract recovered the highest of the cellular damage caused by Lead acetate. Biochemical and histopathological screening, therefore, demonstrate that Water hyacinth could be one of the promising sources of normalizing the Pb-poisoning and enhancing the thrombolysis in an animal model. Therapeutic prospects of Water hyacinth could be further studied through highlighting a dose-response study.

        Kinetics of Base Hydrolysis of Some Chromen-2-one Indicator Dyes in Different Solvents at Different Temperatures

        7-hydroxy-2H-chromen-2-one(HC)와 7-hydroxy-2H-chromen-2-one-4-acetic acid(HCA)의 염기성 가수분해반응을 aqueous-methanol과 aqueous-acetone 혼합물에서 283 K에서 313 K의 온도 범위에서 속도론적으로 연구하였다. 반응의 활성화 파라미터를 구하고 토의하였다. 게다가, 물, 물-에탄올, 물-아세톤 혼합물 내에서 화합물들에 대한 활성화 에너지 장벽의 변화를 속도론적 데이터로부터 추정하였다. 활성화 장벽의 변화는 HC and HCA의 가수분해 반응과 거의 같았다. HC와 HCA의 염기성 가수분해는 $k_{obs}=k_2[OH^-]$와 같은 속도법칙을 따른다. 메탄올 또는 아세톤의 비가 증가함에 따라 HC 와 HCA의 속도 상수들이 감소하는 것은 $OH^-$ 이온이 불안정해지기 때문이다. 활성화 엔트로피가 큰 음의 값을 갖는 다는 것은 반응이 중간 착물의 형성을 경유하며 진행된다는 것을 의미하며, 또한 중간 착물이 경직성과 안정도를 갖는다는 것을 의미한다. 그러므로, 중간 착물의 고리 열림이 속도 조절 단계가 될 것이다. Base hydrolysis of 7-hydroxy-2H-chromen-2-one (HC) and 7-hydroxy-2H-chromen-2-one-4-acetic acid (HCA) in aqueous-methanol and aqueous-acetone mixtures were studied kinetically at temperature range from 283 to 313 K. The activation parameters of the reactions were evaluated and discussed. Moreover, the change in the activation energy barrier of the investigated compounds from water to water-methanol and water-acetone mixtures was estimated from the kinetic data. It is observed that the change in activation barriers is more or less the same for the hydrolysis of HC and HCA. Base hydrolysis of HC and HCA follows a rate law with $k_{obs}=k_2[OH^-]$. The decrease in the rate constants of HC and HCA as the proportion of methanol or acetone increases is due to the destabilization of $OH^-$ ion. The high negative values of entropy of activation support the proposal mechanism, i.e. the investigated reaction takes place via the formation of an intermediate complex. Moreover, these values refer to the rigidity and stability of the intermediate complex. Thus, the ring opening of the intermediate complex would be the rate controlling step.

        Preemptive pregabalin for postoperative analgesia during minimally invasive hysterectomy: a systematic review and meta-analysis of randomized controlled trials

        We aimed to perform a systematic review and meta-analysis of all randomized placebo-controlled trials (RCTs)that examined the analgesic benefits of preemptive pregabalin among patients undergoing minimally invasivehysterectomy. Five major databases were systematically screened from inception until August 29, 2021 Relevantstudies were evaluated for risk of bias. Endpoints were analyzed using the random-effects model and pooled asthe mean difference or risk ratio with a 95% confidence interval. Four studies with seven treatment arms met theinclusion criteria. The total sample size was 304 patients: 193 and 111 patients were allocated to the pregabalin andplacebo groups, respectively. Overall, the included studies revealed a low risk of bias. The summary results revealedthat the mean postoperative pain scores at rest were significantly lower in the pregabalin group than in the controlgroup at 0, 2, 4, 6, 12, and 24 hours. Moreover, the mean postoperative pain scores on movement/coughing weresignificantly lower in the pregabalin group than in the control group at 12 and 24 hours. The rate of patients whowere opioid-free postoperatively was significantly higher in the pregabalin group than in the control group. Therewas no significant difference between the groups in terms of the mean postoperative time to first rescue analgesicand the rates of adverse events. Compared with placebo, preemptive pregabalin was largely safe, and was correlatedwith superior analgesic effects in terms of lower postoperative pain scores and higher opioid-sparing effects. Additional RCTs are needed to confirm these findings.

        Prophylactic tranexamic acid to reduce blood loss and related morbidities during hysterectomy: a systematic review and meta-analysis of randomized controlled trials

        To perform a systematic review and meta-analysis of all randomized controlled trials (RCTs) that evaluated the efficacyand safety of prophylactic tranexamic acid (TXA) versus a control (placebo or no treatment) during hysterectomy forbenign conditions. Six databases were screened from inception to January 23, 2022. Eligible studies were assessed forrisk of bias. Outcomes were summarized as weighted mean differences and risk ratios with 95% confidence intervalsin a random-effects model. Five studies, comprising six arms and 911 patients were included in the study. Two andthree studies had an overall unclear and low risk of bias, respectively. Estimated intraoperative blood loss, requirementfor postoperative blood transfusion, and requirement for intraoperative topical hemostatic agents were significantlyreduced in a prophylactic TXA group when compared with a control group. Moreover, postoperative hemoglobinlevel was significantly higher in the prophylactic TXA group than in the control group. Conversely, the frequencyof self-limiting nausea and vomiting was significantly higher in the prophylactic TXA group than in the control group. There were no significant differences between the groups in terms of surgery duration, hospital stay, and diarrhearate. All the RCTs reported no incidence of major adverse events in either group, such as mortality, thromboembolicevents, visual disturbances, or seizures. There was no publication bias for any outcome, and leave-one-out sensitivityanalyses demonstrated stability of the findings. Among patients who underwent hysterectomy for benign conditions,prophylactic TXA appeared largely safe and correlated with substantial reductions in estimated intraoperative bloodloss and related morbidities.

        Synergistic of a Coumarin Derivative with Potassium Iodide on the Corrosion Inhibition of Aluminum Alloy in 1.0 M H2SO4

        Synergistic effects of the addition of KI on the corrosion inhibitive performance of a coumarin derivative on analuminum alloy in 1.0 M H2SO4 at different temperatures were studied using various electrochemicalmeasurements. Density functional theory was used to calculate the quantum chemical parameters of thecoumarin derivative. The experimental results showed that the coumarin derivative is considered as a mixedtypeinhibitor. The corrosion potential values were almost unchanged upon the addition of PBBC to the acidicsolution. The inhibition efficiency increases with increasing inhibitor concentration and increases further inthe presence of 6.02 mM KI but decreases significantly at higher temperature. The adsorption of PBBC obeyedthe Langmuir isotherm, and being chemically adsorbed at lower temperatures, while physical adsorption isfavoured at higher temperature. The theoretical results indicated that the coumarin derivative was adsorbedonto the surface of Al2024 through the sulfur, oxygen and nitrogen atoms.

        Highlights on selected microscopy techniques to study zebrafish developmental biology

        Bio-imaging is a tedious task when it concerns exploring cell functions, developmental mechanisms, and other vital processes in vivo. Single-cell resolution is challenging due to different issues such as sample size, the scattering of intact and opaque tissue, pigmentation in untreated animals, the movement of living organs, and maintaining the sample under physiological conditions. These factors might lead researchers to implement microscopy techniques with a suitable animal model to mimic the nature of the living cells. Zebrafish acquired its prestigious reputation in the biomedical research field due to its transparency under advanced microscopes. Therefore, various microscopy techniques, including Multi-Photon, Light-Sheet Microscopy, and Second Harmonic Generation, simplify the discovery of different types of internal functions in zebrafish. In this review, we briefly discuss three recent microscopy techniques that are being utilized because they are non-invasive in investigating developmental events in zebrafish embryo and larvae.

      • One-step facile route to copper cobalt sulfide electrodes for supercapacitors with high-rate long-cycle life performance

        <P><B>Abstract</B></P> <P>The impressive electrochemical energy storage performance of Earth-abundant ternary copper cobalt sulfide (CCS) thin film electrodes that are prepared on stainless steel substrates via a simple and cost-effective hydrothermal process is demonstrated. The optimized CCS electrode shows a high specific capacitance of ∼516 F/g at a current density of 10 A/g, a good rate capability of ∼72% at a high current density of 50 A/g, and a good cycling retention of ∼66% with a coulombic efficiency of ∼99% after 10,000 charge-discharge cycles. The CCS electrode exhibits a high energy density of ∼35.2 Wh/kg at a power density of ∼6.6 kW/kg. The excellent electrochemical supercapacitor properties of the CCS electrode are a result of a synergetic effect between the uniform full coverage, robust adhesion, and desired chemical composition. A low charge transfer resistance, resulting from the large electrochemically active surface area (ECSA) and good diffusion, significantly contributes to the enhanced electrochemical supercapacitor performance. This excellent CCS electrode material has the potential to become a low-cost and long-cycle life electrode for the next-generation high-power-capacity supercapacitors.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A binder-free CuCo<SUB>2</SUB>S<SUB>4</SUB> electrode is synthesized using one-pot hydrothermal deposition. </LI> <LI> Superior electrochemical energy storage-deliver performance is demonstrated. </LI> <LI> High specific capacitance, excellent rate performance, and high rate long-term cyclic stability are obtained. </LI> </UL> </P>

        Galvanic corrosion of aluminum alloy (Al2024) and copper in 1.0M hydrochloric acid solution

        The corrosion of an aluminum alloy (Al2024) and copper in 1.0M HCl solution was investigated at 30,40, 50 and 60 oC using open circuit potential (OCP), electrochemical impedance spectroscopy (EIS) and potentiodynamic polarization (PDP) measurements. The galvanic corrosion of Al2024 and copper was studied using the zero resistance ammeter (ZRA) method. Galvanic current densities (Ig) and galvanic potential (Eg) were measured at 30 oC in 1.0M HCl solution. Thermodynamic parameters, such as activation energy (Ea), enthalpy of activation (ΔHa) and entropy of activation (ΔSa), were calculated and discussed. The results indicated that the corrosion rates of both Al2024and copper increased with temperature. The ZRA results demonstrated that Al2024 is a sacrificial anode in 1.0M HCl solution when coupled with copper.
