      • Mouse Pulmonary Cytochrome P-450 Naphthalene Hydroxylase : cDNA Cloning, Sequence, and Expression in Saccharomyces cerevisiae

        We have isolated a cDNA clone, Nah-2, encoding the cytochrome P-450^Nah(naphthalene hydroxylase) from a mouse lung λZAP cDNA library using anti-cytochrome P-450^Nah IgG as a probe. This same antibody selectively blocked[Nagata, K.,Martin, B. M., Gillette, J. R., & Sasame, H. A.(1990) Drug Metab. Dispos. 18, 557-564] the cytochrome P-450 in mouse lung microsomes that catalyzed the conversion of naphthalene to (1R,2S)- naphthalene 1,2-oxide, which has been postulated as a causative agent in the naphthalene-induced tissue-specific necrosis of Clara cells in mouse lung. The toxic effect is seen in mouse and not in rat. The cDNA encodes a polypeptide of 491 amino acids with a molecular mass of 50 kDa. Northern blot analysis with an Nah-2specific probe revealed that the mRNA is expressed in a species-and tissue-specific manner, present only in mouse lung and liver and not in that of rat. The mRNA encoding Nah-2 is constitutively expressed and is not induced by either phenobarbital, pyrazole, pregnenolone 16∂-carbonitrile, or 3-methylcholanthrene. Comparative amino acid sequence analyses with other documented members of the P-450 gene superfamily revealed that this encoded protein is in the IIF subfamily. To analyze its substrate specificity, the cDNA was inserted into the vector, pAAH5, and expressed in the Saccharonyces cerevisiae strain, AH_22. The presence of cytochrome P-450_Nah in the micreosomes isolated from transformed cell and analyzed by Western blot was confirmed by immunocomplexing product with anti-cytochrome P450_Nah IgG. Furthermore, activity toward naphthalene in the microsomes from the transformed cells established that this clone encodes a naphthalene hydroxylase. Like lung microsomes and purified and reconstituted cytochrome P450_Nah, transformed yeast microsomes convert naphthalene primarily to the trans-(1R)-hydroxy-(2R)-glutathionyl-1, 2-dihydronaphthalene conjugate, a stable form of the putative toxicant(1R,2S) oxide in the presence of glutathione and a mixture of glutathione S-transferases. Results of immunochemical studies support a role of this cytochrome P-450 in lung toxicity in mice exposed to high doses of naphthalene.

        비열대성 스프루(Celiac disease)와 감별이 어려웠던 장병증-형태 장관 T-세포 악성 림프종 1예

        저자들은 최근 장병증으로 내원한 환자에서 임상적, 병리학적으로 비열대성 스프루와 감별이 힘들었던 소장의 T-세포 악성 림프종을 경험하였기에 이를 보고하는 바이다. T-세포 림프종은 상당히 드물지만, 예후가 극히 불량하므로 비특이적이고 빠른 임상 경과를 취하는 장관병증을 보이는 환자가 있다면 비록 상하부 내시경상 특이한 점막 소견이 없더라도 심이지장 제2부를 포함한 위장관 각 부위의 점막 생검은 감별 진단을 위하여 꼭 필요하며 경우에 따라서는 보다 적극적으로 개복 소장 생검술 등도 고려해야 할 것으로 생각된다. Primary gastrointestinal lymphomas are commonly B-cell type in contrast to the rare T-cell type, which has been noted as a complication of celiac disease that has not been reported in Korea so far. Primary T-cell lymphoma is commonly associated with enteropathy, and we report a case of small bowel T-cell lymphoma associated with enteropathy, with minimal mucosal lesion lesions as flattening of folds, which was difficult to differentiate with celiac disease clinically and pathologically.

        Utilization of Cotton Stalks-Biomass Waste in the Production of Carbon Adsorbents by KOH Activation for Removal of Dye-Contaminated Water

        Four activated carbons were produced by two-stage process as followings; semi-carbonization of indigenous biomass waste, i.e. cotton stalks, followed by chemical activation with KOH under various activation temperatures and chemical ratios of KOH to semi-carbonized cotton stalks (CCS). The surface area, total pore volume and average pore diameter were evaluated by N2-adsorption at 77 K. The surface morphology and oxygen functional groups were determined by SEM and FTIR, respectively. Batch equilibrium and kinetic studies were carried out by using a basic dye, methylene blue as a probe molecule to evaluate the adsorption capacity and mechanism over the produced carbons. The obtained activated carbon (CCS-1K800) exhibited highly microporous structure with high surface area of 950 m2/g, total pore volume of 0.423 cm3/g and average pore diameter of 17.8 a. The isotherm data fitted well to the Langmuir isotherm with monolayer adsorption capacity of 222 mg/g for CCS-1K800. The kinetic data obtained at different concentrations were analyzed using a pseudo-first-order, pseudo-second-order and intraparticle diffusion equations. The pseudo-second-order model fitted better for kinetic removal of MB dye. The results indicate that such laboratory carbons could be employed as low cost alternative to commercial carbons in wastewater treatment.

        EFFECTS OF POLYPHENOLS OF Cocos Nucifera HUSK FIBRE ON SELECTED KIDNEY FUNCTION INDICES IN MICE

        Decoction of Cocos nucifera husk fibre is used indigenously in Nigeria for malaria treatment. Polyphenols have been identified as the phytochemicals responsible for the antimalarial activity of Cocos nucifera husk fibre, though their toxicity has not been evaluated. The polyphenols of Cocos nucifera husk fibre were therefore evaluated for their effects on selected kidney function indices in mice. Fifty mice were randomly divided into five groups (A-E) of ten mice each. Mice in group A were orally administered 5% DMSO solution while those in groups B, C, D and E were orally administered 31.25, 62.5, 125 and 250 mg/Kg body weight of the polyphenols respectively for seven days. Serum urea, creatinine and uric acid concentrations were determined. Serum levels of sodium, potassium, chloride and calcium ions and kidney alkaline phosphatase (ALP), glutamate dehydrogenase (GDH) and gamma-glutamyltransferase (GGT) activities were also determined. The results showed that the polyphenols significantly reduced (p<0.05) urea concentration at 250 mg/Kg body weight and creatinine concentration at all doses compared to controls. The polyphenols caused no significant alteration (p>0.05) in serum uric acid concentration and kidney ALP, GGT and GDH activities compared to controls. There was significant increase (p<0.05) in serum sodium ion concentration at 31.25, 125 and 250 mg/Kg body weight of polyphenols whereas significant increase (p<0.05) in serum potassium and chloride ions was observed at 62.5 and 250 mg/Kg body weight compared to controls. Thus, polyphenols of Cocos nucifera husk fibre may adversely affect some osmoregulatory functions of the kidney, especially at higher concentrations.

        저분자 유기산에 의한 중금속 오염토양의 복원

        For the remediation of the contaminated soil with heavy metals, Cd, Cr, Cu, and Pb, the reaction parameters were optimized. Tartaric acid (TA) and oxalic acid(OA) as a washing agent were evaluated as a function of concentration, reaction time, mixing ratio of washing agent and recovery of metals. The optimum washing conditions of TA and OA were in the ratio of 1 : 20 between soil and acid solution during 2 hr reaction under unbuffered pH solutions. At the optimized reaction conditions, the removal efficiencies were compared with that of 0.1 M HC1 and ethylenediamine tetraacetic acid(EDTA). TA showed higher efficiency on the removal of Pb than that of EDTA, which established for the remediation of contaminated soil with Pb and Cd metals. The recovery of metal ions from washing solution was achieved by adding calcium hydroxide and sodium sulfide by forming the precipitation of metal hydroxide and metal sulfide. Optimum amounts of sodium sulfide and calcium hydroxide were Cd = 25g/ℓ, Cu = 5∼10g/ℓ and Pb = 5∼10g/ℓ for the washing solution of OA and 2∼5g/ℓ for the washing solution of TA, respectively. The amounts of Na_2S and Ca(OH)_2 for the tartaric acid was less than that of oxalic acid.

        Sequential Optimization Approach for Enhanced Production of Glutamic Acid from Corynebacterium glutamicum 2262 Using Date Juice

        To improve glutamic acid production from Corynebacterium glutamicum 2262 using date juice, a culture medium was screened and optimized using the statistical experimental designs of Plackett-Burman and response-surface methodology. In the first step, a two-level Plackett-Burman design was adopted to select the most important nutrients influencing the glutamic acid production,which showed that the date juice sugars, urea, peptone, and glycine betaine were the most significant ingredients (P <0.05). Finally, response surface Box-Behnken design was employed to develop a mathematical model to identify the optimum concentrations of key components for higher glutamic acid production, which revealed the following:date juice (45 g/L), urea (16.9 g/L), peptone (15 g/L), and glycine betaine (12 g/L). The high correlation between the predicted and observed values indicated the validity of the model. Glutamic acid concentration increased significantly with optimized medium (33.2 g/L) when compared with non-optimized medium (12 g/L).

        난관수종액이 초기 임신 과정에 태반 세포 증식에 미치는 영향

        Objective : To assess the effect of hydrosalpingeal fluid on trophoblast cell proliferation and evaluate the cytokine level in hydrosalpingeal fluid Materials and Methods : Hydrosalpingeal fluid samples obtained from 10 women at the time of laparoscopic surgery. Trophoblast cell proliferation was assayed by a colorimetric immunoassay, based on the measurement of BrdU incorporation during DNA synthesis in response to hydrosalpingeal fluid and the levels of the Th-1 and Th-2 type cytokine determined by sandwich ELISA. Statistical analysis was performed by regression analysis and nonparametric testing(Mann-Whitney U test). Results : Samples from 7 out of 10 patients significantly suppressed trophoblast proliferation in a dose- dependent manner compared to controls(r=-0.0673, p$lt;0.05). IFN-γ and IL-6 were present in 2 out of 10 hydrosalpingeal fluid samples. IL-10 was not detected in all 10 samples. TNF-α, Th-1 type cytokine, was checked in all samples [median(range); 13.12 pg/ml(8.27 pg/ml~19.67 pg/ml)], and the mean concentration of TNF-α was significantly greater in group that suppressed trophoblast proliferation than in group that did not(14.74 pg/ml ± 1.05 pg/ml versus 9.33 pg/ml ± 0.55 pg/ml, p=0.0164). Conclusions : These findings suggest that hydrosalpingeal fluid may negatively affect trophoblast cell proliferation leading to implantation failure and also high level of the Th-1 type cytokine, TNF-α, in hydrosalpingeal fluid may be a potential mechanism for early pregnancy loss.

      • A systems level predictive model for global gene regulation of methanogenesis in a hydrogenotrophic methanogen

        <P>Methanogens catalyze the critical methane-producing step (called methanogenesis) in the anaerobic decomposition of organic matter. Here, we present the first predictive model of global gene regulation of methanogenesis in a hydrogenotrophic methanogen, <I>Methanococcus maripaludis</I>. We generated a comprehensive list of genes (protein-coding and noncoding) for <I>M. maripaludis</I> through integrated analysis of the transcriptome structure and a newly constructed Peptide Atlas. The environment and gene-regulatory influence network (EGRIN) model of the strain was constructed from a compendium of transcriptome data that was collected over 58 different steady-state and time-course experiments that were performed in chemostats or batch cultures under a spectrum of environmental perturbations that modulated methanogenesis. Analyses of the EGRIN model have revealed novel components of methanogenesis that included at least three additional protein-coding genes of previously unknown function as well as one noncoding RNA. We discovered that at least five regulatory mechanisms act in a combinatorial scheme to intercoordinate key steps of methanogenesis with different processes such as motility, ATP biosynthesis, and carbon assimilation. Through a combination of genetic and environmental perturbation experiments we have validated the EGRIN-predicted role of two novel transcription factors in the regulation of phosphate-dependent repression of formate dehydrogenase—a key enzyme in the methanogenesis pathway. The EGRIN model demonstrates regulatory affiliations within methanogenesis as well as between methanogenesis and other cellular functions.</P>

        The impact of major league baseball on the incidence of operative hand and facial trauma at a level 1 trauma center

        Background Professional affiliation between medical centers and professional sports teams can be mutually beneficial in the provision of healthcare and marketing strategy. Anecdotal evidence suggests a link between trauma volume and sporting events; however, there is limited data. This study aims to characterize the incidence of operative hand and facial trauma during professional baseball home games. Methods A retrospective review of surgical cases for traumatic hand or facial injuries at a level 1 center between 1999 and 2012 was performed. Demographic information including date of injury, admission status, and operative repair were collected. Patients were grouped based on whether their trauma occurred on the date of a home game. Results Operative hand and facial trauma occurred at a rate of 33.4 injuries per 100 days with home games, compared to 22.2 injuries per 100 days (incidence rate ratios, 1.50; 95% confidence interval, 1.34–1.69). When home games were played, patients were more likely to present as a result of motorcycle accidents (3.1% vs. 1.5%; P=0.04) or bicycle accidents (5.0% vs. 2.6%; P=0.01). Other mechanisms of trauma were not statistically different. There was an increase incidence of injuries during home games in August; weekly variability showed an increased incidence during the weekends. Conclusions There was an increased rate of operative hand and facial injuries on dates with professional home games. The incidence of injuries during home games was higher in the late summer and on the weekends. Further analysis may allow improved resource allocation and strategies for injury prevention and treatment.
